Verfahren zur Herstellung von neuen Thiosemicarbazonen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Thiosemicarbazonen mit wertvollen pharmakologischen Eigenschaften. Insbesondere sind sie wirksam gegen Viren. Sie können z.B. zur Behandlung von Virus-Infektionen, wie Infektionen von Mäusen mit dem Vaccinia-Virus, verwendet werden. Zudem zeigen die Verbindungen der vorliegenden Erfindung Antitumor Eigenschaften, antibakterielle Effekte, z.B. gegen P. septica, M. tuberculosis oder Staph. aureus, oder zentralstimulierende Effekte sowie antihistaminische oder analgetische Eigenschaften. Sie können daher entsprechend verwendet werden, in erster Linie zur Behandlung von Virus-Krankheiten oder als Antitumor-Mittel sowie als zentralstimulierende, antibakterielle, antihistaminische oder analgetische Mittel.
Ein weiterer Vorteil der neuen Verbindungen ist deren Bildung von wasserlöslichen Salzen; zusätzlich zeigen sie auch eine relativ niedrige Toxizität.

Die neuen Verbindungen werden erfindungsgemäss erhalten, wenn man eine Verbindung der Formel 11

oder ein Salz davon mit Dithiocarbaminsäure oder einem Derivat davon oder mit Thiocyansäure oder einem Salz davon behandelt. Wenn erwünscht, kann ein erhaltenes Salz in die freie Verbindung oder eine erhaltene freie Verbindung in ein Salz umgewandelt werden.
Die obige Reaktion wird zweckmässig nach allgemein gebräuchlichen Methoden durchgeführt; wenn erwünscht, werden die Ausgangsstoffe in der Form von Salzen verwendet.
Eine Verbindung der Formel II mit einer unsubstituierten Hydrazonogruppe wird durch Behandeln mit Dithiocarbaminsäure oder einem reaktionsfähigen Derivat, wie einem Ester oder einem Amid davon, oder mit Thiocyansäure oder einem Salz, wie einem Ammonium- oder Alkalimetallsalz davon, in die erwünschte Thiosemicarbazonverbindung übergeführt.
Die oben erwähnte Reaktion wird in bekannter Weise durchgeführt, in Gegenwart oder Abwesenheit von Verdünnungsmitteln und/oder Kondensationsmitteln, bei Zimmertemperatur oder unter Kühlen oder Erhitzen, unter atmosphärischem oder erhöhtem Druck.
Je nach den Reaktionsbedingungen und der Wahl der Ausgangsmaterialien erhält man die Verfahrensprodukte in freier Form oder in Form ihrer Salze, welche ebenfalls von der vorliegenden Erfindung umfasst sind.
Die Salze der Verfahrensprodukte können in an sich bekannter Weise in die freien Verbindungen, z.B. durch Behandeln mit alkalischen Mitteln oder mit Anionenaustauschern, übergeführt werden. Werden die freien Verbindungen mit organischen oder anorganischen Säuren, besonders mit solchen, welche pharmazeutisch verwend bare Salze ergeben, behandelt, so erhält man ihre Salze.
Als Säuren seien zB. die Halogenwasserstoff-, Schwefeloder Phosphorsäuren, Salpeter- oder Perchlorsäure, sowie aliphatische, alicyclische, aromatische oder heterocyclische Carboxyl- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Malein-, Hydroxymalein-, Brenztrau- ben-, Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, SaIicyl-, p-Aminosalicyl-, Embon-, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfon-, Halogenobenzolsulfon-, Toluolsulfon-, Naphthalinsulfon- oder Sulfanilsäure, sowie Ascorbinsäure, Methionin, Tryptophan, Lysin.: oder Arginin, erwähnt.
Die oben genannten sowie andere Salze der neuen Verbindungen, zB. deren Pikrate,-können auch zur Reinigung der erhaltenen freien Verbindungen verwendet werden, zB. durch Überführen der letzteren in deren Salze, Abtrennen dieser Salze und Freisetzen der freien Verbindungen. Im Hinblick auf die enge Verwandtschaft zwischen den neuen Verbindungen in freier Form oder deren Salzen ist, was für die freien Verbindungen ausgesagt worden ist und wird, im jeweiligen Fall auch auf die entsprechenden Salze anzuwenden.
Die Erfindung umfasst auch Abänderungen des vorliegenden Verfahrens, nach welchen man die Ausgangsstoffe in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches umsetzt oder in Form von Salzen verwendet.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden hergestellt werden. So können z.B. l-unsubstituierte Isatine der Mannich-Reaktion mit Formaldehyd und einem Amin der Formel Z-H unterworfen werden oder können in Form ihrer Alkalimetallsalze oder in Gegenwart von Kondensationsmitteln, welche zur BiIdung von solchen Salzen geeignet sind, mit einem Toluolsulfonsäureester eines Chloralkohols umgesetzt und das Halogenatom durch die Gruppe Z ausgetauscht werden.
Die neuen Verbindungen können in der Form von pharmazeutischen Präparaten verwendet werden. bi den nachfolgenden Beispielen werden die Temperaturen in Celsiusgraden angegeben.
Beispiel
Eine Lösung von 8,5 g 1-(3-Methylpiperidinomethyl)- isatin-3-hydrazon in 120 ml absolutem Äthanol wird tropfenweise bei 100 mit 5 ml konzentrierter Salzsäure, gefolgt von 3,8 g Ammoniumthiocyanat, behandelt. Das Gemisch wird während 6 Stunden unter Rüclffluss gekocht, dann auf ein Volumen von ewa 50 ml konzentriert und filtriert. Das Filtrat verdünnt man mit 50 ml Isopropanol und filtriert erneut, konzentriert dann auf etwa 30 ml und lässt während 2 Tagen bei 0 stehen.
Der kristalline Niederschlag wird dann abfiltriert und aus einem Gemisch von Methanol und Isopropanol umkristallisiert; das so erhältliche 1-(3-Methylpiperidinome thyl)-isatin-3-thiosemicarbazon schmilzt bei 172- 1730.
Das Ausgangsmaterial kann wie folgt hergestellt werden: Eine Lösung von 7,47 g l-(3-Methylpiperidinomethyl)-isatin in 200 ml Äthanol und 10 ml Methylenchlorid wird auf 50 abgekühlt und tropfenweise mit 0,96 g igem Hydrazin in 50 ml Äthanol versetzt.
Man rührt das Reaktionsgemisch während 2 Stunden bei 100 und während 18 Stunden bei Zimmertemperatur und dampft dann zur Trockne ein. Der Rückstand wird mit 5 ml Isopropanol trituriert, abfiltriert und aus einem Gemisch von Chloroform und Isopropanol umkristallisiert; das so erhältliche l-(3-Methylpiperidinomethyl)- isatin-3-hydrazon schmilzt bei 2450.
Die folgenden Verbindungen können bei Auswahl der geeigneten Ausgangs stoffe in analoger Weise erhalten werden: 1- Dibenzylamino - methyl-isatin-3-thiosemicarbazon, F.
1600 nach Umkristallisieren aus Methanol; 1-(N-Methyl-N-benzyl-aminomethyl)-isatin-3 -thiosemicar bazon, F. 1500 als Monohydrat; 1 -Hexamethylenamino-methyl-isatin-3 -thiosemicarbazon,
F. 1550 nach Umkristallisieren aus absolutem Äthanol; l-Octahydroazocino-methyl-isatin-3-thiosemicarbazon, F.
1650 nach Umkristallisieren aus absolutem Äthanol.
Process for the production of new thiosemicarbazones
