EMI4.1
Process for the preparation of new phenothiazine derivatives
The present invention relates to a process for the preparation of phenothiazine derivatives of the formula I (see formula sheet), in which R, hydrogen, halogen, the trifluoromethyl, the cyano group or a lower alkanoyl, alkoxy or alkylthio group, R2 is hydrogen or the methyl group and R3 and R4 each represent a lower alkyl group or, together with the nitrogen atom, a pyrrolidine, piperidine, morpholine or N-methyl-piperazine ring, and their salts with inorganic or organic acids.
According to the invention, the compounds of the formula I are obtained by adding compounds of the formula II in which R 1 and R 2 have the above meanings and X represents chlorine, bromine, iodine or the residue of an organic sulfonic acid in the presence of an acid-binding agent in an inert under the reaction conditions Solvents with piperidine derivatives of the formula III, in which Rs and R4 have the above meanings, reacted and the compounds of the formula I obtained, if desired, converted into their acid addition salts.
The procedure is carried out as follows, for example:
A compound of the formula II is heated in an anhydrous organic solvent, preferably abs. Toluene or abs. Xylene, for a long time (10 to 50 hours) with a piperidine derivative of the formula III with stirring at normal pressure to boiling or possibly in a sealed tube to 130-180. The acid-binding agent used is e.g. B. an inorganic or tertiary organic base or a second mole of the compound III. The product obtained is isolated from the reaction mixture in a known manner and purified by chromatographic adsorption, crystallization or by conversion into a suitable salt.
The compounds prepared according to the invention have not yet been described in the literature.
They are basic substances and, with inorganic or organic acids, form stable salts crystallized at room temperature, which are also the subject of the present invention. Examples of such salts are the hydrochlorides, hydrobromides, sulfates or the malonates, succinates, fumarates, hydrogen fumarates, maleinates, tartrates, p-toluenesulfonates, methanesulfonates, etc.
The compounds of formula I are characterized by sedative and neuroleptic properties, such as, for. B. narcosis-potentiating effect, inhibition of conditioned and emotional reactions and motor activity, etc. In addition, the compounds have pronounced adrenolytic, also hypotensive or antihypertensive, bradycardic and analgesic properties.
The daily dose should be 10-500 mg. The new compounds can be used in internal medicine as sedatives, for the treatment of circulatory diseases, in particular of the hypertonic type, and for the treatment of vegetative dystonia; preferably in the form of their physiologically compatible water-soluble salts.
The compounds can be used as medicaments alone or in corresponding medicament forms for enteral or parenteral administration. For the purpose of producing suitable dosage forms, the active ingredients are processed with inorganic or organic, pharmacologically indifferent auxiliaries. One uses, for example, for tablets and dragees: lactose, starch, talc, stearic acid, etc.; For injection preparations: water, alcohols, glycerine, vegetable) oils and the like. In addition, the preparations can contain suitable preservatives, stabilizers and wetting agents, solubilizers, sweeteners and colorants, flavorings, etc.
The piperidine derivatives of the formula III required as starting materials are new. For their preparation, an acetamide of the formula IV, in which R3 and R4 have the meaning mentioned at the beginning, is used in the presence of an alkali amide, such as lithium amide, in a solvent which is inert under the reaction conditions, e.g. B. in liquid ammonia or abs. Ether, abs. Dioxane etc. or in mixtures of liquid ammonia with abs.
Ether, abs. Dioxane, etc., with 1-benzyl-4-piperidone and splits off the benzyl group hydrogenolytically in the condensation product obtained.
In practice, for example, a suspension of lithium amide in liquid ammonia is slowly mixed with an acetamide of the formula IV and, after stirring for about 30 minutes, 1-benzyl-4-piperidone is added dropwise. After about two hours of stirring, the reaction mixture is decomposed by adding solid ammonium chloride, the ammonia is allowed to evaporate, the residue is treated with water while cooling with ice and extracted with a water-immiscible solvent, e.g. B. benzene. The organic phases are combined, washed with water, dried and evaporated.
For hydrogenolytic cleavage of the benzyl group, the compound obtained as a residue is z. B. hydrogenated in glacial acetic acid over a palladium catalyst at 50 and about 5 atmospheres in a pressure vessel.
The compounds of the formula II are already known or easily accessible from the corresponding phenothiazines of the formula V, in which Ri has the meaning mentioned at the beginning, for example by reaction with 1, 3-dihalopropanes or 3-halo-1-propanols, which in 2-position can carry a methyl group in the presence of a strong base such as sodium amide in liquid ammonia, and subsequent treatment of the 3-hydroxypropyl-phenothiazines obtained in the latter case with thionyl chloride, phosphorus tribromide or p-toluene, methane, ethane sulfonyl chloride, etc. .
In the following examples, which explain the implementation of the process but are not intended to restrict the scope of the present invention in any way, all temperatures are given in degrees Celsius; the melting and boiling points are not corrected.
(Numbering of the phenothiazine structure according to the rules of IUPAC.)
Example 1 2-Chloro-10- [3-gy-4-dimethylcarbamoyl-methyl-1-piperidyl) propyl] phenothiazine
A mixture of 9.6 g of 2-chloro-10- (3-chloropropyl) phenothiazine and 11.6 g of 4-hydroxy-4-dimethylcarba- moylmethylpiperidine is heated to 170 in 40 ml of xylene in a sealed tube for 48 hours. The tube contents are rinsed out with 100 ml of benzene and washed neutral with water. It is dried over sodium sulfate and the solvent is evaporated.
The residue obtained is dissolved in 100 ml of acetone, filtered through activated charcoal and the filtrate is treated with a hot solution of the calculated amount of fumaric acid in ethanol. After standing in the refrigerator for a long time, the pure 2-chloro-10- [3- (4-hydroxy-4-dimethylcarbamoylmethyl-1-piperidyl) propyl] phenothiazine hydrogen fumarate crystallizes out; 197-200 (dec.).
The 4-hydroxy-4dimethylcarbamoylmethyl-piperidine required as starting material is z. B. prepared as follows: a) 1-Benzyl-4-hydroxy-4-dimethylcarbamoylmethyl piperidine
1.4 g of lithium are converted to lithium amide in 300 ml of liquid ammonia in the presence of iron (III) nitrate. 18.3 g of N, N-dimethylacetamide are then added dropwise to this lithium amide suspension.
The mixture is stirred for 30 minutes at the temperature of the liquid ammonia and 18.9 g of 1-benzyl-4-piperidone are slowly added dropwise. After stirring for two hours, 10 g of ammonium chloride are carefully added, the ammonia is evaporated and the residue is treated with 100 ml of water while cooling with ice.
It is extracted several times with benzene, the combined extracts are washed twice with water, dried over sodium sulfate and the benzene is evaporated. The residue is distilled under greatly reduced pressure, the pure 1-benzyl-4-hydroxy-4-dimethylcarbamoylmethyl-piperidine passing over at 164¯167 / 10-4 mm Hg. b) 4-Hydroxy-4-dimethylcarbamoyltnetlayl-piperidine
23, 0 g of 1-benzyl-4-hydroxy-4-dimethylcarbamoylmethyl-piperidine are dissolved in 170 ml of glacial acetic acid and in the presence of 4.5 g of a palladium catalyst (10 / o on carbon) at an initial pressure of 5 atm and a temperature of 50 hydrogenated. The amount of hydrogen calculated for the debenzylation is absorbed after 2 hours.
