Verfahren zur Herstellung neuer 4ss,19-Oxido-steroide Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 4ss,19-Oxido-steroiden aus 19-unsubstituierten 4ss-Hydroxy-5a@steroiden. Die genannten Oxidosteroide können z.
B. zur Herstellung pharmakologisch wic'h'tiger 19-Nor-steroide (Anabo lika oder Progestativa), wie Derivaten des 19-Nor- testosterons und 19-Nor-progesterons und deren 3 Desoxoverbindungen, wie d4 -17ss - Hydroxy -17a- äthinyI-19 nor-androsten, verwendet werden.
Das erfindungsgemässe Verfahren besteht darin, däss man 19 unsubstitüierte 4ss-Hydroxy-5a-steroide mit oxydierend wirkenden Bleiacylaten umsetzt. Falls erwünscht, können die erhaltenen 4ss,19-Oxido- steroide oxydiert werden.
Als Ausgangsstoffe für das vorliegende Verfahren eignen sich entsprechende 4ss-Hydroxy-5a-steroide der Androstan-, Pregnan-, Ch.olan-, Chol'estan-, Stigma- stan-, Siprostan- und Cardanol'idreihe, welche im Ringsystem, insbesondere in einer oder mehreren der Stellungen 1, 2, 3, 5, 6, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 und 21 weitere Substituenten aufweisen können,
wie freie oder funktionell abgewandelte Oxogruppen, veresterte oder verätherte Hydroxyl- gruppen, niedere Alkyl- (z. B. Methyl-)gruppen und/ oder Halogenatome. Als funktionell abgewandelte Oxogruppen sind ketalisierte oder in Enolderivate, z. B. Enoläther oder Enolester, übergeführte Oxo- gruppen zu nennen.
Ausserdem können die Aus gangsstoffe auch Doppelbindungen oder Oxidogrup- pen aufweisen, z. B. in 9,11- oder 16,17-Stellung.
Besonders wichtige Ausgangsstoffe sind solche entsprechende 4ss-Hydroxy-5a-steroide, welsche in S und gegebenenfalls auch in 5-Stellung solche Sub- stituenten aufweisen, welche nach der Öffnung der 4f,19-Oxido-Brücke die Ausbildung einer d4-3-Oxo- gruppierung ermöglichen, z.
B. geschützte 3a,5a-Di- hydroxyverbindungen, wie cyclische Carbonate, Sul- fite, Acetonide oder Benzalverbindungen oder ins besondere 3-Ester und 3-Äther von 3-Hydroxy-5a- halogen-steroiden oder Ketale von 3-Oxo-5a halogen- steroiden.
Spezifische Ausgangsstoffe sind z. B. die folgen den Verbindungen: 3ss,17ss-Diacetoxy-4ss-hydroxy-5a-androstan, 3ss,17ss-Diacetoxy-4P-hydroxy-17a-methyl- 5a-androstan, das 3,5-Carbonat des 3a,4ss,5a-Trihydroxy-17-oxo-androstans, 3ss,17ss-Diacetoxy-ss-hydroxy-5a-chlor- androstan, 3ss,17ss-Diacetoxy-4ss-hydroxy-5a-brom- androstan, 3ss,17ss-Diacetoxy-4ss-hydroxy-5a-chlor-17a- methyl-androstan, 3ss,
17ss-Diacetoxy-4ss hydroxy-5a-brom-17a- methyl-androstan, 3P,20ss-Diacetoxy-4ss nydroxy-5a-chfor-pregnan, 3ss,20ss-DipTopionyloxy-4fl-hydroxy-5ahrom- pregnan, das 3ss-Acetoxy-4ss-hydioxy-5a-chlor-20-oxo- pregnan, oder 3ss-Acetoxy-4ss-hydroxy-5a-chlor-spirostan. Besonders wichtige Ausgangsstoffe zur Gewin nung der in 3-Stellung unsubstituierten 4ss,
19-Oxido- steroide sind z. B. die in 3-Stellung unsubs!tituierten 4ss-Hydroxy-5a-steroide, wie 4ss-Hydroxy-5a-halogen- androstane, welche in 17-Stellung eine Oxogruppe oder eine veresterte oder verätherte Hydroxygruppe aufweisen, z.
B. 4ss-Hydroxy-Sa-chlor-17-oxo-andro- stan, 17 Ester des 4f,17ss Dihydroxy-5a-chlor-andro- stans, wie das 17-Acetat, 17-Propionat oder 17- Benzoa't und 17-Ester des 4ss,17ss-Di'hydroxy-5a- chlor-17a-methyl-androstans. Die genannten Aus gangsstoffe sind bekannt oder können, falls neu, nach bekannten Methoden hergestellt werden.
Die 4f-Hydroxy-a-halogensteroide können z. B. durch Anlagerung von unterhalogeniger Säure an die ent sprechenden 4,5-ungesättigten Verbindungen herge- stellt werden. 4ss-Hydroxy-steroide lassen sich auch besonders leicht durch katalytische Hydrierung der entsprechenden 4-Ketone mit einem Platinkatalysator in saurer Lösung, z.
B. in Eisessig oder durch hydro- lytische Spaltung von 4,5-Epoxyden herstellen.
Die für das erfindungsgemässe Verfahren ver wendeten oxydierend wirkenden B'leiacylate sind z. B. Bleitetraacylate, deren Säurekomponente sich z.
B. von einer niederen aliphatischen, cycloaNphati- schen, araliphatischen oder aromatischen Säure ab leitet, wie der Essig-. Propion-, Trifluoressig-, Hexa- hydrobenzoe-, Phenylessig oder Benzoesäure. Ferner können Dialkylbleidiacylate, wie Diäthylbleidiacetat,
oder Anhydride der Metableisäure mit den obigen Carbonsäuren, wie Diacetoxybleioxyd, verwendet werden.
Die verfahrensgemässe Umsetzung wird z. B. so vorgenommen, dass man .die Ausgangsstoffe, gege benenfalls unter Zugabe einer schwachen anorga nischen oder organischen Base, z.
B. von Erdalkal'i- carbonaten, wie Calcium-, Barium- oder Ströntium- carbonat, oder von tertiären Aminen, wie Pyridin, zusammen mit einem Überschoss .an oxydierendem Bleiacylat, vorzugsweise in einem apalaren, gegen über dem Oxydationsmittel inerten Lösungsmittel, z.
B. auf dessen Siedetemperatur, vorzugsweise aber über 60 C, erwärmt und nach üblichen Methoden das als Verfahrensprodukt anfallende 4p,19-Oxido- steroid isoliert.
Als Lösungsmittel eignen sich insbesondere ali- phatische, cycloaliph.atische und aromatische Kohlen wasserstoffe, wie z. B. Hexan, Heptan, Cyclohexan, Methylcyclohexan, Dimethylcyclohexan oder Benzol.
Die Reaktionsdauer ist im allgemeinen von der Reaktionstemperatur abhängig und beträgt vorzugs weise 4-20 Stunden.
Falls erwünscht, kann in den verfahrensgemäss erhaltenen 4ss,19-Oxyden die oxigenierte C-19- Methylengruppe unter drastischeren Bedingungen weiter oxydiert werden. Dies kann z. B. geschehen, in dem man die genannten 4ss,19-Oxyde mit starken Oxydationsmitteln, z.
B. mit Rutheniumtetroxyd oder besonders mit Derivaten des 6-wertigen Chroms, wie Chromsäure oder tertiär-Butylchroma4 in Lösungs mitteln, z. B. niederen Fettsäuren, wie Essigsäure, Propionsäure, oder in chlorierten Kohleriwasserstof- fen, wie Tetrachlorkohlenstoff, insbesondere bei er höhter Temperatur, z. B. zwischen 50 und 100 C, behandelt.
Man erhält so durch Einführung einer Oxogruppe in 19-Stellüng Loktone von 4ss-Hydroxy- steroid-19-säuren.
Die Temperaturen sind .in den nachfolgenden Beispielen in Celsiusgraden angegeben. <I>Beispiel 1</I> Zu einer kurz auf 80 erwärmten Suspension von 5,0 g vorgetrocknetem Blei-(IV)-acetat und 1,50 g Calciumcarbonat in 150 ml Cyclohexan wer den 500 mg 4ss Hydroxy-17ss-propionyloxy-5a-andro- stan zugegeben.
Das Reaktionsgemisch wird anschlie ssend unter Rühren während 16 Stunden am Rück floss gekocht, dann abgekühlt, von anorganischen Anteilen abfiltriert, das Filtrat mit 50 ml einer 10 igen Kaliumjodid- und mit 50 ml einer 10 % igen Natriumthiosulfatlösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Man erhält 490 mg eines kristallinen Rohproduktes, das nach Dünnschichtchromatogramm aus drei Komponenten besteht.
Durch Chromatographie des Gemisches an Aluminiumoxyd (Aktivität II) werden 216 mg rei nes 4ss,19-Oxido-17ss-propionyloxyd-5a-androstan er halten, das nach Umlösen aus Methanol bei 109 bis 110g C schmilzt. [a]D = +22 ; im IR-Spektrum der Verbindung treten unter anderem Absorptions banden bei 5,78, 8,37, 9,26, 9,66, 9,82, 9,94, 10,02 und 11,90,u auf. Das NMR-Spektrum ist im Einklang mit der genannten Struktur der Verbindung.
Durch erneute Chromatographie der Mutterlaugen an Sili- cagel und anschliessende Kristallisation der entspre chenden Fraktionen werden weitere 130 mg des gleichen Produktes vom F. 112-114 neben etwa 10 % von 4-Oxo-17ss-propionyloxy-5a-androstan (F. 119-121') und etwa 15 % des amorphen 4a,9a- Oxido-17ss-propionyloxy-5fl-androstans gewonnen.
Das als Ausgangsstoff verwendete 4ss-Hydroxy- 17ss-propionyloxy-5a-androstan kann wie folgt her gestellt werden: 5,0 g 44-17ss-Propionyloxy-androstan werden in 75 ml Eisessig gelöst, mit 30 ml konzentrierter Sal petersäure und 3,0g Natriumnitrit umgesetzt, das an fallende d4-4-Nitro-17ss-propionyloxy-andros'tan ohne Reinigung in 34 ml Eisessig und 5 ml Wasser mit 10 g Zink während 4 Stunden bei 100 reduziert.
Durch chromatographische Reinigung des anfallen- den Rohproduktes an Aluminiumoxyd wird das reine, bei 119-121' schmelzende 4-Oxo@17ss-propiony 1'oxy- 5a-androstan ([ab = +3,5 ) gewonnen. Die kata lytische Hydrierung der Verbindung mit Platinoxyd in Eisessig liefert das 4ss-Hydroxy-17ss-propionyl'oxy- 5a-androstan,
das nach Umlösen aus Äther-Petrol- äther bei 157-159 schmilzt; [w]D = +5,1 .
<I>Beispiel 2</I> Unter den in Beispiel 1 angegebenen Bedingun gen wird aus 1,0 g 4ss-Hydroxy-5a brom-20ss-acet- oxy-pregnan neben etwa 5 % 4-Oxo-5a-brom-20ss- acetoxy-pregnan in 65-70%iger Ausbeute das 4ss,19- Oxido-5abrom-20ss-acetoxy-pregnan erhalten.
Das als Ausgangsstoff verwendete 4ss-Hydroxy- 5a-brom-20ss-acetoxy-pregnan wird z. B. aus d4-3- Oxo-20ss-acetoxy-pregnen durch Umsetzung mit Äthylendithioglykol, Entschwefelung des erhaltenen 3-Thioketals mit Natrium in flüssigem Ammoniak,
Nachacetylierung und Anlagerung von unterbromiger Säure an die d4-Doppelbindung durch Einwirkung von N-Brom-succinimid hergestellt.
<I>Beispiel 3</I> 500 mg 4ss,19-Oxido-17ss-propionyloxy-5a-andro- stan werden in 10 ml Eisessig während 30 Minuten bei 90 mit einer Lösung von 700 mg Chrom-(VI)- oxyd in 0,7 ml Wasser und 6 ml Eisessig behandelt. Das abgekühlte Reaktiohsgemisch wird anschliessend mit Wasser verdünnt und mit Methylenchlorid extra hiert.
Die Methylenchloridlösung wäscht man nach einander mit Wasser, gesättigter Natriumbicarbonat- lösung und mit Wasser, trocknet und dampft sie im Wasserstrahlvakuum ein.
Nach einmaligem Uirikri- stallisieren des Rohproduktes aus Methyfenchl'orid- Petroläther erhält man 210 mg reines 4ss,19-Lakton der 4ss-Hydroxy-17ss-propionyloxy-5a-androstan-19- säure vom F. 188-190 ; [a]D = -17 .
<I>Beispiel 4</I> 2,2 g vorgetrocknetes Blei-(VI)-acetat, 700 mg Bariumcarbonat und 200 mg 3ss,17ss-Diacetoxy-4ss- hydroxy-5a-chlor-androstan in 50 m1 Benzol werden unter Rühren während 16 Stunden am Rückfluss ge kocht.
Die abgekühlte Reaktionslösung wird an schliessend filtriert, der Rückstand mit etwa 10 ml Benzol nachgewaschen, die vereinigten Filtrate mit 10%iger Kaliumjodid- und Natriumthiosulfatlösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft.
Das erhaltene Rohprodukt wird an 30- facher Menge neutralem Aluminiumoxyd (Aktivität II) chromatographiert, wobei 125 mg reines 3ss,17ss- Diacetoxy-4ss,19-oxido-5a-chlor-androstan (charakte ristische IR Banden u. a. bei 5,78, 7,28, 8,15, 9,71 und 9,90 ,u) anfallen.

The process according to the invention consists in reacting 19 unsubstituted 4ss-hydroxy-5a-steroids with oxidizing lead acylates. If desired, the 4ss, 19-oxido steroids obtained can be oxidized.
Suitable starting materials for the present process are corresponding 4ss-hydroxy-5α-steroids of the androstan, pregnan, cholan, cholestan, stigma- stan, siprostan and cardanolid series, which are in the ring system, in particular may have further substituents in one or more of the positions 1, 2, 3, 5, 6, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 and 21,

B. the following compounds: 3ss, 17ss-diacetoxy-4ss-hydroxy-5a-androstane, 3ss, 17ss-diacetoxy-4P-hydroxy-17a-methyl-5a-androstane, the 3,5-carbonate of 3a, 4ss, 5a-trihydroxy-17-oxo-androstane, 3ss, 17ss-diacetoxy-ss-hydroxy-5a-chloro-androstane, 3ss, 17ss-diacetoxy-4ss-hydroxy-5a-bromo-androstane, 3ss, 17ss-diacetoxy-4ss- hydroxy-5a-chloro-17a-methyl-androstane, 3ss,
17ss-diacetoxy-4ss-hydroxy-5a-bromo-17a-methyl-androstane, 3P, 20ss-diacetoxy-4ss-hydroxy-5a-chfor-pregnan, 3ss, 20ss-DipTopionyloxy-4fl-hydroxy-5ahrom-pregnane, the 3ss-acetoxy -4ss-hydioxy-5a-chloro-20-oxo-pregnane, or 3ss-acetoxy-4ss-hydroxy-5a-chloro-spirostane. Particularly important starting materials for obtaining the 4ss unsubstituted in the 3-position,
19-oxido steroids are z. B. the 4ss-hydroxy-5a-steroids unsubstituted in the 3-position, such as 4ss-hydroxy-5a-halogeno-androstane, which have an oxo group or an esterified or etherified hydroxy group in the 17-position, e.g.
B. 4ss-Hydroxy-Sa-chloro-17-oxo-androstane, 17 esters of 4f, 17ss dihydroxy-5a-chloro-androstane, such as 17-acetate, 17-propionate or 17-benzoeth and 17-ester of 4ss, 17ss-di'hydroxy-5a-chloro-17a-methyl-androstane. The starting materials mentioned are known or, if new, can be prepared by known methods.
The 4f-hydroxy-a-halogen steroids can e.g. B. be produced by the addition of hypohalous acid to the corresponding 4,5-unsaturated compounds. 4ss-Hydroxy-steroids can also be particularly easily obtained by catalytic hydrogenation of the corresponding 4-ketones with a platinum catalyst in an acidic solution, e.g.
B. in glacial acetic acid or by hydrolytic cleavage of 4,5-epoxides.
The ver used for the inventive method oxidizing B'leiacylate are z. B. lead tetraacylates, the acid component of which is z.
B. derives from a lower aliphatic, cycloaNphatic, araliphatic or aromatic acid, such as acetic acid. Propionic, trifluoroacetic, hexahydrobenzoic, phenylacetic or benzoic acid. Furthermore, dialkyl lead diacylates, such as diethyl lead diacetate,
or anhydrides of metableic acid with the above carboxylic acids, such as diacetoxy lead oxide, can be used.
The procedural implementation is z. B. made so that .die starting materials, if necessary with the addition of a weak inorganic or organic base, z.
B. of Erdalkal'i- carbonates, such as calcium, barium or Ströntium carbonate, or of tertiary amines such as pyridine, together with an excess .an oxidizing lead acylate, preferably in an apalarous solvent inert to the oxidizing agent, eg .
B. heated to its boiling point, but preferably above 60 C, and the 4p, 19-oxido steroid obtained as process product is isolated by customary methods.
Particularly suitable solvents are aliphatic, cycloaliphatic and aromatic hydrocarbons, such as. B. hexane, heptane, cyclohexane, methylcyclohexane, dimethylcyclohexane or benzene.
The reaction time is generally dependent on the reaction temperature and is preferably 4-20 hours.
If desired, the oxygenated C-19 methylene group in the 4ss, 19-oxides obtained according to the process can be further oxidized under more drastic conditions. This can e.g. B. happen in which the said 4ss, 19-oxides with strong oxidizing agents, z.
B. with ruthenium tetroxide or especially with derivatives of 6-valent chromium, such as chromic acid or tertiary butyl chroma4 in solvents, z. B. lower fatty acids, such as acetic acid, propionic acid, or in chlorinated hydrocarbons, such as carbon tetrachloride, especially at he increased temperature, eg. B. between 50 and 100 C treated.
In this way, by introducing an oxo group in the 19-position, loktones of 4ss-hydroxy-19-acid are obtained.
In the examples below, the temperatures are given in degrees Celsius. <I> Example 1 </I> To a suspension, briefly heated to 80, of 5.0 g of predried lead (IV) acetate and 1.50 g of calcium carbonate in 150 ml of cyclohexane, 500 mg of 4ss hydroxy-17ss-propionyloxy- 5a-androstane was added.
The reaction mixture is then refluxed with stirring for 16 hours, then cooled, inorganic components are filtered off, the filtrate is washed with 50 ml of 10% potassium iodide and 50 ml of 10% sodium thiosulphate, dried and evaporated in a water-jet vacuum. 490 mg of a crystalline crude product are obtained which, according to thin-layer chromatography, consists of three components.
By chromatography of the mixture of aluminum oxide (activity II), 216 mg of pure 4ss, 19-oxido-17ss-propionyloxyd-5a-androstane are kept, which melts at 109 to 110g C after dissolving from methanol. [a] D = +22; absorption bands at 5.78, 8.37, 9.26, 9.66, 9.82, 9.94, 10.02 and 11.90 u occur in the IR spectrum of the compound. The NMR spectrum is consistent with the stated structure of the compound.
By renewed chromatography of the mother liquors on silica gel and subsequent crystallization of the appropriate fractions, a further 130 mg of the same product from F. 112-114 in addition to about 10% of 4-oxo-17ss-propionyloxy-5a-androstane (F. 119- 121 ') and about 15% of the amorphous 4a, 9a-oxido-17ss-propionyloxy-5fl-androstane obtained.
The 4ss-hydroxy-17ss-propionyloxy-5a-androstane used as starting material can be made as follows: 5.0 g of 44-17ss-propionyloxy-androstane are dissolved in 75 ml of glacial acetic acid, with 30 ml of concentrated nitric acid and 3.0 g Sodium nitrite reacted, the d4-4-nitro-17ss-propionyloxy-andros'tan which is obtained is reduced in 34 ml of glacial acetic acid and 5 ml of water with 10 g of zinc for 4 hours at 100 without purification.
The pure 4-oxo @ 17ss-propiony 1'oxy-5a-androstane ([ab = +3.5), which melts at 119-121 ', is obtained by chromatographic purification of the resulting crude product on aluminum oxide. The catalytic hydrogenation of the compound with platinum oxide in glacial acetic acid yields 4ss-hydroxy-17ss-propionyl'oxy-5a-androstane,
which melts at 157-159 after dissolving from ether-petroleum ether; [w] D = +5.1.
<I> Example 2 </I> Under the conditions given in Example 1, 1.0 g of 4ss-hydroxy-5a becomes bromo-20ss-acetoxy-pregnane in addition to about 5% 4-oxo-5a-bromo-20ss Acetoxy-pregnane obtained in 65-70% yield the 4ss, 19-oxido-5abromo-20ss-acetoxy-pregnane.
The 4ss-hydroxy-5a-bromo-20ss-acetoxy-pregnane used as starting material is z. B. from d4-3- Oxo-20ss-acetoxy-pregnen by reaction with ethylene dithioglycol, desulfurization of the 3-thioketal obtained with sodium in liquid ammonia,
Post-acetylation and addition of hypobromous acid to the d4 double bond produced by the action of N-bromosuccinimide.
<I> Example 3 </I> 500 mg of 4ss, 19-oxido-17ss-propionyloxy-5a-androstane are in 10 ml of glacial acetic acid for 30 minutes at 90 with a solution of 700 mg of chromium (VI) oxide in Treated 0.7 ml of water and 6 ml of glacial acetic acid. The cooled reaction mixture is then diluted with water and extracted with methylene chloride.
The methylene chloride solution is washed successively with water, saturated sodium bicarbonate solution and with water, dried and evaporated in a water-jet vacuum.
After a single uricrystallization of the crude product from methylene chloride petroleum ether, 210 mg of pure 4ss, 19-lactone of 4ss-hydroxy-17ss-propionyloxy-5a-androstane-19-acid with a melting point of 188-190; [a] D = -17.
<I> Example 4 </I> 2.2 g of predried lead (VI) acetate, 700 mg of barium carbonate and 200 mg of 3ss, 17ss-diacetoxy-4ss-hydroxy-5a-chloro-androstane in 50 ml of benzene are added with stirring refluxed for 16 hours.
The cooled reaction solution is then filtered, the residue is washed with about 10 ml of benzene, and the combined filtrates are washed with 10% strength potassium iodide and sodium thiosulphate solution, dried and evaporated in a water jet vacuum.
The crude product obtained is chromatographed on 30 times the amount of neutral aluminum oxide (activity II), with 125 mg of pure 3ss, 17ss-diacetoxy-4ss, 19-oxido-5a-chloro-androstane (characteristic IR bands, inter alia at 5.78, 7 , 28, 8.15, 9.71 and 9.90, u).
