Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate des p-Aminoalkyl-benzolsulfonamids.

Wie nun gefunden wurde, besitzen die neuen Verbindungen, insbesondere das 1 -[p-(2-Butyramidoäthyl)-phenylsulfonyl]-2-imino-3 -propyl-imidazolidin, 1 -[p- < 2-(m-Methoxy-benzamido)-äthyl,-phenylsulfonyl]- -2-imino-3-propyl-imidazolidin, 1 -[p-(2-Propionamido-äthyl)-phenylsulfonyl]-2-imino-3 -isopropyl-imidazolidin, 1-[p-(2-Butyramidoäthyl)-phenylsulfonyl]-20imino-3-isopropyl-imidazolidin, 1-[p-(2-Acetamido-äthyl)-pyenysulfonyl]-2-imino-3-butyl-imidazolidin, 1-[p-2-(2-Methyl-butyramido)-äthyl-phenylsulfonyl]-2 imino-3-n-butyl-imidazolidin, 1 -[p.(2-Valeramino-äthyl)-phenylsulfonyl]-2-imino-3- -butyl-imidazolidin,

Die hypoglykämische Wirkung wurde an Standardversuchen an Warmblütern, z.B. an Ratten, nachgewiesen.
In den Verbindungen der allgemeinen Formel I kann R beispielsweise folgende Bedeutungen haben: Als Alkylgruppe: die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, sec. Butyl-, tert. Butyl-, Isobutyl-, Pentyl-, Isopentyl-, 2,2-Dimcthyl-propyl-, 1-Methyl-butyl-, 1-Äthyl-propyl-, 1,2-Dimethyl-propyl-, die Hexyl-, Heptyl-, Octyl-, tert. Octyl-, Nonyl-, Decyl- oder die Dodecylgruppe; als Alkenylgruppe: die Allyl-, 1 -Methyl-allyl-, 2-Methyl-allyl-, Butenyl- oder Pentenyl; als Cycloalkylgruppe: die Cyclopropyl-, Cyclobutyl-, Cyclopentyl-, 2- und 4-Methylcyclohexyl, Cyclohexyl- oder die Cycloheptylgruppe ; als Cycloalkenylgruppe : die 2-Cyclopenten-1-yl-, 2-Cyclohexen-1-yl-, 3-Cyclohexen-1-yl-, 2-Methyl-2-cyclohexen -1-yl oder die 2-Cyclohepten-1-ylgruppe ; als Phenylalkylgruppe : die Benzyl-, Phenäthyl- oder die x-Methylphen äthylgruppe.
Der Substituent R2 als Alkylgruppe kann die Methyloder Äthylgruppe bedeuten und der Substituent R als Alkylgruppe die unter R1 aufgezählten Alkylgruppen mit höchstens 7 Kohlenstoffatomen; als Chloralkylgruppe beispielsweise: die 1-Chloräthyl-, 1 -Chlorpropyl-, 1-Chlorbutyl-, 1-Chlorpentyl-, 1-Chlorhexyl-, 2-Chlorhexyl- oder die 1-Chlorheptylgruppe; als Cycloalkylgruppe;

Der als Substituent R2 vorkommende Phenylrest oder die im R3 vorliegende Phenylgruppe können einfach bis dreifach substituiert sein.
Dieser Substituent oder diese Substituenten können folgende Gruppen sein: Als Halogen: Chlor, Fluor oder Brom, als niedere Alkylgruppen: Die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, sec.Butyl- oder die tert.Butylgruppe und als Alkoxy- oder Alkylthiogruppe: die Methoxy-, Äthoxy-, Methylthio- oder die Äthylthiogruppe.
Nach dem erfindungsgemässen Verfahren stellt man Verbindung der allgemeinen Formel I her. indem man eine Verbindung der allgemeinen Formel II

in welcher R, und R2 die unter Formel I angegebene Bedeutung haben, unter Ringschluss kondensiert und gegebenenfalls die erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
Geeignete reaktionsfähige Ester von Hydroxyverbindungen der allgemeinen Formel III sind beispielsweise Halogenide, insbesondere Chloride oder Bromide, ferner Sulfonsäureester, z.B. der o- oder p-Toluolsulfonsäureester oder der Methansulfonsäureester. Die Kondensation erfolgt vorzugsweise in einem mit Wasser mischbaren oder nichtmischbaren Lösungsmittel in An- oder Abwesenheit von Wasser. Als Lösungsmittel können Alkanole, z.B. Butanol, ätherartige Flüssigkeiten, z.B. Dioxan, Diäthylenglykolmonomethyläther, Carbonsäureamide, wie N,N-Dimethylformamid, oder Sulfoxide, wie Dimethylsulfoxid, verwendet werden. Es ist vorteilhaft, die Kondensation in Gegenwart eines säurebindenden Mittels vorzunehmen. Als solche können anorganische Basen und Salze, z.B.
Alkalimetallhydroxide, -hydrogencarbonate, -carbonate und -phosphate verwendet werden, ferner auch tertiäre organische Basen, wie z.B. N,N-Diisopropyl -äthylamin.
Als Ausgangsprodukte des vierten Verfahrens können z.B. die weiter vorne aufgezählten reaktionsfähigen Ester einer Hydroxyverbindung der allgemeinen Formel III verwendet werden, deren Symbole R1 und R mit den Symbolen übereinstimmen, die anschliessend an Formel I aufgezählt sind. Eine Gruppe von Bromiden der allgemeinen Formel III können z.B. erhalten werden, wenn man 1-Alkyl-aziridine [vgl. A. Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, Part One, John Wiley Sons Inc., London (1964)] mit Bromcyan in Dioxan umsetzt.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel I mit der äquivalenten Menge einer Säure in einem geeigneten wässrigorganischen oder organischen Lösungsmittel, wie z.B.
Methanol, Äthanol, Diäthyläther, Chloroform oder Methylenchlorid.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, -Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure, sowie Salze mit blutzuckersenkenden Sulfonylharnstoffen, wie z.B. p -Toluolsulfonyl - butyl - harnstoff, p - Chlorbenzolsulfonyl -propylharnstoff, p - [2 - (2 - Methoxy - 5 -chlorbenzamido) -äthyl] -phenylsulfonyl-cyclohexylharnstoff.
Die neuen Wirkstoffe werden vorzugsweise peroral verabreicht. Die täglichen Dosen bewegen sich zwischen 30 und 300 mg für erwachsene Patienten mit normalem Gewicht. Geeignete Doseneinheitsformen, wie Dragees, Tabletten enthalten vorzugsweise 30-300 mg eines erfindungsgemässen Wirkstoffes und zwar 20 bis 80% einer Verbindung der allgemeinen Formel I.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzige Ausführungsform desselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
Zu einem Gemisch von 4 g Natriumhydroxyd, 15 ml Wasser und 300 ml Dimethylsulfoxyd löst man 28,4 g p-(2-Isovaleramido-äthyl)-benzolsulfonamid und 16g N -(2-Chlor-äthyl)-N-butyl-cyanamid. Die erhaltene Lösung kocht man 1 Stunde am Rückfluss (Badtemperatur: 1100).
Das Dimethylsulfoxyd wird dann im Vakuum abdestilliert und der Rückstand, ein braunes öl, in Methylenchlorid aufgenommen. Man wäscht dreimal mit Wasser, trocknet und filtriert die organische Phase, die nach Eindampfen braune Kristalle ergibt. Nach Kristallisation aus Essigester erhält man das 1-[p-(2-Isovaleramido-äthyl)-phenylsulfonyl-2-imino-2-butyl-imidazolidin vom Smp. 130 bis 130,50.
In analoger Weise erhält man aus: 14,7 g N-(2-Chlor-äthyl)-N-propyl-cyanamid und 27,0 g p-(2-Butyramido-äthyl)-benzolsulfonamid das 1-[p-(2 -Butyramido-äthyl)-phenylsulfonyl]-2-imino-3-propyl- -imidazolidin, Smp. 114 - 1160; 14,7 g N-(2-Chlor-äthyl)-N-propyl-cyanamid und 33,4 g p-[2-(3-Methoxy-benzamido)-äthyl]-benzolsulfonamid das l;[p- < 2-(3-Methoxy-benzamido)-äthyl > -phenylsulfonyl]-2- -imino-3-propyl-imidazolidin, Smp. 144 - 1470; 16,0 g N-(2-Chloräthyl)-N-tert.butyl-cyanamid und 39,6 g p-[2-(3,4,5-Trimethoxy-benzamido)-äthyl]-benzolsulfon- amid das 1 -[p- < 2-(3,4,5-Trimethoxy-benzamido)-äthyl > - -phenylsulfonyl]-2-imino-3-tert.butyl-imidazolidin, Smp.

Beispiel 2

Nach Kristallisation aus Essigester erhält man das 1-[p-(2-Isovaler- amido-äthyl)-benzolsulfonyl]-2-imino-3-tert. butyl-imidazolidin vom Smp. 152 - 154 .
In analoger Weise erhält man aus: 16,0 g N-(2-Chloräthyl)-N-tert.butyl-cyanamid und 31,8 g p-[2-(2-Toluylamido)-äthyl]-benzolsulfonamid das 1-[p -2-(2-Toluylamido)-äthyl-phenylsulfonyl]-2-imino-3 -tert, butyl-imidazolidin. in which R 1 and R 2 have the meaning given under formula I, condensed with ring closure and, if appropriate, the compound obtained is converted into an addition salt with an inorganic or organic acid.
Suitable reactive esters of hydroxy compounds of the general formula III are, for example, halides, in particular chlorides or bromides, and also sulfonic acid esters, e.g. the o- or p-toluenesulfonic acid ester or the methanesulfonic acid ester. The condensation is preferably carried out in a water-miscible or immiscible solvent in the presence or absence of water. Alkanols, e.g. Butanol, ethereal liquids, e.g. Dioxane, diethylene glycol monomethyl ether, carboxamides such as N, N-dimethylformamide, or sulfoxides such as dimethyl sulfoxide can be used. It is advantageous to carry out the condensation in the presence of an acid-binding agent. As such, inorganic bases and salts, e.g.
Alkali metal hydroxides, hydrogen carbonates, carbonates and phosphates are used, and also tertiary organic bases, such as e.g. N, N-diisopropylethylamine.
As starting products of the fourth process, e.g. the reactive esters of a hydroxy compound of the general formula III listed above are used, the symbols R1 and R of which correspond to the symbols which are listed after formula I. A group of bromides of general formula III can e.g. be obtained when 1-alkyl-aziridines [cf. A. Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, Part One, John Wiley Sons Inc., London (1964)] with cyanogen bromide in dioxane.
The compounds of general formula I obtained by the process according to the invention are then, if desired, converted into their salts with inorganic and organic acids. These salts are produced e.g. by reacting the compounds of general formula I with the equivalent amount of an acid in a suitable aqueous, organic or organic solvent, such as e.g.
Methanol, ethanol, diethyl ether, chloroform or methylene chloride.
For use as medicinal substances, their pharmaceutically acceptable salts with acids can be used instead of the free compounds of the general formula I. Suitable addition salts are e.g. Salts with hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulphonic acid, ethanesulphonic acid, -hydroxyethanesulphonic acid, acetic acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, malic acid, tartaric acid, citric acid, salicylic acid, such as sulphonic acid, phenyl acetic acid, and alicylic acid, such as sulphonic acid, phenyl acetic acid, and emanoic acid, such as sulphonic acid, phenyl acetic acid, such as sulphonic acid, phenyl acetic acid e.g. p -toluenesulfonylbutylurea, p -chlorobenzenesulfonylpropylurea, p - [2 - (2 - methoxy - 5-chlorobenzamido) ethyl] phenylsulfonylcyclohexylurea.
The new active ingredients are preferably administered orally. Daily doses range from 30 to 300 mg for adult patients of normal weight. Suitable dosage unit forms such as dragees, tablets preferably contain 30-300 mg of an active ingredient according to the invention, namely 20 to 80% of a compound of the general formula I.
The following examples explain the preparation of the new compounds of general formula I and of hitherto not described intermediates in more detail, but by no means represent the only embodiment of the same. The temperatures are given in degrees Celsius.
Example I.
28.4 g of p- (2-isovaleramido-ethyl) -benzenesulfonamide and 16 g of N- (2-chloro-ethyl) -N-butyl-cyanamide are dissolved in a mixture of 4 g of sodium hydroxide, 15 ml of water and 300 ml of dimethyl sulfoxide. The solution obtained is refluxed for 1 hour (bath temperature: 1100).
The dimethyl sulfoxide is then distilled off in vacuo and the residue, a brown oil, is taken up in methylene chloride. It is washed three times with water, dried and filtered, the organic phase, which gives brown crystals after evaporation. After crystallization from ethyl acetate, 1- [p- (2-isovaleramido-ethyl) -phenylsulfonyl-2-imino-2-butyl-imidazolidine with a melting point of 130 to 130.50 is obtained.

28.4 g of p- (2-isovaleramido-ethyl) -benzenesulfonamide and 16 g of N- (2-chloro-ethyl) -N-tert-butyl are dissolved in a mixture of 4 g of sodium hydroxide, 15 ml of water and 300 ml of dimethyl sulfoxide - The solution obtained is refluxed for 1 hour (bath temperature: 110). The dimethyl sulfoxide is then distilled off in vacuo and the residue, a brown oil, is taken up in methylene chloride. It is washed three times with water, dried and filtered, the organic phase, which gives brown crystals after evaporation.
