Verfahren zur Herstellung neuer quaternärer Amnoniumsalze
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer quaternärer Ammoniumsgalze mit wertvollen mikrobioziden Eigenschaften.
Quaternäre TetraalkylXammonium-Salze sind als antimikrobielle Wirkstoffe im Pflanzen- und Materialschutz sowie in der Human-Medizin wohlbekannt. Demgegenüber haben Aralkyl- und Aryloxyalkyl-trialkylammonium-Salze nur im Mbatefialschutz und zur medizinischen Desinfektion Bedeutung erlangt. Für den Pflanzenschutz sind diese quaternären Ammoniumsalze wegen ihrer Phytotoxizität und ihrer geringen Witterungsbeständigkeit nicht geeignet.
Überraschenderweise wurde nun gefunden, dass quaternäre Ammoniumsalze der Formel

ausgezeichnete mikrobizide Eigenschaften aufweisen und sich insbesondere zur Bekämpfung phytopathogener Pilze eignen. Diese Wirkstoffe sind nicht phytotoxisch und gegenüber Witterungseinflüssen weitgehend beständig. Sie besitzen auch eine gute Wirksamkeit gegen solche Pilze und Bakterien, die organische Materialien und Gebrauchsgegenstände schädigen und zerstören.
In der Formel I bedeuten die Symbole R1, R2, R4 je einen unsubstituierten oder substituierten aliphatischen Kohlenwasserstoffrest, wobei mindestens einer dieser Reste 8 bis 24 Kohlenstoffatome aufweist, Rq eine Aikanoyl-, Carboxyl-, Alkoxy-carbonylgfuppe, einen a-Hydroxyalkyirest, einen acylierten a-Hydroxyal- kylrest oder einen durch Hydrolyse in eine Alkanoyl- gruppe überführbaren Rest, R5 Wasserstoff oder ein Halogenatom mit einem Atomgewicht unter 100, R6 Wasserstoff, ein Halogenatom mit einem Atonigewicht unter 100, die Nitro-, Amino-, Cyano- oder Hydroxyl Gruppe, einen unsubstituierten oder substituierten aliphatischen Kohlenwasserstoffrest,
einen über Sauerstoff oder Schwefel gebundenen, unsubstituierten oder substituierten aliphatischen Kohlenwasserstoffrest, eine Alkylamino-, Dialkyiamino- oder Acylaminogruppe, oder R5 und RG zusammen einen ankondensierten Benzolkern, X ein Sauerstoff- oder Schwefelatom, Z ein aliphatisches Kohlenwasserstoffkettenglied mit 2 bis 6 Kohlenstoffatomen, wovon mindestens 2 in der direkten Kette, A ein Äquivalent eines Anions einer organischen oder anorganischen Säure und n die Zahl 1 oder 2 ist.
Die Kohlenwasserstoffreste Rt, R2 und R3 sind geradkettige oder verzweigte Alkyl- oder Alkenylreste.
Diese Reste können z. B. durch Halogen, die Hydroxyl-, Mercapto-, Amino- oder Cyanogruppe und/oder Alkoxy-, Alkylthio-, Alkyl- oder Dialkylaminogruppe substituiert sein. Die Alkanoylgruppe R4 bedeutet insbesondere den Formyl-, Acetyl-, Propionyl- oder Butyryl Rest. Als ein durch Hydrolyse in eine Alkanoylgruppe überführbaren Rest bedeutet R4 einen Acetal-, Acyial oder Ketal-Rest. Als Acetal-Rest kommen die Reste von Hemiacetalen, monomeren und polymeren Acetalen in Betracht. Das aliphatische Brückenglied, über das die genannten Alkanoyl-, Carboxyl- und Alkoxycarbonyl- gruppen gebunden sein können, ist insbesondere ein niederer Alkylen- oder Alkenylen-Rest.
Ein aliphati- scher Kohlenwasserstoffrest Rg weist vorzugsweise 1 bis 4 Kohlenstoffatome auf; er kann z. B. durch Halogen substituiert sein. Mehrere Substituenten können untereinander gleich oder verschieden sein. Z als aliphati sches Kohlenwasserstoffkettenglied bedeutet insbesondere einen Alkylen- oder Alkenylen-Rest. Das Symbol A steht für ein Anion einer anorganischen oder organischen Säure. In Betracht kommen beispielsweise die Anionen der folgenden Säuren:
Halogenwasserstoffsäuren, Schwefelsäure,
Phosphorsäure, Perchlofsäure, Alkyl-Schwefelsäuren, Naphthoes äuren,
Benzoesäure, Essigsäure, Aminoessigsäure,
Buttersäure, Stearinsäure, Oxalsäure,
Adipinsäure, Maleinsäure, Weinsäure,
Milchsäure usw.
Das erfindungsgemässe Verfahren zur Herstellung von quaternären Ammoniumsalzen der Formel I ist dadurch gekennzeichnet, dass man eine Verbindung der Formel II

mit äquivalenten Mengen eines quaternären Ammoniumsalzes der Formel III

umsetzt, in welchen Formeln B und D bei der Umsetzung sich - mit Ausnahme eines in einem der beiden enthaltenen 0- oder S-Atoms - abspaltende Reste sind, und R1, R2, R , R.1, R5, R6, Z, A und n die unter Formel I angegebenen Bedeutungen haben.
In den allgemeinen Formeln II und III können von den Symbolen B und D, z. B. eines die Hydroxyl- oder Mercaptogruppe, und das andere ein Halogenatom, oder eines die Gruppe 0- oder S-Me, worin Me ein Metallatom darstellt, und das andere ein Halogenatom bedeuten.
Es ist vorteilhaft, die Umsetzung bei einer Temperatur zwischen 110 und 1500 C in Gegenwart eines gegenüber den Reaktionsteilnehmern inerten Lösungsoder Verdünnungsmittels und eines säurebindenden Mittels vorzunehmen. Die so erhaltenen quaternären Salze können zwecks Austausch des Anions mit anderen nichtphytotoxischen anorganischen oder organischen Säuren umgesetzt werden.
Die als Ausgangsmaterial dienenden substituierten Phenole und Thiophenole der allgemeinen Formel II sind bekannt und die Ammonium-Verbindungen der allgemeinen Formel III werden in einfacher Weise durch Zusammengeben äquimolarer Mengen eines tertiären Amins der Formel IV

und eines disubstituierten Kohlenwasserstoffes der Formel V D-Z-A (v) in welchen Formeln R1, R , Rs, A, D und Z die obige Bedeutung haben, in einem inerten organischen Lösungsmittel, erhalten.
Aufgrund ihrer ausgezeichneten Wirkung gegenüber zahlreichen phytopathogenen Pilze besitzen die quaternären Ammoniumsalze der Formel VI

in der R1, R , RQ, R5, R6, A, X und Z die unter Formel I angegebenen Bedeutungen haben und R4, eine Alkanoylgruppe oder einen durch Hydrolyse in eine solche überführbaren Rest darstellt, grosse Bedeutung.
Bevorzugte quaternäre Ammoniumsalze sind jene Verbindungen der Formel VI, in der R4' eine in o Stellung zum Brückenglied X stehende Formylgruppe und R5 und R6 je ein Halogenatom bedeuten.
Die quaternären Ammoniumsalze der Formel VI, in denen R4' den Formyfrest, einen Acetal- oder Acylrest darstellt, sind beispielsweise gegen folgende, Pflanzenkrankheiten erzeugende Pilze wirksam:
Phytophthora infestans Altefnaria solaní
Botrytis cinerea
Uromyces appendicuiatus
Venturia inaequalis
Von grosser Bedeutung ist die Wirksamkeit gegen Pilze, die als Erreger echter Mehltaukrankheiten bekannt sind;, wie z. B. Arten der Gattung Erysiphe. Es ist durch die Anwendung dieser Wirkstoffe möglich, Nicht-Mehltaupilze und an den gleichen Wirtspflanzen auftretende MehltawTnfektionen zu bekämpfen oder einzudämmen.
Die neuen Mittel können zum Schützen von Pflanzen und Pflanzenteilen (Blüten, Samen, Früchten, Laubwerk, Stengeln) vor dem Befall durch Pilze verwendet werden. Als Saatgutbeizmittel verleihen die neuen Mittel dem behandelten Saatgut einen guten Schutz, ohne dass Keimungshemmung auftritt. Diese Salze sind auch unterschiedlich bakterizid wirksam, ihr Hauptwert liegt aber in der hohen fungiziden Wirksamkeit. Die fungizide Wirksamkeit der erfindungsgemäss verwendbaren quaternären Ammoniumsalze wird an:
1. Botrytis cinerea auf Bohnen (Vicia faba) und
2. Aiternaria solani auf Tomaten geprüft.
Bei diesen Versuchen wurde das aus dem belgischen Patent Nur. 631 160 bekannte N-PlrenoxyäthyiN-dodecyl-N,N-dimethyl- ammoniumbromid als Vergleichssubstanz mitgeprüft. Während keines der erfindungsgemässen Salze die behandelten Pflanzen schädigte, verursachte die bekannte Verbindung starke Verbrennungen, die eine Beurteilung des Pilzwachstums nicht mehr ermöglichten.
Botrytis cinerea auf Bohnen (Vicia Faba)
Petrischalen werden mit feuchtem Filterpapier ausgelegt und mit je drei frischen abgeschnittenen Bohnenblättern (Vicia faba) beschickt (Biattoberseite nach unten).
Die Blätter werden dann mit einer 0,1 ,% igen wässerigen Wirkstoffsuspension besprüht. Nach dem Abtrocknen des Sprühbelages werden sie mit einer frischen Sporensuspension von Botrytis cinerea infiziert. Die Schalen werden abgedeckt und 1 bis 2 Tage bei Zimmertemperatur stehengelassen. Dann wird der Versuch nach der Skala 10 = Totalbefall; 9 bis 1 = Befallsverminderung je nach Ausmass; 0 = kein Pilzwachstum, ausgewertet. Die Kontrollpflanzen, die nun mit der Sporensuspension infiziert, nicht aber mit einem Wirkstoff behandelt wurden, zeigten Totalbefall und erhielten die Bewertung 10.

Die Blätter von 3 bis 4 Wochen alten Tomatenpflanzen (roter Gnom) werden mit einer wässrigen 0,1 % igen Wirkstoffsuspension gespritzt.
Nach dem Abtrocknen des Wirkstoffbelages werden die Blätter mit einer frischen Sporensuspension von Alternafia solani infiziert, und die Pflanzen 5 bis 6 Tage im Gewächshaus bei etwa 200 und 95 bis 100S Luftfeuchtigkeit aufgestellt. Die Versuche werden nach folgender Skala
10 = Totalbefall; 9 bis 1 = Befallsverminderung je nach Ausmass; O = kein Pilzwachstum, ausgewertet.
Die Kontrollpflanzen waren total befallen und er hielten die Bedeutung 10.

Für die Verwendung im Pflanzenschutz werden die neuen Wirkstoffe in üblicher Weise mit Verteilungsund/oder Trägerstoffen aufgearbeitet und so angewendet, dass die Wirkstoffkonzentration im Bereich von 0,01 bis 2% liegt.
Die folgenden Beispiele beschreiben die Herstellung der neuen Verbindungen der Formel I. Teile bedeuten, sofern nichts anderes vermerkt ist, Gewichtsteile, die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
1,58 Teile o-Hydroxyacetophenon-Na-Salz, 4,01 Teile α-Brom-ss-(dimethyl-n-dodecylammonio) äthyl-bromid und 50 Volumteile 96 % iger Äthylalkohol werden unter gutem Rühren während 10 Stunden am Rückfluss erhitzt. Danach wird der Äthylalkohol am Rotationsverdampfer restlos eingedampft. Der Rückstand wird in 70 Volumteilen siedendem Essigester aufgerührt und zur Entfernung von Salzen genutscht. Die klare Lösung wird eingedampft, und der klebrige Rückstand wird aus Aceton/Äther umkristallisiert. Man erhält das
N-(2-Acetyl-phenoxy-äthyl)-N,N-dimethyl-N dodecylmmonium-bromid vomFp. 91 bis 920.
Beispiel 2
5,1 Teile analog zu Beispiel 1 hergestelltem
N-(2-Formyl-4,6-dichlor-phenoxy-äthyl)-N,N dimethyl-N-dodecyl-ammonium-bromid werden in 70 Volumteilen 96 SOigem Äthanol gelöst und nach Zugabe von 1,5 Teilen Silberchlorid 25 Stunden lang bei 200 kräftig gerührt. Danach nutscht man den entstandenen Niederschlag und dampft das Filtrat zur Trockene ein. Das zurückgebliebene Ö1 wird durch Lösen in Aceton und durch Fällen mit wasserfreiem Ather gereinigt. Das erhaltene
N-(2-Formyl-4,6-dichlor-phenoxy-äthyl)-N,N dimethyl-N-dodecyl-ammonium-chlorid hat den Fp. 750.
Beispiel 3
5,1 Teile
N-(2-Formyl-4,6-dichlor-phenoxy-äthyl)-N,N dimethyl-N-dodecyl-ammonium-bromid werden in der Wärme in 50 Volumteilen Wasser gelöst. Dazu gibt man bei 70 bis 800 vorsichtig 5 Volumteile konzentrierte Schwefelsäure und lässt das Reaktionsgemisch anschliessend 15 Minuten stehen. Danach wird abgekühlt, worauf man das abgeschiedene Öl durch
Extraktion in Chloroform aufnimmt. Anschliessend trocknet man die Chloroformlösung über Calciumchlo rid und destilliert das Chloroform unter Vakuum ab.
Der Rückstand wird mit Aceton umkristallisiert. Das erhaltene
N-(2-Formyl-4,6-dichlor-phenoxy-äthyl)-N,N dimethyl-N-do decyl-ammonium-hydrosulfat hat den Fp. 1100.
Die in der nachstehenden Tabelle aufgeführten Ver bindungen werden in analoger Weise hergestellt.

Process for the preparation of new quaternary ammonium salts
The present invention relates to a process for the production of new quaternary ammonium salts with valuable microbiocidal properties.
Quaternary tetraalkyl-xammonium salts are well known as antimicrobial agents in the protection of plants and materials as well as in human medicine. In contrast, aralkyl and aryloxyalkyl trialkylammonium salts have only gained importance in the field of anti-bacterial protection and for medical disinfection. These quaternary ammonium salts are not suitable for plant protection because of their phytotoxicity and their poor weather resistance.
Surprisingly, it has now been found that quaternary ammonium salts of the formula

have excellent microbicidal properties and are particularly suitable for combating phytopathogenic fungi. These active ingredients are not phytotoxic and are largely resistant to the effects of the weather. They also have a good effectiveness against those fungi and bacteria that damage and destroy organic materials and everyday objects.

Of great importance is the effectiveness against fungi, which are known to cause powdery mildew diseases; B. Species of the genus Erysiphe. By using these active ingredients, it is possible to combat or contain non-powdery mildew fungi and powdery mildew infections that occur on the same host plants.
The new agents can be used to protect plants and parts of plants (flowers, seeds, fruits, foliage, stems) from being attacked by fungi. As a seed dressing agent, the new agents give the treated seed good protection without inhibiting germination. These salts also have different bactericidal effects, but their main value lies in their high fungicidal effectiveness. The fungicidal effectiveness of the quaternary ammonium salts which can be used according to the invention is determined from:
1. Botrytis cinerea on beans (Vicia faba) and
2. Aiternaria solani tested for tomatoes.

Petri dishes are lined with moist filter paper and filled with three fresh, cut bean leaves (Vicia faba) each (top side facing down).

1.58 parts of o-hydroxyacetophenone Na salt, 4.01 parts of α-bromo-ss- (dimethyl-n-dodecylammonio) ethyl bromide and 50 parts by volume of 96% ethyl alcohol are refluxed for 10 hours with thorough stirring .
