Verfahren zur Herstellung bisher unbekannter Polypeptide
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von bisher unbekannten Polypeptiden bzw.
Polypeptidderivaten der allgemeinen Formel (3 -Thio-2-X-propionyl) -L-seryl-L-asparaginyl-L4eucyl L-seryl-L-threonyl-L-hemicystinyl-L-valyl-L-leucyl- glycyl-L-lysyl-L-leucyl-L-seryl-L-glutaminyl- L-glutamyl-L-leucyl-L-histidyl-L-lysyl-L-leucyl L-glutaminyl-L-threonyl-L-tyrosyl-L-prolyl-L-arginyl- L-threonyl-L-asp araginyl-L-threonyl-glycyl-L-serylglycyl-L-threonyl-Y, worin X die Bedeutung -H, -NH3, R-CO-NH- (R= Alkyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst. Aryl) oder R'-O-CO-NH- (R' = Alkyl, Alkenyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst.
Aryl, wobei der Rest R' nicht durch Trifluoressigsäure abspaltbar ist) und Y die Bedeutung L-Prolinamid oder L-Prolin-L-valinamid besitzt und in der die einzelnen Aminosäuren der obigen Sequenz durch andere, natürliche a-Aminosäuren oder deren Homologe ausgetauscht sein können, sofern die biologische Wirkung erhalten bleibt, ihren therapeutisch wirksamen Säureadditionssalzen und Schwermetallkomplexen.
Hat X - Jie Bedeutung -NHv, R-CO-NH- (R = Alkyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst. Aryl), oder R'-O-CO-NH- (R' = Alkyl, Alkenyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst.
Aryl), so liegt die Aminosäure (HS-CH-CHX-COOH) in der L-Form vor.
Beispielsweise kann R die Methyl-, Athyl-, Propyl-, tert.-Butylgruppe oder aber einen gegebenenfalls durch ein Halogenatom, eine niedere Alkoxy- oder die Nitrogruppe substituierten Phenyl- oder Benzylrest bedeuten.
Beispielsweise kann R' die Propyl-, Allyl-, Benzyl-, p-Nitrobenzyl-, p-Chlorbenzyl-, p-Brombenzylgruppe, den p-Phenylazobenzyl- oder den p-(p'-Methoxy-phenylazobenzyl)rest bedeuten.
In den Peptidsequenzen der obigen allgemeinen Formel können einzelne Aminosäuren durch andere, natürliche Aminosäuren oder deren Homologe ausgetauscht sein, sofern dadurch die biologische Wirkung im wesentlichen erhalten bleibt.
Zum Beispiel kann in Stellung 2, 5, 13 und 29 L Seryl gegen L-Threonyl oder L-Alanyl, in Stellung 3 und 26 L-Asparaginyl gegen L-Aspartyl, L-Glutaminyl oder L-Glutamyl, in Stellung 6, 21, 25, 27 und 31 L-Threonyl gegen L-Seryl oder L-Alanyl, in Stellung 11 und 18 L-Lysyl gegen BOC-L-Lysyl, L-Ornithyl oder L-Arginyl, in Stellung 14 und 20 L-Glutaminyl gegen L-Glutamyl, L-Asparaginyl oder L-Aspartyl, in Stellung 15 L-Glutamyl gegen OTB-Glutamyl, L-Glutaminyl, L-Asparaginyl oder L-Aspartyl, in Stellung 19 L-Leucyl gegen L-Tyrosyl, in Stellung 22 L-Tyrosyl gegen L-Phenylalanyl und in Stellung 24 L-Arginyl gegen L-Ornithyl oder L-Lysyl ersetzt werden.
Die bisher unbekannten Polypeptide bzw. Polypeptidderivate der allgemeinen Formel können nach für die Synthese von Verbindungen dieser Art allgemein bekannten Methoden hergestellt werden, indem man die zu ihrem Aufbau nötigen Verbindungen unter Bildung von CONH-Bindungen in beliebiger zeitlicher Reihenfolge miteinander kondensiert, wobei nicht an der Reaktion teilnehmende freie funktionelle Gruppen durch geeignete Schutzgruppen geschützt werden, zu einem beliebigen Zeitpunkt der Synthese unter Beibehaltung einer gegebenen falls in Stellung 1 vorhandenen R'-O-CO-Gruppe, worin R' obige Bedeutung hat, die Schutzgruppen abspaltet, die Mercaptogruppe nach Bildung der Sequenz 1 bis 7 zum Disulfid oxydiert, die Carboxylgruppe des endständigen Prolyl- bzw. Valylrestes in die Amidgruppe überführt und gegebenenfalls anschliessend die Säureadditionssalze herstellt, indem man die Polypeptide bzw.
Polypeptidderivate der obigen Formel durch Umsetzung mit organischen oder anorganischen Säuren in die entsprechenden Salze überführt.
Die Einführung einer gegebenenfalls am Endprodukt am endständigen L-Hemicystin-Molekül gewünschten Schutzgruppe kann in einer beliebigen Stufe vor der letzten Stufe erfolgen. Die Umwandlung einer geschütz ten Aminogruppe in eine freie Gruppe am L-Hemicystin-Molekül zur Erlangung einer am endständigen L-Hemicystin-Molekül nicht geschützten Verbindung erfolgt nach an sich bekannten Methoden durch Behandlung mit hydrolysierenden bzw. reduzierenden Mitteln.
In der letzten Stufe der Kondensation sind jedoch Methoden zu verwenden, bei denen Racemisierung nicht auftritt oder gering gehalten werden kann, vorzugsweise durch Verwendung der Azide oder der aktivierten Ester, wobei die Aktivierung vorzugsweise mit N-Hydroxysuccinimid vorgenommen wird.
Für die Blockierung der Guanidogruppe des Argininrestes in der nachstehend beschriebenen Teilsequenz B wurde die Nitrogruppe verwendet, doch können auch andere geeignete Schutzgruppen, wie die Tosylgruppe, die p-Nitrocarbobenzoxygruppe oder die 2-(Isopropyl oxycarbonyl)-3,4,5,6-tetrachlorobenzoylgruppe verwendet werden. Man kann auch den Schutzeffekt der Protonisierung der Guanidogruppe bei der Synthese verwenden.
Beim Aufbau der bisher unbekannten Polypeptide bzw. Polypeptidderivate hat sich für die Blockierung der y-Carboxylgruppe, beispielsweise in der nachstehend beschriebenen Teilsequenz D, die tert.-Butyloxygruppe bewährt, doch können auch andere Schutzgruppen, wie die Methoxy-, die iSXthoxy-, die tert. Amyloxy-, die Amid- oder die Benzyloxygruppe verwendet werden.
Für die Blockierung der W-Aminogruppe des Lysinrestes, beispielsweise in der nachstehend beschriebenen Teilsequenz C, kann eine Carbo-tert.-alkoxygruppe, vorzugsweise die Carbo-tert.-butoxygruppe verwendet werden.
Als Mercaptoschutzgruppe in den nachstehend beschriebenen Teilsequenzen Es und E7 verwendet man vorzugsweise die Benzyl- oder Tritylgruppe. tJblicher- weise werden die zum Schutz der SH-Gruppen verwendeten Benzyl- bzw. Tritylreste am Ende der Synthese durch Behandlung mit Natrium in flüssigem Ammoniak abgespalten. Es wurde nun gefunden, dass die Abspaltung der Benzylschutzgruppen sowie die Herstellung der S-S-Bindung vor der letzten Stufe zu besonders guten Ausbeuten an Endprodukt führt.
Die Ausgangsprodukte zur Herstellung der neuen Polypeptide bzw. Polypeptidderivate können, sofern sie bisher nicht bekannt waren, nach den für die Peptidchemie bekannten Methoden erhalten werden, wobei die Aminosäuren einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden.
Die neuen Polypeptide bzw. Polypeptidderivate lassen sich auch in Form ihrer Salze gewinnen bzw. verwenden. Als Salze kommen solche mit organischen Säuren. wie Essigsäure. Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure. Malonsäure, Bernsteinsäure. Maleinsäure, Fumarsäure. Weinsäure. Zitronensäure. Benzoesäure. Zimtsäure, Salicylsäure, 2-Phenoxy- oder 2-Acetoxybenzoesäure, Mandelsäure. Methansulfonsäure, Äthansulfonsäure, lIydroxyäthansul- fonsäure, Benzol- oder Toluolsulfonsäure. Naphthalinsulfonsäure. Sulfanilsäure sowie polymere Säuren, wie Gerbsäure, Alginsäure. Polygalacturonsäure, Polyphloretinphosphat oder Carboxymethylcellulose und Salze mit anorganischen Säuren. wie Halogenwasserstoffsäure, z. B. Salzsäure oder Bromwasserstoffsäure. Salpetersäure, Thiocyansäure. Schwefelsäure und Phosphorsäure in Frage. Die Polypeptide bzw.
Polypeptidderivate der obigen Formel können durch Umsetzung mit Schwermetallsalzen in die entsprechenden Schwermetallkomplexe überführt werden. Als Schwermetallkomplex kommt z. B. derjenige von Zinks36 in Frage.
Die erfindungsgemäss hergestellten Verbindungen stellen ein wichtiges therapeutisches Prinzip dar. Sie senken den Calciumplasmaspiegel und bewirken als Antagonisten des Parathormons eine positive Calciumbilanz im Knochen. Die biologische Wirkung der neuen Verbindungen wurde an Ratten geprüft, wobei sich aufgrund der Senkung des Blut-Calciums ein Wert von 3000-5000 MRC-Einheiten/mg Peptid ergab.
Die zu verabreichende Dosis hängt von der gewünschten Wirkung sowie von der Applikationsart ab.
Im allgemeinen werden jedoch mit einer einmal zu verabreichenden Tagesdosis von 1 bis 10 Einheiten pro kg Tiergewicht zufriedenstellende Resultate erzielt. Für grössere Säugetiere beträgt die Tagesdosis etwa 70 bis 700 Einheiten, die auf einmal oder in mehreren Anteilen verabreicht werden können. Für die intramuskuläre Applikation enthält eine geeignete Verabreichungsform etwa 70 bis 700 Einheiten der Wirksubstanz, vermischt mit einem flüssigen Träger.
Die erfindungsgemäss hergestellten Verbindungen sind somit indiziert bei allen Zuständen, bei welchen eine Senkung des Plasmacalciumspiegels bzw. Beeinflussung des Knochenstoffwechsels erwünscht ist, z. B.
Hypercalcämien infolge Mangels des endogenen Thyreocalcitonins durch Ausfall von Schilddrüsengewebe oder Hyperfunktion der Nebenschilddrüsen. Sie sind ferner indiziert bei allen Knochenaffektionen, die auf einem vermehrten Abbau beruhen oder bei welchen eine Calciumfixation im Knochen erwünscht ist, z. B. Osteoporose verschiedener Genese (z. B. postklimakterisch, posttraumatisch, bedingt durch Corticosteroidtherapie oder Inaktivität usw.), Frakturen, Osteomalacie, Rachitis und renal bedingte Osteodystrophie sowie insbesondere zur Kombinationstherapie mit Calcium bzw.
Phosphat.
Ein Vorteil der erfindungsgemässen Polypeptide bzw. Polypeptidderivate der allgemeinen Formel, worin X für R-CO-NH- (R = Alkyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst. Aryl) oder R'-O-CO-NH- (R' = Alkyl, Alkyl, gegebenenfalls subst. Aralkyl, gegebenenfalls subst. Aryl) steht, ist ihre Beständigkeit gegenüber der abbauenden Wirkung von Aminopeptidasen.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die genannten Verbindungen in Mischung mit einem für die parenterale Applikation geeigneten organischen oder anorganischen Trägermaterial. Die neuen Verbindungen können auch in Form eines Depotpräparates verabreicht werden.
Die neuen Verbindungen und ihre Salze können auch als Zwischenprodukt zur Herstellung von pharmazeutischen Präparaten Verwendung finden.

Teilsequenz A L-Seryl-glycyl-L-threonyl-L-prolinamid (H-S er-Gly-Thr-Pro-NH2) a) L-Threonyl-L-prolinamid . hydrochlorid (H-Thr-Pro-NH2. HCl)
Man löst bei 5 134 g Z-Thr-NH-NH. in 2 1 1n Salzsäure und versetzt mit 0,55 1 1n Natriumnitrit.
Nach 5 Minuten wird Kaliumcarbonat bis pH 9 zugegeben, das entstandene Azid mit Äthylacetat extrahiert und eine Lösung von 80 g H-Pro-NH2 - hydro- chlorid in 100 ml Wasser, 500 ml Dimethylformamid und 77 ml Triäthylamin hinzugefügt. Man verdampft das Äthylacetat bei 200 im Vakuum und lässt über Nacht bei 250 stehen. Die restliche Lösung wird im Vakuum verdampft, der Rückstand in Äthylacetat gelöst, die Lösung mit Wasser, verdünnter Salzsäure und einer wässrigen Kalciumcarbonatlösung gewaschen und über Natriumsulfat getrocknet. Man verdampft im Vakuum, löst in warmem Äthylacetat und kühlt ab.
Man erhält Z-Thr-Pro-NH-,; Smp. 1480, [a] D =-72 in 95 % Essigsäure. Man löst hierauf 90 g Z-Thr-Pro-NH, in 2 1 Dioxan und 260 ml in Salzsäure und hydriert bei 200 und Normaldruck in Gegenwart eines Palladiumkatalysators. Man filtriert, verdampft die Lösung im Vakuum, wäscht den kristallinen Rückstand mit Athylacetat und erhält H-Thr-Pro-NH3 - HCl; Smp. 2160, [aJ D0 -640 in 95 % Essigsäure.
b) N-B enzyloxycarbonyl-L-seryl-glycin-äthylester (Z-Ser-Gly-OEt)
Man löst 14,5 g Benzyloxycarbonyl-serin in 100 ml Chloroform und gibt 6,1 g N-Methylmorpholin dazu; danach tropft man 8,2 g Chlorameisensäureisobutylester hinzu. Nach 10 Minuten wird eine Lösung von 6,6 g Glycin-äthylester in 50 ml Chloroform hinzugefügt und 1 Stunde bei Raumtemperatur gerührt. Das Reaktionsgemisch wird mit verdünntem Ammoniak, dann mit Salzsäurelösung gewaschen, über Natriumsulfat getrocknet und die organische Phase abgedampft.
Man kristallisiert das erhaltene Z-Ser-Gly-OEt aus Essigester um. Smp. 1030, [a] =+30 in Dimethylformamid.
c) N-Benzyloxycarbonyl-L-seryl-glycin-hydrazid (Z-Ser-Gly-NHNH)
Man löst 19,4 g Z-Ser-Gly-OEt in 270 ml Athyl- alkohol, gibt 27 ml Hydrazinhydrat hinzu und lässt 2 Taee bei Raumtemperatur stehen. Man filtriert die krist'iiine.Masse ab,- wäscht mit Methanol und trocknet.
Man erhält Z-Ser-Gly-NHNH2, Smp. 1800, [a] D0 = + 2 in Dimethylformamid.
d) N-Benzyloxycarbonyl-L-seryl-glycyl-L-threonyl-
L-prolinamid (ZSer-Gly-Thr-Pro-NH)
Zu 200 ml Dimethylformamid werden 25 ml einer 4n Chlorwasserstofflösung in Äthyl äther gegeben und darin 11 g Z-Ser-Gly-NHNHo gelöst. Hierauf tropft man bei -10 5.4 ml tert.-Butylnitrit hinzu, fügt der Lösung 18 ml Triäthylamin bei und filtriert anschlie ssend. Gleichzeitig löst man 12 g H-Thr-Pro-NH.2 HCl in 100 ml Dimethylformamid, gibt 6.2 ml Triäthylamin zu, filtriert vom ausgeschiedenen Triäthylaminhydrochlorid ab und fügt zu dem Filtrat die obenerhaltene Dipeptidazid-Lösung hinzu.
Man lässt 2 Stunden bei 0 stehen, dampft zur Trockne ab löst den Rückstand in einem Gemisch von 500 ml Methanol und 100 ml Wasser und lässt die erhaltene Lösung durch eine 100 ml Dowex-50-Säule (H + -Form). Man dampft ein und kristallisiert den Rückstand aus Methanol/Essigester. Man erhält Z-Ser-Gly-Thr-Pro-NHn vom Smp. 1320 (mit Zersetzung), [a] 2D0 = 220 in Dimethylformamid.
e) L-Seryl-glycyl-L-threonyl-L-prolinamid (H-Ser-Gly-Thr-Pro-N-)
Man löst 22 g Z-Ser-Gly-Thr-Pro-NH- in einem Gemisch von 440 ml Methanol und 88 ml Wasser und hydriert bei 200 und Normaldruck in Gegenwart von Palladiumkohle. Danach wird filtriert, zur Trockne eingedampft, der Rückstand mit Äther gewaschen und getrocknet. Man erhält H-Ser-Gly-Thr-Pro-NH2 vom Smp. 950 (mit Zersetzung), [a] 2,0 = -270 in Dimethylformamid.
Teilsequenz B tert.-Butyloxycarbonyl-L-arginyl-L-threonyl L-asparaginyl-L-threonyl-glycinhydrazid (BO C-Arg-Thr-Asn-Thr-Gly-NHNH2) a) N-Benzyloxyzarbonyl-L-asparaginyl-L-threonyl- glycinäthylester (Z-Asn-Thr-Gly-OEt)
Man löst 43 g Z-Thr-Gly-OEt in 500 ml Dimethylformamid und hydriert bei Normaldruck und Zimmertemperatur in Anwesenheit von Palladiumkohle. Man filtriert, gibt 60 g Z-Asn-OCP zu, lässt 16 Stunden bei 250 stehen, konzentriert auf etwa 250 ml, gibt Essigester zu, wäscht mit verdünnter Salzsäure, trocknet über Natriumsulfat und dampft zur Trockne ein. Der Rückstand wird mit Essigester gewaschen und getrocknet.
Man erhält Z-Asn-Thr-Gly-OEt. Smp. 2250, [a] r) = -40 in Dimethylformamid.
b) N-Benzyloxycarbonyl-L-threonyl-L-asparaginyl
L-threonyl-glycinäthylester (Z-Thr-Asn-Thr-Gly-OEt)
Man löst 29 g Z-Asn-Thr-Gly-OEt in 700 ml Dimethylformamid, hydriert bei Normaldruck und Zimmertemperatur in Anwesenheit von Palladiumkohle, filtriert und versetzt das Filtrat mit Z-Thr-N2, das aus 30 g Z-Thr-NHNHs hergestellt wurde [siehe Teilsequenz A a)]. Man löst 22 g Z-Thr-Asn-Thr-Gly-OEt in 700 ml Dimethylformamid, hydriert bei Normaldruck und Zimmertemperatur in Anwesenheit von Palladiumkohle und filtriert. Man löst 23 g BOC-Arg(NO.,)-OH in 500 ml Dimethylacetamid, gibt 12 ml Triäthylamin zu, kühlt auf -100, gibt 9,4 ml Chlorameisensäureisobutylester hinzu, lässt 10 Minuten reagieren und gibt die obenerhaltene Tetrapeptidlösung zu. Nach 30 Minuten bei 250 fügt man 100 ml Wasser hinzu, behandelt die erhaltene Lösung mit Amberlit-IRA-410 (OH-Form) bis zur negativen Chlorreaktion, filtriert und dampft zur Trockne ab. Man wäscht den Rückstand mit Chloroform, Essigester und Äther und trocknet.
Man erhält BOC-Arg(NOs)-Thr-Asn-Thr-Gly-OEt. Smp. X 100, [a] 2r) =-4 in Dimethylformamid.

A protective group which may be desired on the end product on the terminal L-hemicystine molecule can be introduced in any stage before the last stage. The conversion of a protected amino group into a free group on the L-hemicystine molecule to obtain a compound that is not protected on the terminal L-hemicystine molecule is carried out according to methods known per se by treatment with hydrolyzing or reducing agents.
In the last stage of the condensation, however, methods are to be used in which racemization does not occur or can be kept low, preferably by using the azides or the activated esters, the activation preferably being carried out with N-hydroxysuccinimide.
The nitro group was used to block the guanido group of the arginine residue in the partial sequence B described below, but other suitable protective groups such as the tosyl group, the p-nitrocarbobenzoxy group or the 2- (isopropyl oxycarbonyl) -3,4,5,6- tetrachlorobenzoyl group can be used. The protective effect of protonation of the guanido group can also be used in the synthesis.
In the construction of the previously unknown polypeptides or polypeptide derivatives, the tert-butyloxy group has proven useful for blocking the γ-carboxyl group, for example in the partial sequence D described below, but other protective groups, such as the methoxy, the iSXthoxy, the tert. Amyloxy, the amide or the benzyloxy group can be used.
A carbo-tert-alkoxy group, preferably the carbo-tert-butoxy group, can be used to block the W-amino group of the lysine residue, for example in the partial sequence C described below.
The benzyl or trityl group is preferably used as the mercapto protecting group in the partial sequences Es and E7 described below. The benzyl or trityl radicals used to protect the SH groups are usually split off at the end of the synthesis by treatment with sodium in liquid ammonia. It has now been found that the splitting off of the benzyl protective groups and the production of the S-S bond before the last stage lead to particularly good yields of the end product.
The starting products for the production of the new polypeptides or polypeptide derivatives can, if they were not previously known, be obtained by the methods known for peptide chemistry, the amino acids being linked to one another individually or after prior formation of smaller peptide units.
The new polypeptides or polypeptide derivatives can also be obtained or used in the form of their salts. The salts used are those with organic acids. like acetic acid. Propionic acid, glycolic acid, lactic acid, pyruvic acid. Malonic acid, succinic acid. Maleic acid, fumaric acid. Tartaric acid. Citric acid. Benzoic acid. Cinnamic acid, salicylic acid, 2-phenoxy- or 2-acetoxybenzoic acid, mandelic acid. Methanesulfonic acid, ethanesulfonic acid, hydroxyethanesulfonic acid, benzene or toluenesulfonic acid. Naphthalenesulfonic acid. Sulfanilic acid and polymeric acids such as tannic acid and alginic acid. Polygalacturonic acid, polyphloretin phosphate or carboxymethyl cellulose and salts with inorganic acids. such as hydrohalic acid, e.g. B. hydrochloric acid or hydrobromic acid. Nitric acid, thiocyanic acid. Sulfuric acid and phosphoric acid in question. The polypeptides or
Polypeptide derivatives of the above formula can be converted into the corresponding heavy metal complexes by reaction with heavy metal salts. As a heavy metal complex z. B. that of Zinks36 in question.
The compounds prepared according to the invention represent an important therapeutic principle. They lower the calcium plasma level and, as antagonists of the parathyroid hormone, bring about a positive calcium balance in the bones. The biological activity of the new compounds was tested on rats, which resulted in a value of 3000-5000 MRC units / mg peptide due to the reduction in blood calcium.
The dose to be administered depends on the desired effect and the type of application.
In general, however, satisfactory results are achieved with a once daily dose of 1 to 10 units per kg of animal weight. For larger mammals, the daily dose is about 70 to 700 units, which can be administered all at once or in several portions. For intramuscular administration, a suitable form of administration contains about 70 to 700 units of the active substance mixed with a liquid carrier.
The compounds prepared according to the invention are thus indicated in all conditions in which a reduction in the plasma calcium level or an influence on the bone metabolism is desired, e.g. B.
Hypercalcaemia as a result of a deficiency in endogenous thyreocalcitonin due to the failure of thyroid tissue or hyperfunction of the parathyroid glands. They are also indicated for all bone affections that are based on increased degradation or in which calcium fixation in the bone is desired, e.g. B. Osteoporosis of various origins (e.g. post-climacteric, post-traumatic, caused by corticosteroid therapy or inactivity, etc.), fractures, osteomalacia, rickets and renal osteodystrophy and especially for combination therapy with calcium or

An advantage of the polypeptides or polypeptide derivatives according to the invention of the general formula in which X stands for R-CO-NH- (R = alkyl, optionally substituted aralkyl, optionally substituted aryl) or R'-O-CO-NH- (R '= Alkyl, alkyl, optionally substituted aralkyl, optionally substituted aryl) is their resistance to the degrading action of aminopeptidases.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations, use. These contain the compounds mentioned in a mixture with an organic or inorganic carrier material suitable for parenteral administration. The new compounds can also be administered in the form of a depot preparation.
The new compounds and their salts can also be used as intermediates for the production of pharmaceutical preparations.

Partial sequence A L-Seryl-glycyl-L-threonyl-L-prolinamide (H-S er-Gly-Thr-Pro-NH2) a) L-threonyl-L-prolinamide. hydrochloride (H-Thr-Pro-NH2. HCl)
Dissolve in 5,134 g of Z-Thr-NH-NH. in 2 1 1N hydrochloric acid and mixed with 0.55 1 1N sodium nitrite.
After 5 minutes, potassium carbonate is added to pH 9, the azide formed is extracted with ethyl acetate and a solution of 80 g of H-Pro-NH2 hydrochloride in 100 ml of water, 500 ml of dimethylformamide and 77 ml of triethylamine is added. The ethyl acetate is evaporated at 200 in vacuo and left to stand at 250 overnight. The remaining solution is evaporated in vacuo, the residue is dissolved in ethyl acetate, the solution is washed with water, dilute hydrochloric acid and an aqueous calcium carbonate solution and dried over sodium sulfate. It is evaporated in vacuo, dissolved in warm ethyl acetate and cooled.
Z-Thr-Pro-NH-, is obtained; M.p. 1480, [a] D = -72 in 95% acetic acid. 90 g of Z-Thr-Pro-NH are then dissolved in 2 l of dioxane and 260 ml in hydrochloric acid and hydrogenated at 200 and normal pressure in the presence of a palladium catalyst. It is filtered, the solution is evaporated in vacuo, the crystalline residue is washed with ethyl acetate and H-Thr-Pro-NH3-HCl is obtained; M.p. 2160, [aJ D0-640 in 95% acetic acid.
b) N-B enzyloxycarbonyl-L-seryl-glycine-ethyl ester (Z-Ser-Gly-OEt)
14.5 g of benzyloxycarbonyl-serine are dissolved in 100 ml of chloroform and 6.1 g of N-methylmorpholine are added; then 8.2 g of isobutyl chloroformate are added dropwise. After 10 minutes, a solution of 6.6 g of glycine ethyl ester in 50 ml of chloroform is added and the mixture is stirred at room temperature for 1 hour. The reaction mixture is washed with dilute ammonia and then with hydrochloric acid solution, dried over sodium sulfate and the organic phase is evaporated.
The Z-Ser-Gly-OEt obtained is recrystallized from ethyl acetate. M.p. 1030, [a] = + 30 in dimethylformamide.
c) N-Benzyloxycarbonyl-L-seryl-glycine-hydrazide (Z-Ser-Gly-NHNH)
19.4 g of Z-Ser-Gly-OEt are dissolved in 270 ml of ethyl alcohol, 27 ml of hydrazine hydrate are added and 2 teas are left to stand at room temperature. The crystalline mass is filtered off, washed with methanol and dried.
Z-Ser-Gly-NHNH2, mp. 1800, [a] D0 = + 2 in dimethylformamide is obtained.
d) N-Benzyloxycarbonyl-L-seryl-glycyl-L-threonyl-
L-prolinamide (ZSer-Gly-Thr-Pro-NH)
To 200 ml of dimethylformamide, 25 ml of a 4N hydrogen chloride solution in ethyl ether are added and 11 g of Z-Ser-Gly-NHNHo are dissolved therein. 5.4 ml of tert-butyl nitrite are then added dropwise at -10, 18 ml of triethylamine are added to the solution and the mixture is then filtered. At the same time, 12 g of H-Thr-Pro-NH.2 HCl are dissolved in 100 ml of dimethylformamide, 6.2 ml of triethylamine are added, the precipitated triethylamine hydrochloride is filtered off and the dipeptide azide solution obtained above is added to the filtrate.
The mixture is left to stand at 0 for 2 hours, evaporated to dryness, the residue is dissolved in a mixture of 500 ml of methanol and 100 ml of water and the resulting solution is passed through a 100 ml Dowex 50 column (H + form). It is evaporated and the residue is crystallized from methanol / ethyl acetate. Z-Ser-Gly-Thr-Pro-NHn of melting point 1320 (with decomposition), [a] 2D0 = 220 in dimethylformamide is obtained.
e) L-Seryl-glycyl-L-threonyl-L-prolinamide (H-Ser-Gly-Thr-Pro-N-)
22 g of Z-Ser-Gly-Thr-Pro-NH- are dissolved in a mixture of 440 ml of methanol and 88 ml of water and hydrogenated at 200 and normal pressure in the presence of palladium carbon. It is then filtered, evaporated to dryness, the residue washed with ether and dried. H-Ser-Gly-Thr-Pro-NH2 is obtained with a melting point of 950 (with decomposition), [a] 2.0 = -270 in dimethylformamide.
Partial sequence B tert-Butyloxycarbonyl-L-arginyl-L-threonyl L-asparaginyl-L-threonyl-glycine hydrazide (BO C-Arg-Thr-Asn-Thr-Gly-NHNH2) a) N-Benzyloxycarbonyl-L-asparaginyl-L -threonyl- glycine ethyl ester (Z-Asn-Thr-Gly-OEt)
43 g of Z-Thr-Gly-OEt are dissolved in 500 ml of dimethylformamide and hydrogenated at normal pressure and room temperature in the presence of palladium carbon. It is filtered, 60 g of Z-Asn-OCP are added, left to stand at 250 for 16 hours, concentrated to about 250 ml, ethyl acetate is added, washed with dilute hydrochloric acid, dried over sodium sulfate and evaporated to dryness. The residue is washed with ethyl acetate and dried.
Z-Asn-Thr-Gly-OEt is obtained. M.p. 2250, [a] r) = -40 in dimethylformamide.
b) N-Benzyloxycarbonyl-L-threonyl-L-asparaginyl
L-threonyl-glycine ethyl ester (Z-Thr-Asn-Thr-Gly-OEt)
29 g of Z-Asn-Thr-Gly-OEt are dissolved in 700 ml of dimethylformamide, hydrogenated at normal pressure and room temperature in the presence of palladium carbon, filtered and the filtrate is treated with Z-Thr-N2, which is produced from 30 g of Z-Thr-NHNHs was [see partial sequence A a)]. After 12 hours at 0 it is evaporated to dryness, the residue is washed with water, ether and ethanol. You get

c) tert-butyloxycarbonyl-nitro-L-arginyl-L-threonyl
L-asparaginyl-L-threonyl-glycine ethyl ester (BOC-Arg (NO2) -Thr-Asn-Thr-Gly-OEt)
22 g of Z-Thr-Asn-Thr-Gly-OEt are dissolved in 700 ml of dimethylformamide, hydrogenated at normal pressure and room temperature in the presence of palladium-carbon and filtered. 23 g of BOC-Arg (NO.,) - OH are dissolved in 500 ml of dimethylacetamide, 12 ml of triethylamine are added, the mixture is cooled to -100, 9.4 ml of isobutyl chloroformate is added, left to react for 10 minutes and the tetrapeptide solution obtained above is added. After 30 minutes at 250, 100 ml of water are added, the solution obtained is treated with Amberlite-IRA-410 (OH form) until the chlorine reaction is negative, filtered and evaporated to dryness. The residue is washed with chloroform, ethyl acetate and ether and dried.
BOC-Arg (NOs) -Thr-Asn-Thr-Gly-OEt is obtained. M.p. X 100, [a] 2r) = -4 in dimethylformamide.
