<tb> Dieses Verfahren ist z.B. im Englischen Patent 1 080 327 beschrieben, wobei dort die Verbindungen der Formel III des obigen Schemas als Ausgangsprodukte figurieren.
Ein besonderer Vorteil dieses Verfahrens ist, dass die genannten Ausgangsstoffe III in situ unter Verwendung des gleichen Reagenz (z.B. Diisobutylaluminiumhydrid) hergestellt werden können, das auch bei der weiteren Stufe III-IV zur Anwendung gelangt, sodass die Stufenfolge II-IV als Eintopf-Verfahren ausgeführt werden kann. Die Überführung des 20,21-Enoläthers IV in das Aldosteron-3-ketal V wird gemäss dem Verfahren des genannten Englischen Patentes durch Behandlung mit hydroxylierenden Mitteln, wie Osmiumtetroxid, Rutheniumtetroxid oder Mangandioxid, Wolframtrioxid oder Vanadiumpentoxid, oder mit epoxidierenden Mitteln, wie organischen Persäuren, und nachfolgende Hydrolyse oder Acylolyse, vorgenommen.
Es wurde nun gemäss der vorliegenden Erfindung gefunden, dass z.B. das Steroid der Formel IV durch Behandeln mit acyloxylierend wirkenden Schwermetallsalzen vom Typ des Bleitetraacetats direkt und in guter Ausbeute in das 3-Aethylenketal VI übergeführt werden kann, was einen weiteren Fortschritt in der technischen Herstellung von Aldosteron und dessen Derivaten mit sich bringt. Die Verwendung der acyloxylierend wirkenden Schwermetallsalze, insbesondere von Bleitetraacetat, anstelle des teuren, flüchtigen und giftigen Osmiumtetroxids ist sowohl in allgemein betriebstechnischer Hinsicht wie auch in bezug auf die Oekologie von grossem Vorteil. Ferner ist die Aufarbeitung der Reaktionsprodukte sehr leicht und bequem, da keine schwer zu trennenden Gemische entstehen.
Eine glatte Überführung der Verbindung IV in das 21-Acylaldosteronketal VI gemäss dem vorliegenden Verfahren ist in Anbetracht der Verschiedenartigkeit, mit welcher die involvierten Oxydationsmittel mit Substraten dieser Art reagieren können, überraschend.
Das erfindungsgemässe Verfahren ist somit dadurch gekennzeichnet, dass man auf eine 11ss, 18; 18, 20-Bisepoxypregn-20-en-Verbindung der Formel

worin Rl und die übrigen Zeichen die für Formel (A1) erläuterte Bedeutung besitzen, in einem inerten Lösungs- oder Suspensionsmittel, ein acyloxylierend wirkendes Schwermetallacylat einwirken lässt. Falls erwünscht, kann in der so erhaltenen 21 Acyloxy- 1 8-hydroxy- 11(3-1 8-epoxy-20-oxo-pregnan-Verbin- dung die 21-Acyloxygruppe in die freie Hydroxygruppe übergeführt werden.
Veresterte Hydroxygruppen R1 sind vorzugsweise solche, die sich von Carbonsäuren mit 1-18 C-Atomen ableiten, z.B.
von aliphatischen, aromatischen, araliphatischen oder heterocy clischen Säuren, und insbesondere von niederen aliphatischen Carbonsäuren mit 1-7 C-Atomen, wie Essigsäure, Propionsäure, den Buttersäuren, Valeriansäuren oder Capronsäuren.
Unter den von aromatischen Säuren abgeleiteten Estergruppen sind insbesondere die Benzoate zu nennen. Unter den Aethergruppen sind insbesondere diejenigen hervorzuheben, die sich von niederen aliphatischen Alkanolen mit 1-7 C-Atomen ableiten, oder von monocyclischen Aryl-mederaliphatischen Alkoholen, wie den Phenyl- niederalkyl-Alkoholen, z.B. dem Benzyl- oder Phenäthylalkohol, oder von heterocyclischen Alkoholen vom Tetrahydrofuranol- oder Tetrahydropyranol Typ, oder von Tri-niederalkylsilanolen.
Unter den ketalisierten Oxogruppen sind insbesondere diejenigen hervorzuheben, die sich von niederen Alkandiolen ableiten, insbesondere Aethylendioxy- und Propylendioxygruppen.
Zur Darstellung von Aldosteron und seinen Derivaten, oder von 11-Dehydroaldosteron und seinen Estern, dienen vor allem Verbindungen der Formel

und insbesondere deren 3-Ketale, in erster Linie das 3-Aethylenketal, die durch Anwendung des Verfahrens der vorliegenden Erfindung zu Aldosteronestern bzw. deren Ketalen führen.
Als acyloxylierend wirkende Schwermetallacylate kommen z.B. Blei (IV)-, Quecksilber (II)-, Cer (IV)- und Thallium (III)acylate in Betracht. Man verwendet insbesondere die Acylate von niederen aliphatischen Carbonsäuren mit 1-7 C-Atomen, wie die Acetate, Propionate, Butyrate, Valerianate oder Capronate oder insbesondere auch Trihalogenacetate, wie Trifluoracetate, oder Acylate von monocyclischen aromatischen Carbonsäuren, wie der Benzoesäure oder Homologen. Ein bevorzugtes Reagenz ist das Bleitetraacetat.
Die Umsetzung der genannten Ausgangsstoffe mit den oben genannten Agenzien erfolgt in einem inerten Lösungs- oder Suspensionsmittel, gegebenenfalls unter Zusatz einer Säure oder einer Base.
Als Säuren verwendet man eine Brönstedt- oder Lewissäure oder Gemische derselben; vorzugsweise verwendet man die gleiche Carbonsäure, welche dem Säurerest des verwendeten Acylats entspricht, also z.B. Essigsäure, wenn die Umsetzung mit Bleitetraacetat vorgenommen wird. Als Basen werden vorzugsweise tertiäre Stickstoffbasen aromatischen Charakters, wie z.B. Pyridin, Kollidin oder Morpholin verwendet. In vielen Fällen genügen auch schwächere Basen, wie einige der unten angeführten Lösungsmittel, z.B. Aether, wie Dioxan oder Tetrahydrofuran.
Passende inerte Lösungsmittel sind z.B. aliphatische oder aromatische oder cycloaliphatische Kohlenwasserstoffe oder ihre halogenierten, insbesondere chlorierten Derivate, wie z.B.
die höheren flüssigen Paraffine, chlorierte Paraffine, wie Methylenchlorid, Aethylenchlorid oder Chloroform, Cyclohexan und seine Homologen, Benzol und seine Homologen, Chlorbenzol und Chlortoluole, Aether, wie die niederen aliphatischen Aether, Dioxan, Tetrahydrofuran, Ester, wie diejenigen von niederen aliphatischen Säuren und niederen aliphatischen Alkoholen, z.B. Essigester, Butylacetat oder Amylacetat oder Ketone, wie die niederen aliphatischen Ketone, z.B. Aceton oder Methylketon oder die üblichen als Lösungsmittel verwendeten cycloaliphatischen Ketone, wie Cyclopentanon oder Cyclohexanon.
Die Reaktion wird vorteilhaft bei Raumtemperatur oder bei tieferer Temperatur, z.B. bis zu - 20", durchgeführt. Sie findet jedoch auch bei erhöhter Temperatur statt, sodass man das Verfahren auch z.B. bis zu ca. 100" ausführen kann. Die Reaktionszeit dauert je nach Ausgangsstoff von einigen Minuten bis zu mehreren Stunden.
Die Aufarbeitung findet in der Regel in an sich bekannter Weise statt: nach vollendeter Reaktion wird. z.B. vom Oxydationsmittel abfiltriert, die organische Lösung mit Wasser oder wässerigen Salz-Lösungen bzw. zur Neutralisation vorhandener Säuren mit Basen gewaschen, das Lösungsmittel eingedampft und den Rückstand in üblicher Weise, z.B. durch Chromatographie oder Kristallisation, isoliert.
Bei schonender Aufarbeitung der Reaktionsgemische unter Vermeidung alkalischer Bedingungen werden im allgemeinen die gewünschten 21-hydroxylierten Verfahrensprodukte in Form der 21-Acylate erhalten. Aus den 21-Acylaten kann die 21-Hydroxygruppe, vorzugsweise nach Freisetzung der Ketalgruppe in 3-Stellung, z.B. durch saure Hydrolyse in an sich bekannter Weise, oder in ebenfalls bekannter Weise, z.B. durch alkalische Verseifung, in Freiheit gesetzt werden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden hergestellt werden.
Die folgenden Beispiele erläutern die Erfindung. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Man löst 520 mg Bleitetraacetat (enthaltend 10-15% freie Essigsäure) in 10 ml abs. Benzol, denen man 1 ml Eisessig zugesetzt hat, und fügt unter Rühren bei Zimmertemperatur 200 mg 3,3-Aethylendioxy-llss,18; 18, 20-bisepoxy-pregna5,20-dien (Smp. 212-216") zu, wobei eine klare Lösung entsteht. Nach einigen Minuten fällt ein weisser Niederschlag aus.
Nach ca. 10 Minuten giesst man auf Eis, extrahiert mit Essigester, trennt die organische Phase ab und wäscht sie je 3 mal mit gesättigter Kochsalz-Lösung und dann mit Wasser. Nach dem Abtrennen der wässrigen Phase trocknet man den organischen Extrakt über Natriumsulfat, dampft im Vakuum ein und führt das Produkt durch saure Hydrolyse in 21-O-Acetyl-aldosteron (Smp. 184-188, 50) über, welches in an sich bekannter Weise in Aldosteron umgewandelt wird.
Beispiel 2
Eine wasserfreie Lösung von 185 mg 3,3-Aethylendioxy 11,ss,18; 18,20-bisepoxy-pregna-5,20-dien in 10 ml Benzol versetzt man in trockener Stickstoffatmosphäre, unter Nachspülen mit 1 ml 100-proz. Essigsäure, mit 500 mg Bleitetraacetat. Nach 4-stündigem Rühren bei Raumtemperatur wird die Reaktionslösung vom ausgeschiedenen Bleidiacetat abgegossen, überschüssiges Bleitetraacetat nach Verdünnen mit Benzol und viel Aether durch Schütteln mit wässriger 0,1-molarer Oxalsäurelösung zerstört, die organische Phase weiter mit Wasser gewaschen, mit Natrium-Sulfat getrocknet und eingedampft. Der nahezu farblose, kristalline Rückstand stellt rohes 3,3-Aethy lendioxy-llss,18-epoxy-18,21-dihydroxy-pregn-5-en-20-on- 21-acetat dar, das ohne weitere Reinigung der Ketalspaltung unterworfen wird.
235 mg rohes 3,3-Aethylendioxy-1 18,18-epoxy-18,21- dihydroxy-pregn-5-en-20-on-21-acetat werden in 5 ml 90-proz.
wässriger Essigsäure 10 Minuten auf 97-100" erhitzt. Man kühlt die Lösung alsdann auf ca. 25" ab und dampft die Reaktionslösung unter Zusetzen von Toluoyl im Vakuum ein. Der säurefreie Rückstand wird in Chloroform gelöst und an 12,5 g Kieselgel (enthaltend 10% Wasser) mit Chloroform-Aceton-Gemischen von steigendem Acetongehalt nach der Durchlaufmethode aufgetrennt.
Durch Umlösen der dünnschichtchromatographisch praktisch einheitlichen mittleren Fraktionen aus Aceton gewinnt man das 11B,18-Epoxy-18,21 -dihydroxy-pregn-4-en- 3 ,20-dion-2 1 -acetat (21 -O-Acetyl-aldosteron) vom F. 195 198ob
Beispiel 3
In trockener Stickstoffatmosphäre vorgelegte 370,5 mg 3,3 Aethylendioxy-11S, 18 ;18,20-bisepoxy-pregna-5,20-dien wer den unter Feuchtigkeitsausschluss in 5 ml wasserfreiem Methy lenchlorid gelöst und alsdann unter raschem Rühren 560 mg essigsäurefreies Bleitetraacetat zugegeben. Es bildet sich sofort eine milchige Trübung, die nach wenigen Minuten in eine nahezu farblose, kristalline Deposition übergeht.
Nach 35 Minuten langem Rühren giesst man das Reaktionsgut auf eine sowohl an Natriumthiosulfat als auch an Natriumjodid 0,1molare wässrige Lösung und Eis und extrahiert mit Essigsäure äthylester. Die mit 1-molarer Ammoniumhydrogencarbonat und Wasser gewaschenen Auszüge werden vereinigt, mit Natriumsulfat getrocknet, filtriert und eingedampft. Das zurückbleibende rohe Oxydationsprodukt, das 3,3-Aethylen- dioxy-ll B, 18-epoxy-18,21 -dihydroxy-pregn-5-en-20-on- 21-acetat, kristallisiert vollständig aus Aether und wird ohne weitere Reinigung der Ketalspaltung unterworfen.
Das gemäss den obigen Angaben erhaltene rohe Oxydationsprodukt (495 mg) übergiesst man mit 7,5 ml 67-proz.wässriger Essigsäure und erwärmt zunächst 1 Stunde unter Rühren in einem Bad von 62-65 . Unter fortgesetztem Rühren verdünnt man alsdann die Reaktionslösung bei 62-650 im Verlauf von 1Y2 Stunden mit 17,5 ml Wasser und kühlt hierauf auf 12-15 ab.
Das dabei auskristallisierende rohe Hydrolyseprodukt wiegt nach dem Trocknen 315 mg. Durch Säulenchromatographie an 30 g Kieselgel (enthaltend 10% Wasser), Eluieren mit Methylenchlorid-Aceton-Gemischen von steigendem Acetongehalt und Umkristallisieren der dünnschichtchromatographisch einheitlichen Fraktionen aus Methylenchlorid-Aether gewinnt man 267 mg reines 118,18-Epoxy-18,21-dihydroxy-pregn-4-en- 3,20-dion-21-acetat (21 -O-Acetyl-aldosteron) vom F. 195 198".
Die extraktive Aufarbeitung der wässrigen Mutterlauge der vorstehend beschriebenen Ketalspaltung ergibt eine weitere Menge an kristallinem Hydrolyseprodukt welche zusammen mit den noch unreinen Anteilen au.r obigem Chromatogramm nach erneuter säulenchromatographischer Auftrennung an Kieselgel, unter Verwendung von Toluol-Chloroform-Gemischen, weitere Mengen reines 11ss, 18-Epoxy-18,21-dihydroxy-pregn-4-en3,20-dion-21-acetat (21-O-Acetyl-aldosteron) vom F. 195 198 liefert.

Heavy metal acylates with an acyloxylating effect are e.g. Lead (IV) -, mercury (II) -, cerium (IV) - and thallium (III) acylate into consideration. In particular, the acylates of lower aliphatic carboxylic acids with 1-7 carbon atoms, such as the acetates, propionates, butyrates, valerianates or capronates, or especially trihaloacetates, such as trifluoroacetates, or acylates of monocyclic aromatic carboxylic acids, such as benzoic acid or homologs, are used. the higher liquid paraffins, chlorinated paraffins such as methylene chloride, ethylene chloride or chloroform, cyclohexane and its homologues, benzene and its homologues, chlorobenzene and chlorotoluenes, ethers such as the lower aliphatic ethers, dioxane, tetrahydrofuran, esters such as those of lower aliphatic acids and lower aliphatic alcohols, e.g. Ethyl acetate, butyl acetate or amyl acetate or ketones, such as the lower aliphatic ketones, e.g. Acetone or methyl ketone or the usual cycloaliphatic ketones used as solvents, such as cyclopentanone or cyclohexanone.
The reaction is advantageously carried out at room temperature or at lower temperature, e.g. up to - 20 ". However, it also takes place at an elevated temperature so that the process can also be carried out, for example, up to approx. 100". The reaction time takes from a few minutes to several hours, depending on the starting material.
The work-up usually takes place in a manner known per se: after the reaction has ended, becomes. e.g. the oxidizing agent is filtered off, the organic solution is washed with water or aqueous salt solutions or with bases to neutralize any acids present, the solvent is evaporated and the residue is removed in the usual manner, e.g. by chromatography or crystallization.
If the reaction mixtures are worked up gently while avoiding alkaline conditions, the desired 21-hydroxylated process products are generally obtained in the form of the 21-acylates. From the 21-acylates, the 21-hydroxyl group can, preferably after liberation of the ketal group in the 3-position, e.g. by acid hydrolysis in a manner known per se, or in a manner also known, e.g. by alkaline saponification.
The starting materials are known or can be prepared by methods known per se.
The following examples illustrate the invention. The temperatures are given in degrees Celsius.
example 1
520 mg of lead tetraacetate (containing 10-15% free acetic acid) are dissolved in 10 ml of abs. Benzene, to which 1 ml of glacial acetic acid has been added, and 200 mg of 3,3-ethylenedioxy-llss, 18; 18, 20-bisepoxy-pregna5,20-diene (melting point 212-216 "), a clear solution being formed. After a few minutes, a white precipitate separates out.
After about 10 minutes, it is poured onto ice, extracted with ethyl acetate, the organic phase is separated off and washed 3 times each with saturated sodium chloride solution and then with water. After the aqueous phase has been separated off, the organic extract is dried over sodium sulfate, evaporated in vacuo and the product is converted into 21-O-acetyl-aldosterone (melting point 184-188, 50) by acid hydrolysis, which in a manner known per se is converted into aldosterone.
Example 2
An anhydrous solution of 185 mg of 3,3-ethylenedioxy 11, ss, 18; 18,20-bisepoxy-pregna-5,20-diene in 10 ml of benzene is added in a dry nitrogen atmosphere, while rinsing with 1 ml of 100 percent. Acetic acid, with 500 mg lead tetraacetate. After 4 hours of stirring at room temperature, the reaction solution is poured off from the precipitated lead diacetate, excess lead tetraacetate is destroyed after dilution with benzene and a lot of ether by shaking with 0.1 molar aqueous oxalic acid solution, the organic phase is washed further with water, dried with sodium sulfate and evaporated. The almost colorless, crystalline residue is crude 3,3-Aethy lendioxy-llss, 18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate, which is subjected to ketal cleavage without further purification .
235 mg of crude 3,3-ethylenedioxy-1 18,18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate are dissolved in 5 ml of 90 percent strength.
aqueous acetic acid heated to 97-100 "for 10 minutes. The solution is then cooled to about 25" and the reaction solution is evaporated in vacuo with the addition of toluoyl. The acid-free residue is dissolved in chloroform and separated on 12.5 g of silica gel (containing 10% water) with chloroform-acetone mixtures of increasing acetone content by the continuous flow method.
The 11B, 18-epoxy-18,21-dihydroxy-pregn-4-en-3, 20-dione-2 1 -acetate (21-O-acetyl-aldosterone) is obtained by redissolving the middle fractions from acetone, which are practically uniform by thin-layer chromatography. from F. 195 198ob
Example 3
370.5 mg of 3,3 ethylenedioxy-11S, 18; 18,20-bisepoxy-pregna-5,20-diene placed in a dry nitrogen atmosphere were dissolved in 5 ml of anhydrous methylene chloride with the exclusion of moisture and then 560 mg of acetic acid-free lead tetraacetate with rapid stirring admitted. A milky cloudiness forms immediately, which changes into an almost colorless, crystalline deposit after a few minutes.
After stirring for 35 minutes, the reaction mixture is poured into a 0.1 molar aqueous solution of both sodium thiosulphate and sodium iodide and ice and extracted with ethyl acetate. The extracts washed with 1 molar ammonium hydrogen carbonate and water are combined, dried with sodium sulfate, filtered and evaporated. The remaining crude oxidation product, the 3,3-ethylene-dioxy-II B, 18-epoxy-18,21-dihydroxy-pregn-5-en-20-one-21-acetate, crystallizes completely from ether and becomes without further purification subjected to ketal cleavage.
The crude oxidation product (495 mg) obtained according to the above information is poured over with 7.5 ml of 67 percent aqueous acetic acid and initially heated for 1 hour in a bath of 62-65 with stirring. The reaction solution is then diluted with 17.5 ml of water at 62-650 in the course of 1½ hours with continued stirring and then cooled to 12-15.
The crude hydrolysis product which crystallizes out weighs 315 mg after drying. Column chromatography on 30 g of silica gel (containing 10% water), eluting with methylene chloride-acetone mixtures of increasing acetone content and recrystallization of the fractions uniformly based on thin-layer chromatography from methylene chloride-ether give 267 mg of pure 118,18-epoxy-18,21-dihydroxy pregn-4-ene-3,20-dione-21-acetate (21-O-acetyl-aldosterone) dated 195,198 ".
The extractive work-up of the aqueous mother liquor from the above-described ketal cleavage gives a further amount of crystalline hydrolysis product which, together with the still impure components from the above chromatogram, after renewed separation by column chromatography on silica gel, using toluene-chloroform mixtures, further amounts of pure 11ss, 18-epoxy-18,21-dihydroxy-pregn-4-en3,20-dione-21-acetate (21-O-acetyl-aldosterone) from F. 195 198 provides.
