Verfahren zur Herstellung neuer Polypeptide Die vorliegende Erfindung betrifft Verfahren zur Herstellung neuer Pentacosapeptide der allgemeinen Formel I D-Seryl-L-tyrosyl-L-seryl-L-methionyl-X-L-histidyl-Lphenylalanyl-L-arginyl-L-tryptophanyl-glycyl-L-lysyl-L- prolyl-L-valyl-glycyl-L-lysyl-L-lysyl-L-arginyl-L-arginyl- L-prolyl-L-valyl-L- lysyl-L-valyl-L- tyrosyl - L-prolyl - L- valinamid, worin X einen L-Glutamyl- oder L-Glut- aminylrest bedeutet, dadurch gekennzeichnet, dass man entsprechende,
zu ihrem Aufbau nötige Aminosäuren unter Bildung von CONH-Bindungen in beliebiger zeitlicher Reihenfolge miteinander kondensiert, wobei man nicht an der Reaktion teilnehmende Gruppen durch Einführung von zum Schutz solcher Gruppen geeigneten Schutzgruppen schützt, die nach erfolgter Kondensation abgespalten werden, die Carboxylgruppe des endständigen Valinrestes zu Beginn oder in einem beliebigen Zeitpunkt der Synthese in die Amidgruppe überführt und gegebenenfalls die Säureadditionssalze herstellt, indem man das Pentacosapeptid durch Umsetzung mit organischen oder anorganischen Säuren in die entsprechenden Salze überführt.
Das so erhaltene Pentacosapeptid kann zur Herstellung von Schwermetallkomplexen verwendet werden, indem man das Pentacosapeptid mit entsprechenden Schwermetallsalzen umsetzt.
Diese neuen Pentacosapeptide der allgemeinen Formel I und ihre Säureadditionssalze besitzen eine hohe adrenocorticotrope Wirksamkeit. Die unter die allgemeine Formel 1 fallenden Pentacosapeptide können in abgekürzter Schreibweise auch wie folgt bezeichnet werden: D-Serl-Val-NH225-Pentacosapeptid und D-Serl-Gln5- Val-NH2a5_pentacosapeptid.
Es war bereits aus dem belgischen Patent Nr. 636 667 bekannt, dass man ein Pentacosapeptid der Sequenz L-Seryl-L-tyrosyl-L-seryl-L-methionyl-L-glutamyl-Lhistidyl-L-phenylalanyl-L-arginyl-L-tryptophanyl-glycylL-lysyl-L-prolyl-L-valyl-glycyl-L-lysyl-L-lysyl-L-argi- nyl-L-arginyl-L-prolyl-L-valyl-L-lysyl-L-valyl-L-tyrosyl- L-prolyl-L-valinamid mit corticotroper Wirkung, im nachstehenden als Pentacosapeptid bezeichnet, synthetisieren kann.
Das Pentacosapeptid besitzt gegenüber dem natürlichen ACTH den Vorteil, dass es keine antigenen Eigenschaften aufweist. Ein weiterer Vorteil des Pentacosa- peptids ist, dass es in Stellung 25 einen Valinamidrest aufweist, der beim natürlichen ACTH an dieser Stelle nicht vorkommt. Der Valinamidrest an Carboxylende schützt die Peptidkette gegen abbauende Enzyme.
Gleich dem natürlichen ACTH weist jedoch das Pentacosapeptid in Stellung 1 einen L-Serinrest auf, der bekanntlich gegenüber der abbauenden Wirkung von Aminopeptidasen sehr empfindlich ist.
Es wurde aus diesem Grunde versucht, den L-Serinrest des Pentacosapeptids durch einen Rest zu ersetzen, der gegenüber der abbauenden Wirkung der Aminopeptidasen stabil ist. Tatsächlich gelang es, durch den Ersatz des L-Serinrestes mit einem D-Serinrest beim Pentacosapeptid, das gegenüber Aminopeptidasen unempfindliche D-Serl- Pentacosapeptid zu erhalten.
Da aber bekanntlich D- Aminosäurereste in den natürlichen, biologisch aktiven Peptidhormonen nicht vorhanden sind, war es überraschend und nicht vorauszusehen, dass durch den Ersatz eines natürlichen Aminosäurerestes durch seinen in der Natur nicht vorkommenden Antipoden eine Verbindung erhalten wird, die dieselben biologischen und therapeutischen Eigenschaften wie das natürliche ACTH aufweist.
Überraschenderweise wurde nun auch festgestellt, dass beim D-Serl-Val-NH22s_pentacosapeptid der Glutaminsäurerest in Stellung 5 durch einen Glutaminrest ohne Verlust der biologischen Wirkung ersetzt werden kann. D-Serl-Val-NH225-pentacosapeptid und D-Serl-Gln5-Val- NH22s-pentacosapeptid weisen die oben beschriebenen Vorteile des Pentacosapeptids gegenüber dem natürlichen ACTH auf.
Hinzu kommt die Beständigkeit der beiden Pentacosapeptide gegenüber den abbauenden Amino- peptidasen.

Die neuen Pentacosapeptide der allgemeinen Formel I können nach für die Synthese von Verbindungen dieser Art allgemein bekannten Methoden hergestellt werden, wobei die Aminosäuren in der, in der obigen Formel festgelegten Reihenfolge einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden.
Erfindungsgemäss können die neuen Pentacosapeptide der Formel 1 beispielsweise hergestellt werden, indem man L-Valyl- e -N-R-L-lysyl-L-valyl-L-tyrosyl -L-prolyl- Lvalinamid, worin R eine Carbo-tert.-butoxy- oder eine Carbo-tert:
amyloxy-, eine Toluolsulfonyl-, eine Phthalyl-, eine Formyl- oder eine Trifluoracetyl-Gruppe bedeutet, mit dem N-Carbobenzoxy-L-valyl-glycyl-e-N-R-L-lysyl- s-N-R-L-lysyl-nitro-L-arginyl-nitro-L-arginyl-L-prolin, worin R obige Bedeutung hat, kondensiert, das erhaltene N-Carbobenzoxy-L-valyl-glycyl-e-N-R-L-lysyl-e-N-R-L- lysyl-nitro-L-arginyl-nitro-L-arginyl-L-prolyl-L-valyl-e- N-R-L-lysyl-L-valyl-L-tyrosyl-L-prolyl-L-valinamid, worin R obige Bedeutung hat, nach Abspaltung der Carbobenzoxygruppe und der Nitrogruppen mit dem N-Triphenylmethyl-L-glutaminyl (oder y-O-tert.-butyl-L- glutamyl)
-Im-triphenylmethyl-L-histidyl-L-phenylalanyl- L-arginyl-L-tryptophanyl-glycyl-e-N-R-L-lysyl-L-prolin, worin R obige Bedeutung hat, kondensiert, das erhaltene N-Triphenylmethyl-L-glutaminyl (oder y-O-tert.-butyl-Lglutamyl)-Im-triphenyhnethyl-L-histidyl-L-phenylalanylL-arginyl-L-tryptophanyl-glycyl-e-N-R-L-lysyl-L-prolylL-valyl-glycyl-e-N-R-L-lysyl-L-arginyl-L-arginyl-L-pro- lyl-L-valyl-e-N-R-L-lysyl-L-valyl-L-tyrosyl-L-prolyl-L- valinamid, worin R obige Bedeutung hat, nach Abspaltung der N-Triphenylmethyl-Gruppe mit dem N-R'-D- Seryl-L-tyrosyl-L-seryl-L-methionylazid, worin R' eine Triphenylmethyl-, eine Carbo-tert.-butoxy- oder eine Carbo-tert.-amyloxy-,
eine Carbobenzoxy-, eine Trifluor- acetyl-, eine Acetyl-, eine Chloracetyl- oder eine Formyl- gruppe bedeutet, kondensiert und die gesamten Schutzgruppen der erhaltenen neuen, geschützten Pentacosa- peptide N-R'-D-Seryl-L-tyrosyl-L-seryl-L-methionyl-L- glutaminyl (oder y-O-tert.-butyl-L-glutamyl)-Im-triphenylmethyl-L-histidyl-L-phenylalanyl-L-arginyl-L-tryptophanyl-glycyl-e-N-R-L-lysyl-L-prolyl-L-valyl-glycyl-e-NR-L-lysyl-e-N-R-L-lysyl-L-arginyl-L-arginyl-L-prolyl-L- valyl-e-N-R-L-lysyl-L-valyl-L-tyrosyl-L-prolyl-L-valin- amid, worin R und R' obige Bedeutung haben,
in einer oder in mehreren Stufen im sauren Milieu abspaltet.
Die Ausgangsprodukte zur Herstellung der neuen Pentacosapeptide der allgemeinen Formel I können, sofern sie bisher noch nicht bekannt waren, nach den für die Peptidchemie bekannten Methoden erhalten werden, wobei die Aminosäuren einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden.
Die neuen Pentacosapeptide der allgemeinen Formel I lassen sich auch in Form ihrer Salze gewinnen bzw. verwenden. Als Salze kommen solche mit organischen Säuren, wie Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure, Weinsäure, Zitronensäure, Benzoesäure, Zimtsäure, Salicylsäure, 2-Phenoxy- oder 2-Acetoxy-benzoesäure, Mandelsäure, Methansulfon- säure, Äthansulfonsäure, Hydroxyäthansulfonsäure,
Benzol- oder Toluolsulfonsäure, Naphthahnsulfonsäure, Sulf- anilsäure sowie polymere Säuren wie Gerbsäure, Algin- säure, Polygalacturonsäure, Polyphloretinphosphat oder Carboxymethylcellulose und Salze mit anorganischen Säuren, wie Halogenwasserstoffsäure, z. B. Salzsäure oder Bromwasserstoffsäure, Salpetersäure, Thiocyansäure, Schwefelsäure und Phosphorsäure in Frage. Als Schwermetallkomplex kommt z. B. derjenige des Zinks in Frage.
Die neuen Pentacosapeptide der allgemeinen Formel I, ihre Salze und Schwermetallkomplexe können beispielsweise für die Behandlung der folgenden Erkrankungen verwendet werden: Akuter und chronischer Gelenkrheumatismus, Colitis ulcerosa, Nephrose, Kollagenosen, wie z. B. Lupus erythematodes, Skleroder- mie usw., Allergische Erkrankungen der verschiedenen Organsysteme, wie Asthma bronchiale, Ekzem, Urticaria, Dermatitis exfoliativa, anaphylaktischer Schock usw., Intoxikationen verschiedener Genese, wie z. B. bei chronischem Alkoholismus, Tumoren, wie z. B.
Leukämien, Lymphosarcomen, Reti- culosarcomen usw., zur Verhinderung der Nebennierenrindenatrophie bei der Therapie mit Corticosteroiden, Insuffizienzerscheinungen der Hypophyse.
Ein wesentlicher Vorteil gegenüber natürlichem, aus tierischem Material extrahierten Hormon liegt darin, dass den synthetischen Pentacosapeptiden antigene Eigenschaften fehlen. Es kann somit bei den oben erwähnten Erkrankungen auch dann unbedenklich verwendet werden, wenn beim Patienten im Verlauf einer früheren Behandlung mit natürlichem ACTH allergische Erscheinungen auftraten.
In den für die Standardisierung üblichen und anerkannten Tests besitzen die neuen Pentacosapeptide der allgemeinen Formel I im Vergleich zum natürlichen Corticotropin eine sehr hohe Aktivität. So dienten zur Testung des verfahrensgemäss hergestellten Pentacosa- peptids der 3. Internationale Standard für Corticotropin, welcher in Form des International Standard for Corti- cotropin zur Verfügung steht und die Einstellung von ACTH-Präparaten auf Internationale Einheiten ermöglicht.
Die Dosierung der verfahrensgemäss hergestellten Pentacosapeptide entspricht derjenigen vom ACTH, d.h. sie variiert ungefähr zwischen 40 und 60 IE pro Tag, in besonderen Fällen zwischen 10 und 100 IE pro Tag.
Die neuen Pentacosapeptide der allgemeinen Formel I können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die genannten Verbindungen in Mischung mit einem für die parenterale Applikation geeigneten organischen oder anorganischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi arabicum, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.
B. in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe wie Konservierungs-, Stabilisierungs-, Netzmittel oder Emulgiermittel. Sie können auch noch andere, therapeutisch wertvolle Stoffe enthalten. Die neuen Verbindungen können wie natürliches ACTH auch in Form eines Depotpräparates verabreicht werden.
Die neuen Pentacosapeptide der allgemeinen Formel I und ihre Salze können auch als Zwischenprodukt zur Herstellung von pharmazeutischen Präparaten Verwendung finden.

Beim Aufbau der neuen Pentacosapeptide haben sich für die Blockierung der Aminogruppe des Serinrestes die Triphenylmethyl-, die Carbo-tert:
butoxy- und die Carbo- tert.-amyloxy-Gruppe bewährt, doch können auch andere geeignete Schutzgruppen wie die Carbobenzoxy-, die Trifluoracetyl-, die Acetyl-, die Chloracetyl-, die Formyl- gruppe verwendet werden.
Für die Blockierung der s-Aminogruppe des Lysin- restes hat sich die Carbo-tert.-butoxy- und die Carbo- tert: amyloxy-Gruppe bewährt, doch können auch andere geeignete Schutzgruppen wie die Carbobenzoxy-, die Toluolsulfonyl-, die Phthalyl-, die Formyl- und die Tri- fluor-acetyl-Gruppe verwendet werden.
Für die Blockierung der y-Carboxyl-Gruppe des Glutaminsäurerestes hat sich die tert.-Butyloxy-Gruppe bewährt, doch können auch andere geeignete Schutzgruppen, wie die Methoxy, die Äthoxy, die tert: Amyloxy-, die Amid- oder die Benzyloxygruppe verwendet werden.
Für die Blockierung der Imidazolgruppe des Histidin- restes hat sich die Triphenylmethylgruppe bewährt, doch können auch andere geeignete Schutzgruppen, wie die Carbo-tert.-butoxy-, die Carbo-tert: amyloxy-, die Carbo- benzoxy- oder die Benzylgruppe verwendet werden.
Für die Blockierung der Guanidogruppe der Arginin- reste wurde die Nitro-Gruppe verwendet, doch können auch andere geeignete Schutzgruppen, wie die Tosyl- gruppe, die p-Nitrocarbobenzoxygruppe oder die 2-(Iso- propyloxycarbonyl)-3,4,5,6-tetrachlorobenzoylgruppe verwendet werden. Man kann auch den Schutzeffekt der Protonisierung der Guanidogruppe bei der Synthese verwenden.

Man löst 88 g CBO-(N02)Arg-(N02)Arg-Pro-OMe in einem 90%igen Dioxan-Wasser Gemisch und versetzt mit 220 cm3 2n Natronlauge. Nach 2 Std. wird mit 1 1 Wasser verdünnt und wiederholt mit Essigester ausgewaschen. Anschliessend säuert man die wässerige Lösung mit 4n Salzsäure an, löst das ausgefallene Produkt in einem Gemisch von Methanol-Azeton 1:
1 auf und fällt durch Zugabe von Äthyläther. Man erhält 70 g CBO- (N02)Arg-(N02)Arg-Pro-OH vom Smp. 108 (mit Zersetzung).

Man lässt die Lösung 16 Stunden stehen und dampft das Lösungsmittel ein. Der Rückstand wird in einem Gemisch von n-Butanol-Essig- ester 2: 8 gelöst und wiederholt mit verdünnter Schwefelsäure nachgewaschen.
Man konzentriert die Lösung im Vakuum ein und fällt mit Äther aus. Man erhält 90 g CBO-Val-Gly-(CTB)Lys-(CTB)Lys-(N02)Arg-(N02)Arg- Pro-OH vom Smp. 151 (mit Zersetzung).

Man löst 56 g CBO-Val-Gly-(CTB)Lys-(CTB)Lys- (N02)Arg-(N02)Arg-Pro-OH in 900 ml Dimethylform- amid und 900 ml Tetrahydrofuran. Nach Zugabe von 6,2 ml Triäthylamin wird die Lösung auf -10 C abgekühlt und bei dieser Temperatur mit 4,2 ml Chlorameisensäure- äthylester versetzt. Nach 10 Min. wurden 36 g H-Val- (CTB)Lys-Val-Tyr-Pro-Val-NH2 in 160 nil Dimethyl- formamid zugegeben und über 16 Stunden bei 20 weiter gerührt. Das Lösungsmittel wurde im Vakuum verdampft und der Rückstand mit Wasser ausgewaschen.
Man löst das Peptid in heissem Äthanol und fällt mit Essigester aus. Nach Abnutschen und Trocknen erhält man 72 g CBO- Val-Gly-(CTB) Lys-(CTB) Lys-(N02)Arg-(N02)Arg-Pro- Val-(CTB)Lys-Val-Tyr-Pro-Val-NH2 vom Smp 190 (mit Zersetzung).

Nach 24stündigem Schütteln bei 20 wird der Harnstoff abfiltriert und die Lösung mit Äther gefällt. Das Produkt wird wiederholt in Methanol gelöst und mit Essigester gefällt.
Man erhält 90 g Trit-(CTB)Glu-(Trit)His-Phe-Arg- Try-Gly-(CTB)Lys-Pro-Val-Gly-(CTB)Lys-Arg-Arg-Pro- Val-(CTB)Lys-Val-Tyr-Pro-Val-NH, Tritoluolsulfonat. Smp. 184 (mit Zersetzung).

Man löst das oben erhaltene Tripeptid in 300 ml Methanol und 33 ml 2 N Salzsäure, hydriert in Gegenwart von 5 g 10 % Palladium-Kohle, filtriert, dampft ab, löst den Rückstand in 150 ml Methylenchlorid, kühlt auf 0 , gibt 21 ml Triäthylamin und anschliessend 27 g Triphenylchlormethan zu, lässt 16 Std. bei 20 stehen, wäscht mit verdünnter Essigsäure, Wasser und 1 N Na- triumbicarbonatlösung, trocknet und dampft ab. Man löst den Rückstand in Diäthyläther und fällt mit Petrol- äther. Man erhält 49 g Trit-Gln-(Trit)His-Phe-OMe, das man in 100 ml Methanol löst.
