Verfahren zur Herstellung von 2,2-disubstituierten 3-Acyl-5ffi-azido- -thiazolidin-4-carbonsäuren oder Säurederivaten davon
Gegenstand der vorliegenden Erfindung ist ein Methodikverfahren zur Herstellung von Azidoverbindungen, das zur Darstellung von wertvollen Zwischenprodukten und insbesondere bei der erstmaligen synthetischen Herstellung der 7-Amino-cephalosporansäure und ihrer Derivate Anwendung fand und zu dieser eigenartigen Synthese besonders geeignet ist.

Derivate sind in erster Linie N-Acylverbindungen, worin Acylreste insbesondere diejenigen von wirksamen N Acylderivaten der 7-Amino-cephalosporansäure, wie der Thienylacetyl-, Cyanacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z.B. der tert. Butyloxycarbonylrest, bedeuten.
Die Synthese dieser für die Herstellung wertvoller Arzneimittel wichtigen Verbindung und ihrer Derivate beruht auf der Idee, von einer 3,5-unsubstituierten 2,2disubstituierten Thiazolidin-4-carbonsäure, z.B. einer Verbindung der Formel I

Zu den oben erwähnten, als Zwischenprodukte wertvollen 2,2-disubstituierten 3 -Acyl-5azido-thiazolidin-4- -carbonsäuren oder Säurederivaten davon, wie z.B. zur Verbindung der Formel VII, gelangt man überraschenderweise, indem man eine, in 5,ss-Stellung eine reaktionsfähige durch eine anorganische oder eine starke organische Säure veresterte Hydroxygruppe enthaltende 2,2disubstituierte 3-Acyl-thiazolidin-4-carbonsäure oder ein Säurederivat davon mit einem Salz der Stickstoffwasserstoffsäure umsetzt. Wenn erwünscht, kann in einer erhaltenen Verbindung ein Substituent in einen anderen übergeführt, und/oder, wenn erwünscht, ein erhaltenes Gemisch von Isomeren in die einzelnen Isomeren aufgetrennt werden.
Eine reaktionsfähige, wie oben erwähnt veresterte Hydroxygruppe ist eine durch eine starke Mineralsäure, wie eine Halogenwasserstoffsäure, z.B. Chlorwasserstoff-, Bromwasserstoff- oder Jodwasserstoffsäure, oder Schwefelsäure, oder durch eine starke organische Carbonsäure, z.B. 4-Nitrobenzoesäure, oder eine starke organische Sul fonsäure, z.B. Methansulfon-, p-Toluolsulfon-, p-Brombenzolsulfon- oder p- oder m-Nitrobenzolsulfonsäure, veresterte Hydroxygruppe.
Die Reaktion des Ausgangsmaterials mit Salzen der Stickstoffwasserstoffsäure, z.B. mit einem Metallazid, wie einem Alkalimetallazid, z.B. Natriumazid oder Lithiumazid wird vorteilhafterweise in Gegenwart eines geeigneten Verdünnungsmittels, besonders eines polaren, wasserfreien oder wasserhaltigen Lösungsmittels, wie Dimethylformamid oder Diäthylformamid, N-Methylpyrrolidon, Acetonitril oder Phosphorsäure-tri-dimethylamid, sowie Dimethylsulfoxyd, Dimethylacetamid oder eines Niederalkanols wie Methanol, Äthanol, Isopropanol oder tert. Butanol, wenn notwendig unter Kühlen oder Erhitzen und/oder in einer Inertgasatmosphäre durchgeführt.
In erhaltenen Verbindungen können Substituenten nach an sich bekannten Methoden in andere übergeführt werden, So können erhaltene Säurederivate, wie Ester, in die freie Säure umgewandelt werden, ohne dass die Acylgruppe, besonders eine leicht abspaltbare Acylgruppe, wie die tert.Butyloxycarbonylgruppe, in 3-Stellung entfernt wird. So lässt sich eine Carbodiphenylmethoxy- gruppe unter sauren Bedingungen, z.B. in Gegenwart von katalytischen Mengen einer Säure, wie Trifluoressigsäure, in eine freie Carboxylgruppe umwandeln.
Erhaltene Gemische von Isomeren können nach an sich bekannten Methoden, z.B. durch fraktioniertes Kristallisieren, Adsorptionschromatographie (Kolonnen- od.
Dünnschichtchromatographie) oder anderen Verfahren in die einzelnen Isomeren getrennt werden: erhaltene Racemate können durch Bilden eines Gemisches von diastereoisomeren Salzen mit optisch aktiven salzbildenden Mitteln, Trennen des Gemisches in die diastereoisomeren Salze und Überführen der abgetrennten Salze in die freien Verbindungen in die Antipoden getrennt werden.
Das Verfahren umfasst auch diejenigen Ausführungsformen, wonach Ausgangsstoffe in Form von während der Reaktion gebildeten rohen Reaktionsgemischen verwendet werden.
Die erfindungsgemäss erhaltenen Verbindungen sind 2,2- disubstituierte 3-Acyl-Ssc-azido-thiazolidin-4-carbon- säuren und deren funktionellen Derivate, insbesondere die Verbindungen der Formel VIIa

Acylreste Ac sind in erster Linie solche, welche in pharmakologisch wirksamen N-Acylderivaten der 7-Aminocephalosporansäure vorkommen, wie der Thienylacetyl-, z.B. 2-Thienylacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z.B. der tert. Butyloxycarbonylrest, oder irgendsolche andere geeignete Acylreste organischer Säuren.

Die Gruppe R1 steht für eine freie oder vorzugsweise funktionell abgewandelte Carboxylgruppe, insbesondere eine veresterte Carboxylgruppe. Letztere ist mit zur Ver- esterung von Carbonsäuren geeigneten Hydroxyverbindungen jeglicher Art, insbesondere mit aliphatischen Alkoholen wie Alkanolen, insbesondere Niederalkanolen, z.B. Methanol, Äthanol, n-Propanol oder tert.Butanol, cycloaliphatischen Alkoholen, wie Cycloalkanolen, z.B. Cyclohexanol, oder araliphatischen Alkoholen, wie Phenylalkanolen, z.B.
Benzylalkohol, oder Diphenylmethanol, sowie mit Phenolverbindungen verestert, wobei die obgenannten Hydroxyverbindungen unsubstituiert sind oder Niederalkyl-, Niederalkoxy- oder Trifluormethylgruppen oder insbesondere Halogenatome sowie andere Gruppen als Substituenten enthalten können; be besonders geeignet als die Carbonsäure veresternde substituierte Hydroxyverbindungen sind halogenierte Niederalkanole, wie 2,2,2-Trichloräthanol.
Andere funktionell abgewandelte Carboxylgruppen R sind z.B. stickstoffhaltige funktionell abgewandelte Carboxylgruppen, wie Carbamylgruppen, welche am Stick- stoffatom unsubstituiert oder durch gegebenenfalls Niederalkyl-, freie, veresterte oder verätherte Hydroxy-, wie Niederalkoxy-, Aralkoxy-, Niederalkanoyloxy- oder Aroyloxygruppen oder Halogenatome, Nitro- oder Tri- fluormethylgruppen enthaltende aliphatische, alicyclische, aromatische oder araliphatische Kohlenwasserstoff- reste oder heterocyclische Reste aromatischen Charakters, wie Niederalkyl-, Cycloalkyl-, Phenyl-, Phenyl-niederalkyl-, Phenyl-niederalkyliden- oder Pyridylreste, sowie durch freie, verätherte oder veresterte Hydroxylgruppen, wie den oben erwähnten Gruppen dieser Art,

Die im obigen Verfahren verwendeten Ausgangsstoffe sind insbesondere die Verbindungen der Formel

worin Ac, R, und X die oben gegebene Bedeutung haben und Y für eine reaktionsfähige veresterte Hydroxylgruppe steht. Letztere ist eine der oben erwähnten veresterten Hydroxylgruppen, insbesondere eine mit einer der genannten starken organischen Carbon- oder Sulfonsäuren oder einer Halogenwasserstoffsäure veresterten Hydroxylgruppen.
Die als Ausgangsstoffe verwendeten Verbindungen können z.B. nach dem in der Patentschrift Nr. 497 449 beschriebenen Verfahren erhalten werden.
Die erfindungsgemäss erhaltenen Verbindungen können, wie im Formelschema gezeigt wird, in 7-Amino cephalosporansäure und deren Derivate umgewandelt werden; diese Umwandlung kann z.B. nach dem in den Patentschriften Nrn. 497 451, 497 456, 497 371, 497 369.
497 457, 497 460, 497 379 und 497 461 beschriebenen Verfahren erfolgen.
In den folgenden Beispielen werden die Temperaturen in Celsiusgraden angegeben.
Beispiel I
Eine Lösung von 66 g L-2,2-Dimethyl-3-tert.butyloxy- carbonyl - 5,8 - hydroxy-thiazolidin - 4 - carbonsäuremethyl- ester in 900 ml trockenem Dimethylformamid wird mit 141,5 ml Äthyl-diisopropylamin versetzt und unter kräftigem Rühren und bei einer Temperatur von 15 bis l8 tropfenweise mit 51,7 ml Methan-sulfonylchlorid behandelt.
Nach 25 Minuten ist die Zugabe beendet; das Reaktionsgemisch enthaltend den L-2,2-Dimethyl-3-tert.butyl- oxycarbonyl - 5,B- methylsulfonyloxy-thiazolidin-4-carbonsäuremethylester der Formel

wird bei Zimmertemperatur während einer Stunde gerührt und dann tropfenweise unter kräftigem Rühren mit einer Lösung von 73,7 g Natriumazid in 181 ml Wasser behandelt. Die Zugabe, welche bei 20 erfolgt, ist nach 45 Minuten beendet, und das Reaktionsgemisch wird in 6000 ml Äther ausgegossen.
Die organische Lö- sung wird zweimal mit je 100 ml 20 lOiger wässriger Zitronensäurelösung extrahiert, und einmal mit 500 ml kalter gesättigter Natriumhydrogencarbonatlösung und 6 Mal mit je 2000ml kaltem Wasser gewaschen. Nach dem Trocknen und Eindampfen unter Wasserstrahl vakuum bei Raumtemperatur erhält man ein braunes Rohprodukt, welches sofort an der 40fachen Gewichtsmenge Silicagel enthaltend 5% Wasser chromatographiert wird.
Die Kolonne wird in reinem Benzol aufgezogen; mit einem 9: 1-Gemisch von Benzol und Essigsäure- methylester wird ein gelbes öl eluiert, welches in Pentan gelöst und durch eine Kolonne enthaltend 54g eines Aktivkohlepräparats filtriert wird; das nach dem Verdampfen des Pentans erhaltene farblose viskose Material kristallisiert unter Tiefkühlung aus dem gleichen Volumen Pentan und man erhält den L-2,2-Dimethyl-3-tert.butyloxycarbonyl-5a - azido-thiazolidin-4-carbonsäuremethylester der Formel

Beispiel 2
Eine Lösung von 33 g L.2,2.Dimethyl.3.tert.butyloxy.
carbonyl - 5p- hydroxy-thiazolidin - 4 - carbonsäuremethyl ester in 250ml trockenem Dimethylformamid wird mit 48 ml Triäthylamin versetzt, auf 150 gekühlt und tropfenweise innert 25 Minuten unter Rühren und Feuchtigkeitsausschluss mit 26 mol Methansulfonylchlorid behandelt, wobei die Temperatur durch Kühlen bei 15- 200 gehalten wird. Nach 90minütigem Rühren bei Raumtemperatur lässt man zum Reaktionsgemisch, enthaltend L- 2,2 - Dimethyl-3-tert. butyloxycarbonyl.5p.methylsulfo.
nyloxy-thiazolidin-4-carbonsäuremethylester, unter starkem Rühren 36,9 g Natriumazid in 90 ml Wasser zufliessen; man rührt während 45 Minuten bei 200 weiter und giesst dann auf ein Gemisch von 250 ml gesättigter Kochsalzlösung und 5 g Zitronensäure aus. Das Gemisch wird dreimal mit je 250 ml Cyclohexan ausgeschüttelt, die erhaltenen organischen Extrakte mit je 150ml Wasser gewaschen, vereinigt und getrocknet und unter Wasserstrahlvakuum eingedampft. Das nach dem Entgasen erhaltene Rohprodukt wird in 600ml Pentan gelöst, mit 16 g eines Aktivkohlepräparats während 15 Minuten verrührt, filtriert und die Aktivkohle zweimal mit je 300 ml Pentan gewaschen.
Das erhaltene farblose Produkt wird aus Pentan kristallisiert, und man erhält den erwünschten reinen L-2,2-Dimethyl-3-tert.butyloxycarbo nyl-5,-azido-thiazolidin-4-carbonsäuremethylester, F. 55 bis 560. Die Mutterlauge enthält eine weitere Menge des gewünschten Produkts, sowie eine kleine Menge des 2,2- Dimethyl -3. tert.butyloxycarbonyl-4-thiazolin-4-car- bonsäuremethylesters.
Beispiel 3
Durch Behandeln von L-2,2-Dimethyl-3-tert.butyloxy- carbonyl-5-(4 - nitro-benzoyloxy)-thiazolidin-4-carbonsäu.
remethylester mit Natriumazid erhält man wie im Beispiel 1 den L-2,2-Dimethyl-3-tert.butyloxycarbonyl-5α- -azido-thiazolidin-4-carbonsäuremethylester.
Beispiel 4
0,525g des rohen L-2,2-Dimethyl-3-tert.butyloxycar- bonyl - 5,ss - chlor - thiazolidin-4-carbonsäuremethylester in 5 ml absolutem Dimethylformamid werden mit 0,5 g pulverisiertem Natriumazid versetzt und nach 30minütigem Rühren werden 100 mol Wasser und 100 mol Äther zugegeben. Die wässrige Phase wird mit Äther ausgewaschen und die vereinigten organischen Extrakte mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird in 10 ml Methylenchlorid gelöst, mit 1 g eines Diatomeenerdepräparats (Florisil) behandelt und filtriert.
Der Rückstand wird bei -150 aus Pentan kristallisiert und man erhält so den L-2,2-Dimethyl-3 -tert.butyloxy- 5a- azido-thiazolidin-4-carbonsäuremethylester, F. 52 - 540.
Beispiel 5
Durch Behandeln von L-2,2-Dimethyl-3.tert.butyloxy.
carbonyl-4-cyano-5ss-chloro-thiazolidin mit Natriumazid nach dem im Beispiel 1 beschriebenen Verfahren erhält man das L-2,2-Dimethyl-3-tert.butyloxycarbonyl-4-cyano- -5a-azido-thiazolidin der Formel

Eine Lösung von 0,435 g L-2,2-Dimethyl-3-tert.butyloxycarbonyl-5ss-hydroxy-thiazolidin-4-carbonsäure 2,2,2 -trichloräthylester in 9 mol wasserfreiem Dimethylform amid wird mit 0,7 g N,N-Dilsopropyl-N-äthylamin und dann mit 0,35g Methan-sulfonylchlorid in 2 mol Dimer thylformamid versetzt.
Nach 50minütigem Rühren bei Zimmertemperatur wird das Gemisch, enthaltend den 1-2,2 - Dimethyl-3-tert.butyloxycarbonyl-5p-methyl-sulfo- nyloxy-thiazolidin - 4- carbonsäure-2,2,2-trichloräthylester der Formel

mit etwa 0,5 g Natriumazid behandelt und 21/2 Stunden gerührt. Das Gemisch wird auf 20%ige wässrige Zitronensäure gegossen und mit Äther extrahiert; die organische Lösung wird verdampft und 0,08 g des erhaltenen Rohprodukts wird an 20 g eines Diatomeenerdepräparats chromatographiert.

Beispiel 7
Eine Lösung von 1,1 g L-2,2-Dimethyl-3-tert.butyl- oxycarbonyl- thiazolidin - 4 - carbonsäure- methylester in 15 ml Bromtrichlorkohlenstoff wird, nach Zugabe von 2,35 g Di-tert.butyl-peroxalat. bis zum Aufhören der Gasentwicklung (etwa 75 Minuten) unter einer Stickstoffatmosphäre bei einer Badtemperatur von 700 erwärmt.
Nach dem Eindampfen unter vermindertem Druck (zuerst Wasserstrahlpumpe, dann Hochvakuum) wird der viskose Rückstand, enthaltend den L-2,2-Dimethyl-3 tert. butyloxycarbonyl - - brom-thiazolidin - 4- carbonsäure-methylester der Formel

in 8 mol Dimethylformamid gelöst und, unter Kühlen mit Wasser, mit einer gesättigten wässrigen Lösung von 0,75 g Natriumazid versetzt und eine Stunde bei Zimmertemperatur stehengelassen. Nach Zusatzen von 30 ml Wasser wird der organische Anteil zweimal mit je 25 ml Cyclohexan extrahiert, die organischen Extrakte werden getrocknet und eingedampft und der viskose Rückstand an 60g gereinigtem Silicagel mit einem 3: 1-Gemisch von Benzol und Essigsäureäthylester chromatographiert, wobei man Fraktionen zu 40ml abtrennt.
Durch Eindampfen der Fraktionen 6 - 9 erhält man ein Rohprodukt, das, wie aus dem Dünnschichtchromatogramm (Silicagel; 9:l-Gemisch von Benzol und Essigsäure äthylester; charakteristischer Fleck mit Rf = 0,5) und aus den NMR- und UV-Spektren ersichtlich, den L-2,2 Dimethyl-3-tert.butyloxycarbonyl - 5 - azido-thiazolidin -4-carbonsäure-methylester sowie den 2,2-Dimethyl-3 -tert.butyloxycarbonyl.4.thiazolin.4-carbonsäure - methyl- ester enthält.
Beispiel 8
Eine Lösung von 0,418 g L.2,2-Dimethyl-3-tert.butyl- oxycarbonyl-5,8-hydroxy-thiazolidin-4-carbonsäure-diphe- nylmethylester in 1,6 ml kochendem Dimethylformamid wird mit 0,4 ml N,N-Diisopropyl-N-äthylamin versetzt und dann mit einer Lösung von 0,17 ml Methansulfonsäurechlorid in 0,2 ml Dimethylformamid tropfenweise behandelt. Nach 2stündigem Rühren bei Raumtemperatur wird das Reaktionsgemisch, enthaltend den L-2,2 - Dimethyl-3-tert.butyloxycarbonyl-5p-methylsulfonyloxy -thiazolidin-4-carbonsäure-diphenylmethylester der Formel

mit einer gesättigten wässrigen Lösung von 0,5g Natriumazidlösung behandelt. Das Gemisch wird während 21/2 Stunden bei Zimmertemperatur gerührt und auf 30 ml Wasser gegossen.

welcher im Infrarot-Absorptionsspektrum (in Methylen chlorid) Banden bei 4,76,u, 5,70 , 5,85y und 5,92 zeigt, während Fraktionen 18-30 unveränderten 1-2,2- -Dimethyl-3-tert.butyloxycarbonyl - 5,8 - hydroxy - thiazolidin-4-carbonsäure-diphenylmethylester enthalten.
Beispiel 9

Process for the preparation of 2,2-disubstituted 3-acyl-5ffi-azido-thiazolidine-4-carboxylic acids or acid derivatives thereof
The present invention relates to a methodological process for the preparation of azido compounds which was used for the preparation of valuable intermediates and in particular in the first synthetic preparation of 7-amino-cephalosporanic acid and its derivatives and which is particularly suitable for this peculiar synthesis.

Example I.
A solution of 66 g of L-2,2-dimethyl-3-tert-butyloxycarbonyl-5,8-hydroxy-thiazolidine-4-carboxylic acid methyl ester in 900 ml of dry dimethylformamide is mixed with 141.5 ml of ethyl diisopropylamine and treated dropwise with 51.7 ml of methanesulfonyl chloride with vigorous stirring and at a temperature of 15 to 18.

Example 3
By treating L-2,2-dimethyl-3-tert-butyloxycarbonyl-5- (4-nitro-benzoyloxy) -thiazolidine-4-carboxylic acid.
Remethyl ester with sodium azide is obtained as in Example 1, L-2,2-dimethyl-3-tert.butyloxycarbonyl-5α-azido-thiazolidine-4-carboxylic acid methyl ester.
Example 4
0.525 g of the crude L-2,2-dimethyl-3-tert.butyloxycarbonyl-5, ss-chloro-thiazolidine-4-carboxylic acid methyl ester in 5 ml of absolute dimethylformamide are mixed with 0.5 g of powdered sodium azide and after stirring for 30 minutes 100 mol of water and 100 mol of ether were added. The aqueous phase is washed out with ether and the combined organic extracts are washed with water, dried and evaporated. The residue is dissolved in 10 ml of methylene chloride, treated with 1 g of a diatomaceous earth preparation (Florisil) and filtered.
