Verfahren zur Herstellung von tricyclischen Derivaten des Acetamids
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen tricyclischen Derivaten des Acetamids der Formel I

worin X eine gesättigte oder ungesättigte, geradkettige oder verzweigte Alkylengruppe mit 1 bis 3 Kohlenstoffatomen, eine Oxymethylen-, Thiomethylen-, Thioäthylengruppe, eine gegebenenfalls alkylierte bzw. acylierte Iminomethylen- oder gegebenenfalls alkylierte Carboximidogruppe, R1 Wasserstoff, ein Halogenatom, eine Niederalkyl-, Niederalkoxy-, Trifluormethyl- oder Niederalkylmercaptogruppe, R Wasserstoff oder die Hydroxygruppe, R3 Wasserstoff oder eine Niederalkylgruppe, R4 Wasserstoff oder zusammen mit R2 eine Doppelbindung bedeuten.
Es wurde gefunden, dass die Substanzen der Formel I wertvolle pharmakologische Eigenschaften, z. B. muskelrelaxierende, tranquilisierende und antikonvulsive Wirkungen, aufweisen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Carbonsäure der Formel II

oder deren reaktionsfähige Derivate mit Ammoniak umsetzt und die so erhaltenen Produkte für den Fall, dass Re eine Hydroxygruppe bedeutet, gewünschtenfalls nachträglich dehydratisiert und für den Fall, dass R; und R4 zusammen eine Doppelbindung darstellen, gewünsch tenfalls nachträglich hydriert.
Als reaktionsfähige Derivate der Carbonsäuren der Formel II kommen insbesondere die Säurehalogenide und Ester in Frage. Für den Fall, dass als Ausgangsprodukte die freien Säuren der Formel II verwendet werden, entstehen mit Ammoniak zunächst die Ammoniumsalze, die pyrolytisch oder katalytisch, vorzugsweise durch Zusatz eines sauren Katalysators, wie p-Toluolsulfonsäure, oder durch Zusatz eines Dehydratisierungsmittels, wie Carbodiimid, zu den Amiden der Formel I dehydratisiert werden. Wenn man von den reaktiven Derivaten der Carbonsäuren der Formel II ausgeht, werden diese vorzugsweise in einem inerten Lösungsmittel mit Ammoniak umgesetzt.
Bei einer bevorzugten Variante des erfindungsgemässen Verfahrens werden die freien Säuren der Formel II mit einem Halogenierungsmittel wie Thionylchlorid umgesetzt und die so entstandenen Säurechloride ohne Isolierung mit überschüssigem Ammoniak umgesetzt.
Die gegebenenfalls gewünschte nachträgliche Dehydratisierung der Verbindungen der Formel I erfolgt zweckmässig durch Erwärmen eines Gemisches von Eisessig und Mineralsäuren, vorzugsweise Salzsäure, oder Stehenlassen bei Raumtemperatur in alkoholischer Salzsäure. Eine gewünschtenfalls nachträglich durchzuführende Hydrierung von ungesättigten Verbindungen der Formel I (R2 und R4 bilden zusammen eine Doppelbindung) erfolgt vorzugsweise durch katalytische Hydrierung oder mit nascierendem Wasserstoff, der z. B. bei der Umsetzung von Wasser mit amalgamiertem Aluminium entwickelt wird.
Die als Ausgangsstoffe verwendeten Carbonsäuren der Formel II erhält man durch Umsetzung von Ketonen der Formel III

mit einem Fettsäureester (beispielsweise tert. Butylacetat) in Gegenwart eines alkalischen Kondensationsmittels. Die hierbei als Zwischenprodukte gebildeten Hydroxyester der Formel IV

lassen sich durch vorsichtige saure Hydrolyse in die gewünschten Carbonsäuren der Formel II umwandeln, in denen R. eine Hydroxygruppe bedeutet. Unter kräftigen Hydrolysebedingungen (z. B. Eisessig/6n Salzsäure) erreicht man eine Wasserabspaltung und Verseifung zu ungesättigten Carbonsäuren der Formel II (R2 und R4 bilden eine Doppelbindung), welche gewünschtenfalls katalytisch oder mit amalgamiertem Aluminium zu den gesättigten Säuren der Formel II (RD und R4 bedeuten Wasserstoff) hydriert werden.
Es ist aber erfindungsgemäss auch möglich, die Ester der Formel IV unmittelbar mit Ammoniak umzusetzen und auf die vorhergehende Isolierung der freien Carbonsäuren der Formel II zu verzichten.
In den nachstehenden Beispielen ist das erfindungsgemässe Verfahren näher erläutert.
Beispiel 1 11-Carboxamido-methylen-6, 1 1-dihydro-dibenz- [b,e]oxepin
25,4 g (0,1 Mol) 6,11 -Dihydro-dibenzo[b,e]oxepin- 1 1-yliden-essigsäure (Fp. 1l4I15o C) werden mit 40 ml Thionylchlorid 2 Stunden unter Rückflusskühlung erhitzt. Anschliessend dampft man ein, nimmt in 100 ml äther oder Tetrahydrofuran auf und versetzt mit etwa 50 ml flüssigem Ammoniak. Danach lässt man über Nacht das überschüssige Ammoniak verdampfen, saugt vom ausgeschiedenen Ammonchlorid ab und entfernt im Vakuum das Lösungsmittel. Der Rückstand wird dann aus Isopropanol oder Alkohol umkristallisiert. Die Ausbeute an 1 1-Carboxamido-methylen-6,1 1-dihydro-di- benz[b,e]oxepin beträgt 79 S der Theorie; Fp. 2050 C.
Beispiel 2
11 -Carboxamido-methyl-6, 11 -dihydro-dibenzb,e] oxepin
25,3 g (0,1 Mol) des gemäss Beispiel 1 hergestellten
11 -Carboxamido-methylen-6, 1 l-dihydro-dtibenz- [b,e]oxepin werden in 100 ml Tetrahydrofuran gelöst und nach Zugabe der gleichen Gewichtsmenge amalgamiertem Aluminium-Griess innerhalb von 3 Stunden tropfenweise mit 50 ml Wasser versetzt. Anschliessend lässt man über Nacht weiter reagieren, saugt ab und dampft die Lösung im Vakuum ein. Der Eindampfrückstand kann aus Essigester oder Isopropanol umkristallisiert werden. Die Ausbeute an 11 -Carboxamido-methyl-6, 11 -dihydro-di- benz[b,e]oxepin beträgt 86% der Theorie; Fp. 140 bis 1410 C.
Beispiel 3
S-Carboxamido-methyl-10,11-dihydro-SH-dibenzo- [a,d]cyclohepten Variante a:
In analoger Weise wie im Beispiel 1 beschrieben erhält man unter Verwendung von 10,1 1-Dihydro-5H-di- benzo[a,d]cyclohepten-5-yl-essigsäure (Fp. 163-164 C) mit einer Ausbeute von 82 % der Theorie
5-Carboxamido-methyl- 10,11 -dihydro-5H-dibenzo [a,d]cyclohepten; Fp. 137-1380 C.
Variante b:
In analoger Weise wie im Beispiel 2 beschrieben erhält man aus 0,1 Mol 5-Carboxamido-methylen-10,1 1- dihydro-5H-dibenzo[a,d]cyclohepten mit einer Ausbeute von 90% der Theorie 5-Carboxamido-methyl-10,1 1-dihydro-5H-dibenzo[a,d]cyclohepten; Fp. 137-138 C.
Das
5-Carboxamido-methylen-1 0,11 -dihydro 5 H-dibenzoa,dj cyclohepten wird wie folgt erhalten:
23,1 g (0,1 Mol) 5-Cyanomethylen-10,1 1-dihydro- 5H-dibenzo[a,d]cyclohepten (Fp. 105-106 C) werden mit der gleichen Gewichtsmenge Kaliumhydroxid in einem Gemisch aus 100 ml Wasser und 100 ml Alkohol vier Stunden zum Sieden erhitzt. Anschliessend verdünnt man mit Wasser, saugt den Niederschlag ab und kristallisiert aus Isopropanol um. Die Ausbeute an
5-Carboxamido-methylen- 10,11 -dihydro 5H-dibenzo[a,d] cyclohepten beträgt 85 % der Theorie; Fp. 166-167 C.
Beispiel 4
5 -Carboxamido-methyien-5H-dibenzoa,d] cyclohepten
In analoger Weise wie im Beispiel 1 beschrieben erhält man aus 24,8 g (0,1 Mol) SH-Dibenzo[a,d]cyclo- hepten-5-yliden-essigsäure (Fp. 207-209 C) mit einer Ausbeute von 87 % der Theorie 5-Carboxamido-methy len-5H-dibenzo[a,d]cyclohepten; Fp. 144-145 C.
Beispiel 5 S-Carboxamido-methyl-SH-dibenzo[a,d]cyclohepten
In analoger Weise wie im Beispiel 2 beschrieben erhält man aus 0,1 Mol des gemäss Beispiel 4 hergestellten 5-Carboxamido-methylen-5H-dibenzo[a,d]- cycloheptens mit einer Ausbeute von 81 % der Theorie S-Carboxamido-methyl-SH-dibenzo[a,d]- cyclohepten; Fp. 207-208 C.
Beispiel 6
In analoger Weise wie im Beispiel 1 beschrieben erhält man die folgenden Substanzen:
11 -Carboxamido-methylen-6,11 -dihydro-dibenzo- [b,e]thiepin; Fp. 209-210 C (Ausbeute 80 % d. Th.),
11 -Carboxamido-methyl-6, 11 -dihydro- dibenzo[b,e]thiepin; Fp. 194-195 C (Ausbeute 78 % d. Th.), 1 2-Carboxamido-methylen-7, 1 2-dihydro-6H- dibenzo[b,e]thiocin; Fp. 171-173 C (Ausbeute 72 % d. Th.),
11 -Carboxamido-methylen-2-methyl-6, 11 -dihydro- dibenzo[b,ejoxepin; Fp. 2020 C (Ausbeute 78 % d. Th.).
Als Ausgangsprodukt werden hierfür die folgenden
Säuren verwendet: 26,8 g 6,11 -Dihydro-dibenzo[b,e]thiepin- 11 -yliden- essigsäure (Fp. 180-1810 C); 27,0 g 6,11-Dihydro-dibenzo[b,e]thiepin-l l-yl essigsäure (Fp. 208-2090 C); 28,2 g 7,1 2-Dihydro-6H-dibenzo[b,e]thiocin 1 2-yliden- essigsäure (F. 188-1900 C); 26,6 g 2-Methyl-6,1 1-dihydro-dibenz[b,e]oxepin-1 1- yliden-essigsäure (Fp. 209-2110 C).
Beispiel 7
11 -Carboxamido-methyl-2-methyl-6, 11 -dihydro- dibenzb,e]oxepin
In analoger Weise wie im Beispiel 2 beschrieben erhält man aus 0,1 Mol des gemäss Beispiel 6 hergestellten
11 - Carboxamido - methylen - 2 - methyl - 6,11 - dihydrodibenz[b,e]oxepin mit einer Ausbeute von 76 % der Theorie 11 - Carboxamido - methyl - 2 - methyl - 6,1 1-di- hydro-dibenz[b,e]-oxepin; Fp. 1460 C.
Beispiel 8 trans- 11 -(1 -Carboxamido-äthylen- 1 )-6, 11 -dihydro dibenz[b,e]oxepin
In analoger Weise wie im Beispiel 1 beschrieben erhält man unter Verwendung von Il-(l-Carboxy-äthy- len-1)-6,11-dihydro-dibenz[b,e]oxepin in 42%iger Ausbeute das oben genannte Amid vom Fp. 188-1890 C (aus Isopropanol/Ligroin); aus der Mutterlauge wird die entsprechende cis-Form vom Fp. 218-2190C (31 % d. Th.) isoliert.
Die als Ausgangsprodukt verwendete Säure (cis/ trans-Gemisch) 11 -(1 -Carboxy-äthylen- 1 )-6, 11 -dihydrodibenz [b,c]oxepin hat einen Schmelzpunkt von 175 bis 1780 C.
Beispiel 9 ll-Carboxamido-methyl-5,6-dihydro- morphanthridinon-6
8 g 5,6-Dihydro-morphanthridin-6-on- 11 -acetylchlo- rid in 50 ml Benzol werden mit 200 ml konzentriertem Ammoniakwasser verrührt. Man saugt ab, wäscht ausgiebig mit Wasser und trocknet. Ausbeute: 5,3 g (72% d. Th.) 11 - Carboxamidomethyl -5,6- dihydro - morph- anthridinon-6 vom Schmp. 254-2550 C.
Das als Ausgangsmaterial verwendete 5,6-Dihydro morphanthridinon-6-on- 11 -acetylchlorid wird wie folgt erhalten:
11,5 g 5,6-Dihydro-morphanthridin-6,1 1-dion, 16,8 g Carbäthoxymethyl-phosphonsäure-diäthylester und 200 ml Dimethylformamid werden unter Rühren mit einer Lösung von 2,7 g Natrium in 20 ml Methanol und 20 ml Dimethylformamid versetzt. Nach zweistündigem Rühren neutralisiert man mit Essigsäure, giesst in Wasser und saugt den kristallinen Niederschlag ab. Ausbeute: 11 g (75 % d. Th.) 1 l-Carbäthoxymethylen-5,6-dihydromorphanthridinon-6 vom Schmp. 154-1550 C (Isopropanol). Dieses Produkt (9 g) wird in 200 ml Methanol gelöst und mit Raney-Nickel drucklos hydriert. Man filtriert, dampft ein und kristallisiert den Rückstand aus Isopropanol um. Ausbeute: 7 g (77 % d.
Th.) ll-Carb äthoxymethyl -5,6- dihydro-morphanthridinon - 6 vom Schmp. 150-1510 C. Die Verseifung des Esters mit kalter alkoholischer Natronlauge ergibt die 5,6-Dihydro morphanthridin-6-on-11-essigsäure vom Schmp. 267 bis 2680 C. Mit Oxalylchlorid bei Raumtemperatur erhält man aus dieser Säure das 5,6-Dihydro-morphanthridin 6-on-1 1-acetylchlorid vom Schmp. 102-1030 C.
In analoger Weise erhält man folgende Produkte: 11 -Carboxamido-methylen-5,6-dihydro morphanthridinon-6 Fp. 315-3180 C (Ausbeute 60 % der Theorie); 5-Methyl-l -carboxamido-methylen-5 ,6-dihydro- morphanthridinon-6 Fp. 250-2520 C (Ausbeute 70% der Theorie); 5-Carboxamido-methylen-1 0,1 l-dihydro-5H- dibenzo[a,d]cyclohepten Fp. 166-1670 C (Ausbeute 85 % der Theorie).
Process for the preparation of tricyclic derivatives of acetamide
The present invention relates to a process for the preparation of new tricyclic derivatives of acetamide of the formula I.

It has been found that the substances of formula I have valuable pharmacological properties, e.g. B. muscle relaxing, tranquilizing and anticonvulsant effects have.
The process according to the invention is characterized in that a carboxylic acid of the formula II

or the reactive derivatives thereof are reacted with ammonia and the products thus obtained are, if desired, subsequently dehydrated in the event that Re is a hydroxyl group, and in the event that R; and R4 together represent a double bond, if desired subsequently hydrogenated.
Particularly suitable reactive derivatives of the carboxylic acids of the formula II are the acid halides and esters. In the event that the free acids of the formula II are used as starting materials, ammonia first forms the ammonium salts, which are pyrolytically or catalytically, preferably by adding an acidic catalyst such as p-toluenesulfonic acid, or by adding a dehydrating agent such as carbodiimide the amides of the formula I are dehydrated. If the reactive derivatives of the carboxylic acids of the formula II are used, they are preferably reacted with ammonia in an inert solvent.
In a preferred variant of the process according to the invention, the free acids of the formula II are reacted with a halogenating agent such as thionyl chloride and the acid chlorides formed in this way are reacted with excess ammonia without isolation.
Any subsequent dehydration of the compounds of the formula I, which may be desired, is expediently carried out by heating a mixture of glacial acetic acid and mineral acids, preferably hydrochloric acid, or by allowing it to stand at room temperature in alcoholic hydrochloric acid. A hydrogenation to be carried out subsequently, if desired, of unsaturated compounds of the formula I (R2 and R4 together form a double bond) is preferably carried out by catalytic hydrogenation or with nascent hydrogen, the z. Example 1 11-Carboxamido-methylene-6,11-dihydro-dibenz- [b, e] oxepin
25.4 g (0.1 mol) 6,11-dihydro-dibenzo [b, e] oxepin-1 1-ylidene-acetic acid (melting point 114 ° C.) are refluxed with 40 ml of thionyl chloride for 2 hours. It is then evaporated, taken up in 100 ml of ether or tetrahydrofuran and treated with about 50 ml of liquid ammonia. The excess ammonia is then allowed to evaporate overnight, the ammonium chloride which has separated out is suctioned off and the solvent is removed in vacuo. The residue is then recrystallized from isopropanol or alcohol. The yield of 1 1-carboxamido-methylene-6,1 1-dihydro-dibenz [b, e] oxepine is 79% of theory; Mp. 2050 C.
Example 2
11-carboxamido-methyl-6, 11-dihydro-dibenzb, e] oxepin
25.3 g (0.1 mol) of that prepared according to Example 1
11-carboxamido-methylene-6, 1 l-dihydro-dtibenz- [b, e] oxepin are dissolved in 100 ml of tetrahydrofuran and, after adding the same amount by weight of amalgamated aluminum grit, 50 ml of water are added dropwise over the course of 3 hours. The reaction is then allowed to continue overnight, and the solution is filtered off with suction and evaporated in vacuo. The evaporation residue can be recrystallized from ethyl acetate or isopropanol. The yield of 11-carboxamido-methyl-6, 11-dihydro-dibenz [b, e] oxepine is 86% of theory; Mp. 140 to 1410 C.
Example 3
S-carboxamido-methyl-10,11-dihydro-SH-dibenzo- [a, d] cycloheptene variant a:
In a manner analogous to that described in Example 1, 1-dihydro-5H-dibenzo [a, d] cyclohepten-5-yl-acetic acid (melting point 163-164 ° C.) is obtained in a yield of 82 using 10.1 % of theory

Variant b:
In a manner analogous to that described in Example 2, 5-carboxamido-methylene-10,1 1-dihydro-5H-dibenzo [a, d] cycloheptene is obtained from 0.1 mol of 5-carboxamido-methylene in a yield of 90% of theory -10.1 1-dihydro-5H-dibenzo [a, d] cycloheptene; M.p. 137-138 C.

5-carboxamido-methylene-1 0,11 -dihydro 5 H-dibenzoa, dj cycloheptene is obtained as follows:
23.1 g (0.1 mol) of 5-cyanomethylene-10.1 1-dihydro-5H-dibenzo [a, d] cycloheptene (melting point 105-106 C) are mixed with the same amount by weight of potassium hydroxide in a mixture of 100 ml Water and 100 ml of alcohol heated to boiling for four hours. It is then diluted with water, the precipitate is filtered off with suction and recrystallized from isopropanol. The yield at
5-carboxamido-methylene-10,11-dihydro 5H-dibenzo [a, d] cycloheptene is 85% of theory; M.p. 166-167 C.
Example 4
5 -Carboxamido-methyl-5H-dibenzoa, d] cycloheptene
In a manner analogous to that described in Example 1, 24.8 g (0.1 mol) of SH-dibenzo [a, d] cyclohepten-5-ylidene-acetic acid (melting point 207-209 C) are obtained in a yield of 87% of theory 5-carboxamido-methylene-5H-dibenzo [a, d] cycloheptene; M.p. 144-145 C.
Example 5 S-carboxamido-methyl-SH-dibenzo [a, d] cycloheptene
In a manner analogous to that described in Example 2, 0.1 mol of the 5-carboxamido-methylene-5H-dibenzo [a, d] -cycloheptene prepared according to Example 4 is obtained with a yield of 81% of theory S-carboxamido-methyl- SH-dibenzo [a, d] - cycloheptene; Mp 207-208 C.
Example 6
In a manner analogous to that described in Example 1, the following substances are obtained:
11-carboxamido-methylene-6,11-dihydro-dibenzo- [b, e] thiepin; Mp. 209-210 C (yield 80% of theory),

11-carboxamido-methyl-2-methyl-6, 11-dihydro-dibenzb, e] oxepine
In a manner analogous to that described in Example 2, 0.1 mol of that prepared according to Example 6 is obtained
11 - carboxamido - methylene - 2 - methyl - 6.11 - dihydrodibenz [b, e] oxepine with a yield of 76% of theory 11 - carboxamido - methyl - 2 - methyl - 6,1 1-dihydrodibenz [ b, e] -oxepine; M.p. 1460 C.
Example 8 trans-11 - (1-carboxamido-ethylene-1) -6, 11 -dihydro dibenz [b, e] oxepin
In a manner analogous to that described in Example 1, using II- (l-carboxy-ethylene-1) -6,11-dihydro-dibenz [b, e] oxepine, the above-mentioned amide from is obtained in 42% yield M.p. 188-1890 C (from isopropanol / ligroin); the corresponding cis form of melting point 218-2190C (31% of theory) is isolated from the mother liquor.
The acid (cis / trans mixture) 11- (1-carboxy-ethylene- 1) -6, 11 -dihydrodibenz [b, c] oxepine used as the starting product has a melting point of 175 to 1780 C.
Example 9 II-Carboxamido-methyl-5,6-dihydromorphanthridinone-6
8 g of 5,6-dihydro-morphanthridin-6-one-11-acetyl chloride in 50 ml of benzene are stirred with 200 ml of concentrated ammonia water. It is suctioned off, washed extensively with water and dried. Yield: 5.3 g (72% of theory) 11-carboxamidomethyl -5,6-dihydro-morph-anthridinone-6 of melting point 254-2550 C.
