Verfahren zur Herstellung neuer 6ss,19-Oxido-steroide Gegenstand der vorliegenden Erfindung ist ein neues Verfahren zur Herstellung von 6f,19-Oxido-steroiden aus 19-unsubstituierten 6ss-Hydroxy-steroiden. Die ge nannten Oxidosteroide sind wichtige Zwischenprodukte zur Gewinnung pharmakologisch wichtiger 19-Nor- steroide, z.
B. von gewissen Derivaten des 19-Nor- testosterons und 19-Nor-progesterons. So haben bei spielsweise das 19-Nor-17a-methyl-testosteron, das 19 Nor-17a, äthinyl-testosteron und Ester des 19-Nor-testo- sterons therapeutische Verwendung gefunden.
Alle diese Verbindungen waren bisher nur durch Reduktion von Steroidverbindungen mit einem aroma tischen Ring A zugänglich, welche ihrerseits aus unge sättigten 3-Keto-steroiden durch thermische Eliminierung der angularen C-19-Methylgruppe und gleichzeitige Aromatisierung gewonnen werden mussten. Die gemäss der vorliegenden Erfindung nunmehr leicht zugäng lichen 6ss,19-Oxidosteroide ermöglichen nun die Her stellung von 19-Norsteroiden auf äusserst einfache Weise, ohne dass dabei der Ring A zuerst aromatisiert werden muss.
Das erfindungsgemässe Verfahren besteht darin, dass man auf 19-unsubstituierte 6ss-Hydroxy-steroide oxy dierend wirkende Schwermetallacylate einwirken lässt.
Als Ausgangsstoffe für das vorliegende Verfahren eignen sich 6ss-Hydroxyverbindungen der Androstan-, Pregnan-, Cholan-, Cholestan-, Stigmastan-, Spirostan- und Cardanolidreihe, welche im Ringsystem, insbeson dere in einer oder mehreren der Stellungen 1, 2, 3, 4, 5, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 und 21 westere Sub- stituenten aufweisen können, wie freie oder funktionell abgewandelte Oxogruppen,
veresterte oder verätherte Hydroxylgruppen, Alkyl- (z. B. Methyl)- -Gruppen und/ oder Halogenatome. Unter funktionell abgewandelten Oxogruppen kommen ketalisierte oder in Enolderivate, z. B. Enoläther oder Enolester, übergeführte Oxo- gruppen in Frage. Ausserdem können die Ausgangsstoffe auch Doppelbindungen oder Oxsdogruppen aufweisen, z.
B. in 4,5-, 9,11- oder 16,17-Stellung. Besonders wichtige Ausgangsstoffe sind solche 6fl- Hydroxyverbindungen, welche eine 44-3-Oxogruppierung oder in 3- und 5-Stellung solche Substituenten auf weisen, welche die Ausbildung einer d4-3-Oxogruppie- rung ermöglichen, z. B. geschützte 3a,5a-Dihydroxy- verbindungen, z.
B. cyclische Carbonate, Sulfite, Aceto- nide oder Be.nzalverbindungen, oder veresterte oder verätherte 44-3-Hyd'roxyverhindungen oder insbesondere Ester und Äther von 3-Hydroxy-5a-halogensteroiden, oder Ketale von 3-Oxo-5a@halogen-steroide, welche z. B.
durch Anlagerung von unterhalogeniger Säure an die entsprechenden 5,6-ungesättigten Verbindungen ent stehen.
Spezifische Ausgangsstoffe sind z. B. die folgenden Verbindungen: 3ss-Acetoxy-6ss hydroxy-cholestan, 3,17-Dioxo-6ss hydroxy-5a- bzw. 5ss-androstan, das 3,5-Carbonat oder 3,5-Sulfit des 3a,5a,6ss-Trihydroxy-20-oxo-pregnans, das 3,5-Carbonat oder 3,5-Sulfit des 3a,5a@,6ss-Trihydroxy 17-oxo-androstans oder des 3a,5a,6ss-Trihydroxy-17ss-acetoxy-androstans oder die entsprechenden Verbindungen,
in denen die freien Oxogruppen ketalisiert sind, sowie 3ss-Acetoxy-5a-chlor- oder 3ss-Acetoxy-5 a-'brom-6ss-hydroxy-cholestan, 3ss-Acetoxy-5a-chlor- oder 3ss-Acetoxy-5a-brom-6ss-hydroxy-spirostan, 3ss-Acetoxy-5a-cblor- bzw. 3ss-Acetoxy-5a-brom-6ss-hydroxy-17-oxo-androstan, 3ss,17ss Diacetoxy-5a-chlor- bzw. 3ss,17,6-Diacetoxy-5a-brom-6ss-hydroxy-androstan, 3ss,17ss-Diacetoxy-5a-chlor- bzw.
3ss,17ss-Diacetoxy-5a-brom-6ss-hydroxy-17a- methyl-androstan, d4-3,17-Dioxo-6f hydroxy-and'rosten, 3ss-Acetoxy-5a-chlor-6ss hydroxy 16,17a-oxido- 20-oxo-pregnan; die 17,20;20,21-Bismethylendioxyverbindung des 3ss Acetoxy-5a-chlor-6ss,17a,21-trihydroxy- 20-oxa-pregnans; 3ss,20ss-Diacetoxy-5a-chlor-6ss-hydroxy-5a-pregnan;

3fl-Acetoxy-5a-chlor-6ss hydroxy-17a-valerianoyl- oxy-20-oxo-pregnan, 3ss Acetoxy-5a,chlor-6ss-hydroxy-17a-methoxy- 20-oxo-pregnan usw. In den genannten Ausgangsstoffen werden die freien Ketogruppen vor der verfahrensgemässen Reaktion vor teilhaft, z. B. durch Ketalisierung mit Äthylenglykol, geschützt.
Die als Ausgangsstoffe verwendeten 6ss-Hydroxy- Verbindungen werden mit oxydierend! wirkenden Schwermetallacylaten, insbesondere mit Bleitetraacy- laten, wie Bleitetraacetat, Bleitetrapropionat, Bleitetra- benzoat und dergleichen, behandelt.
Die Reaktion, z. B. mit Bleitetraacetat, kann auch unter Zusatz einer schwachen Base vorgenommen wer den. Das Verfahren lässt sich beispielsweise so durch führen, dass man den Ausgangsstoff :in einem gegen über dem Oxydationsmittel inerten Lösungsmittel, z. B. einem Kohlenwasserstoff, löst, Bleitetraacetat und eine schwache Base, z. B. Calciumcarbonat, zugibt und die Reaktionsmischung unter Rühren bei Normal- oder er höhtem Druck erhitzt.
Besonders geeignete Lösungsmittel sind gesättigte, cyclische Kohlenwasserstoffe, wie Cyclohexan, Methyl- cyclohexan, Dimethylcyclohexan, doch können auch aromatische Kohlenwasserstoffe, wie Benzol, oder halo- genierte Kohlenwasserstoffe, z. B. Tetrachlorkohlenstoff oder Hexachlorbutadien, oder Gemische dieser Lösungs mittel Verwendung finden. Die benötigte Reaktions dauer hängt von der Temperatur bzw. von dem ver wendeten Lösungsmittel ab.
Beim Arbeiten mit Blei tetraacetat, z. B. in siedendem Methyleyclohexan, ist die Reaktion in der Regel nach etwa 1,5 bis 3 Stunden beendet.
In den verfahrensgemäss erhaltenen 6ss,19-Oxyden vorhandene geschützte Hydroxygruppen, z. B. in S und 17- oder 20-Stellung, können durch Einwirkung von basischen Mitteln hydrolysiert werden.
In den oben genannten Estern bedeuten die Säure reste, insbesondere solche von aliphatischen, cycloali- phatischen, araliphatischen und aromatischen Carbon- säuren mit 1-15 Kohlenstoffatomen, z.
B. Formiate, Acetate, Propionate, Butyrate, Trimethylacetate, Oenan- thate, Capronate, Decanoate, Cyclopentyl'propionate, Valzrianate, Benzoate, Furoate, Hexahydrobenzoate, Phenylpropionate, Trifluoracetate, Äthyl- und Methyl- carbonate usw.
Die erfindungsgemäss erhaltenen 6ss,19-Oxido- steroide lassen sich nach den Verfahren der deutschen Patente Nrn. <B>1192</B> 195 oder 1 192196 in 44- bzw. d5 19-oxygenierte Steroide und diese nach bekannten Methoden leicht in 19 Nor-steroide umwandeln.
Die Temperaturen sind in den nachfolgenden Bei spielen in Celsiusgraden angegeben. <I>Beispiel 1</I> 1 g Calciumcarbonat und 3 g Bleitetraacetat werden in 100 ml Methylcyclohexan 1/,4 Stunde auf etwa 100 erwärmt. Nach Zugabe von 373 mg 3ss Acetoxy-5a- chloro-6ss-hydroxy-cholestan wird das Reaktionsgemisch weitere 31/2 Stunden unter Rühren am Rückfluss ge kocht.
Die abgekühlte Lösung wird von anorganischen Salzen abgenutscht, der Rückstand erschöpfend mit absolutem Äther gewaschen und :die vereinigten Filtrate im Scheidetrichter mit 10ä iger Natriumthiosulfatlösung und mit Wasser gewaschen. Die mit Natriumsulfat ge trocknete Lösung wird im Vakuum eingedampft. Aus dem Eindampfrückstand werden nach einmaliger Um kristallisation aus Äther-Methanol 218 mg 3ss-Acetoxy- 5a-chloro-6ss,19-oxido-cholestan mit F. 134-139 er halten.
Aus der Mutterlauge können weitere 34 mg eines etwas, unreineren Produktes (F. 129-132 ) isoliert werden.
<I>Beispiel 2</I> 2,5 g Calciumcarbonat und 6,0 Bleitetraacetat wer den in 200 ml Cyclohexan erhitzt. Zur siedenden U sung gibt man 950 mg 3ss,17ss-Diacetoxy-5a-chlor-6ss- hydroxy-androstan und hält die Lösung unter Rühren während 14 Stunden im Sieden. Dann kühlt man ab, filtriert die ungelösten Salze ab, wäscht mit Äther nach und schüttelt das Filtrat mit Wasser aus.
Aus der ge trockneten organischen Lösung erhält man beim Ein dampfen im Wasserstrahlvakuum 1,17 g eines Rück standes. Dieser wird durch Chromatographie an Alu miniumoxyd' gereinigt. Durch Kristallisation der mit Benzol eluierten Substanz aus Alkohol- erhält man das 3ss,17ss-Diacetoxy-5a-chlor-6ss,19-oxido-androstan vom F. 160-1610.
<I>Beispiel 3</I> Zu einer Suspension von 7,5g Calciumcarbonat und 20 g Bleitetraacetat in 800 ml Methylcyclohexan gibt man 4,0 g 3ss,20ss-Diacetoxy-5a-chlor-6fl-hydroxy-5a- pregnan und kocht das Reaktionsgemisch unter Rühren während 3,5 Stunden unter Rückfluss. Dann kühlt man ab, saugt die unlöslichen Salze ab und wäscht den Niederschlag mit Äther nach. Das Filtrat wird mit Kaliumjodid- und verdünnter Natriumthiosulfatlösung gewaschen, getrocknet und im Wasserstrahlvakuum ein gedampft.
Man erhält 5,2 g eines teilweise kristalli sierten Rückstandes. Durch Umlösen aus Aceton-Hexan gewinnt man 2,8 g 3ss,20ss-Diacetoxy-5a-chlor-6ss,19- oxido-5a4pregnan vom F. 148-150 ; [a]D = +25 (in Chloroform).
<I>Beispiel 4</I> Eine Suspension von 3,0 g Bleitetraacetat und 1,4 g Calciumcarbonat werden in 120 ml Cyclohexan während 30 Minuten zum Sieden erhitzt. Dann gibt man 600 mg 3ss,17ss-Diacetoxy-Sa-brom - 6ss - hydroxy - androstan zu und rührt die siedende Lösung noch 14 Stunden weiter. Schliesslich kühlt man ab und arbeitet wie in Beispiel 2 beschrieben auf. Man erhält 820 mg Rohprodukt, wel ches durch Chromatographie an 20g Aluminiumoxyd gereinigt wird.
Aus den mit Benzol-Hexan-1 : 1-Gemisch und mit Benzol eluierten Fraktionen gewinnt man durch Kristallisation aus Äther-Hexan 250 mg 3ss,17ss-Diacet- oxy-5a-brom-6ss,19-oxido-androstan von F. 178-180 ; [a']D = - 5,4 (in Chloroform).
<I>Beispiel 5</I> 1 g 3ss,17ss-Diacetoxy-6ss-hydroxy-5a-androstan, 6 g Bleitetraacetat und 2 g Calciumcarbonat werden in 250 ml Cyclohexan unter Rühren während 14 Stunden am Rückflusskühler gekocht. Das abgekühlte Reaktions gemisch filtriert man durch Celite und wäscht den Rückstand mit Cyclohexan nach.
Die vereinigten Filtrate werden nacheinander mit 5 % iger Kaliumjodid- und Natriumthiosulfatlösung sowie mit Wasser ausgeschüttelt, getrocknet und im Wasserstrahlvakuum eingedampft. Der erhaltene Rückstand (1,2 g) wird aus Äther-Petrol- äther kristallisiert und -liefert 600 mg 3ss,17ss-Diacetoxy- 6/3,19-oxido-5a-androstan vom F. 145 .
<I>Beispiel 6</I> 2,0 g 3/3,11 a,20ss-Triacetoxy-5 a-chlor-6ss - hydroxy- pregnan werden in 500 cm3 Cyclohexan nach Zugabe von 15,0 g Bleitetraacetat während 14 Stunden unter Rühren und Feuchtigkeitsausschluss gekocht. Dann kühlt man ab, trennt die ungelösten Salze durch Filtration ab und wäscht den Filterkuchen mit Essigester nach. Das Filtrat wird mit Wasser gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft.
Man erhält 2,328 g eines Teils kristallisierten Rückstandes. Er wird in Benzol gelöst und die Lösung durch 30 g Aluminiumoxyd filtriert. Aus den mit insgesamt 600 cm3 Benzol eluierten Fraktionen erhält man nach Kristallisation aus Äther- Pentan 1,03 g des 3ss,lla,20ss-Triacetoxy-5a-chlor- 6ss,19-oxid'o-pregnans vom F. 228-230 .
Aus den fol genden, mit 200 cm3 Benzol-Essigester-1:1-Gemisch eluierten Fraktionen erhält man durch Kristallisation der Eindampfrückstände aus Äther 452 mg der als Aus gangsstoff verwendeten 5a-Chlor-6ss-hydroxy-Verbin- dung zurück.
<I>Beispiel 7</I> Eine Suspension von 500 mg 3ss,11ss.20ss-Triacetoxy- 6ss-hydroxy-5a-pregnan und 4,0 g Bleitetraacetat in 150 cm3 Cyclohexan erhitzt man unter Rühren während 14 Stunden zum Sieden. Nach dem Abkühlen trennt man die ungelösten Salze durch Filtration ab, wäscht mit Essigester nach und schüttelt das Filtrat mit Wasser aus. Aus der getrockneten organischen Lösung erhält man 578 mg eines kristallisierten Rückstandes.
Dieser wird in Benzol-Hexan-1 : 1-Gemisch gelöst und an 30 g Aluminiumoxyd' chromatographiert. Aus den ersten mit Benzol-Essigester-9:1-Gemisch eluierten Fraktionen (353 mg) erhält man durch Kristallisation aus Äther das reine 3ss,11ss,20fl-Triacetoxy-6ss,19-oxid'o-5a-pregnan vom F. 192-195 /l98-200 .
Aus den späteren, eben falls mit Benzol-Essigester-9: 1-Gemisch eluierten Frak tionen (50 mg) isoliert man durch Kristallisation aus Äther das 3ss,11ss,20ss-Triacetoxy-6-oxo-5ss-pregnan vom F. 220-222 .
<I>Beispiel 8</I> Eine Suspension von 10,0 g 3ss-Acetoxy-5a-chlor-6ss- hydroxy-17-oxo-androstan und 65 g Bleitetraacetat in 3,3 Liter Cyclohexan wird unter Rühren 14 Stunden am Rückfluss gekocht. Man kühlt, filtriert, wäscht den Filterrückstand mit Äther und schüttelt das Filtrat mit verdünnter Thiosulfatlösung und mit Wasser aus. Nach dem Verdampfen des organischen Lösungsmittels hinter bleiben 12,1 g eines rohen Produktes, das aus Äther Methanol umkristallisiert wird.
Man erhält 6,3 g 3ss- Acetoxy-5a-chlor-6ss,19-oxido-17 - oxo - androstan vom F.180-182 .
In völlig analoger Weise erhält man aus 2,5 g 3ss- Acetoxy-5a-brom-6ss-hydroxy-17-oxo-androstan <B>1,51</B> g 3ss - Acetoxy - 5a -Brom- 6ss,19 -oxido-17-oxo-androstan vom F.
174-178 /184-187 . Beispiel <I>9</I> Eine Mischung von 2,5 g 3ss Acetoxy-5a-chlor-6ss- hyd'roxy-20-oxo-pregnan, 5 g Calciumcarbonat und 10 g Bleitetraacetat in 450 ml Cyclohexan wird 16 Stunden am Rückfluss gekocht. Nach dem Abkühlen und Fil trieren wird die Lösung mit verdünnter Natriumsulfit lösung und Wasser gewaschen, getrocknet und unter vermindertem Druck eingedampft.
Durch Umkristalli Bieren des Rohproduktes aus Äther erhält man 1,47 g 3ss-Acetoxy-5a-chlor-6ss,19-oxido-20-oxo-pregnan vom F. 150-153 ; [a]D - +65 (in Chloroform). IR Banden bei 5,78, 5,88, 6,70, 8,13, 9,12, 9,66, 10,60, 10,86 und 11,75,u. In analoger Weise erhält man aus 3ss,17a-Diacetoxy-5a-chlor-6ss-hydroxy- 20-oxo-pregnan das 3ss,17a-Diacetoxy-5a-chlor-6ss,19-oxido- 20-oxo-pregnan vom F. 187-187,5 .
Zu einer gerührten Suspension von 10,0 g Blei- (IV)-acetat und 3,0 g Calciumcarbonat in 250 ml Cyclo- hexan werden 1,5 g 3fl-Acetoxy-5a-chlor-6ss-hydroxy- 17a@-valerianyl-oxy-20-oxo-pregnan zugegeben. Das Ge misch wird anschliessend 7 Stunden am Rückfluss ge kocht.
Nach Abkühlen auf 20 filtriert man von anorga nischen Anteilen ab, wäscht den Rückstand mit Cyclo- hexan nach und schüttelt die vereinigten Filtrate nach einander mit Wasser, 5 % iger Kaliumjodidlösung, <I>5</I> %iger Natriumthiosulfatlösung und erneut mit Wasser .aus. Der durch Eindampfen der getrockneten organischen Lösung im Vakuum erhaltene Rückstand beträgt etwa 2,1 g.
Durch anschliessende Filtration einer Lösung dieses Rückstandes in Petroläther-Benzol-Gemisch durch die 30fache Gewichtsmenge Aluminiumoxyd (Aktivi tät 1I - neutral) wird zuerst ein aromatisch riechendes Öl und dann mit reinem Benzol insgesamt 0,9 g reines 3ss-Ace@toxy-5a-chlor-6ss,19-oxido-17cc- valerianyloxy-20-oxo-pregnan erhalten.
Die als Ausgangsstoffe verwendeten Chlorhydrine können in der folgenden Weise hergestellt werden: Man behandelt eine Lösung von 150 g Pregnenolon acetat an 5 Liter Äther mit<B>150</B> Chlorkalk (mit einem Gehalt von<B>30%</B> aktivem Chlor) und 8,2 Liter Wasser. Das Gemisch wird kräftig gerührt, und nach 5 Minuten gibt man 105 ml Eisessig und nach weiteren 25 Minuten 500 ml einer 10 % igen Kaliumjodidlösung zu.
Die wäss- rige Phase wird abgetrennt, die ätherische Phase mit einer 10%igen Natriumthiosulfatlösung, dann mit ver dünnter Natriumbicarbonatlösung und mit Wasser ge waschen. Man trocknet und dampft die Lösung hierauf im Wass@erstrahlvakuum ein. Man behandelt den Rück stand mit 800 ml Aceton und 1'ässt bei 0 über Nacht stehen. Es kristallisieren so 47,5 g 3ss-Acetoxy-5a-chlor- 6fl-hydroxy-20-oxo-pregnan aus; aus den Mutterlaugen können nochmals 58 g gewonnen werden.
Nach dem Umkristallisieren aus Aceton schmilzt das Produkt bei l96-197 . Optische Drehung [a]D = 25,5 (in Chloro form. Im IR-Spiektrum sind Banden u. a. bei 2,75, 5,78, 5,88, 8,12, 8,68, 9,68 und 9,72 ,u vorhanden.
In analoger Weise erhält man durch Anlagerung von unterchloriger Säure an das 17a-Acetoxy-pregnenolon- acetat das 3ss,17a-Diacetoxy-5a-chlor-6ss-hydroxy-20- oxo-pregnan und an das 17a-Valeriany1.oxy-20-oxo- pregnenolonacetat das 3ss-Acetoxy-5a-chlor-6ss-hydroxy- 17a-valerianyloxy-20-oxo-pregnan. <I>Beispiel 10</I> 2,0 g 3ss,11a,
20fl-Triacetoxy-5a-chlor-6ss-hydroxy- pregnan werden in 500 ml Cyclohexan nach Zugabe von 15,0 g Bleitetraacetat während 14 Stunden unter Rühren und Feuchtigkeitsausschluss gekocht. Dann kühlt man ab, trennt die ungelösten Salze durch Filtration ab und wäscht den Filterkuchen mit Essigester nach. Das Filtrat wird mit Wasser gewaschen, getrocknet und im Wasser strahlvakuum eingedampft. Man erhält 2,328 g eines teils kristallisierten Rückstandes.
Er wird in Benzol gelöst und die Lösung durch 30 g Aluminiumoxyd filtriert. Aus den mit insgesamt 600 ml Benzol eluierten Fraktionen erhält man nach Kristallisation aus Äther- Pentan 1,03 g des 3ss,11a,20ss-Triäcetoxy-5a-chlor- 6ss,19-oxido-pregnans vom F. 228-230 .
Aus den fol genden, mit 200 ml Benzol-Essigester-1:1-Gemisch eluierten Fraktionen erhält man durch Kristallisation der Eindampfrückstände aus Äther 452 mg der als Ausgangsstoff verwendeten 5a-Chl#or-6ss-hydroxyverbin dung zurück.

Nach dem Abkühlen trennt man die ungelösten Salze durch Filtration ab, wäscht mit Essigester nach und schüttelt das Filtrat mit Wasser aus.
Aus der getrockneten organischen Lösung erhält man 578 mg eines kristallisierten Rückstandes. Dieser wird in Benzol-Hexan-1 : 1-Gemisch gelöst und an 30 g Aluminiumoxyd chromatographiert. Aus den ersten mit Benzol-Essigester-9: 1-Gemisch eluierten Fraktionen (353 mg) erhält man durch Kristallisation aus Äther das reine 3ss,11ss,20ss-Triacetoxy-6ss,19-oxido-5a-pregnan vom F. 192-195 /198-200 .

B. of certain derivatives of 19-nor-testosterone and 19-nor-progesterone. For example, 19-nor-17a-methyl-testosterone, 19-nor-17a, äthinyl-testosterone and esters of 19-nor-testosterone have found therapeutic use.
All these compounds were previously only accessible by reducing steroid compounds with an aromatic ring A, which in turn had to be obtained from unsaturated 3-keto-steroids by thermal elimination of the angular C-19 methyl group and simultaneous aromatization. The 6ss, 19-oxidosteroids, which are now easily accessible according to the present invention, now enable the manufacture of 19-norsteroids in an extremely simple manner, without ring A having to be aromatized first.
The method according to the invention consists in that 19-unsubstituted 6ss-hydroxy steroids oxidizing heavy metal acylates are allowed to act.
Suitable starting materials for the present process are 6ss-hydroxy compounds of the androstane, pregnan, cholan, cholestan, stigmastan, spirostane and cardanolide series, which are in the ring system, in particular in one or more of the positions 1, 2, 3, 4, 5, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 and 21 may have western substituents, such as free or functionally modified oxo groups,
esterified or etherified hydroxyl groups, alkyl (e.g. methyl) groups and / or halogen atoms. Functionally modified oxo groups include ketalized or enol derivatives, e.g. B. enol ethers or enol esters, converted oxo groups in question. In addition, the starting materials can also have double bonds or oxzo groups, e.g.

3fl-Acetoxy-5a-chloro-6ss hydroxy-17a-valerianoyl-oxy-20-oxo-pregnane, 3ss-acetoxy-5a, chlor-6ss-hydroxy-17a-methoxy-20-oxo-pregnane etc. In the starting materials mentioned the free keto groups before the process according to some kind of reaction, z. B. protected by ketalization with ethylene glycol.
The 6ss-hydroxy compounds used as starting materials become oxidizing! acting heavy metal acylates, in particular with lead tetraacylates such as lead tetraacetate, lead tetrapropionate, lead tetra benzoate and the like.
The reaction, e.g. B. with lead tetraacetate can also be made with the addition of a weak base who the. The process can be carried out, for example, by the starting material: in a solvent which is inert towards the oxidizing agent, e.g. B. a hydrocarbon, dissolves, lead tetraacetate and a weak base, e.g. B. calcium carbonate, is added and the reaction mixture is heated with stirring at normal or he increased pressure.
Particularly suitable solvents are saturated, cyclic hydrocarbons, such as cyclohexane, methylcyclohexane, dimethylcyclohexane, but aromatic hydrocarbons, such as benzene, or halogenated hydrocarbons, eg. B. carbon tetrachloride or hexachlorobutadiene, or mixtures of these solvents find medium use. The required reaction time depends on the temperature and the solvent used.
When working with lead tetraacetate, z. B. in boiling methyleyclohexane, the reaction is usually complete after about 1.5 to 3 hours.
Protected hydroxyl groups present in the 6ss, 19-oxides obtained according to the process, e.g. B. in the S and 17- or 20-position, can be hydrolyzed by the action of basic agents.
In the above-mentioned esters, the acid radicals, in particular those of aliphatic, cycloaliphatic, araliphatic and aromatic carboxylic acids with 1-15 carbon atoms, eg.
B. formates, acetates, propionates, butyrates, trimethylacetates, oenanthates, capronates, decanoates, cyclopentyl propionates, valzrianates, benzoates, furoates, hexahydrobenzoates, phenylpropionates, trifluoroacetates, ethyl and methyl carbonates, etc.
The 6ss, 19-oxido steroids obtained according to the invention can easily be converted into 44- or d5 19-oxygenated steroids by the processes of German patent nos. 1192 195 or 1 192196 and these can easily be converted into 19-oxygenated steroids by known methods Convert nor-steroids.
The temperatures are given in degrees Celsius in the following examples. <I> Example 1 </I> 1 g calcium carbonate and 3 g lead tetraacetate are heated to about 100 for 1/4 hour in 100 ml methylcyclohexane. After adding 373 mg of 3pp acetoxy-5a-chloro-6ss-hydroxy-cholestane, the reaction mixture is refluxed for a further 31/2 hours with stirring.
The cooled solution is suction-filtered to remove inorganic salts, the residue is washed exhaustively with absolute ether and: the combined filtrates are washed in a separating funnel with 10% sodium thiosulphate solution and with water. The solution, dried with sodium sulfate, is evaporated in vacuo. After a single order of crystallization from ether-methanol, 218 mg of 3ss-acetoxy-5a-chloro-6ss, 19-oxido-cholestane with a temperature of 134-139 are obtained from the evaporation residue.
A further 34 mg of a somewhat impure product (F. 129-132) can be isolated from the mother liquor.
<I> Example 2 </I> 2.5 g calcium carbonate and 6.0 lead tetraacetate are heated in 200 ml cyclohexane. 950 mg of 3ss, 17ss-diacetoxy-5α-chloro-6ss-hydroxy-androstane are added to the boiling solution and the solution is kept at the boil for 14 hours while stirring. It is then cooled, the undissolved salts are filtered off, washed with ether and the filtrate is shaken out with water.
From the dried organic solution, 1.17 g of a residue is obtained when evaporating in a water jet vacuum. This is purified by chromatography on aluminum oxide '. Crystallization of the substance eluted with benzene from alcohol gives the 3ss, 17ss-diacetoxy-5a-chloro-6ss, 19-oxido-androstane with a melting point of 160-1610.
<I> Example 3 </I> To a suspension of 7.5 g calcium carbonate and 20 g lead tetraacetate in 800 ml methylcyclohexane are added 4.0 g 3ss, 20ss-diacetoxy-5a-chloro-6fl-hydroxy-5a-pregnane and boiled reflux the reaction mixture with stirring for 3.5 hours. It is then cooled, the insoluble salts are filtered off with suction and the precipitate is washed with ether. The filtrate is washed with potassium iodide and dilute sodium thiosulfate solution, dried and evaporated in a water-jet vacuum.
5.2 g of a partially crystallized residue are obtained. 2.8 g of 3ss, 20ss-diacetoxy-5a-chloro-6ss, 19-oxido-5a4pregnane with a melting point of 148-150 are obtained by dissolving from acetone-hexane; [a] D = +25 (in chloroform).
<I> Example 4 </I> A suspension of 3.0 g of lead tetraacetate and 1.4 g of calcium carbonate are heated to the boil in 120 ml of cyclohexane for 30 minutes. Then 600 mg of 3pp, 17pp-diacetoxy-sa-bromo-6pp-hydroxy-androstane are added and the boiling solution is stirred for a further 14 hours. Finally, it is cooled and worked up as described in Example 2. 820 mg of crude product are obtained, which is purified by chromatography on 20 g of aluminum oxide.
From the fractions eluted with a benzene-hexane 1: 1 mixture and with benzene, 250 mg of 3ss, 17ss-diacetoxy-5a-bromo-6ss, 19-oxido-androstane of F. 178 are obtained by crystallization from ether-hexane -180; [a '] D = -5.4 (in chloroform).
<I> Example 5 </I> 1 g of 3ss, 17ss-diacetoxy-6ss-hydroxy-5a-androstane, 6 g of lead tetraacetate and 2 g of calcium carbonate are refluxed in 250 ml of cyclohexane with stirring for 14 hours. The cooled reaction mixture is filtered through Celite and the residue is washed with cyclohexane.
The combined filtrates are shaken out successively with 5% potassium iodide and sodium thiosulfate solution and with water, dried and evaporated in a water jet vacuum. The residue obtained (1.2 g) is crystallized from ether-petroleum ether and delivers 600 mg of 3ss, 17ss-diacetoxy-6 / 3,19-oxido-5a-androstane with a melting point of 145.
<I> Example 6 </I> 2.0 g 3 / 3.11 a, 20ss-triacetoxy-5α-chloro-6ss-hydroxy-pregnane are added in 500 cm3 of cyclohexane after adding 15.0 g of lead tetraacetate for 14 hours cooked while stirring and excluding moisture. It is then cooled, the undissolved salts are separated off by filtration and the filter cake is washed with ethyl acetate. The filtrate is washed with water, dried and evaporated in a water jet vacuum.
2.328 g of a portion of the crystallized residue are obtained. It is dissolved in benzene and the solution is filtered through 30 g of aluminum oxide. From the fractions eluted with a total of 600 cm3 of benzene, after crystallization from ether-pentane, 1.03 g of 3ss, 11a, 20ss-triacetoxy-5a-chloro-6ss, 19-oxide'o-pregnans from F. 228-230 are obtained.
452 mg of the 5α-chloro-6ss-hydroxy compound used as the starting material are recovered from the following fractions eluted with 200 cm3 of benzene-ethyl acetate 1: 1 mixture by crystallizing the evaporation residues from ether.
<I> Example 7 </I> A suspension of 500 mg 3ss, 11ss.20ss-triacetoxy-6ss-hydroxy-5a-pregnane and 4.0 g lead tetraacetate in 150 cm3 cyclohexane is heated to boiling with stirring for 14 hours. After cooling, the undissolved salts are separated off by filtration, washed with ethyl acetate and the filtrate is shaken out with water. 578 mg of a crystallized residue are obtained from the dried organic solution.
This is dissolved in a benzene-hexane 1: 1 mixture and chromatographed on 30 g of aluminum oxide. From the first fractions (353 mg) eluted with benzene-ethyl acetate 9: 1 mixture, the pure 3ss, 11ss, 20fl-triacetoxy-6ss, 19-oxide'o-5a-pregnane of m. 192 is obtained by crystallization from ether -195 / l98-200.
The 3ss, 11ss, 20ss-triacetoxy-6-oxo-5ss-pregnane with a melting point of 220- is isolated from the later fractions (50 mg) that were also eluted with a benzene-ethyl acetate 9: 1 mixture by crystallization from ether. 222.
<I> Example 8 </I> A suspension of 10.0 g of 3ss-acetoxy-5a-chloro-6ss-hydroxy-17-oxo-androstane and 65 g of lead tetraacetate in 3.3 liters of cyclohexane is refluxed with stirring for 14 hours cooked. It is cooled, filtered, the filter residue is washed with ether and the filtrate is extracted with dilute thiosulphate solution and with water. Example <I> 9 </I> A mixture of 2.5 g of 3ss acetoxy-5a-chloro-6ss-hydroxy-20-oxo-pregnane, 5 g of calcium carbonate and 10 g of lead tetraacetate in 450 ml of cyclohexane is used for 16 hours Boiled under reflux. To a stirred suspension of 10.0 g of lead (IV) acetate and 3.0 g of calcium carbonate in 250 ml of cyclohexane, 1.5 g of 3fl-acetoxy-5a-chloro-6ss-hydroxy-17a @ -valerianyl- oxy-20-oxo-pregnane added. The mixture is then refluxed for 7 hours.
After cooling to 20, inorganic components are filtered off, the residue is washed with cyclohexane and the combined filtrates are shaken one after the other with water, 5% potassium iodide solution, 5% sodium thiosulphate solution and again with water .out. The chlorohydrins used as starting materials can be prepared in the following way: A solution of 150 g of pregnenolone acetate in 5 liters of ether is treated with <B> 150 </B> chlorinated lime (with a content of <B> 30% </B> active chlorine) and 8.2 liters of water. The mixture is stirred vigorously and, after 5 minutes, 105 ml of glacial acetic acid and, after a further 25 minutes, 500 ml of a 10% potassium iodide solution are added.
