R1 kann beispielsweise bedeuten Propyl, Isopropyl, Butyl, Isobutyl, sec. Butyl, geradkettiges oder verzweigtes Amyl (Pentyl) oder Hexyl sowie die den genannten Kohlenwasserstoffresten entsprechenden Reste mit einer äthylenischen Doppelbindung wie Allyl oder Crotyl.
Besonders bevorzugt sind im Sinne der Erfindung solche Verbindungen, die als R1 einen cycloaliphatischen, gegebenenfalls mit Alkyl substituierten oder über Alkylen an das Stickstoffatom gebundenen Kohlenwasserstoffrest enthalten. Als solche Reste seien beispielsweise genannt Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Methyl-cyclohexyl, Äthylcyclohexyl, Propyl- und Isopropylcyclohexyl, wobei die Alkylgruppen vorzugsweise in 4-Stellung sowohl in cis- als auch in trans-l-Position vorliegen können, Endomethylencyclohexyl (2,2,1 -Bicycloheptyl), Endoäthylencyclohexyl (2,2,2 Bicyclooctyl), Endomethylencyclohexenyl, Endoäthylencyclohexenyl, Endomethylencyclohexylmethyl, Endo äthylencyclohexylmethyl, Endomethylencyclohexenylmethyl oder Endoäthylencyclohexenylmethyl.

tragende Benzolsulfonylisoharnstoffäther, -isothioharnstoffäther, -halogenameisensäureamidine oder -parabansäuren hydrolysiert und die Verfahrensprodukte gegebenenfalls zur Salzbildung mit alkalischen Mitteln behandelt.
Die Hydrolyse der als Ausgangsstoffe genannten Benzolsulfonyl-parabansäuren, -isoharnstoffäther, -iso- thioharnstoffäther oder -halogenameisensäureamidine er folgt zweckmässig in alkalischem Medium. Isoharnstoff äther und Halogenameisensäureamidine können auch in einem sauren Medium mit gutem Erfolg hydrolysiert werden.
Die Ausführungsformen des Verfahrens gemäss der
Erfindung können im allgemeinen hinsichtlich der Reak tionsbedingungen weitgehend variiert und den jeweiligen
Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden.
Beim Vorliegen bestimmter Substituentenkombina tionen kann das erfindungsgemässe Verfahren in einzel nen Fällen ungeeignet sein oder den gewünschten individuellen Benzolsulfonylharnstoff nur in geringen Ausbeuten liefern. Diese Fälle treten verhältnismässig selten auf und können vom Fachmann ohne Schwierigkeiten erkannt werden.
Die nach dem Verfahren gemäss der Erfindung er hältlichen Benzolsulfonyl-harnstoff-Derivate stellen wertvolle Arzneimittel dar, die sich durch eine starke und lang anhaltende blutzuckersenkende Wirkung auszeichnen. Ihre blutzuckersenkende Wirkung konnte z. B. am Kaninchen dadurch festgestellt werden, dass man die Verfahrensprodukte in einer Dosis von 10 mg/kg verfütterte und den Blutzuckerwert nach der bekannten Methode von Hagedorn-Jensen oder mit einem Autoanalyzer über eine längere Zeitdauer bestimmte. So wurde z. B. ermittelt, dass der N-4-(ssjndolino-carbonamidoäthyl)-benzolsuffonyll N'- (4-methyl-cyclohexyl) -harnstoff am Kaninchen eine Blutzuckersenkung von 32 % bewirkt, die nach 24 Stunden noch 29 % beträgt und sogar nach 48 Stunden noch 13 % ausmacht.
Der N-[4-(ss-Indolino-carbonamidoäthyl)-benzolsulfonyl]
N'-cycolhexylharnstoff bewirkt unter den angegebenen Versuchsbedingungen sogar eine Blutzuckersenkung von 38%, die nach 24 Stunden noch 20% beträgt.
Ferner wurde ermittelt, dass der N-[4-(ss- {N-Methyl-N-phenylureido} -äthyl)-benzol- sulfonyl]-N'-(4-methyl-cyclohexyl)-harnstoff am Kaninchen nach 3 Stunden eine Blutzuckersenkung von 30% bewirkt, die nach 24 Stunden sogar 49 % beträgt. Der N-[4-(ss-{N-Methylphenyl-N-methylureido}-äthyl)- benzolsulfonyl]-N'-(4-methyl-cyclohexyl)-harnstoff bewirkt unter den angegebenen Versuchsbedingungen nach 6 Stunden eine Blutzuckersenkung von 32%, die nach 24 Stunden noch 27 % beträgt und nach 48 Stunden immer noch 25 % ausmacht. In gleicher Weise führt der N-[4-(B- (2-Chlorphenyl-N-methylureido )-äthyl)- benzolsulfonyl] -N'-cyclohexyl-harnstoff nach 3 Stunden zu einer Blutzuckersenkung von 30%, die nach 24 Stunden 39 % beträgt.
Demgegenüber ist der als orales Antidiabetikum bekannte N-(4-Methyl-benzolsulfonyl)-N'-butyl-harnstoff bei Dosen von weniger als 25 mg/kg am Kaninchen unwirksam.
Die beschriebenen Benzolsulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise alkalische Mittel wie Alkali- oder Erdalkalihydroxyde, -carbonate oder -bicarbonate herangezogen werden.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe wie Talkum, Stärke, Milchzucker, Tragant oder Magnesiumstearat enthalten.
Ein Präparat, das die beschriebenen Benzolsulfonylharnstoffe als Wirkstoff enthält, z. B. eine Tablette oder ein Pulver mit oder ohne die genannten Zusätze, ist zweckmässig in eine geeignete dosierte Form gebracht.
Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Benzolsulfonylharnstoffes und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 0,5 bis 100 mg, vorzugsweise 2 bis 10 mg, jedoch können auch erheblich darüber oder darunter liegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.
