Beispiel 1
Man löst 520 mg Bleitetraacetat (enthaltend 10-15% freie Essigsäure) in 10 ml abs.

13. The method according to claim 12, characterized in that at least one of the tautomers is isolated from the tautomer mixture obtained.
The present invention relates to a new process for the preparation of 11ss, 1 8-epoxy-18,21-dihydroxy-20-oxo-pregnane compounds or their 21-esters. This class of compounds, which can also be mentioned as those of the 11ss, 21-dihydroxy-20-oxo-pregnan-18-aldehydes, of which the 11ss, 18-epoxy-18,21-dihydroxy-20-oxo-pregnanes mentioned Compounds which are tautomeric forms are known pharmacologically active compounds, such as aldosterone and its derivatives, especially its esters.
Among the various syntheses of aldosterone and its derivatives described are those based on the lactone of the formula
EMI1.3
of particular importance, since this compound can be prepared relatively easily from the 11c-hydroxy-progesterone available in technical quantities. Of these syntheses, the one which is particularly suitable for the technical preparation of aldosterone is the one which leads to the end product via the 20,21-enol ether of the formula IV and the secondary products V-VII, in accordance with the following reaction scheme:

This method is e.g. in English Patent 1,080,327, where the compounds of the formula III of the above scheme figure as starting products. A particular advantage of this process is that the starting materials III mentioned can be prepared in situ using the same reagent (for example diisobutylaluminum hydride) which is also used in the further stage III-IV, so that the stage sequence II-IV as One-pot procedure can be performed.
The conversion of the 20.21 enol ether IV into the aldosterone-3-ketal V is carried out according to the process of the English patent mentioned by treatment with hydroxylating agents, such as osmium tetroxide, ruthenium tetroxide or manganese dioxide, tungsten trioxide or vanadium pentoxide, or with epoxidizing agents, such as organic peracids, and subsequent hydrolysis or
Acylolysis.
It has now been found that e.g. the compound IV can be converted directly and in good yield into the 3-ethylene ketal VI by treatment with acyloxylating heavy metal salts of the type of lead tetraacetate, which brings about further progress in the industrial production of aldosterone and its derivatives. The use of the heavy metal salts with an acyloxylation effect, in particular lead tetraacetate, instead of the expensive, volatile and toxic osmium tetroxide is of great advantage both in terms of operational engineering and in terms of ecology. Working up the reaction products is also very easy and convenient since no difficult-to-separate mixtures are formed.
A smooth transfer of IV into the 21 acylaldosterone ketal VI according to the present process is surprising in view of the diversity with which the oxidizing agents involved can react with substrates of this type.
The process according to the invention for the preparation of a tautomer mixture comprising an 11ss, 18-epoxy-18 hydroxy-2 1 -acyloxy-2O-oxo-pregnane compound and an 11 (3-hydroxy-2 1 -acyloxy-20-oxo-pregnane 1 8-aldehyde compound, is characterized in that a corresponding 11 (3.18; 11 ss, 18; 18.20-bisepoxy-pregn-20-ene compound in an inert solvent or suspending agent The corresponding 21 hydroxy compounds, which include aldosterone and derivatives thereof, can be obtained from the 21-acylates thus obtained by alkaline hydrolysis of the 21-acyloxy group in a manner known per se.
118.18 claims particular interest as starting materials; 18,20-Bisepoxy-pregn-20-ene compounds which have further substituents and / or double bonds in the steroid skeleton.
Particularly preferred starting materials are those of the formula (I)
EMI3.1
into consideration, in which the H atom is in the 5-position in a or (3-position and also a double bond, either in the 4.5- or in the 5.6-position, and in which R is a free or protected hydroxyl group with hydrogen or a free or protected oxo group, and in particular the known compounds of the formula
EMI3.2
and in particular their 3-ketals, primarily the 3-ethylene ketal, which lead to aldosterone esters or their ketals by using the process of the present invention.
A protected hydroxy group is in particular an esterified or etherified hydroxy group, a protected oxo group in particular a ketalized oxo group. Preferred among the esters are those derived from carboxylic acids with 1-18 C atoms, e.g. of aliphatic, aromatic, araliphatic or heterocyclic acids, and in particular of lower aliphatic carboxylic acids with 17 carbon atoms, such as acetic acid, propionic acid, butyric acids, valeric acids or caproic acids. Among the ester groups derived from aromatic acids, benzoates should be mentioned in particular.
Among the ether groups, particular mention should be made of those derived from lower aliphatic alkanols with 1-7 C atoms, or from monocyclic aryl-lower aliphatic alcohols, such as the phenyl-lower alkyl alcohols, e.g. the benzyl or phenethyl alcohol, or of heterocyclic alcohols of the tetrahydrofuranol or tetrahydropyranol type, or of tri-lower alkylsilanols.
Among the ketalized oxo groups, those which derive from lower alkanediols, in particular ethylenedioxy and propylenedioxy groups, are to be emphasized.
Heavy metal acylates with acyloxylating activity include Lead (IV), mercury (II), cerium (IV) and thallium (III) acylates are considered. In particular, the acylates of lower aliphatic carboxylic acids with 1-7 C atoms, such as the acetates, propionates, butyrates, valerianates or capronates or in particular also trihaloacetates, such as trifluoroacetates, or acylates of monocyclic aromatic carboxylic acids, such as benzoic acid or homologs, are used.
A preferred reagent is lead tetraacetate.
The reaction of the starting materials mentioned with the above-mentioned agents takes place in an inert solvent or suspension medium, optionally with the addition of an acid or a base.
Brnstedt or Lewis acid or mixtures thereof are used as acids; preferably the same carboxylic acid is used which corresponds to the acid residue of the acylate used, e.g. Acetic acid if the reaction is carried out with lead tetraacetate. Tertiary nitrogen bases of aromatic character, such as e.g. Pyridine, collidine or morpholine are used. In many cases weaker bases, such as some of the solvents listed below, e.g. Ethers such as dioxane or tetrahydrofuran.
Suitable inert solvents are e.g. aliphatic or aromatic or cycloaliphatic hydrocarbons or their halogenated, especially chlorinated derivatives, such as e.g.
the higher liquid paraffins, chlorinated paraffins, such as methylene chloride, ethylene chloride or chloroform, ¯ cyclohexane and its homologues, benzene and its homologues, chlorobenzene and chlorotoluenes, ethers, such as the lower aliphatic ethers, dioxane, tetrahydrofuran, esters, such as those of lower aliphatic acids and lower aliphatic alcohols, e.g. Ethyl acetate, butyl acetate or amyl acetate or ketones such as the lower aliphatic ketones e.g. Acetone or methyl ketone or the usual cycloaliphatic ketones used as solvents, such as cyclopentanone or cyclohexanone.
The reaction is advantageously carried out at room temperature or at a lower temperature up to 200. However, it also takes place at elevated temperature, so that the method can also be used e.g. can run up to about 100o. Depending on the starting material, the reaction time lasts from a few minutes to several hours.
The workup is expediently carried out in a manner known per se: after the reaction is complete, e.g. filtered off from the oxidizing agent, the organic solution washed with water or aqueous salt solutions or with bases to neutralize existing acids, the solvent evaporated and the residue in a conventional manner, e.g. isolated by chromatography or crystallization.
If the reaction mixtures are worked up carefully while avoiding alkaline conditions, the desired 21-hydroxylated process products are generally obtained in the form of the 21-acylates. From the 21-acylates
21-hydroxy group, preferably after the ketal group has been released in the 3-position, e.g. by acid hydrolysis in a manner known per se, in a manner also known, e.g. be set free by alkaline saponification.
The starting materials are known or can be prepared by methods known per se.
The following examples illustrate the invention. The temperatures are given in degrees Celsius.
example 1
520 mg of lead tetraacetate (containing 10-15% free acetic acid) are dissolved in 10 ml of abs. Benzene, to which 1 ml of glacial acetic acid has been added, and adds with stirring at room temperature
200 mg 3,3-ethylenedioxy llss, 18; 18, 20-bisepoxy-pregna5,20-diene (mp. 2122160), whereby a clear solution is formed. After a few minutes, a white precipitate falls out.
After about 10 minutes, the mixture is poured onto ice, extracted with ethyl acetate, the organic phase is separated off and washed 3 times with saturated sodium chloride solution and then with water. After the aqueous phase has been separated off, the organic extract is dried over sodium sulfate, evaporated in vacuo and the product is converted into 21-O-acetal aldosterone (mp. 1N84-188.50) by acid hydrolysis, which is known per se in aldosterone is converted.
Example 2
An anhydrous solution of 185 mg of 3,3-ethylenedioxy 118.18; 18.20-bisepoxy-pregna-5.20-diene in 10 ml of benzene is added in a dry nitrogen atmosphere, with rinsing with 1 ml of 100 percent. Acetic acid, with 500 mg lead tetraacetate. After stirring for 4 hours at room temperature, the reaction solution is poured off from the excreted lead diacetate, excess lead tetraacetate after dilution with benzene and a lot of ether is destroyed by shaking with aqueous 0.1 molar oxalic acid solution, the organic phase is further washed with water, dried with sodium sulfate and evaporated. The almost colorless, crystalline residue is crude 3,3-ethylenedioxy-11 ss, 18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate, which is subjected to ketal cleavage without further purification .
235 mg of crude 3,3-ethylenedioxy-1 1 (3,18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate are dissolved in 5 ml of 90 percent.
aqueous acetic acid heated to 97-100o for 10 minutes. The solution is then cooled to about 25 ° and the reaction solution is evaporated in vacuo with the addition of toluene. The acid-free residue is dissolved in chloroform and 12.5 g
Silica gel (containing 10% water) with chloroform-acetone
Mixtures of increasing acetone content separated by the continuous method. The 11B, 18-epoxy-18,21 11 (3,18-epoxy-18,21-dihydroxy-pregn-4-en-3, 20-dione-2) is obtained by redissolving the thin-layer chromatographically practically uniform middle fractions from acetone 1-acetate (2 1-O-acetyl-aldosterone) from
F. 195-1980.
Example 3
370.5 mg of 3,3-ethylenedioxy-11 1 (3.18; 18.20-bisepoxy-pregna-5.20-diene) placed in a dry nitrogen atmosphere are anhydrous in 5 ml with exclusion of moisture
Dissolved methylene chloride and then with rapid stirring
560 mg of lead tetraacetate free of acetic acid are added. A milky cloudiness forms immediately, which changes to an almost colorless, crystalline deposition after a few minutes. After 35
For minutes, the reaction mixture is poured onto a 0.1 molar aqueous solution of both sodium thiosulfate and sodium iodide and ice and extracted with ethyl acetate. The extracts washed with 1 molar ammonium hydrogen carbonate and water are combined with
Dried sodium sulfate, filtered and evaporated.
The crude oxidation product which remains, the 3,3-ethylenedioxy-1 1 ss, 18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate, crystallizes completely from ether and becomes without further Cleaning subjected to ketal splitting.
The crude oxidation product obtained according to the above information (495 mg) is poured with 7.5 ml of 67 percent.
aqueous acetic acid and first heated under 1 hour
Stir in a bath of 62-650. The reaction solution is then diluted at 62-65o im with continued stirring
Over a period of 11/2 hours with 17.5 ml of water and then cools down to 12-15o. The crude hydrolysis product which crystallized out weighed 315 mg after drying.
Column chromatography on 30 g of silica gel (containing 10% water), elution with methylene chloride-acetone mixtures of increasing acetone content and recrystallization of the thin-layer chromatographically uniform fractions from methylene chloride ether gives 267 mg of pure 11 (3,18-epoxy-18,21-dihy - droxy-pregn-4-en-3,20-dione-21-acetate (21-O-acetylaldosterone) of F. 195-1980.
The extractive work-up of the aqueous mother liquor of the ketal cleavage described above results in a further one
Amount of crystalline hydrolysis product which, together with the still impure fractions from the above chromatogram after renewed column chromatography on silica gel, using toluene-chloroform mixtures, further amounts of pure 11pp, 18-epoxy-18,21-dihydroxypregn-4-en-3 , 20-dione-21-acetate (2 1-O-acetyl-aldosterone) from F. 195-19801.
