Verfahren zur Herstellung von neuen basisch substituierten Naphthalin-Derivaten und ihre Verwendung zur Herstellung ihrer quartären Ammoniumverbindungen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen basisch substituierten N;aphthalinbDefivaten der Formel I

R1 und R Wasserstoff, Halogen oder Nitro-, Al kyl-, Alkoxy- bzw. Trifluormethylgruppen, X einen gesättigten oder ungesättigten, geradkettigen oder verzweigten Alkylenrest, der gegebenenfalls durch eine Oxo- oder Hydroxygruppe substituiert sein kann, A einen geradkettigen oder verzweigten Alkylrest, und Y einen basischen organischen Rest.
Es wurde gefunden, dass die Verbindungen der Formel I hervorragende fungizide und/oder bakterizide Eigenschaften aufweisen.
Als basische Substituenten Y kommen gegebenenfalls alkylierte Aminogruppen oder die Reste von N Heterocyclen (z. B. Pyrrolidino-, Piperidino-, Morpholino-, Piperazino-, Imidazolidinoreste) in Frage.
Das erfindungsgemässe \Verfahren zur Herstellung der neuen basisch substituierten Naphthalin-Denvate ist dadurch gekennzeichnet, dass man ein Naphthoi-Deri- vat der Formel II

mit Verbindungen der Formel III
U-A-Y, III in welcher U eine gegebenenfalls mit einem reaktiven Säurerest veresterte Hydroxylgruppe bedeutet, umsetzt.
Die erhaltenen Verbindungen der Formel I können - für den Fall, dass X ein durch eine Oxogruppe substituierter Alkylenrest ist - gewünschtenfalls nachträglich zu den entsprechenden Hydroxyverbindungen reduziert, diese zu den ungesättigten Alkylenverbindun- gen dehydratisiert, letztere zu den gesättigten Alkylenverbindungen reduziert und gewünschtenfalls in ihre Salze überführt werden.
Ausserdem können die erhaltenen Verbindungen zur Herstellung ihrer quaternären Ammoniumverbindungen verwendet werden.
Die Umsetzung der Verbindungen der Formeln II und III erfolgt nach den üblichen Verätherungsmetho den, zweckmässig durch Erhitzen der Komponenten in einem geeigneten Lösungsmittel in Gegenwart von Alkali (bzw. Alkalialkoholaten oder -amiden). Als reaktive Säurereste U kommen vorzugsweise Halogen- und aromatische Sulfonsäurereste (Tosyl-, Mesyl-, Brosyl) in Frage. Anstelle der Aminoalkylverbindungen U-A-Y lassen sich auch die Alkylverbindungen U-A-U (z.B.
Alkyldihlalogenide bzw. entsprechende Alkylenoxide) verwenden, wobei man Zwischenprodukte der Formel IV erhält

weiche anschliessend durch Behandlung mit Basen H-Y in die Verfahrensprodukte I übergeführt werden können; für den Fall, dass U in der obigen Formel IV eine freie OH-Gruppe ist, wird man diese zweckmässig vor der Aminierung in eine reaktive Estergruppe umwandeln.
Für den Fall, dass X in den Ausgangsprodukten der Formel II ein durch eine Oxogruppe substituierter Alkylenrest ist, können die erhaltenen Verbindungen der Formel I nachträglich in üblicher Weise zu den ent sprechenden Hydroxyverbindungen (X = OH-substituier ter Alkylenrest) reduziert werden, wobei man Wfzugs- weise ein komplexes Metallhydrid (z. B. Natriumbor hydrid, Lithiumaluminiumhydrid) verwendet Die ge nannten Hydroxyverbindungen lassen sich mit üblichen Dehydratisierungsmitteln (z. B. alkoholischer Salzsäure) in die ungesättigten Alkylenverhindungen überführen, die wiederum (vorzugsweise durch katalytische Hydrierung) zu den gesättigten Alkylenderivaten reduziert werden können.
Die basischen Verfahrensprodukte lassen sich mit
Hilfe anorganischer oder organischer Säuren in bekann ter Weise in die entsprechenden Salze (z. B. die Hy drohalogenide) überführen. Zur Herstellung der quartä ren Ammoniumverbindungen werden die Verbindungen der Formel I mit den üblichen Quaternärisierungsmitteln (z.B. Alkyl- bzw. Aralkylhalogeniden) behandelt.
Die als Ausgangsverbindungen der Formel II benutzten Naphthol-Derivate erhält man z. B. durch Umsetzung von Alkoxy-naphthalinen mit einer Verbindung der Formel V

unter den Bedingungen der Friedel-Crafts-Synthese, wo bei gleichzeitig oder anschliessend die Alkoxygruppe zur freien OH-Gruppe lentalkyliert wird.
In den nachstehenden Beispielen ist das erfindungs gemässe Verfahren näher erläutert.
Beispiel 1 2-(ss-Diäthyiamino äthoxy)- 6-(4'-chlorphenacetyl)- naphthalin
2,3 g Natrium (0,1 Mol) werden in 250 ml Iso propanol gelöst und mit 29,6 g (0,1 Mol) 6-(4'-Chlor phenacetyl)-naphthol-(2) 10 Minuten zum Sieden er hitzt. Anschliessend kühlt man ab, lässt 14,8 g Di äthylaminoäthylchlorid (0,11 Mol) zutropfen und er hitzt 3 Stunden unter Rückfluss. Der Niederschlag wird abgesaugt und das Lösungsmittel im Vakuum entfernt.
Danach nimmt man den Rückstand in Ather auf und fällt durch Zugabe von ätherischer Salzsäure das Hy drochlorid des
2-(ss-Diäthylaminoäthoxy)-6-(4'-chlor- phenacetyl)-naphthalins aus, das aus Isopropanol umkristallisiert werden kann.
Ausbeute 29,5 g (68,5% d. Th); Fp. 186 bis 1870C.

Beispiel 2 Phenyl-2-(ss-diäthylaminoäthoxy)-naphthyl-6]- carbinol
6 g des gemäss Beispiel 1 hergestellten 2-(ss-Di äthylaminoäthoxy)-6-benzoyl-naphthalins (0,0177 Mol) werden mit 0,68 g <RTI
ID=3.1> Lithiumaluminiurahydrid (0,0177 Mol) in 50 mol äther 2 Stunden zum Sieden erhitzt.
Anschliessend zersetzt man durch Zugabe von gesättigter wässriger Natriumchloridlösung, trennt die organische Schicht ab, trocknet sie mit Natriumsulfat und dampft ein. Es werden nach dem Umkristallisieren aus ,einer bei 100 bis 1400 C siedenden Petrolfraktion 5 g (81 % d. Th.) Phenyl-[2-(ss-diäthylaminoäthoxy)-naph- thyl-6]-carbinol vom Fp. 82 bis 830 C erhalten.

1-[2'-(γ-Dimethylaminopropoxy)-naphthyl-6']- 2-(4'-chlorphenyl)-äthylen
3 g 4-Chlorbenzyl-[2'-(γ-dimethylaminopropoxy)- naphthyl-6']-carbinol-hydrochlorid (0,0072 Mol) werden in 30 mi alkoholischer Salzsäure 1 Stunde zum Sieden erhitzt. Nach dem Abkühlen kristallisieren 2,5 g (86,5%) 1-[2'-(γ-Dimethylaminopropoxy)-naphthyl-6']-
2- (4"-chlorphenyl) -äthylen in Form des Hydrochlorids vom Fp. 275 bis 2770 C aus.

Beispiel 4 2-(ss-Diäthylaminoäthoxy)-6-(4'-chlorphenacetyl)- naphthalin-methojodid
9,9 g des gemäss Beispiels 1 hergestellten
2-(ss-Diäthylaminoäthoxy)-6-(4'-chlorphenacetyl)- naphthalins (0,025 Mol) werden mit 4,2 g Methyljodid (0,03 Mol) in 50 mi Aceton 2 Stunden zum Sieden erhitzt. An schliessend saugt man die in der Kälte ausgeschiedenen Kristalle ab und kristallisiert aus Methyläthylketon um.
Es werden 12,0 g (86,5 % d. Th.) Methojodid vom Fp. 152 bis 1530 C isoliert.
In analoger Weise erhält man durch Umsetzung mit Äthylbromid das entsprechende Athobromid (Fp.
176 bis 1770 C, Ausbeute 56 % d. Th.) oder durch Umsetzung mit Propyljodid das entsprechende Propiojodid (Fp. 160 bis 1610 C, Ausbeute: 49 % d. Th.).
Beispiel 5 2-(ss-Diäthylaminoäthoxy)-6-[ss'-(2'-chlorphenyl)- äthyl]-naphthalin
10,4 g von analog Beispiel 3 hergestelltem
1 -[2'-(ss-Diäthylaminoäthoxy)-naphthyl-6']- 2-(2"-chlorphenyl)-äthylenhydrochlorid (0,025 Mol) werden in 100 mi Methanol gelöst und nach Zugabe von 0,5 g 5 % iger Palladiumkohle drucklos hydriert. Anschliessend saugt man ab, dampft ein und erhält durch Umfällen aus I & propanol/Äther das zum Teil amorphe Hydrochlorid (72% Ausbeute) des 2-(ss-Diäthylaminoäthoxy- 1 )-6 -[ss'-(2'-chlor- phenyl)-äthyl]-naphthalins vom Fp. etwa 1230 C.
In analoger Weise erhält man durch Hydrierung des 1 -[2'-(y-Diäthylaminopropoxy)-naphthy1-6']-
2-(4"-chlorphenyl)-äthylen-hydrochlorids in 83 % iger Ausbeute das Hydrochlorid des 2-(;'-Diäthyiaminoprnpoxy)-6-[ss'-(4'-chior- phenyl) -äthyl] -naphthalins vom Fp. 154 bis 1550 C (aus Isopropanol).
