Verfahren zur Herstellung neuer Thiosemicarbazone
Gegenstand der Erfindung ist die Herstellung von ss-Thiosemicarbazonen mit wertvollen pharmakologischen Eigenschaften. Insbesondere zeigen sie eine Wirkung gegen Viren. So sind z. B. bei Virus-Infek- tionen, wie bei der durch Vaccinia-Virus an der Maus hervorgerufenen Infektion, wirksam. Sie können dementsprechend als Heilmittel, vor allem bei Virus-Er krankungen, verwendet werden. Ein besonderer Vorteil der neuen Verbindungen liegt noch dlarin, da# sie Säuresalze bilden und als solche in wässrige Lösung gebracht werden können. Zudem besitzen sie eine ver gleichsweise geringe Toxizität.
Die Erfindung betrifft die Herstellung von Verbin dungen der Formel

Substituenten im Rest Ph sind vorzugsweise in 4, 6 und/oder 7-Stellungen, während z. B. Athylen oder einen geraden oder verzweigten Propylen-, Butylen-oder Pentylenrest, wie Propylen- (1, 3), Butylen- (l, 4), Pentylen-(1,5) oder einen Methyläthylen-, Methylenpropylen-, Dimethyläthylen-, Methylbutylen- oder Dimethylpropylenrest, vor allem aber Methylen, darstellt. B. die Pyrrolidino-, Piperidino-oder Morpholinogruppe, und eine Piperazino-z. B. die N-Methyl-oder Hydroxyäthyl-piper- azinogruppe darstellt.

oder ein Salz davon mit Dithiocarbaminsäure oder einem reaktionsfähigen Derivat davon, oder mit Rhodani- wasserstoffsäure oder einem Salz davon umsetzt.
Reaktionsfähige Derivate von Dithiocarbaminsäure sind deren Ester oder Amide ; Salze von Rhodanwasser- stoffsäure sind Ammonium-oder Alkalirhodanide.
Diese Umsetzung wird in üblicher Weise, gegebenenfalls unter Verwendung der Ausgangsstoffe in Form von Salzen, durchgeführt.
Die genannten Reaktionen werden in an sich bekannter Weise, in An-oder Abwesenheit von Ver diinnungs-und/oder Kondensationsmitteln, bei tiefer, gewöhnlicher oder erhöhter Temperatur, im offenen oder im geschlossenen Gefäss unter Druck durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Salze. Die Salze der Endstoffe können in an sich be kannter Weise, z. B. mit Alkalien oder Ionenaus tauschern, in die freien Basen übergeführt werden. Von den letzteren lassen sich durch Umsetzung mit organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen.
Als solche Säuren seien beispielsweise genannt : Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure ; aliphatische, alicyclische, aromatische oder heterocyclische Carbon'-oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Apfel-, Wein-, Zitronen-, Ascorbin-, Malin-, Hydroxy malein-oder Brenztraubensäure, Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthrani-, p-Hydroxybenzoe-, Salicyloder p-Aminosalicylsäure, Embonsäure, Methansuffon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure, Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehung zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Basen sinn-und zweckmässig gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungs- formen des Verfahrens, nach denen man von einer auf irgendeiner Stufe des Verfahrens als Zwischenprodukt erhältlichen Verbindung ausgeht und die fehlenden Ver fahrensschiitte durchführt, oder das Verfahren auf irgendeiner Stufe abbricht, oder bei denen man die Ausgangsstoffe unter den Reaktionsbedingungen bildet, oder bei denen die Reaktionskomponenten gegebenenfalls in Form ihrer Salze vorliegen.
Für die erfindungsgemässen Reaktionen werden vornehmlich solche Ausgangsstoffe verwendet, die die oben erwähnten bevorzugten Verbindungen ergeben.
Die Ausgangsstoffe sind bekannt oder. können nach an sich bekannten Methoden gewonnen werden.
So kann man N-unsubstituierte Isatine gemäss der Mannich-Reaktion mit Formaldehyd und Aminen der Formel H-Z umsetzen oder man kann sie in Form ihrer Alkalimetallsalze oder in Gegenwart solcher bilden- der Kondensationsmittel mit reaktionsfähigen Estem, wie Halogeniden von Aminoalkanolen oder mit einem Toluolsulfonsäureester eines Chloralkanols, umsetzen und das Halogenatom gegen die Grappe Z austauschen.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden.
Im folgenden Beispiel sind Temperaturen in Celsius- graden angegeben.
Beispiel
Eine Lösung von 13 g 1-Morpholinomethyl-isatin- 3-hydrazon in 130 ml trockenem Tetrahydrofuran wird mit 4 g Ammoniumthiocyanat behandelt ; das Reaktionsgemisch wird gerührt und tropfenweise mit 33 ml einer 2n Lösung von trockener Chlorwasserstoffsäure in Isopropanol behandelt. Nach 15minutigem Rühren bei Zimmertemperatur und 6stündigem Kochen am Rück flu# wird abgekühlt, der Niederschlag (Ammoniumchlorid) abfiltriert und das Filtrat auf dem Dampfbad unter vermindertem Druck eingedampft. Der Rückstand wird mit 30 ml Tetrahydrofuran trituriert, wobei sich ein festes Material abscheidet, welches abfiltriert und aus einem Gemisch von kaltem Dimethylformamid und Wasser umkristallisiert wird.
Das so erhaltene 1-Mor- pholinomethyl-isatin-3-thiosemicarbazon der Formel

schmilzt bei 215 bis 216 (mit Zersetzen). Sein Monohydrat schmilzt nach Umkristallisieren aus wässrigem Äthanol bei 206 bis 207 .
Das oben verwendete Ausgangsmaterial wird wie folgt hergestellt :
14, 7 g Isatin, 8, 7 g Morpholin und 9 ml einer 27% igen wässrigen Formaldehyd. losung werden mit 1150 ml Benzol vermischt und das Gemisch während 8 Stunden unter Rückfluss gekocht. Das 1-Morpholino- methyl-isatin kristallisiert nach dem Abkühlen aus und schmilzt nach Umkristallisieren aus einem Gemisch von Äthanol, Essigsäureäthylester und Methyläthylketon bei 203 .
Eine Lösung von 12, 3 g 1-Morpholinomethyl-isatin in 150 ml Chloroform wird tropfenweise mit 1, 76 g wasserfreiem Hydrazin in 10 ml absolutem Äthanol behandelt und während 3 Stunden unter Rückfluss er hitzt. Das Lösungsmittel wird unter vermindertem Druck auf dem Dampfbad abgedampft und das rohe 1-Mor- pholinomethyl-isatin-hydrazon ohne weitere Reinigung verwendet.
