Verfahren zur Herstellung von Pyrimidin.Derivaten
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Pyrimidin-Derivaten der allgemeinen Formel:

worin R gerades oder verzweigtes Alkyl mit höchstens 9 C-Atomen bedeutet, sowie von Säure-Additionssalzen davon.
Die genannten Verbindungen zeigen im Tierversuch, z. B. bei Mäusen, Ratten und Hunden, eine narkotische und hypnotische Wirkung. Die nachfolgende Tabelle enthält einen Wirkungsvergleich zwischen er findungsgemäss erhältlichen Verbindungen und dem als bestwirksam bekannten Narkotikum Pentobarbital Natrium. Als Mass für die hypnotische bzw. narkotische Wirkung wird diejenige iv. Dosis (ED 50) angegeben, bei welcher 5Q /o der Versuchsmäuse in Seitenlage geraten. In der Tabelle sind ferner die intravenösen Toxizitäten bei der Maus als DL 50 statuiert.

Aus der Tabelle geht hervor, dass die erfindungsgemäss erhältlichen Verbindungen in wesentlich niedrigeren Dosen wirksam sind als das Pentobarbital-Natrium und zudem eine wesentlich grössere therapeutische Breite (Quotient zwischen LD 50 und ED 50) aufweisen.
Die Verbindungen gemäss Formel I sowie ihre Säure-Additionssalze sollen als Schlaf- und Narkosemittel in der Human- und Veterinärmedizin Verwendung finden.
Die gewünschten Verbindungen werden erhalten, wenn man eine Verbindung der allgemeinen Formel:

worin R die genannte Bedeutung hat und X eine reaktive, mit Wasserstoff von Morpholin abspaltbare Gruppe bedeutet, mit Morpholin umsetzt. Als reaktive Gruppe X eignet sich insbesondere ein Halogenatom, vorzugsweise ein Chloratom, die Sulfhydrilgruppe, eine niedrige Alkylthiogruppe, beispielsweise die Methylthiogruppe, oder eine gegebenenfalls durch Substituenten aktivierte Aralkyltbiogruppe, z. B. die p-Nitrobenzylthiogruppe.
Die Umsetzung wird vorzugsweise während 3 bis 15 Stunden in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. Benzol oder Toluol oder insbesondere einem Alkohol, z. B. einem Alkohol der Formel R-OH, wobei R die genannte Bedeutung hat, und bei einer Temperatur zwischen Zimmertemperatur und Rückflusstemperatur durchgeführt.
Als besonders vorteilhaft hat sich erwiesen, wenn das Reaktionsgemisch während ca. 8 Stunden auf Rückflusstemperatur erhitzt wird. Die Umsetzung kann unter den obenerwähnten Bedingungen auch ohne Lösungsmittel durchgeführt werden.
Falls man von Verbindungen der Formel II ausgeht, worin X Halogen bedeutet, und zu den freien Basen der Formel I gelangen will, soll die Umsetzung in Gegenwart eines säurebindenden Mittels, beispielsweise rriäthylamin oder unter Verwendung eines zumindest 1000/oigen Überschusses an Morpholin durchgeführt werden.
Falls man von Verbindungen der Formel II ausgeht. worin X Halogen bedeutet, und keine säurebindenden Mittel bzw. kein Überschuss an Morpholin zusetzt, erhält man Salze von Verbindungen der Formel I, aus welchen die Basen auf an sich bekannte Weise freigesetzt werden können.
Die nach dem beschriebenen Verfahren erhaltenen Verbindungen, welche auf an sich bekannte Weise isoliert und gereinigt werden, sind bei Zimmertemperatur teils feste, gegebenenfalls kristalline, teils flüssige basische Verbindungen, die durch Umsetzen mit geeigneten anorganischen oder organischen Säuren in ihre Säure Additionssalze übergeführt werden können. Hierfür haben sich als anorganische Säuren z. B. Halogenwasserstoffsäuren, Salpetersäure, Phosphorsäure und als organische Säuren z. B. Methansulfonsäure und Pikrinsäure als geeignet enviesen.
Zu den als Ausgangsmaterialien erwähnten Verbindungen der Formel II, worin X für ein Halogenatom steht, kann man z. B. gelangen, wenn man eine Verbindung der Formel:

worin Hal Halogen bedeutet, mit einem Alkohol der Formel R-OH, wobei R die genannte Bedeutung hat, umsetzt. Verbindungen der Formel III sind bekannt oder können auf an sich bekannte Weise hergestellt werden.
Ausgangsverbindungen der Formel II, worin X die Sulfhydrilgruppe bedeutet, erhält man z. B. durch Umsetzen von Uracilcarbonsäure (5) der Formel:

mit Phosphorpentasulfid und Verestern der erhaltenen Verbindung bzw. eines reaktionsfähigen Säurederivates davon, insbesondere eines Halogenides, mit einem Alkohol der Formel R-OH. Durch Alkylierung oder entsprechende Aralkylierung des so erhaltenen Produktes kann man zu den Ausgangsmaterialien der Formel II, worin X eine Alkylthio- oder eine gegebenenfalls aktivierte Aralkyltbiogruppe bedeutet, gelangen.
Diejenigen Ausgangsverbindungen der Formel II, worin der Rest X eine andere Bedeutung besitzt als die eines Halogenatoms, der Sulfhydrilgruppe, einer Alkylthiogruppe oder einer gegebenenfalls aktivierten Aralkylthiogruppe können in analoger Weise hergestellt werden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert.
Beispiel 1
11,7 g 2,4-Dichlor-5-carbisobutoxy-pyrimidin werden in 100 ml Isobutanol gelöst und mit 11 g Tri äthylamin und 8,8 g Morpholin versetzt. Das Gemisch wird während 8 Stunden am Rückfluss erhitzt und hierauf zur Trockne eingedampft. Der Rückstand wird in Chloroform gelöst und mit Wasser unter Zusatz von Eis gewaschen, mit Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wird in Hexan gelöst und durch Aluminiumoxyd filtriert. Nach Abdampfen des Lösungsmitteis erhält man 2,4-bis-Mopholino-5-carbisobutoxy-pyrimidin in Form eines Öles, welches nach längerem Stehenlassen Kristalle vom Schmelzpunkt 64-68 "C liefert.
Den in diesem Beispiel verwendeten Ausgangsstoff erhält man wie folgt:
Eine Lösung von 10 g 2,4-Dichlor-uracil-5-carbonsäurechlorid in 100 ml absolutem Toluol wird unter Rühren und Eiskühlung zu einem Gemisch von 3,7 g Isobutanol, 8 g Triäthylamin und 120 ml absolutem Toluol getropft, worauf man während 2 Stunden am Rückfluss erhitzt. Das Reaktionsgemisch wird hiermit mit Wasser und 2N-Kaliumbicarbonatlösung gewaschen, mit Natriumsulfat getrocknet und am Vakuum eingedampft.
Man erhält 2,4-Dichlor-5-carbisobutoxy-pyrimidin in Form einer rötlich gefärbten Flüssigkeit vom Siedepunkt 140-150 ob/0,4 mm Hg.
Beispiel 2
Bei gleichem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-carbisopropoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbisopropoxy-pyrimidin in Form eines Öles. Dises wird in Äther gelöst und mit ätherischer Pikrinsäure versetzt. Der nach Eindampfen des Reaktionsgemisches erhaltene Rückstand wird aus Methanol/Wasser kristallisiert. Man erhält 2,4-bis-Morpholino-5-carbiso propoxy-pyrimidin-Pikrat vom Schmelzpunkt 170-172 "C.
Beispiel 3
Bei gleichem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-carbobutoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbobutoxy-pyrimidin in Form eines nicht destillierbaren Öls.
Beispiel 4
Bei gleichem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbo-sek.-butoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbosek.-butoxy-pyrimidin-Pikrat vom Schmelzpunkt 158-160 "C.
Beispiel 5
Bei gleichem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbo-tert.-pentoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbo tert.-pentoxy-pyrimidin-Pikrat vom Schmelzpunkt 163-166 CC.
Beispiel 6
Bei gleichem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbohexoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5carbohexoxy-pyrimidin-Pikrat vom Schmelzpunkt 121-125 OC.
Beispiel 7
Bei gleichem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbisopentoxy-pyrimi- din, erhält man 2,4-bis-Morpholino-5 carbisopentoxy-pyrimidin-Pikrat vom Schmelzpunkt 160-164 "C.
Beispiel 8
Bei gleichem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-carbäthoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbäthoxy-pyrimidin vom Schmelzpunkt 92-94 "C.
Beispiel 9
Bei analogem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-5-carbomethoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbomethoxy-pyrimidin vom Schmelzpunkt 117-120 "C.
Beispiel 10
Bei analogem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-5-carbopropoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbopropoxy-pyrimidin vom Schmelzpunkt 68-75 "C.
Beispiel 11
Bei analogem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbopentoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5 carbopentoxy-pyrimidin-Pikrat vom Schmelzpunkt 156159C C.
Beispiel 12
Bei analogem Vorgehen wie in Beispiel 2, jedoch ausgehend von 2,4-Dichlor-5-carbo-tert.-butoxy-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbo tert.-butoxy-pyrimidin-Pikrat vom Schmelzpunkt 188-191 OC.

Bei analogem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-5-carbo-(2-methyl-butoxy)-pyrimidin, erhält man 2,4-bis-Morpholino-5-carbo- (2-methyl-butoxy)-pynmidin vom Schmelzpunkt 90-94 "C.
Beispiel 18
Bei analogem Vorgehen wie in Beispiel 1, jedoch ausgehend von 2,4-Dichlor-5-carbo-(2-äthyl-butoxy)-pynmidin, erhält man 2,4-bis-Morpholino-5-carbo (2-äthyl-butoxy)-pyrimidin vom Schmelzpunkt 113-116 "C.

The compounds according to formula I and their acid addition salts are intended to be used as sleeping and anesthetic agents in human and veterinary medicine.
