Verfahren zur Herstellung von tricyclischen organischen Verbindungen
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung substituierter Diazepine, Oxazepine und Thiazepine, und zwar insbesondere substituierter 11 -Aminodibenz-[b,f]l1,4]-oxazepine oder -thiazepine und substituierter 1 1-Aminodibenz-[b,f]fi,4]-diazepine ausentsprechenden substituierten Diphenylthioharnstoffen durch Ringschluss. Die Verfahrensprodukte sind wegen ihrer Zentralnervensystem(CNS)- Aktivität als Tranquilizer und Antidepressiva sowie als Analgetica verwendbar.
Die mittels des neuen Verfahrens gemäss der Erfindung erhältlichen Diazepine, Oxazepine und Thiazepine lassen sich durch folgende Formel darstellen:

4 -Morpholinorest oder 2,2-Polymethylenhydrazinorest und Z Sauerstoff, Schwefel oder niederalkylsubstituierten Stickstoff bedeuten.
Die erfindungsgemässen Verbindungen sind im allgemeinen weisse kristalline Feststoffe, die nur wenig löslich in Wasser, aber mässig löslich in organischen Lösungsmitteln, zum Beispiel Methanol, Äthanol und dergleichen sind. Sie sind basische Substanzen, die gewöhnlich in wässrigen Mineralsäuren bei Zimmertemperatur löslich sind. Sie bilden im wesentlichen unlösliche Säureadditionssalze, zum Beispiel Hydrochloride, Sulfate, Phosphate, Citrate, Taktrate, Maleate, Fumarate und dergleichen. Die erfindungsgemässen Verbindungen können im allgemeinen in Form ihrer Salze oral oder parenteral verabreicht werden und sind wirksame Zentralnervensy- stemmittel, wenn sie so verabreicht werden.
Für die orale Verabreichung können die neuen Verbindungen gemäss der Erfindung mit den üblichen pharmazeutischen Trägern vereinigt und beispielsweise in Form von Tabletten, Kapseln, Dragees, Flüssigkeiten, die tropfenweise zu verabreichen sind, Emulsionen, Suspensionen und Sirupen sowie in Schokolade, Süssigkeiten, Kaugummi und dergleichen angewandt werden. Sie können ferner in Form von Suppositorien und als wässrige Lösungen für parenterale Injektion verabreicht werden.
Das neue Verfahren gemäss der Erfindung besteht in der Cyclisierung substituierter Thioharnstoffe (II) zu Verbindungen (I) durch Erwärmen mit Phosphorpentoxyd oder Phosphoroxychlorid gemäss dem folgenden Reaktionsschema:

Besonders vorteilhaft für die Umwandlung von II zu I ist die Verwendung von Phosphorpentoxyd in Verbindung mit Phosphoroxychlorid. Ein Überschuss des letztgenannten Reagens kann auch als zweckmässiges Lösungsmittel dienen. Die Cyclisierungsreaktionen werden im allgemeinen bei Temperaturen zwischen etwa 50 und 1500C durchgeführt. Die bevorzugte Temperatur beträgt etwa 80- 1100C. Der Zusatz anderer Lösungsmittel, die unter den Reaktionsbedingungen inert sind, zum Beispiel Benzol, Toluol, Xylol und dergleichen, kann ebenfalls vorteilhaft sein. Nach erfolgter Cyclisierung, gewöhnlich nach etwa 30 Minuten bis etwa 24 Stunden langem Erwärmen, werden die Produkte (I) im allgemeinen durch Behandlung der Reaktionslösung mit Wasser und anschliessende Reinigung des Rohprodukts durch bekannte Massnahmen erhalten.
Die neuen Thioharnstoffe (II), die bei diesem neuen Verfahren als Ausgangsstoffe für die Verbindungen (I) dienen, können aus substituierten Diphenylestem, substituierten Diphenylsulfiden und substituierten Diphenylaminen gemäss dem folgenden Reaktionsschema hergestellt werden:

Gemäss diesem Reaktionsschema führt die Acylierung der substituierten Diphenylester, substituierten Diphenylsulfide und substituierten Diphenylamine mit Phenoxythio carbonylchlorid zu den Thiocabanilaten (IV). Die Behandlung der letztgenannten Verbindungen mit einem Mono- oder Diamin liefert die Thioharnstoffe (II).
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen weisen physiologische Wirkung auf das Zentralnervensystem auf. Sie zeigen hohe Aktivität als Tranquilizer bei nichttoxischen Dosen und in manchen Fällen Antidepressiva-Eigenschaften bei Dosierungswerten, die weder offenkundige Stimulierung noch Depression erzeugen.
Eine geeignete Prüfung auf Tranquilizeraktivität besteht darin, die Verminderung spontaner motorischer Aktivität bei Tieren mit Hilfe eines Aktophotometers (eine photoelektrische Vorrichtung zur quantitativen Messung locomotorischer Aktivität) zu messen. Abgestufte Dosen der nach dem erfindungsgemässen Verfahren hergestellten aktiven Verbindungen werden an Gruppen von Mäusen verabreicht, und der wirksame Dosierungsbereich für eine signifikante Verminderung motorischer Aktivität (ein Mass für Tranquilizerwirkung) im Vergleich zu Kontrollgruppen wird ermittelt. Die Benutzung der verminderten motorischen Aktivität als Mass für Tranquilizeraktivität wurde von W.D. Gray, A.C. Osterberg und C.E. Rauh, Archives Internationales et de The rapie, Bd. 134, S. 198 (1961) und W.J. Kinnard und C.J.
Carr, Journal of Pharmacology and Experimental Therapeutics, Bd. 121, S. 354 (1957), beschrieben.
Die Antidepressiva-Eigenschaften der nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen treten zutage, wenn man ihre Fähigkeit misst, einer bei Tieren durch Verabreichung von Tetrabenazinhexamat induzierten Depression entgegenzuwirken. Abgestufte Dosen der aktiven Verbindungen gemäss der Erfindung werden an Gruppen von Mäusen verabreicht. Hierauf folgt die Verabreichung einer Tetrabenazindosis, die bekanntermassen das forschende Verhalten normaler Mäuse deutlich unterdrückt. Die mit Antidepressivum behandelten Gruppen zeigen normales forschendes Verhalten, während die Kontrollgruppen und Gruppen, die mit einem als Antidepressivum unwirksamen Mittel behandelt wurden, nicht dieses normale forschende Verhalten, sondern die bekannte tiefe Depression, die durch Tetrabenazin induziert wird, zeigen.
Die Ergebnisse mit verschiedenen Dosierungswerten werden zur Ermittlung wirksamer Dosierungsbereiche verwendet. Die nach dem erfindungsgemässen Verfahren hergestellten Antidepressiva zeigen ihre vorteilhaften Eigenschaften bei dieser Prüfung bei Dosierungswerten, die nur geringe oder keine unerwünschten Nebenreaktionen, zum Beispiel Ataxie oder verminderte spontane motorische Aktivität hervorrufen.
Einige der erfindungsgemäss erhältlichen Verbindungen zeigen ferner andere wertvolle pharmazeutische Eigenschaften, zum Beispiel analgetische Aktivität.
Die folgenden Beispiele erläutern ausführlich die Herstellung repräsentativer substituierter 11-Aminodibenz- b,f][l,4]oxazepine und -thiazepine und substituierter 11 -Aminodibenz[b,fjLl ,4]diazepine nach dieser Erfindung.
Beispiel 1 Herstellung von l 1-(1-Piperidinyl)dibenz[b ,f](1 ,4]oxazepin
Die Verbindung Phenyl-o-phenoxythiocarbanilat und Piperidin werden in Äthanol auf Rückflusstemperatur erwärmt, worauf Entfernung des Lösungsmittels und Kristallisation aus verdünntem Äthanol folgt. Als Produkt wird 2'-Phenoxypiperidinthiocarboxanilid vom F. 135
1360C erhalten.
1,0 g 2'-Phenoxypiperidinthiocarboxanilid werden mit etwa 1 g Phosphorpentoxyd in 5 ml Phosphoroxychlorid 2 Stunden unter Rückfluss gehalten. Nach Abkühlen auf Zimmertemperatur wird die Reaktionsmischung mit Äther verdünnt. Der erhaltene Niederschlag wird mehrere Male mit Äther gewaschen, mit wässriger Kaliumcarbonatlösung (vorzugsweise unter Kühlen) behandelt und dreimal mit Äther extrahiert. Die Ätherlösung wird über wasserfreiem Kaliumcarbonat getrocknet und liefert nach Eindampfen des Lösungsmittels einen gelben Feststoff, der aus Heptan zu weissen Kristallen vom 98 - l000C umkristallisiert wird.
Beispiel 2
Herstellung von ll-Aminalibenz[b,fle1,4loxazepin
Nach der Arbeitsweise von Beispiel 1 wird durch Behandlung von Phenyl-o-phenoxythiocarbanilat mit ammoniakalischem Äther als Produkt l-(2-Phenoxy)-phenylthioharnstoff, vom F. 125 - 1270C nach Umkristallisieren aus verdünntem Methanol erhalten.
Erwärmen des vorstehend hergestellten 1-(2-Phenoxy) -phenylthioharnstoffs mit Phosphorpentoxyd in Phosphoroxychlorid ergibt das kristalline Produkt vom 198 - 2000C.
Beispiel 3
Herstellung von l l -(n-Butylamino)dibenz[b,fl[1,4]oxazepin
16,66 g (0,075 Mol) 2-Aminodiphenylätherhydrochlorid werden durch Schütteln mit verdünntem Ammoniumhydroxyd in die freie Base übergeführt. Die freigesetzte Base wird mit Äther extrahiert, und die ätherische Lösung wird über Natriumsulfat getrocknet. Diese Lösung wird mit einer Lösung von 6,45 g (0,0375 Mol) Phenoxythiocarbonylchlorid in Äther versetzt. Die erhaltene Lösung wird bei der Umgebungstemperatur etwa 21 Stunden mit einem Magnetrührer gerührt. Durch Filtrieren werden weisse Kristalle von 2-Aminodiphenyl ätherhydrochlorid abgetrennt. Durch Entfernung des Lösungsmittels aus dem Filtrat erhält man ein bernsteinfarbenes Ö1, das nach Umkristallisieren aus Hexan 12,4 g Feststoff vom F. 95 - 980C liefert.
Eine Lösung von 3,21 g (10 Mol) des vorstehend hergestellten Phenyl-o-phenoxythiocarbanilats und 1,46 g (20
Mol) n-Butylamin in 50 ml Äthanol wird 50 Minuten auf Rückflusstemperatur erwärmt. Das Lösungsmittel wird entfernt, und der Rückstand kristallisiert beim Verreiben mit Hexan. Durch Umkristallisieren des Produkts 1-Bu tyl-3-(2-phenoxy)phenylthioharnstoff aus Äther-Hexan erhält man weisse Kristalle vom F. 80 - 81 0C.
Nach der Arbeitsweise von Beispiel 1 erhält man durch Umsetzung des vorstehend hergestellten 1-Butyl-3-(2 -phenoxy)- phenylthioharnstoffs mit Phosphorpentoxyd in Phosphoroxychlorid Kristalle vom F. 67 - 710C nach Umkristallisieren aus Heptan.
Beispiel 4
Herstellung von 11 -(4-Methyl-1 -piperazinyl)dibenzC6,ss [1 ,4jexazepin
Nach der Arbeitsweise von Beispiel 1 wird Phenyl-o -phenoxythiocarbanilat mit 1 Moläquivalent 1-Methylpi- perazin zu 4-Methyl-2'-phenoxy- 1 -piperazinthiocarbox anilid umgesetzt, das aus verdünntem Äthanol in Form weisser Kristalle vom F. 139 - 1420C erhalten wird.
Nach der Arbeitsweise von Beispiel 1 wird durch Umsetzung von 4-Methyl-2' phenoxypiperazinthiocarboxani- lid mit Phosphorpentoxyd in Phosphoroxychlorid das oben genannte Produkt in Form von Kristallen vom F.
96- 970C nach Umkristallisieren aus Heptan erhalten.
Beispiel 5
Herstellung von Il-[4-(l-Hydraxyäthyl)-l-piperazinyl dihenzb,J][1 ,4joxyzepin-Dihydrochlond
Nach der Arbeitsweise von Beispiel 1 wird Phenyl -2-(p-chlorphenoxy)thiocarbanilat mit 1 -(p Hydroxyäthyl) piperazin zu 2' -(p-Chlorphenoxy)-4(2'-hydroxyäthyl) - 1- -piperazinthiocarboxanilid vom F. 155 - 1580C nach Umkristallisieren aus Äthylacetat umgesetzt.
Das vorstehend hergestellte 2'-(p-Chlorphenoxy)-4 -(2'-hydroxyäthyl)- 1 -piperazinthiocarboxanilid wird mit Phosphorpentoxyd in Phosphoroxychlorid erwärmt. Nach Auflösen des Produkts in Äther und Behandlung mit Chlorwasserstoff erhält man 11 -f4(8-Hydroxyäthyl)-1- -piperazinyl]dibenz[b,f][1,4]oxazepin-dihydrochlorid als Kristalle vom F. 200 - 232CC.
Beispiel 6
Herstellung von 2-Chlor-11-(4-methyl-1-piperazinyl) d ibenz[b,fl[l ,4Joxazepin
Nach der Arbeitsweise von Beispiel 1 werden durch Umsetzung von Phenyl-2-(pchlorphenoxy)thiocarbanilat mit l-Methylpiperazin Kristalle von 2'-(p-Chlorphenoxy) -4-methyl- 1 piperazinthiocarboxanilid vom F. 145 - 1 470C nach Umkristallisieren aus Aceton-Hexan erhalten.
Wenn man für die Arbeitsweise von Beispiel 1 2'-(p- -Ghlorphenoxy)-4-methyl- 1 -piperazinthiocarboxanilid als Ausgangsmaterial verwendet, erhält man als Produkt bräunliche Kristalle vom F. 109 bis llloC.
Beispiel 7
Herstellung von 2-Chlor-ll- -(4-methyl-1-piperazinyl)dibenz[b,f][1,4]thiazepin
Nach der Arbeitsweise von Beispiel 1 wird Phenyl -(p-Chlorphenylthio)thiocarbanilat mit 1 Moläquivalent l-Methylpiperazin zu 4-Methyl-2'-(p-chlorphenylthio)piperazinthiocarboxanilidhydrochlorid umgesetzt.
Die Umsetzung des vorstehend hergestellten 4-Methyl-2'-(p - chlorphenylthio)piperazinthiocarboxanilid - hy- drochlorids mit Phosphorpentoxyd in Phosphoroxychlo- rid nach der Arbeitsweise von Beispiel 1 liefert weisse Kristalle vom F. 114,5 bis 1 17,00C nach Umkristallisieren aus Heptan.
Beispiel 8
Herstellung von S-Methyl-ll- -(4-Methyl-1-piperazinyl(dibenz[b,f][1,4]diazepin
Durch Umsetzung von 2-Amino-N-methyldiphenylamin mit Phenoxythiocarbonylchlorid nach der Arbeitsweise von Beispiel 1 wird Phenyl-2-(N-methylanilino) thiocarbanilat in Form eines Öls erhalten.
Die Behandlung des vorstehend hergestellten Phenyl -2-(N-methylanilino(thiocarbanilats mit l-Methylpipera- zin nach der Arbeitsweise von Beispiel 1 liefert Kristalle von 4-Methyl-2' -(N-methylanilino)- 1 -piperazinthiocarb- oxanilid vom F. 13 1 - 1320C nach Umkristallisieren aus Heptan-Äthylacetat.
Nach derArbeitsweise von Beispiel 1 wird 4-Methyl-2' -N-methylanilino-l-piperazin-thiocarboxanilid mit Phosphorpentoxyd in Phosphoroxychlorid zu Kristallen vom F. 119-1200C nach Umkristallisieren aus Heptan umgesetzt.
Beispiel 9
Herstellung von 11-(4-Morpholino)dibenz[b,f][1,4]thiazepin Hydrochlorid
Die Umsetzung von 2-Aminodiphenylsulfid mit Phenoxythiocarbonylchlorid nach der Arbeitsweise von Beispiel 1 liefert weisse Kristalle vom F. 86 bis 880C nach Umkristallisieren aus Äthanol.
Nach der Arbeitsweise von Beispiel 1 wird Phenyl-2 -(phenylthio)thiocarbanilat mit Morpholin in Propanol zu Kristallen von 2'-(Phenylthio)-4-morpholinthiocarb- oxanilid vom F. 83 - 840C nach Umkristallisieren aus Äthanol-Heptan umgesetzt.
Eine Lösung von 300 mg 2' -(Phenylthio) 4-morpholino- thiocarboxanilid in 2 mol Phosphoroxychlorid, das einen Tropfen Dimethylformamid enthält, wird 5 Stunden auf Rückflusstemperatur erwärmt. Nach Isolierung des Produkts in der in Beispiel 1 beschriebenen Weise werden cremefarbene Kristalle vom F. 250 - 2600C nach Umkristallisieren aus Äthylacetat-Methanol erhalten.
Beispiel 10
Herstellung von l l -(DimethylaminoäthylaminoJdibenzlb,fl[l ,4]oxazepin
Nach der Arbeitsweise von Beispiel 1 wird durch Umsetzung von Phenyl-o-phenoxythiocarbanilat mit unsym.-Dimethyläthylendiamin in Äthanol 1 -(2-Dimethyl- aminoäthyl)-3 (2-phenoxy)-phenylthiohamstoff als farbloses öl erhalten. Durch Erwärmen von 1-(2-Dimethyl aminoäthyl)-3 -(2-phenoxy)-phenylthiohamstoff mit Phosphoroxychlorid in Gegenwart von Dimethylformamid bei Rückflusstemperatur erhält man 11 -(Dimethylamino- äthylamino)dibenzRb,f][l,4]-oxazepin vom F. 82- 840C.
Beispiel 11
Herstellung von 2-Chlor-11-[(3-dimethylaminoäthyl) methylamino]-dibenz[b,f][1,4]thiazepin-Hydrochlorid
Nach der Arbeitsweise von Beispiel 1 wird 2-(p-Chlorphenylthio)-thiocarbanilat mit Trimethyläthylendiamin zu 1 -f2- (p-Chlorphenylthio)phenyl] -3-methyl-3 -dimethylami- noäthyl-thioharnstoff umgesetzt, der als Hydrochloridsalz vom F. 167 - 1680C isoliert wird.
Das vorstehend hergestellte 1 -f2-(p-Chlorphenylthio)- phenyl]-3-methyl 3 - dimethylaminoäthylthioharnstoffhy drochlorid wird mit Phosphorpentoxyd in Phosphoroxychlorid erwärmt. Durch Auflösen des erhaltenen Produkts in Äther und Behandlung mit Chlorwasserstoff erhält man 2-Chlor- 11-[(3-dimethylaminoäthyl)methyl- amino]-dibenzlb,fll1,4]thiazepinhylrochlorid vom F. 1961970C.
Beispiel 12 Herstellung von 2-Chlor-11-[4-(3-dimethylaminopropyl)- -1-piperazinyl]dibenz[b,f][1,4]oxazepin
Durch Umsetzung von Phenyl-2-(p-chlorphenoxy)thiocarbanilat mit 1-(3-Dimethylaminopropyl)piperazin nach der Arbeitsweise von Beispiel 1 erhält man ein Öl, das durch Behandlung mit ätherischem Chlorwasserstoff in das Dihydrochlorid übergeführt wird. Nach Umkristallisieren aus Methanol wird 2'-(p-Chlorphenoxy)-4-(3-dime thylaminopropyl) -1 - piperazinthiocarboxanilid - dihydrochlorid vom F. 208 - 2100C erhalten.
Nach der Arbeitsweise von Beispiel 1 wird durch Erwärmen des vorstehend hergestellten 2'-(p-Chlorphenoxy)-4-(3 - dimethylaminopropyl)-1-piperazinthiocarboxa- nilid-dihydrochlorids mit Phosphorpentoxyd in Phosphoroxychlorid 2-Chlor-11-[4-(3-dimethylaminopropyl)-1-pi- perazinyl]dibenz[b,f][1,4]oxazepin hergestellt.
Beispiel 13
Herstellung von 2-Chlor-1 1-(4-morpholinyl) dibenz[b,f]U,4]thiazepin
Nach der Arbeitsweise von Beispiel 1 wird Phenyl -2-(p-Chlorphenylthio)thiocarbanilat mit Morpholin zu 2'-(p-Chlorphenylthio)-4morpholin-thiocarboxanilid vom F. 65 - 670C umgesetzt.
Durch Erwärmen des vorstehend hergestellten 2'-(p -Chlorphenylthio)-4-morpholinthiocarboxanilid mit Phos phorpentoxyd in Phosphoroxychlorid erhält man 2 -Chlor-1 1 - (4- morpholinyl)dibenz[b,f][l ,4jthiazepin als Kristalle vom F. 148 - 1500C.
Beispiel 14
Herstellung von 2-Chlor-11 -(1 -piperidinylamino) dibenz[b,f][1 ,4jthiazepin
Nach der Arbeitsweise von Beispiel 1 setzt man Phenyl-2-(p-chlorphenylthio)thiocarbanilat mit N-Aminopiperidin zu 1-r[2-p-Chlorphenylthio)-phenyl]-3-(1-piperidi- no)thioharnstoff vom F. 158,5- 1600C nach Umkristallisieren aus Äthanol zu.
Durch Erwärmen des vorstehend hergestellten 1--(p- - Chlorphenylthio)phenyl] - 3 (1 -piperidino)thioharn stoffs mit Phosphorpentoxyd in Phosphoroxychlorid erhält man 2-Chlor- 11 - (1-piperidinylamino)dibenz[b,f][1,4]thiazepin als Kristalle vom F. 153 - 1540C.
Beispiel 15
Herstellung von 11 -(P-Hydroxyäthylamino)dibe'nz[b,ss [1 4]oxazepin
Nach der Arbeitsweise von Beispiel 1 wird Phenyl-2- phenoxythiocarbanilat mit Äthanolamin zu l-(p-Hydroxy- äthyl)-3-(2'-phenoxy)phenylthioharnstoff umgesetzt.
Durch Erwärmen des vorstehend hergestellten 1-(,8- -Hydroxyäthyl) -3-(2' -phenoxy)phenylthiohamstoffs mit Phosphorpentoxyd in Phosphoroxychlorid erhält man 11 -(ss-Hydroxyäthylamino)dibenz[b,f][[1,4]oxazepin als Kristalle vom F. 136 - 1390C.
Beispiel 16
Herstellung von II -(3-Dimethylaminoprpyl)dibenz[b,f [I,oloxazepin
Nach der Arbeitsweise von Beispiel 1 wird durch Umsetzung von 2-Phenoxythiocarbanilat mit 3-Dimethylaminopropylamin 1-(3 -Dimethylaminopropyl)-3 -(2 -phenoxy)phenylthioharnstoff vom F. 113-1140C nach Umkristallisieren aus Äthanol-Heptan hergestellt.
Durch Erwärmen des vorstehend hergestellten 1-(3-Dimethylaminopropyl)-3-(2-phenoxy) - phenyl-thioharnstoffs mit Phosphorpentoxyd in Phosphoroxychlorid erhält man das Produkt in Form von Kristallen vom F. 108 - 1090C.
Beispiel 17
Herstellung von
11-Allylaminodibenz[b,f][1,4]oxazepin-Hydrochlorid
Nach der Arbeitsweise von Beispiel 1 erhält man durch Umsetzung von Phenyl-2-phenoxythiocarbanilat mit Allylamin als Produkt l-Allyl-3-(2-phenoxy)phenyl- thioharnstoff in Form eines öls.

Das vorstehend hergestellte 5'-Trifluormethyl-2'-phe nylthio-4-methyl- 1 -piperazinthiocarboxanilid wird mit Phosphorpentoxyd in Phosphoroxychlorid erwärmt.
Durch Auflösen des erhaltenen Produkts in Äther und Behandeln in Chlorwasserstoff erhält man 11-(4-Methyl- -1-piperazinyl) -8 - -trifluormethyldibenz[b,f][1,4]thiazepin- dihydrochlorid als Kristalle vom F. 1920C (Zers.).

Herstellung von 2-Nitro-ll- -(4-methyl-1-piperazinyl)dibenz[b,J] [1,4joxazepin
Aus l-Methylpiperazin und Thiophosgen hergestelltes 1 -Methyl-4-piperazinthiocarbonylchlorid wird mit o -Aminophenol zu 1 -Methyl-4-(o-hydroxyphenylthiocarbamoyl)piperazin umgesetzt. Dieses Produkt wird mit 4 -Chlornitrobenzol in Dimethylacetamid in Gegenwart von Kaliumcarbonat behandelt, wodurch 4-Methyl-2'-(p-nitrophenoxy)piperazin-thiocarboxanilid erhalten wird.
Nach der Arbeitsweise von Beispiel 1 wird durch Umsetzung des vorstehend erhaltenen 4-Methyl-2'-(pnitrophenoxy)piperazinthiocarboxanilids mit Phosphorpentoxyd in Phosphoroxychlorid 2-Nitro-l l-(4-methyl-1- piperazinyl)-dibenz[b,fl[1,4]oxazepin in Form von Kristallen vom F. 189 - 1910C nach Umkristallisieren aus Hexan erhalten.
Process for the preparation of tricyclic organic compounds
The invention relates to a process for the preparation of substituted diazepines, oxazepines and thiazepines, in particular substituted 11 -amino-dibenz [b, f] l1,4] -oxazepines or -thiazepines and substituted 1 1-aminodibenz [b, f] fi, 4] -diazepines from corresponding substituted diphenylthioureas by ring closure. Because of their central nervous system (CNS) activity, the products of the process can be used as tranquilizers and antidepressants and as analgesics.
The diazepines, oxazepines and thiazepines obtainable by means of the new process according to the invention can be represented by the following formula:

The compounds according to the invention are generally white crystalline solids which are only slightly soluble in water, but moderately soluble in organic solvents, for example methanol, ethanol and the like. They are basic substances that are usually soluble in aqueous mineral acids at room temperature. They form essentially insoluble acid addition salts, for example hydrochlorides, sulfates, phosphates, citrates, clock rates, maleates, fumarates and the like. The compounds of the invention can generally be administered orally or parenterally in the form of their salts and are effective central nervous system agents when administered in this way.
For oral administration, the new compounds according to the invention can be combined with the usual pharmaceutical carriers and, for example, in the form of tablets, capsules, dragees, liquids to be administered dropwise, emulsions, suspensions and syrups and in chocolate, sweets, chewing gum and the like can be applied. They can also be administered in the form of suppositories and as aqueous solutions for parenteral injection.
The new process according to the invention consists in the cyclization of substituted thioureas (II) to compounds (I) by heating with phosphorus pentoxide or phosphorus oxychloride according to the following reaction scheme:

The use of phosphorus pentoxide in conjunction with phosphorus oxychloride is particularly advantageous for the conversion of II to I. An excess of the latter reagent can also serve as a suitable solvent. The cyclization reactions are generally carried out at temperatures between about 50 and 150.degree. The preferred temperature is about 80-1100C. The addition of other solvents which are inert under the reaction conditions, for example benzene, toluene, xylene and the like, can also be advantageous. After the cyclization has taken place, usually after heating for about 30 minutes to about 24 hours, the products (I) are generally obtained by treating the reaction solution with water and then purifying the crude product by known measures.
The new thioureas (II), which are used as starting materials for the compounds (I) in this new process, can be prepared from substituted diphenyl esters, substituted diphenyl sulfides and substituted diphenyl amines according to the following reaction scheme:

According to this reaction scheme, the acylation of the substituted diphenyl esters, substituted diphenyl sulfides and substituted diphenylamines with phenoxythio carbonyl chloride leads to the thiocabanilates (IV). Treatment of the last-mentioned compounds with a mono- or diamine gives the thioureas (II).
The compounds obtainable by the process according to the invention have a physiological effect on the central nervous system. They show high activity as tranquilizers at non-toxic doses and in some cases antidepressant properties at dose levels that produce neither overt stimulation nor depression.
A suitable test for tranquilizer activity is to measure the reduction in spontaneous motor activity in animals using an actophotometer (a photoelectric device for the quantitative measurement of locomotor activity). Graduated doses of the active compounds prepared by the process of the invention are administered to groups of mice, and the effective dosage range for a significant reduction in motor activity (a measure of tranquilizer effect) compared to control groups is determined. The use of decreased motor activity as a measure of tranquilizer activity was proposed by W.D. Gray, A.C. Osterberg and C.E. Rauh, Archives Internationales et de Therapy, Vol. 134, p. 198 (1961) and W.J. Kinnard and C.J.
Carr, Journal of Pharmacology and Experimental Therapeutics, Vol. 121, p. 354 (1957).
The antidepressant properties of the compounds obtainable by the process according to the invention become evident when their ability to counteract depression induced in animals by administration of tetrabenazine hexamate is measured. Graduated doses of the active compounds according to the invention are administered to groups of mice. This is followed by the administration of a dose of tetrabenazine, which is known to markedly suppress the investigative behavior of normal mice. The groups treated with antidepressant show normal research behavior, while the control groups and groups treated with an agent ineffective as an antidepressant do not show this normal research behavior but the well-known deep depression induced by tetrabenazine.
The results at different dose levels are used to determine effective dose ranges. The antidepressants produced by the process according to the invention show their advantageous properties in this test at dosage values which cause only little or no undesirable side reactions, for example ataxia or reduced spontaneous motor activity.
Some of the compounds obtainable according to the invention also show other valuable pharmaceutical properties, for example analgesic activity.
The following examples explain in detail the preparation of representative substituted 11-aminodibenz-b, f] [1,4] oxazepines and thiazepines and substituted 11-aminodibenz [b, fjLl, 4] diazepines according to this invention.
Example 1 Preparation of l 1- (1-piperidinyl) dibenz [b, f] (1, 4] oxazepine
The compound phenyl-o-phenoxythiocarbanilate and piperidine are heated to reflux temperature in ethanol, followed by removal of the solvent and crystallization from dilute ethanol. The product is 2'-phenoxypiperidinthiocarboxanilid from F. 135
1360C received.
1.0 g of 2'-phenoxypiperidine thiocarboxanilide are refluxed with about 1 g of phosphorus pentoxide in 5 ml of phosphorus oxychloride for 2 hours. After cooling to room temperature, the reaction mixture is diluted with ether. The precipitate obtained is washed several times with ether, treated with aqueous potassium carbonate solution (preferably with cooling) and extracted three times with ether. The ether solution is dried over anhydrous potassium carbonate and, after evaporation of the solvent, yields a yellow solid which is recrystallized from heptane to white crystals of 98-1000C.
Example 2
Production of II-amine libenz [b, fle1,4loxazepine
Following the procedure of Example 1, treatment of phenyl-o-phenoxythiocarbanilate with ammoniacal ether gives l- (2-phenoxy) -phenylthiourea, with a melting point of 125 ° -1270 ° C., after recrystallization from dilute methanol.
Heating the 1- (2-phenoxy) -phenylthiourea prepared above with phosphorus pentoxide in phosphorus oxychloride gives the crystalline product from 198-2000C.
Example 3
Production of l l - (n-butylamino) dibenz [b, fl [1,4] oxazepine
16.66 g (0.075 mol) of 2-aminodiphenyl ether hydrochloride are converted into the free base by shaking with dilute ammonium hydroxide. The released base is extracted with ether and the ethereal solution is dried over sodium sulfate. This solution is treated with a solution of 6.45 g (0.0375 mol) of phenoxythiocarbonyl chloride in ether. The resulting solution is stirred with a magnetic stirrer at ambient temperature for about 21 hours. White crystals of 2-aminodiphenyl ether hydrochloride are separated off by filtration. Removal of the solvent from the filtrate gives an amber-colored oil which, after recrystallization from hexane, yields 12.4 g of solid with a melting point of 95-980C.
A solution of 3.21 g (10 mol) of the phenyl-o-phenoxythiocarbanilate prepared above and 1.46 g (20
Mol) n-butylamine in 50 ml of ethanol is heated to reflux temperature for 50 minutes. The solvent is removed and the residue crystallizes on trituration with hexane. By recrystallizing the product 1-butyl-3- (2-phenoxy) phenylthiourea from ether-hexane, white crystals with a melting point of 80-81 ° C. are obtained.
Following the procedure of Example 1, the reaction of the 1-butyl-3- (2-phenoxy) phenylthiourea prepared above with phosphorus pentoxide in phosphorus oxychloride gives crystals of 67-710C after recrystallization from heptane.
Example 4
Preparation of 11 - (4-methyl-1-piperazinyl) dibenzC6, ss [1,4-jexazepine
Following the procedure of Example 1, phenyl-o -phenoxythiocarbanilate is reacted with 1 molar equivalent of 1-methylpiperazine to give 4-methyl-2'-phenoxy-1 -piperazinthiocarboxanilide, which is obtained from dilute ethanol in the form of white crystals with a temperature of 139-1420C is obtained.

Production of II- [4- (l-Hydroxyethyl) -l-piperazinyl dihenzb, J] [1,4-joxyzepine-Dihydrochlond
Following the procedure of Example 1, phenyl -2- (p-chlorophenoxy) thiocarbanilate with 1 - (p-hydroxyethyl) piperazine is converted to 2 '- (p-chlorophenoxy) -4 (2'-hydroxyethyl) -1-piperazinthiocarboxanilide from F. 155 - 1580C implemented after recrystallization from ethyl acetate.

If 2 '- (p- -chlorophenoxy) -4-methyl-1-piperazinthiocarboxanilide is used as starting material for the procedure of Example 1, the product obtained is brownish crystals with a melting point of 109 to 110C.

Preparation of 2-chloro-II- (4-methyl-1-piperazinyl) dibenz [b, f] [1,4] thiazepine
