Verfahren zur Herstellung neuer Diazacycloalkanverbindungen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 2,4-Dioxo-1 1,3 -diazacycloalkanverbin- dungen der allgemeinen Formel

worin Rt einen niederen aliphatischen Kohlenwasserstoffrest, einen gegebenenfalls substituierten Phenylrest oder insbesondere ein Wasserstoffatom, Z einen niederen unverzweigten Alkylenrest, der die Carbonylgruppe vom Stickstoffatom durch höchstens 4, insbesondere durch 1 bis 3 Kohlenstoffatome trennt und der durch einen oder mehrere gegebenenfalls substituierte Kohlenwasserstoffreste substituiert sein kann, und R ein Wasserstoffatom oder einen gegebenenfalls substituierten aliphatischen Kohlenwasserstoffrest,
vor allem einen unsubstituierten oder einen durch eine Hydroxylgruppe oder eine freie oder substituierte Aminogruppe substituierten niederen Alkylrest, einen niederen Alkenylrest, oder einen araliphatischen Rest, bedeutet, und ihrer Salze.
Als Kohlenwasserstoffreste sind insbesondere niedere Alkylreste, Phenylreste und Phenyl-niederalkylreste, wie Benzyl- oder Phenyläthylreste, zu nennen. Als Substituenten dieser Phenyl- oder Phenyl-niederalkylreste kommen vor allem niedere Alkylreste, niedere Alkoxygruppen, wie Methoxy-, Athoxy-, Propoxy- oder Butoxygruppen, Halogenatome, wie Chlor oder Brom, Trifluoromethylgruppen oder Nitrogruppen in Betracht.
Niedere Alkylreste sind oben und nachfolgend vorzugsweise solche mit höchstens 5 Kohlenstoffatomen, wie Methyl-, Athyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl- oder Pentylreste. Niedere Alkenylreste sind vor allem Allyl- oder Methallylreste. Als araliphatische Reste seien vor allem Phenyl-niederalkylreste, wie Benzyl-, 1-Phenyl-äthyl- oder 2-Phenyl-äthylreste, erwähnt.
Substituierte Aminogruppen sind monosubstituierte, in erster Linie aber disubstituierte Aminogruppen, wobei als Substituenten vor allem Alkyl-, Alkenyl-, Alkylen-, Cycloalkyl-, Oxa- oder Azaalkylenreste in Betracht kommen. Zu nennen sind z. B. Di-niederalkylaminogruppen, wie Dimethyl-, Diäthylamino- oder Dipropylaminogruppen, Pyrrolidino-, Piperidino-, Morpholino-, Hexa- oder Heptamethylenamino-, Piperazino-, N'-Methyl-piperazino- - oder N'-Hydroxyäthyl-piperazinogruppen.
Der substituierte Alkylrest ist besonders ein Hydroxymethylrest oder ein Hydroxy- oder tert.-Aminoäthyloder -propylrest.
Die araliphatischen Reste können an den Kohlenstoffatomen substituiert sein, vor allem an den Arylresten durch Halogenatome, wie Chlor oder Brom, das Pseudohalogen Trifluormethyl, niedere Alkylgruppen, wie Methyl oder Äthyl, niedere Alkoxygruppen, wie Methoxy, Äthoxy oder Methylendioxy, oder auch Nitrogruppen.
Als niedere aliphatische Kohlenwasserstoffreste Rl kommen vor allem niedere Alkylrestee in Frage, wie Methyl-, Athyl-, Propyl-, Tsopropyl-, Butyl-, Isobutyl-oder Pentylreste, ferner auch niedere Alkenylreste, wie Allyloder Methallylreste. Als Substituenten von Phenylresten R1 kommen z. B. die oben angegebenen in Betracht.
Der Rest Z ist insbesondere ein Methylen-, Athy- len-(1,2)-, Propylen-(1,3)- oder Butylen-(1,4)-re-t, der wie angegeben substituiert sein kann, vor allem durch niedere Alkylreste.
Die neuen Verbindungen besitzen wertvolle pharmakologische, insbesondere antiparasitäre und antibakterielle Eigenschaften. Sie zeigen vor allem eine Wirkung gegen Protozoen und Würmer und sind z. B. am infizierten Tier, beispielsweise an Mäusen, gegen gramnegative Bakterien, z. B. Salmonella typhi oder Coli Bazillen, wie Esch. coli, wirksam. Insbesondere wirken die neuen Verbindungen, wie sich z. B. bei Versuchen an Hamstern zeigt, gegen Trichomonaden und Amoeben sowie z. B. an Mäusen und Schafen gegen Schistosomen.
Ferner besitzen sie eine Wirkung gegen Coccidien. Die neuen Verbindungen sind entsprechend als antiparasitäre und antibakterielle Mittel nützlich. Insbesondere eignen sie sich zur Behandlung der durch die genannten Erreger verursachten Erkrankungen. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe.
Besonders hervorzuheben sind die Verbindungen der Formel

Besonders wertvoll bezüglich ihrer biologischen Eigenschaften sind das 1 -[5-Nitrothiazolyl-(2)J-2,4-diox imidazolidin, das I-[5-Nitrothiazolyl-(2)]-2,4-dioxo-3- (hydroxymethyl)-imidazolidin, das 1- [5 -Nitrothiazolyl- (2)]-3-(fl'-diäthylamino-äthyl)-2,4-dicxo-imidazolidin, das 1 - [5 Nitrothiazolyl-(2)] -2 ,4-dioxo-hexahydropyrimidin sowie das 1-[5-Nitrothiazolyl-(2)]-3-(hydroxymethyl)- 2,4-dioxo-hexahydropyrimidin.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel

worin Ri und R die angegebenen Bedeutungen haben, Z einen niederen unverzweigten Alkylenrest, der Y vom Stickstoffatom durch höchstens 4 Kohlenstoffatome trennt und der durch einen oder mehrere gegebenenfalls substituierte Kohlenwasserstoffreste substituiert sein kann, und Y eine freie oder eine eine Oxogruppe enthaltende funktionell abgewandelte Carboxylgruppe bedeutet, intramolekular kondensiert. Eine eine Oxogrup pe enthaltende funktionell abgewandelte Carboxylgruppe ist vor allem eine veresterte Carboxylgruppe, z. B. eine mit einem niederen Alkanol z. B. Methanol oder Athanol veresterte Carboxylgruppe.
Die intramolekulare Kondensation (Ringschluss) erfolgt in üblicher Weise, zweckmässig durch Erhitzen in einem Lösungsmittel und gegebenenfalls in Anwesenheit eines Kondensationsmittels, insbesondere eines sauren Mittels, wie einer Mineralsäure, z. B. einer Halogenwasserstoffsäure oder Schwefelsäure.
In erhaltenen Verbindungen kann man im Rahmen der Endstoffe weitere Substituenten einführen oder vorhandene Substituenten abspalten oder umwandeln. So kann man insbesondere in erhaltenen Verbindungen, in denen R ein Wasserstoffatom bedeutet, einen der eingangs angegebenen Substituenten R einführen. Dies kann in an sich bekannter Weise geschehen, z. B. zur Herstellung von Verbindungen, in denen der Substituent R keine Heteroatome aufweist oder in denen gegebenenfalls in R vorhandene Heteroatome durch mindestens 2 Kohlenstoffatome vom Ringstickstoffatom getrennt sind, durch Reaktion mit reaktionsfähigen Estern von Alkoholen der Formel R-OH. Reaktionsfähige Ester sind dabei z. B. solche mit starken anorganischen Säuren oder organischen Sulfonsäuren, vor allem mit Halogenwasserstoffsäuren, z. B.
Chlor-, Brom- oder Jodwasserstoffsäure, oder Schwefelsäure, oder Aryl- oder Alkansulfonsäuren, vor allem Phenyl-, wie Toluolsulfonsäuren. Dabei kann man, wenn erwünscht, mit einem Metall-, wie Alkalimetallsalz der in 3-Stellung unsubstituierten 2,4-Dioxo-l ,3-diazacycloalkanverbindung oder in Gegenwart eines basischen Kondensationsmittels, besonders eines Metallsalze bildenden Kondensationsmittels, wie Amiden, Hydriden, Kohlenwasserstoffverbindungen, Hydroxyden, Alkoholaten oder Carbonaten von Alkalimetallen arbeiten.
Die Einführung des Restes R der genannten Art kann aber auch gegebenenfalls durch Behandlung mit einer Diazoverbindung der Formel R'=NN erfolgen, worin R' bis auf die Dop pelbindung zum Stickstoff hin dem Alkoholrest R entspricht.
Verbindungen, in denen der Rest R ein eine Hydroxylgruppe tragender Methylrest ist, können durch Reaktion mit Formaldehyd erhalten werden.
Die Einführung der Hydroxymethylgruppe kann durch einfache Reaktion mit Formaldehyd erfolgen, gegebenenfalls in Form eines Formaldehyd-Donators, wie Trioxymethylen oder Paraformaldehyd, vorteilhaft in Gegenwart eines basischen Kondensationsmittels, wie eines Alkalihydroxyds oder -carbonats, oder tertiärer Amine oder quaternärer Ammoniumhydroxyde, wie Tri äthylamin oder Benzyltrimethylammoniumhydroxyd.
Die genannten Reaktionen können in üblicher Weise, in An- oder Abwesenheit von Verdünnungsmitteln, Kondensationsmitteln und/oder Katalysatoren, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, bei normalem oder erhöhtem Druck und/oder unter einer Inertgasatmosphäre durchgeführt werden.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe gegebenenfalls in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Salze. Erhaltene Amine lassen sich in üblicher Weise durch Umsetzung mit organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, in Salze umwandeln. Anderseits lassen sich die erhaltenen Salze in üblicher Weise, z. B. durch Behandlung mit basischen Mitteln oder Ionenaustauschern, in die freien Verbindungen überführen.
Als Säuren, die für die Bildung therapeutisch verwendbarer Salze geeignet sind, seien beispielsweise genannt Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymaleinoder Brenztraubensäure; Phenylessig-, Benzoe-, p-Amino-benzoe-, Anthranil- pHydroxy-benzoe-, Salicyl-, p Amino-salicyl- oder Embonsäure, Methansulfon-, Ät- hansulfon-, Hydroxyäthansulfon-, Athylensulfonsäure, Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure, Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen Basen dienen, indem man diese in Salz überführt, die Salze abtrennt und aus den Salzen die Basen freisetzt. Infolge der engen Beziehungen zwischen den Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Ausgangsstoffe können auch unter den Reaktionsbedingungen gebildet werden.
So kann man beispielsweise eine Verbindung der Formel

worin R1 und R die angegebenen Bedeutungen haben, mit einer Verbindung der Formel X'-Z-Y, worin X' eine reaktionsfähig veresterte Hydroxylgruppe, insbesondere ein Halogenatom, bedeutet, Z einen niederen unverzweigten Alkylenrest, der Y von X' durch höchstens 4 Kohlenstoffatome trennt und der durch einen oder mehrere gegebenenfalls substituierte Kohlenwasserstoffreste substituiert sein kann, und Y die oben angegebene Bedeutung hat, umsetzen, wobei intermediär die Verbindung der Formel II gebildet wird, die dann erfindungsgemäss intramolekular kondensiert wird.
Zweckmässig verwendet man solche Ausgangsstoffe, die zu den eingangs als besonders wertvoll geschilderten Endstoffen führen.
Die verwendeten Ausgangsstoffe sind bekannt oder werden, falls neu, in an sich bekannter Weise hergestellt.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die eingangs genannten Verbindungen können aber auch zusammen mit gebräuchlichen Futter- bzw. Träger- stoffen in Form von Veterinärpräparaten oder als Futter- bzw. Futterzusatzmittel bei der Aufzucht von Tieren Verwendung finden.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
24 g N- [5-Nitro-thiazolyl-(2)} -N-carbäthoxymethyl- N'-(n-butyl)-harnstoff werden in 120 ml alkoholischer Salzsäure während 2 Stunden am Rückfluss gekocht.
Die heisse Lösung wird dann filtriert, eingeengt und das daraus erhaltene Kristallisat isoliert. Man erhält so das 1 [5-Nitro thiazolyl-(2)]-3-(n-butyl)-2, 4-dioxo-imidawl- idin der Formel

Der als Ausgangsmaterial verwendete N-[5-Nitr thiazolyl-(2)j -N-carbäthoxymethyl-N'-(n-butyl)-harn- stoff kann durch Kondensation von 44 g N-[5-Nitro thiazolyl-(2)]-N'-(n-butyl)-harnstoff (hergestellt aus 2 Amino-5-nitrothiazol und n-Butyl-isocyanat in Aceton) mit 33 g Bromessigsäure-äthylester in absolutem Dioxan in Gegenwart von 8,8 g Natriumhydrid in Öl (5001zig) erhalten werden. Er schmilzt bei 143-145".
In ähnlicher Weise können die folgenden Verbindungen hergestellt werden: a) I-[5-Nitrothiazolyl-(2)]-3-äthyl-2,4-dio idin, F. 204-2060; b) l- -Nitrothiazolyl-(2)1-3-methyl-2 ,4-dioxo-imida- zolidin, F. 230-232"; c) I-[5-NitrothiazoIyl-(2)]-2,4-dioxo-imida F.240-242"; d) 1-[5-Nitrothiazolyl-(2)]-2, 4-dioxo-hexahydropyrim- idin, F. 278279C.
Beispiel 2
Zu einer Lösung von 5,0 g 1 -[5-Nitrothiazolyl-(2)]- 2,4-dioxo-imidazolidin in 100 ml absolutem Tetrahydrofuran gibt man eine Lösung von 1 g Diazomethan in 20 ml Ather und lässt 4 Stunden bei Zimmertemperatur stehen. Hierauf wird im Vakuum eingedampft. Den Rückstand kristallisiert man aus Dimethylformamid Wasser um. Man erhält so das l-[5-Nitrothiazolyl-(2)j- 3-methyl-2,4-dioxo-imidazolidin der Formel

Beispiel 3
7,0 g g 1-[5-Nitrothi azolyl-(2)]-2,4-di oxo-hexahydropyrimidin werden in 27 ml Dimethylformamid gelöst.
Unter Rühren gibt man bei Zimmertemperatur 1,45 g einer 500/oigen Natriumhydrid-Dispersion in Mineralöl zu dieser Lösung. Anschliessend werden 4,7 g Methyljodid zugegeben und eine Stunde weitergerührt. Das Reaktionsgemisch wird mit Wasser verdünnt. Der entstandene Niederschlag wird durch Filtrieren isoliert, mit Wasser gewaschen, getrocknet. Die Umkristallisation aus Dimethylformamid liefert das 1-[5-Nitrothiazolyl (2)] -2,4-dioxo-3-methyl-hexahydropyrimidin der Formel

Beispiel 4
Die neuen Verbindungen, insbesondere das 1-[5 Nitrothiazolyl-(2)]-2,4-dioxo-imidazolidin, können als Zusatz zu Tierfutter, z. B. Geflügelfutter, verwendet werden. So kann z.B. das 1-[5-Nitrothiazolyl-(2)]-2,4-di- oxo-imidazolidin mit Cerelose vermischt werden (Gehalt an aktiver Verbindung z. B. 0,1-10:o, vorzugsweise 0,5 O/o). Diese Vormischung kann dann dem Futter in üblicher Weise zugesetzt werden, zweckmässig so, dass der Gehalt an Imidazolderivat ca. 0,01 O/o beträgt.
