Process for the production of new sulfonylureas
The present invention relates to a process for the preparation of new sulfonylureas.
Compounds of general formula I,
EMI1.1
in which Ri is an alkyl or alkenyl group of at most 10 carbon atoms, a cycloalkyl or cycloalkenyl group of at most 12 carbon atoms, or a polycycloalkyl group of at most
11 means carbon atoms, as well as their salts with inorganic or organic bases have not yet become known.
As has now been found, these compounds as well as their pharmaceutically acceptable salts have valuable pharmacological properties. Surprisingly, when administered orally or parenterally, they show long-lasting hypoglycemic action, which characterizes them as being suitable for the treatment of diabetes.
In the compounds of general formula I, Rl can be the following groups: as an alkyl group z. B. the propyl, isopropyl, butyl, isobutyl, sec. Butyl, tert. Butyl, pentyl, isopentyl, tert. Pentyl, 1-methyl-butyl, 2-methyl-butyl, 1,2-dimethyl-propyl, 2,2-dimethyl-propyl, hexyl, 2-methyl-pentyl, heptyl, 4-methyl -hexyl, octyl, nonyl or the decyl group; as an alkenyl group e.g. B. the allyl, crotyl or methallyl group; as a cycloalkyl group e.g.
B. the cyclobutyl, cyclopentyl, cyclohexyl, cycloheptyl, cyclooctyl, cyclononyl, cyclodecyl, cyclododecyl, cyclopentylmethyl, cyclohexylmethyl, cycloheptylmethyl or the cyclooctylmethyl group, optionally by such methyl groups, and derivatives Ethyl or tert. Butyl groups are substituted; also as a cycloalkenyl group, e.g. B. the 2-cyclopenten-1-yl or the 3-cyclohexen-1-yl group and as a polycycloalkyl group z. B. the bornyl, norbornyl, norbornylmethyl, adamantyl or the octahydro-1,2,4-methenopentalenyl group.
The compounds of general formula I are prepared according to the invention by adding a compound of general formula II,
EMI1.2
in which Rl denotes hydrogen or an acyl radical, with an isocyanate derivative of the general formula III, Ri-N = C = O (III) or with a reactive ester of a carbamic acid of the general formula IV,
EMI1.3
in which Ri has the meaning given under formula I, if necessary the reaction product is partially hydrolyzed to split off an acyl radical bonded to the ring nitrogen and, if desired, a compound of the general formula I obtained is converted into a salt with an inorganic or organic base.
Ra can be used as an acyl radical of compounds of the general formula II z. B. a lower alkanoyl group, especially the acetyl group, or a lower alkoxycarbonyl group, such as. B. the methoxycarbonyl or ethoxycarbonyl group. Lower alkyl esters, especially the methyl or ethyl esters, and also phenyl esters, are suitable as reactive esters of carbamic acids of the general formula IV.
The reaction according to the invention is carried out, for example, in the cold or by heating, preferably in an inert organic solvent and in the presence of a condensing agent. Suitable inert organic solvents are, for example, hydrocarbons such as benzene, toluene or xylene, ethereal liquids such as ether, dioxane or tetrahydrofuran, chlorinated hydrocarbons such as methylene chloride, or lower ketones such as acetone or methyl ethyl ketone. An alkali metal hydroxide, such as sodium or potassium hydroxide, or an alkali metal alkoxide, such as sodium or potassium methylate or a corresponding ethylate, can be used as the condensing agent.
Tertiary organic bases can be used as further condensing agents in the conversion of an isocyanate derivative.
Reaction products obtained according to the process which are substituted on the ring nitrogen by an acyl group and preferably of the general formula V
EMI2.1
in which R is a lower alkanoyl or alkoxycarbonyl group and Ri has the meaning given under formula I, are then converted into compounds of general formula I by partial hydrolysis. The partial hydrolysis is preferably carried out in an alkaline medium, for example in methanolic or ethanolic sodium or potassium hydroxide solution.
Starting materials of the general formula II, 10,11-dihydro-5H-dibenz [b, f] azepine-3-sulfonamide and the corresponding 5-acetyl derivative, are described in the literature (cf. JR Geigy AG, Belgian Patent No. 616 134). Further connections of this type can be made in the same way.
The new active ingredients or pharmaceutically acceptable salts thereof are preferably administered orally. Inorganic or organic bases, such as, for example, alkali metal or alkaline earth metal hydroxides, carbonates or bicarbonates, triethanolamine, choline, Nt-dimethyl or N1- (β-phenylethyl) biguanide, can be used for salt formation. The daily doses range between 50 and 2000 mg for adult patients. Suitable dosage unit forms, such as dragees, tablets or push-fit capsules, preferably contain 25-500 mg of an active ingredient according to the invention, namely 20 to 80% of a compound of the general formula I.
For the production of dragees and tablets one combines the active ingredient z. B. with solid powdery carriers such as lactose, sucrose, sorbitol, mannitol; Starches, such as potato starch, corn starch or amylopectin, also laminaria powder or citrus pulp powder; Cellulose derivatives or gelatin, optionally with the addition of lubricants, such as magnesium or calcium stearate or polyethylene glycols, to tablets or to dragee cores. The latter is coated, for example, with concentrated sugar solutions, which z. B. can contain gum arabic, talc and / or titanium dioxide, or with a paint dissolved in volatile organic solvents or solvent mixtures.
Dyes can be added to these coatings, e.g. B. to identify different drug doses.
The mentioned push-fit capsules made of gelatin contain the active ingredient preferably as granules, e.g. B. mixed with fillers such as corn starch and / or lubricants such as talc or magnesium stearate, and optionally stabilizers such as sodium metabisulphite (NagS2OD) or ascorbic acid.
The following instructions are intended to explain the production of tablets and coated tablets in more detail: a) 1000 g of 1- (10, 11-dihydro-5H-dibenz [b, f] -azepin-3-yl-sulfonyl) -3-butyl-urea are used mixed with 550 g of lactose and 292 g of potato starch, the mixture is moistened with an aqueous solution of 8.0 g of gelatin and granulated through a sieve.
After drying, 60.0 g of potato starch, 60.0 g of talc, 10.0 g of magnesium stearate and 20.0 g of colloidal silicon dioxide are mixed in and the mixture is pressed into 10,000 tablets, each weighing 200 mg and containing 100 mg of active ingredient, if desired Partial notches can be provided for finer adjustment of the dosage. b) From 1000 g of sodium salt of 1- (10, 11-dihydro-5H-dibenz- [b, f] azepin-3-ylsulfonyl) -3-butylurea, 379.0 g of lactose and the aqueous solution of 6, 0 g of gelatin is made into granules which, after drying, are mixed with 10.0 g of colloidal silicon dioxide, 40.0 g of talc, 60.0 g of potato starch and 5.0 g of magnesium stearate and pressed to form 10,000 coated tablets.
These are then crystallized with a concentrated syrup of 533.5 g. Sucrose, 20.0 g of shellac, 75.0 g of gum arabic, 250 g of talc, 20 g of colloidal silicon dioxide and 1.5 g of dye coated and dried. The dragees obtained each weigh 240 mg and each contain 100 mg of active ingredient.
The following examples explain the preparation of the new compounds of general formula I and of hitherto not described intermediates in more detail, but are by no means the only embodiment of the same. The temperatures are given in degrees Celsius.
Example 1 a) 31.6 g of 5-acetyl-10,11-dihydro-5H-dibenz [b, f] -azepine-3-sulfonamide (cf. JR Geigy AG, Belgian patent specification No. 616134) are in 25 ml of 5n Sodium hydroxide solution and 200 ml of dioxane dissolved with stirring and mixed with 8.5 g of propyl isocyanate. A slightly exothermic reaction sets in. After this has subsided, the reaction mixture is stirred for one hour at 80, then cooled to 20 and concentrated in vacuo. The residue is dissolved in water, the solution is filtered and poured into ice-cold 2N hydrochloric acid. The crude product precipitates. It is filtered off with suction, washed with water, treated with 2N ammonia, the residue is filtered off and the ammonia solution is poured onto ice-cold 2N hydrochloric acid.
The crude 1- (5-acetyl-10, 11-dihydro-5H-dibenz [b, f] azepin-3-yl-sulfonyl) -3-propyl urea precipitates. The crude product is filtered off with suction, washed thoroughly with water and dried at 60 in a vacuum. b) 40.1 g of crude 1- (5-acetyl-10, 11-dihydro-5H-dibenz [b, f] azepin-3-ylsulfonyl) -3-propylurea are dissolved in 400 ml of ethanol and mixed with 20 , 0 g sodium hydroxide added. The mixture is refluxed for 3 hours, the sodium salt of 1- (10, 11 dihydro-5H-dibenz [b, f] azepin-3-yl-sulfonyl) -3-propylurea crystallizing out. The sodium salt is filtered off, washed with ethanol, recrystallized from 50% ethanol (percent by volume) and dissolved in 300 ml of warm water.
This solution is poured into 2N hydrochloric acid. The free, crude 1- (10, 11-dihydro-5H-dibenz [b, f] 'azepin-3-yl-sulfonyl) -3-propyl-urea precipitates. The crude product is filtered off with suction, washed thoroughly with water and recrystallized from ethanol, whereupon the pure compound melts at 183-185 after previous sintering.

Example 3
27.4 g of 10,11-dihydro-5H-dibenz [b, f] azepine-3-sulfonamide (cf. JR Geigy AG, Belgian patent specification No. 616 134) and 5.6 g of finely powdered potassium hydroxide are dissolved in 250 ml of dioxane dissolved and refluxed with 9.9 g of butyl isocyanate for one hour.
The reaction mixture is then cooled and evaporated in vacuo. The residue is taken up in 300 ml of hot water, little insolubles are filtered off, the filtrate is cooled and poured onto 2N hydrochloric acid with stirring. The crude product precipitates; it is filtered off, washed with water and taken up in 2N ammonia solution. The ammonia solution is separated from little insoluble matter and again poured onto 2N hydrochloric acid. The crude product precipitates; it is filtered off, washed thoroughly with water and recrystallized from acetone. The pure 1- (10, 11-dihydro-SH-dibenz [b, f] azepin-3-ylsulfonyl) -3-butylurea obtained melts at 183.5-185.5.
This urea derivative is converted into the sodium salt as follows: 37.3 g of the compound obtained are refluxed in 400 ml of ethanol containing 5 g of powdered sodium hydroxide. The reaction mixture is then cooled and the precipitated sodium salt of 1- (10, 11-dihydro-5H-dibenz- [b, f] azepin-3-ylsulfonyl) -3-butylurea is filtered off and dried in vacuo.
