Es wurde gefunden, dass die Substanzen der For mel I wertvolle pharmakologische Eigenschaften, z. B. muskelrelaxierende, tranquillisierende und antikonvul sive Wirkungen, aufweisen.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man ein Nitril der Formel II

schonend verseift und die so erhaltenen Produkte für den Fall, dass R2 eine Hydroxygruppe bedeutet, ge wünschtenfalls nachträglich dehydratisiert und für den Fall, dass R2 und R4 zusammen eine Doppelbindung darstellen, gewünschtenfalls nachträglich hydriert.
Die schonende Verseifung der Nitrile der Formel 1I wird beispielsweise durch Erhitzen in alkoholischer Kalilauge oder durch Stehenlassen bei Raumtemperatur in alkoholischer Salzsäure bewirkt. Bei der Verwendung von alkoholischer Salzsäure gehen Verbindungen der Formel II, in denen R2 eine Hydroxygruppe darstellt, unter gleichzeitiger Wasserabspaltung in die Substanzen der Formel I über, in denen R2 und R4 zusammen eine Doppelbindung bilden.
Die gegebenenfalls gewünschte nachträgliche De hydratisierung der Verbindungen der Formel I erfolgt zweckmässig durch Erwärmen eines Gemisches von Eis essig und Mineralsäure, vorzugsweise Salzsäure, oder Stehenlassen bei Raumtemperatur in alkoholischer Salz säure. Eine gewünschtenfalls nachträglich durchzufüh rende Hydrierung von ungesättigten Verbindungen der Formel I (R2 und R4 bilden zusammen eine Doppel bindung) erfolgt vorzugsweise durch katalytische Hydrie rung oder mit nascierendem Wasserstoff, der z. B. bei der Umsetzung von Wasser mit amalgamiertem Alumi nium entwickelt wird.
Die als Ausgangsstoffe verwendeten Nitrile der For mel II sind im Schweizer Patent Nr. 499 487 beschrie ben und werden durch eine Art Aldolkondensation von Ketonen der Formel III

in Gegenwart eines basischen Kondensationsmittels vorzugsweise Lithiumamid in flüssigem Ammoniak und gewünschtenfalls anschliessende Dehydratisierung bzw. Hydrierung gewonnen.
Herstellung der Ausgangsverbindung 11-Carboxamido-methylen-6,11-dihydro- dibenz[b,e]oxepin 25,4 g (0,1 Mol) 6,11-Dihydro-dibenzo[b,e]oxepin- 11-yliden-essigsäure (Fp. 114-115 C) werden mit 40 ml Thionylchlorid 2 Stunden unter Rückflusskühlung erhitzt. Anschliessend dampft man ein, nimmt in 100 ml Äther oder Tetrahydrofuran auf und versetzt mit etwa 50 - ml flüssigem Ammoniak. Danach lässt man über Nacht das überschüssige Ammoniak verdampfen, saugt vom ausgeschiedenen Ammonchlorid ab und entfernt im Vakuum das Lösungsmittel. Der Rückstand wird dann aus Isopropanol oder Alkohol umkristallisiert.
Die Ausbeute an 11-Carboxamido-methylen-6,11-dihydro- dibenz[b,e]oxepin beträgt 79 % der Theorie; Fp. 205 C.
In den nachstehenden Beispielen ist das erfindungs gemässe Verfahren näher erläutert.
<I>Beispiel 1</I> 11-Carboxamido-methyl-6;11-dihydro- dibenzo[b,e]oxepin 25,3 g (0,1 Mol) des oben hergestellten 11-Carbox- amido-methylen-6,11-dihydro-dibenz[b,e]oxepin werden in 100 ml Tetrahydrofuran gelöst und nach Zugabe der gleichen Gewichtsmenge amalgamiertem Aluminium- Griess innerhalb von 3 Stunden tropfenweise mit 50 ml Wasser versetzt. Anschliessend lässt man über Nacht weiter reagieren, saugt ab und dampft die Lösung im Vakuum ein. Der Eindampfrückstand kann aus Essig ester oder Isopropanol umkristallisiert werden.
Die Aus beute an 11-Carboxamido-methyl-6,11-dihydro-dibenz- [b,e]oxepin beträgt 86% der Theorie; Fp. 140-141 C. <I>Beispiel 2</I> 5-Carboxamido-methylen-10,11-dihydro- 5H-dibenzo[a,d]cyclohepten 23,1 g (0,1 Mol) 5-Cyanomethylen-10,11-dihydro- 5H-dibenzo[a,d]cyclohepten (Fp. 105-106 C) werden mit der gleichen Gewichtsmenge Kaliumhydroxid in einem Gemisch aus 100 ml Wasser und 100 ml Alkohol vier Stunden zum Sieden erhitzt.
Anschliessend verdünnt man mit Wasser, saugt den Niederschlag ab und kristal- lisiert aus Isopropanol um. Die Ausbeute an 5-Carbox- amido-methylen-10,11-dihydro -5H-dibenzo[a,d]cyclo- hepten beträgt 85 % der Theorie; Fp. 166-167 C.
<I>Beispiel 3</I> 5-Carboxamido-methyl-10,11-dihydro- 5H-dibenzo[a,d]cyclohepten In analoger Weise wie im Beispiel 1 beschrieben, erhält man aus 0,1 Mol des gemäss Beispiel 2 hergestell ten 5-Carboxamido-methylen-10,11-dihydro-5H-dibenzo- [a,d]cycloheptens mit einer Ausbeute von 90% der Theorie 5-Carboxamido-methyl-10,11-dihydro-5H-di- benzo[a,d]cyclohepten; Fp. 137-138 C.
<I>Beispiel 4</I> 5-Carboxamido-methylen-5H-dibenzo[a,d]cyclohepten In analoger Weise wie im Beispiel 2 beschrieben, erhält man aus 0,1 Mol 5-Cyanomethylen-5H-dibenzo- [a,d]cyclohepten (Fp. 102-103 C) mit einer Ausbeute von 63 % der Theorie 5-Carboxamido-methylen-5H- dibenzo[a,d]cyclohepten; Fp. 144-145 C.
<I>Beispiel 5</I> 5-Carboxamido-methyl-5H-dibenzo[a,d]cyclohepten In analoger Weise wie im Beispiel 1 beschrieben, erhält man aus 0,1 Mol des gemäss Beispiel 4 herge stellten 5-Carboxamido-methylen-5H-dibenzo[a,d]cyclo- heptens mit einer Ausbeute von 81% der Theorie 5-Carboxamido-methyl-5H-dibenzo[a,d]cyclohepten; Fp. 207-208 C.
<I>Beispiel 6</I> 11-Carboxamido-methyl-2-methyl-6,11-dihydro- dibenz[b,e]oxepin In analoger Weise wie im Beispiel 1 beschrieben, erhält man aus 0,1 Mol des oben hergestellten 11-Carboxamido -methylen- 2-methyl-6,11-dihydro-di- benz[b,e]oxepins mit einer Ausbeute von 76% der Theorie 11-Carboxamido-methyl-2-methyl-6,11-dihydro- dibenz[b,e]oxepin; Fp. l46 C.

The gentle saponification of the nitriles of the formula 1I is brought about, for example, by heating in alcoholic potassium hydroxide solution or by allowing it to stand at room temperature in alcoholic hydrochloric acid. When using alcoholic hydrochloric acid, compounds of the formula II in which R2 represents a hydroxyl group are converted into substances of the formula I in which R2 and R4 together form a double bond, with simultaneous elimination of water.
Any desired subsequent dehydration of the compounds of the formula I is conveniently carried out by heating a mixture of glacial acetic acid and mineral acid, preferably hydrochloric acid, or by allowing it to stand at room temperature in alcoholic hydrochloric acid. Preparation of the starting compound 11-carboxamido-methylene-6,11-dihydrodibenz [b, e] oxepin 25.4 g (0.1 mol) 6,11-dihydro-dibenzo [b, e] oxepin-11-ylidene- acetic acid (melting point 114-115 ° C.) are refluxed with 40 ml of thionyl chloride for 2 hours. It is then evaporated, taken up in 100 ml of ether or tetrahydrofuran, and about 50 ml of liquid ammonia are added. <I> Example 1 </I> 11-carboxamido-methyl-6; 11-dihydrodibenzo [b, e] oxepin 25.3 g (0.1 mol) of the 11-carboxamido-methylene-6 prepared above , 11-dihydro-dibenz [b, e] oxepin are dissolved in 100 ml of tetrahydrofuran and, after adding the same amount by weight of amalgamated aluminum grit, 50 ml of water are added dropwise over the course of 3 hours. Example 2 5-Carboxamido-methylene-10,11-dihydro-5H-dibenzo [a, d] cycloheptene 23.1 g (0.1 mol) 5-cyanomethylene -10,11-dihydro-5H-dibenzo [a, d] cycloheptene (melting point 105-106 C) are heated to boiling for four hours with the same amount by weight of potassium hydroxide in a mixture of 100 ml of water and 100 ml of alcohol.
