Verfahren zur Herstellung protrahiert wirksamer Injektionspräparate Die vorliegende Erfindung betrifft Verfahren zur Herstellung stabiler, protrahiert wirksamer Injektionspräparate des Pentacos@apeptids L-Seryl-L-tyrosyl-L-seryl-L-norleucyl-L-glutamyl- L-h.istidyl-L-phenylalanyl'-L-arginyl-L tryptophanyl-glycyl-L-lysyl-L-prolyl-L-valyl-glycylL-lysyl-L-lysyl-L-lysyl-L-prolyl-L-valyl- L-lysyl-L-valyl-L-tyrosyl-L-prolyl- L-valinamid, in Form einer wässrigen Suspension -eines Zinkkomplexes davon, dadurch gekennzeichnet,
dass man die wässrige Lösung eines Salzes des Pentacosapeptids .obiger Formel mit einem :saure Gruppen enthaltenden Polysaccharid und eines physiologisch verträglichen wasserlöslichen Zinksalzes durch Zugabe eines AIkalimetal@lhydroxyds oder eines geeigneten Puffers auf einen pH-Wert von 7,2 bis 8,5 bringt.
Das Pentacosapeptid L-Seryl-L-tyrosyl-L-seryl-L-norleucyl-L-eutamyl- L-histidyl L-phenylalanyl'-L-arginyl-L-tryptophanyl-glycyl-L-lysyl-L-prolyl-L-valyl-glycyl- L-lysyl-Ir1ysyl-L-lysyl-Irprolyl-L-valyl- L-lysyl-L-valyl-L tyrosyl-L-prolyl- L-valinamid, hat strukturelle Ähnlichkeit mit der Aminosäunesequenz 1 bis. 25 des ACTH .und besitzt auch dieselben biologischen und therapeutischen Eigenschaften wie das natürliche ACTH (Corticotropin).
Nach. dem angegebenen Verfahren erhält man eine äusserst stabile Suspension, die nach anschliessender Sterilisierung direkt injizierbar ist. Gegebenenfalls kann das. pH der Suspension durch Zusatz eines geeigneten Puffers, z. B. .eines Phesphatpuffers, stabilisiert werden. Durch Zusatz von geeigneten Stoffen (z. B. Natriumchlorid, Glucose, Glycerin) kann das Präparat isotonisch gemacht werden. Auch Substanzen, die das Wachstum von Mikroorganismen hemmen, wie z.
B. Benzylalkohol, Phenol, Chlorbutanol, können .noch zugesetzt werden. Als saure Gruppen enthaltende Polysaccharide können natürliche Polyuronsäuren (wie z. B. Polygalacturon- säuren, Polym,annuronsäure-n), saure Mucopolysaccha- ride (wie z.
B. Heparin, Hyaluronsäure, Chondroitin- sulfat) sowie synthetische oder mikrobiologisch ge- wonnene saure Gruppen enthaltende Derivate der Cellu- lose, der Stärke oder anderer Polysaecharid@e (wie z. B. Carboxymethyleellulose oder Dextransulfat) verwendet werden.. Als physiologisch wirksame Zinksalze können Zinkacetat, Zinkchlorid, Zinknitrat und Zinksulfat verwendet werden.
Das Verhältnis zwischen Paly- peptidsalz und Zinksalz soll vorzugsweise zwischen 10 und 200 mMol Zinksalz pro 100 I. E. Peptidsalz liegen.
Das Pentacosapeptid obiger Formel kann nach für die Synthese von Polypeptiden allgemein bekannten Methoden hergestellt werden, wobei die Aminosäu:ren in der in der obigen Formel festgelegten Reihenfolge allein oder nach vorheriger-Bildung kleinerer Peptideinheiten miteinander verknüpft werden. Eine Synthesemöglichkeit des synthetischen Peptids ist am Ende des Beispiels 1 näher beschrieben.
Es ist bekannt, dass das natürliche ACTH sowie verschiedene seiner biolögisch aktiven Teilsequenzen im Organismus sehr schnell inaktiviert werden. Es wurde daher empfohlen, die Wirkung von Injektionslösungen von Cor .ticotropsn durch Kombination mit Verbindungen gewisser Schwermetalle oder durch Kombination mit hochmolekularen Stoffen zu verbessern und insbesondere über einen längeren Zeitraum zu erstrecken, doch war der dadurch erreichte Effekt relativ klein.
Das erfindungsgemäss verwendete Polypeptid ist zwar bereits wesentlich stabiler als das natürliche ACTH oder dessen Bruchstücke, wird im Organismus jedoch noch immer relativ rasch abgebaut. Es wurde nun überraschenderweise gefunden, dass das Polypeptid obiger Formel in Form eines Zinkkomplexes eines seiner Salze mit saure Gruppen enthaltenden Polysacchariden bei einem pH von 7,2 bis 8,5 in Form einer wässrigen Suspension, sehr beständig ist.

sion besitzt bei parenteraler Applikation am Tier und am Menschen eine wesentlich stärkere protrahierte ACTH- Wirksamkeit als die bereits bisher bekannten ACTH- Depotpräparate.
So wird durch die erfindungsgemäss hergestellten Präparate bei i. m. oder s. c. Injektion die Sekretion der Corticosteroide während bedeutend mehr als 24 Stunden stimuliert. Dies bedeutet für die Therapie einen grösseren Vorteil, der bisher nicht erreicht werden konnte. Die so erhaltene Suspension ist sehr hitzestabil, was bezüglich Sterilisationsmöglichkeit einen bedeutenden Vorteil darsteht.
Die verfahrensgemäss hergestellten neuen Präparate können beispielsweise für die Behandlung der folgenden Erkrankungen verwendet werden: Akuter und chronischer Gelenkrheumatismus, Colitis ulcerosa, Nephrose, Kollagenosen, wie z. B. Lupus erythematodes, Sklerodermie usw., allergische Erkrankungen der verschiedenen Organsysteme, wie Asthma bronchiale, Ekzem, Urticaria, Dermatitis exfoliativa, anaphylaktischer Schock usw., Intoxycationen verschiedener Genese, wie z.
B. bei chronischem Alkoholismus, Tumoren, wie z. B. Leukämien, Lymphosarcomen, Reti- culosarcomen usw., sowie zur Verhinderung der Nebennierenrindenatrophie bei der Therapie mit Corticos.tero- iden, Insuffizienzerscheinungen der Hypophyse.
Ein wesentlicher Vorteil gegenüber natürlichem, aus tierischem Material extrahiertem Hormon liegt für die Therapie beim Menschen auch darin, dass dem neuen Peutacosapeptidkomplex antigene Eigenschaften fehlen. Es kann somit bei den oben erwähnten Erkrankungen auch dann unbedenklich verwendet werden, wenn beim Patient im Verlauf einer früheren Behandlung mit natürlichem ACHT allergische Erscheinungen auftraten.
In den für die Standardisierung üblichen und anerkannten Tests besitzt -der Pentacosapeptidkomplex im Vergleich zum natürlichen Corticotropin eine sehr hohe Aktivität.
Es werden folgende Abkürzungen verwendet: CBO = Carbobenzoxy Tri = Trityl = Triphenylmethyl CTB = Carbo-tert.-butyloxy OCP = 2,4,
5-Trichlorphenoxy OTB = tert.-Butyloxy OMe = Methoxy OEt = Äthoxy Arg = L-arginyl Glu = L-glutamyl Gly = glycyl His = L-histidyl Lys = Lrfysyl Nle = L-norleucyl Phe = L-phenylalanyl Pro = L-prolyl Ser = L-seryl Try = Lrtryptophanyl Tyr = L-tyrosyl Val = L-valyl In
den folgenden Beispielen, die die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise cinschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 Man löst 6 g des Salzes des Pentacosapeptids L-Seryl-L-tyrosyl-L-seryl L-norleucyl L-glutamyl- L-histidyl L-phenylalanyl'-L-arginyl-L-trypto- phanyl-glycyl-L-lysyl-L-prolyl-L-valyl-glycyl- L-lysyl-L-lysyl-L=lysyl-L-lysyl-L-prolyl-L valyl- L-lysyl-L-valyl-L-tyrosyl-L-prolyl- L-valinamid mit Ca:
rboxymethylcellulose (33 USP E/mg) und 12,6 g Zinkchlorid in 700 cm3 Wasser und tropft die so erhaltene Lösung in eine andre Lösung von 4,6g Natriumchlorid und 18 g Benzylalkohol in 1000 cm3 Wasser, während man das pH des Gemischs durch Zugabe von 1n Natriumhydroxyd während der ganzen Operation bei 8,0 aufhält. Man rührt noch während 10 Minuten die erhaltene Suspension, gibt 3,9g NaH2PO4H20 zu, stellt das pH auf 7,8 ein und füllt zu 2000 cm3 mit Wasser auf.
Man erhält somit eine Suspension, deren Eigenschaften durch Sterilisation auf 120 nicht verändert werden und die folgende Zusammensetzung pro ml besitzt: Pentacosapeptid 100 USP E Carboxymethylcellulose 2,3 mg Zn++ 3 mg Benzylalkohol 9 mg NaCl 8,5 mg P04--- 1,4 mg Das Salz des Pentacosapeptids mit Carboxymethyl- cellulose kann wie folgt hergestellt werden:
a) a-N-Carbobenzoxy-e-N-carbo-teri.-butoxy- L-lysin-2,4,5-trich@orphcnylester (CBO-(CTB)Lys-OCP) Man löst 104 g a-N-Carbobenzoxy-e-N-carbo-ter:t.- butoxy-L-lysin und 70 g 2.,4,5-Trichlorphenof in 900 m1 Chloroform und 90 ml Acetonitril und versetzt bei -10 mit 56g Dicyclohexylcarbodiimid. Man schüttelt während 2 Stunden und filtriert vom Harnstoff ab. Nach Verdampfen des Lösungsmittels wird der Rückstand in Äthanol gelöst und mit Petroläther gefällt.
Nach Umkri stallis:ieren aus Äthanol-Wasser erhält man 140 g a-N Carbobenzoxy-a,a-N-carbo-tert.-butoxy- L-lysin-2,4,5-trichlorphenylester. trich#lorphenylester.
Smp. 99 [,]D = -11 in Dimethylformamid.
b) L-Valyl-a-N-carbo-tert.-butoxy-L-lysyl-L-valyl- L-tyrosyl-L-prolyl-L-valinamid (H-Val-(CTB)Lys- Vaf-Tyr-Pro-Val-NH2) Man löst 50 g L-Propyl-L-valinamid (H-Pro-Val- NH2) und 100 g CBO-(CBO)Tyr-OCP in 100 ml Di- methylformamid, lässt 48 Stunden bei 20 stehen und filtriert. Die Lösung wird mit 500 ml 1n Salzsäure versetzt und der ölige Rückstand in Essigester gelöst. Nach dem Waschen mit verdünnter Salzsäure und: Natrium- bicarbonatlösung wird zur Trockne eingedampft.
Nach Umkristallisierein aus Essigester-Dfäthyläther erhält man 100g CBO-(CBO)Tyr-P-ro-Val-NH2 (Smp. 133 ). Dieses wird mit 90 ml 2n Natronlauge und 200 ml Äthanol versetzt. Man lässt eine Stunde hei Zimmertemperatur stehen und gibt 1000 ml 1n Salzsäure zu. Der Rückstand, wird zuerst mit Wasser und nach Trocknen mit Essigester gewaschen.
Man erhält 60g CBO-Tyr-Pro-

Val-NH2 Smp. 220 . [a]D = -23 in Dimethylform- amid. Das erhaltene Produkt wird in 200 ml einer 4n Bromwasserstofflösung in Eisessig gelöst und eine Stunde bei 20 stehengalassen. Nach Zugabe von Diäthylä'ther- E$sigester fällt das H-Tyr-Pro Val-NH2 als Hydro- bromid aus. Sm#p. 220 .
Man löst 42 g H Tyr-Pro-Val- NH2 - HBr in 200 ml Dimethylformamid und gibt 36 g N-Carbobenzoxy-L-Valin-2,4,5-trichlorphenylesber (CBO-Val-OCP) und 12 ml Triäthyl-amin zu. Man schüttelt 48 Stunden bei Zimmertemperatur und versetzt mit 500 ml In Salzsäure. Das ausgeschiedene Produkt wird in Essigestzr gelöst und mit verdünnter Salzsäure und Natriumbicarbonatlös.ung gewaschen. Die Essigesterlösung wird mit Äther gefällt. Man erhält 39 g CBO-Val-Tyr-Pro-Val-NH2. Smp. 127 [a] D = -26 in Dimethylformamid.
Das erhaltene Produkt wird in 200 ml einer 4n Lösung von Bromwasserstoff in Eisessig gelöst und eine Stunde bei 20 stehengelassen. Nach Zugabe von Diäthyläther-Essigester fällt 32g H-Val-Tyr-Pro-Val- NH2 - HBr aus.
29 g H-Val Tyr-Pro-Val-NH2 - HBr und 33g CBO- (CTB)Lys-OCP (Beispiel 1) werden in 100 ml Dimethyl- formamid gelöst und mit 7,5 ml Triäthylamin versetzt. Man schüttelt 48 Stunden bei 20 , versetzt mit 500 ml In Essigsäure und wäscht das ausgefallene Produkt mit Wasser und anschliessend mit Essigester. Man erhält 35g CBO-(CTB)Lys-Val-Tyr-Pro-Val-NH2. Smp. l25 (Zers.) [a]21 = -25 in Dimethylformamid.
Man löst 39 g CBO(CTB)Lys-Val Tyr-Pro-Val- NH2 in 500 ml Methanol und hydriert bei Normaldruck in Gegenwart eines Palfadiumkatalysators. Man filtriert, verdampft im Vakuum und behandelt den Rückstand mit Äther. Manerhält auf diese Weise 28 g H-(CTB)Lys- Val-Tyr-Pro-Val-NH2. Smp. 145 (Zers). [a]" = -30 in, Dimethylformamid.
Man löst 33 g H-(CTB)Lys-Val-Tyr-Pro-Val-NH2 und 23g CBO-Val-OCP in 150 ml Dimethylformamid und lässt 48 Stunden bei 20 stehen. Die Lösung wird im Vakuum eingedampft und der Rückstand in wenig n-Butanol und Essigester gelöst.
Nach dem Waschen mit verdünnter Schwefelsäure und Natriumbicarbonat- lösung wird zur Trockne eingedampft. Nach Behandlung mit Diäthyläther erhält man 33 g N-CBO Val- = (CTB)Lys-Val-Tyr-Pro-Val-NH2. Smp. 171 [a]" -27 in Dimethylformamid.
Das erhaltene Produkt wird in 500 ml Methanol gelöst und bei Normaldruck in Gegenwart eines Pall'a- diumkatalysators hydriert. Man filtriert, verdampft im Vakuum und trocknet bei 40 . Man erhält 22g H-Val- (CTB)Lys-Val-Tyr-Pro-Val-NH2. [a]D = -26 in Di- methylformamid.
c) L-Valyl-glycyl-s-N-carbo-tert.-butoxy-L-lysyl- N-carbo-tert.-butoxy-L-lysyl-e-N-carbo-tert; butoxy-L-lysyl-e-N-carbo-tert.-butoxy-L 1ysy1- L-prolyl-L-valyl-e-N-carbo-tert.-butoxy-L-lysyl- L-valyl-L-tyrosyl-I-prolyl-L-valinamid (H-Val-Gly-(CTB)Lys-(CTB)Lys-(CTB)Lys- (CTB)Lys-Pro.-Val-(CTB)Lys-Val-Tyr-Pro-Val-NHs) Man löst 470 g CBO-(CTB)Lys-OH und 182 ml Triäthylamin in 5 Liter Essigester, tropft bei -5 125 ml Chlorameisensäureäthylester zu, rührt 30 Minuten bei -5 , gibt 190 g H-Pro-OMe zu, lässt 4 Stunden bei 20 stehen, wäscht mit verdünnter Schwefelsäure und Ammoniak, trocknet und dampft ab.
Man erhält 550 g öliges Dipeptid, welches mag in 3 Liter Dioxan löst, auf 0 kühlt, gibt 3 Liter Natronlauge (In) zu, schüttelt 1 Stunde bei 20 , konzentriert auf 3 Liter, gibt 3 Liter Wasser zu, wäscht mit Diäthyl- äther, kühlt auf 0 , säuert mit 10 % iger Phosphorsäure auf pH 2 an, extrahiert das ausgeschiedene Öl mit Essigester, trocknet, dampft ab, wäscht den Rückstand mit Petroläther und trocknet.
Man erhält 470 g CBO- (CTB)Lys-Pro-OH ([a] D = -431 in Dimethylfarm- amid), das man in 3 Liter Methanol und 300 ml Wasser löst und in Anwesenheit von 30 g 10 % iger Palladium- kohle hydriert. Man filtriert, dampft ab und wäscht den Rückstand mit Diäthyläther. Man erhält 323 g H- (CTB)Lys-Pro-OH. Smp. 114 . [a]" = -25 in Di- methylformamid.
Man löst das oben erhaltene Dipeaptid .in 1000 ml Dimethylformamid, gibt 480 g CBO-(CTB)Lys-OCP zu, lässt 12 Stunden bei 20 stehen, dampft ab, löst,den Rückstand in 1000 ml Natriumbicarbonat auf, wäscht mit Diäthyläther, säuert die wässrige Phase mit 10 % iger Zitronensäurelösung an, extrahiert mit Essigester, trocknet, dampft ab und verarbeitet den Rückstand mit Petroläther. Man erhält 308g CBO-(CTB)Lys-Pro-OH (Smp. 125 ), das man.
in 3 Liter Methanol und 300 ml Wasser löst. Man hydriert in Anwesenheit von 45 g Palladiumkohle, filtriert, dampft ab und kristallisiert den Rückstand aus Dioxan-Diäthyläther um. Man erhält 205 g H-(CTB)Lys-(CTB)Lys-Pro-OH. Smp. 160 .
[a] D = -24 in Dimethylformamid. Man löst 380 g CBO-(CTB)Lys-OH und 260 g H-(CTB)Lys-OMe in einem Gemisch von 1500 ml Acetonitril und 300 ml Dimethylformamid, kühlt auf -20 , gibt 206g Dicyclo- hexylcarbodiimid zu, schüttelt bei -5 während 12 Stunden, filtriert, dampft ab, löst den Rückstand in Essig- ester, wäscht mit verdünnter Schwefelsäure- und Ammoniaklösung, trocknet, dampft ab und kristallisiert den Rückstand aus Diäthyläther-Petrol@äther. Man erhält 410 g CBO-(CTB)Lysi-(CTB)Lys-OMe. Smp. 105 .
= -10 in Methanol.
Der oben erhaltene Dipeptidester wird in 3 Liter Methanol gelöst, in Anwesenheit von 50 g Palladiumkatalysator hydriert und nach Filtration abgedampft. Man löst den Rückstand in 1000 ml Dimethylformamid, gibt 310g CBO-Val-Gly-OCP zu, l'ässt 12 Stunden bei 20 stehen, dampft ab, verarbeitet mit Äthyläther, kristallisiert aus Methanol=Wasser (1 : 1) um und trocknet. Man erhält 448g CBO-Val-Gly-(CTB)Lys-(CTB)Lys-OMe. Smp. 191'. [a110 = -16 in Methanol.
Man löst den so erhaltenen Tetrapeptidester in 600 ml Methanol, gibt 200 ml' Hydrazinhydrat zu, lässt 16 Stunden bei 20 stehen, giesst in 2 Liter Wasser, filtriert, wäscht mit Wasser, trocknet über Schwefelsäure, löst in Methanol, giesst mit Äther, filtriert und trocknet. Man erhält 366 g CBO-Val-Gly-(CTB)Lys-(CTB)Lys-NHNH2. Smp. 204 bis 206 . [a]10 = -110 in Dimethylformamid.
Man löst 48,2 g CBO-Val-Gfy-(CTB)Lys-(CTB)Lys- NHNH2 in einem auf -10 gekühlten Gemisch von 400 ml Dimethylformamid, 100 ml Isopropanol und 60 ml 4n Salzsäure, tropft unter kräftigem Rühren 14 ml einer 5n Natriumnitritlösung zu, rührt noch während 5 Minuten bei -5 , gibt 35 ml Triäthylamin und anschliessend 1000 ml Eiswasser zu, saugt scharf ab, löst den Niederschlag in 200 ml Dimethylformamicl, gibt 28,5g H-(CTB)Lys-Pro-OH und, 7 ml Triäthylamin zu, engt im Hochvakuum auf 180 ml ein, fässt 16 Stunden bei 0 stehen,
gibt 1500 ml Essigester zu, wäscht mit verdünnter Essigsäure, dampft den Essigester ab, giesst den Rückstand in 2 Liter Wasser, filtriert und trocknet

über Natriumhydroxyd. Man erhält 42 g CBO-Val-Gly- (CTB)Lys-(CTB)Lys: (CTB)Ly.s-(CTB)Lys-Pro-OH.Smp. 156 mit Zers. [a121 = -5 in Dimethylformamid.
Man löst das so erhaltene Heptapeptid in 160 ml absolutem Pyridin, dampft ab, löst den Rückstand in 160 ml absolutem Pyridin, gibt 22 g Tri-(2,4,5-trich#lorphenyl) phosphit zu, rührt 16 Stunden bei 20 , dampft ab, löst den Rückstand in Essigester, wäscht mit verdünnter Schwefelsäure und anschliessend mit 1n Natriumbicarbonatlösung, trocknet über Natriumsulfat, dampft ab und kristallisiert den Rückstand aus Essigester-Petroläther. Man erhält 44 g CBO-Val-Gly- (CTB)Lys-(CTB)Lys-(CTB)Lys-(CT'B)Lys-P.ro-OCP, den man sofort in 150 ml Dimethylformamid löst, gibt 38 g H-Val-(CTB)Lys-Val-Tyr-Pro-Val-NH2 zu, lässt 2 Tage bei 20 stehen, fällt durch Zusatz von Diäthyläther, filtriert und trocknet. Man löst das erhaltene Produkt .in Methanol-Wasser (8:2), behandelt mit einem sauren Ionenaustauscher, dampft ab und kristallisiert den Rückstand aus Äthanol-Wasser (8:2).
Marx erhält 45,5 g CBO-Val-Gly-(CTB)Lys-(CTB)Lys-(CTB)Lys- (CTB)Lys-Pro-Val-(CTB)Lys-Val Tyr-ProVal-NH2.
Smp. 189 mit Zers. [a]21 = +5 in. Dimethylformamid. Man löst das erhaltene Tridecapeptid in 500 ml Methanol', hydriert in, Anwesenheit von 5 g Palladiumkohle, filtriert, dampft ab und verarbeitet den Rückstand mit Diäthyläther. Man erhält 26,3 g H-Val-Gly-(CTB)Lys-(CTB)Lys-(CTB)Lys- (CTB)Lys-P.ro-Val-(CTB)Lys-Val Tyr-ProVal-NH2.

d) Trityl-O-tert.-butyl-L-glutamyl-im-tritylLrhistidyl-L-phenylalanyl-IrarginylL-tryptophanyl-glycyl-s-N-carbo-tert.-butaxyLrlysyl-L-prolin-2,4,5,trichlorphenylester (Tri-Glu(OTB)-(Tris)His-Phe-Arg-Try-Gly- (CTB)Lys-Pro-OCP) Man löst 300 g CBO-(N02)Arg Try-Gly-OMe in 5 Liter Dioxan-Wasser (8:2), gibt 720 ml 1n Natronlauge zu, lässt 1 Stunde bei 20 stehen, giesst in 16 Liter 0,05n Schwefelsäure, saugt den Niederschlag ab, wäscht mit Wasser, Alkohol, Diäthyläther und Petroläther und trocknet. Man erhält 285g CBO-(N02)Arg-Try-Gly-OH. (Smp. 225 . [a]21 = -14 in Dimethylformamid), den man in 3,5 Liter absolutem Pyridin löst. Man gibt 250 g Tri-(2,4,5-trichlorphenyl)-phosphit zu, rührt 16 Stunden bei 20 , dampft ab, löst in Essigester, wäscht mit verdünnter Schwefelsäure, trocknet und dampft ab.
Man kristallisiert den Rückstand aus Essigester-Diäthyläther und erhält 370 g CBO-(N02)Arg-Try-Gly-OCP. Smp. 124 [a]21 = -9 in Dimethylformamid.
Man löst den so erhaltenen. Man kristallisiert den Rückstand zweimal aus Essigester. Man erhält 370 g CBO-(NO2)Arg- D Try-Gly-(CTB)Lys-Pro-OH. Smp. 142 . 1a190 =-200 in Dimethylformamid.
Man, löst 245g CBO-(N02)Arg-Try-Gly-(CTB)LysPro-OH in 450 ml Eisessig, gibt 50 ml Wasser und 50 g Palladiumkohle 10% zu, hydriert, filtriert, dampft ab, löst im Wasser, lyophilisiert und trocknet über Natronlauge. Man erhält 135g H-Arb Try-Gly-(CTB)Lys-Pro- OH. [a]21 = -9 in Essigsäure-Wasser (95: 5).
Man löst 457g H-His-Phe-OME - 2HBr und 266 ml Triäthylamim in 1000 ml Dimethylfo.rmamid, rührt 10 Minuten bei 0 , filtriert, gibt dem Filtrat 496 g CBO-Glu(OTB)-OCP zu und lässt 16 Stunden bei 20 stehen. Man dampft das Dimethylformamid ab, löst den Rückstand in Essigester, wäscht mit verdünntem Essigsäure-Wasser .und 1n Natriumbicarbonatlösung, trocknet über Natriumsulfat, dampft ab und kristallisiert den Rückstand aus Essigester.
Man erhält 338 g CBO- Glu(OTB)-H.is-Phe-OMe. Smp. 183 . [a]21 = 21 in Dimethylformami d,.
Man löst das oben erhaltene Tripeptid in 3 Liter Methanol, hydriert in Anwesenheit von 50 g Palladiumkohle 10%, filtriert, dampft ab, löst den Rückstand in 1,5 Liter Methylenchlorid., kühlt auf 0 , gibt 140 ml Triäthylamin und anschliessend 270 g Triphenylchlor- methan zu, lässt 16 Stunden bei 20 stehen, wäscht mit verdünnter Essigsäure, Wasser und 1n Natriumbi- earbonatlösung, trocknet und dampft ab.
Man löst den Rückstand in Diäthyläther und fällt mit Petroläther. Man erhält 483 g Tri-G1u(OTB-(Tri)His-Phe-OMe (Smp. 80 mit Zers.), das man in 1000 ml Methanol löst.
Man gibt 50 ml Hydrazin zu, lässt 24 Stunden bei 20 stehen, konzentriert auf 500 ml, gibt 5 Liter Diäthyläther zu, wäscht mit 0,1n Kochsalzlösung, trocknet, konzentriert auf 500 ml und fällt mit Petrol- äther. Man erhält 390g Tri-Gl'u(OTB)-(Tri)His-Phe- NHNH2. Smp. 80 mit Zers. [a]21 = -14 in Dimethyl- formamid.
Man löst 41,6 g Tri-Glu(OTB)-(Tri)His-Phe-NHNH2 in 100 ml Dimethylformamid und 100 ml Isopropanol, kühlt auf -10 und gibt 40 ml 4n Salzsäure und anschliessend unter Rühren 9 ml 5n Natriumnitritlösung zu.
Nach 5 Minuten fügt man zu 28 ml Triäthylamin und 1000 ml Eiswasser zu, nutscht ab, löst den Nieder- schlag in Essigester, wäscht mit einer gesättigten KochsaMösung, trocknet, dampft bei 0 ab, löst den Rückstand in 100 ml Dimethylformamid, gibt 26g H-Arg- Try-Gly-(CTB)Lys-Pro-OH, 3 AcOH und 4,5 ml Tri- äthylamin zu, fässt 16 Std.

Man löst das oben erhaltene Peptid in 200 ml Pyridin, kühlt auf -20 , gibt 30 ml einer 1n Lösung von Salzsäure in Dioxan zu, rührt 5 Minuten bei -20 , gibt 7,8 ml Triäthylamin zu, konzentriert auf die Hälfte des Volumens, gibt 30 g Tri-(trichlorphenyl)-phosphit zu, lässt 16 Stunden bei 20 stehen, dampft ab, verarbeitet den Rückstand mit Diäthyläther, löst ihn in Essigester, fällt mit Diäthyläther, filtriert und trocknet.

Man lässt 24 Stunden bei 20 stehen, giesst die Lösung in 5 Liter Wasser, zentrifugiert, löst den öligen Rückstand in 2 Liter Essigester .und wäscht die erhaltene Lösung mit Wasser bis zur Neutralität.
