Verfahren zur Herstellung von neuen substituierten Diphenyläthern
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen substituierten Diphenyläthern mit wertvollen biologischen Eigenschaften.

in welcher X und Y unabhängig voneinander Chlor oder Brom bedeuten und Z Wasserstoff, Chlor oder Brom bedeutet, sind bisher nicht bekanntgeworden. Wie nun gefunden wurde, besitzen diese Verbindungen antimikrobielle Eigenschaften von überraschender Intensität und Breite, zugleich ist ihre Toxizität relativ gering.
Sie lassen sich infolge dieser Eigenschaften wie auch ihrer Stabilität, weitgehender Farblosigkeit und fehlender Reizwirkung auf Haut und Schleimhäute als antimikrobielle Wirkstoffe auf den verschiedensten Anwendungsgebieten einsetzen, z. B. zum Schützen von organischen Materialien und Gegenständen vor Mikroorganismen, besonders vor Bakterienbefall, weiter als desinfizierende Zusätze zu Seifen und Waschmitteln sowie zu Hautsalben und andern Körperpflegemitteln.
Besonders wertvoll ist ihre Verwendbarkeit als Wirkstoffe zur Heilung von Krankheitszuständen der Haut, des Intestinalsystems und der Harnwege des Warmblüters, die sich aus der hervorragenden Wirksamkeit gegenüber den weiter unten genannten pathogenen Pilzen und Bakterien, der weitgehend in unveränderter, wirksamer Form erfolgten Ausscheidung aus dem Körper sowie der relativ geringen Toxizität ergibt.
Die Verbindungen der allgemeinen Formel I zeigen beispielsweise gegenüber folgenden grampositiven und gramnegativen Bakterien sowie pathogenen Pilzen eine vorzügliche wachstumshemmende Wirkung: Staphylococcus aureus Smith, Staphylococcus lactis, Bacillus pumilus, Bacillus subtilis, Corynebacterium diphtheriae, Clostridium botulinum, Clostridium butyricum, Clostridium welchii, Clostridium tetani, Klebsiella pneumoniae, Alcaligenes faecalis, Salmonella pullorum, Salmonella typhi, Salmonella paratyphi A und B, Salmonella typhi murium, Salmonella enteritidis, Shigella dysenteriae, Shigella flexneri, Brucella abortus, Proteus mirabilis, Achromobacter spec., Serratia marcescens, Pasteurella pseudotuberculosis;
Trichophyton mentagrophytes, Trichophyton rubrum, Trichophyton tonsurans var. sabourandi, Trichophyton schonleini, Trichophyton quinckeanum, Microsporon canis, Microsporon gypseum, Blastomyces dermatidis, Sporotrichum schenkii, Epidermophyton floccosum. Die Verbindungen der allgemeinen Formel I können beispielsweise als Wirkstoffe von Händedesinfektionsmitteln, Hautpflegemitteln, Wundsalben, Augensalben und weiteren äusserlich anwendbaren Mitteln sowie auch als Wirkstoffe von Mitteln zur inneren Anwendung dienen. Von letzteren seien z. B. Lutschtabletten zur Mund- und Rachendesinfektion sowie Tabletten und Dragees zur Desinfektion des Intestinalsystems und der Harnwege genannt.
Zur erfindungsgemässen Herstellung von Verbindungen der allgemeinen Formel I wird ein o-Alkoxydiphenyläther der allgemeinen Formel II

in welcher R einen niederen Alkylrest, insbesondere den Methylrest, bedeutet und X, Y und Z die oben angegebene Bedeutung haben, entalkyliert. Die Entalkylierung kann nach den für die Spaltung von Aryl-alkyl äthern bekannten Methoden, z. B. durch Behandlung mit Aluminiumchlorid in einem inerten Lösungsmittel, z. B. in Benzol bei dessen Siedetemperatur, weiter durch Erhitzen mit konz. wässriger Bromwasserstoffsäure, mit Bromwasserstoff in Eisessig oder Bromwasseristoffsäure- Essigsäure-Gemisch, erfolgen.
Halogensubstituierte o-Alkoxy-diphenyläther der allgemeinen Formel II können beispielsweise erhalten werden, indem man zunächst
Alkalisalze von 2-Alkoxy-4-halogenphenolen, insbesondere 4hIcr- oder 4-Brom-guaiacol, mit 4-Halogen-1-nitro-benzolen,
3 ,4-Dihalogen-l-nitro-benzolen, 2, 5-Dihalogen-l-nitro-benzolen oder 2,3,5-Trihalogen-1-nitro-benzolen zu
2-Alkoxy-4-halogen-4'-nitro-diphenyläthern,
2-Alkoxy-4, 2-dihalogen-4-nitro-diphenyl-äthern, 2-Alkoxy4n4t-dihalogen-2t-nitro-diphenyläthern bzw.
2-lkoxy-2,4,4¯trihalogen-6"-nitro-diph umsetzt und diese in üblicher Weise, z. B. durch katalytische Hydrierung oder durch Reduktion mit Zinnchlorür und Salzsäure, Zinkstaub und Essigsäure oder Eisen und Salzsäure, in die entsprechenden Aminoverbindungen überführt. Aminoverbindungen mit 4'-ständiger Aminogruppe und gewünschtenfalls auch solche mit 2'-ständiger Aminogruppe können zwecks Ersetzen dieser Gruppe durch Chrom oder Brom in die entsprechenden Diazoniumchloride oder-bromide umgewandelt und diese mit Kupferchlorür bzw. Kupferbromür nach Sandmeyer behandelt werden. Anderseits kann man Aminoverbindungen mit 6'-ständiger Aminogruppe und gewünschtenfalls auch solche mit 2'-ständiger Aminogruppe, d. h.
Verbindungen, die in 4'Stellung bereits das Halogenatom Y enthalten, in die entsprechenden Diazoniumsalze, insbesondere Sulfate, überführen und diese mit Äthanol erwärmen, wobei an die Stelle der intermediären Diazoniumgruppe ein Wasserstoffatom tritt.
Das nachfolgende Beispiel erläutert die Erfindung näher. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel a) In einem mit Rührer und absteigendem Kühler versehenen Dreihalskolben werden 476 g 4-Chlor-2methoxy-phenol (4-Chlor-guaiacol) und 578 g 3,4-Di- chlor-l-nitrobenzol in 400 ml Diäthylenglykoldimethyl- äther geschmolzen und bei etwa 1200 342 g 49,6 ; ige Kalilauge innerhalb etwa 4 Stunden zugetropft. Dann wird die Innentemperatur 12 Stunden auf 140-1500 gehalten, wobei Wasser und geringe Mengen von organischen Stoffen, wie zum Teil schon beim Eintropfen der Kalilauge, abdestillieren. Dann wird die Reaktionsmischung in ein Gemisch von Wasser und Natronlauge gegossen, der Niederschlag abfiltriert, getrocknet und aus Benzol umkristallisiert.
Der erhaltene 2-Methoxy 4,2' - dichlor -4' - nitrodiphenyläther schmilzt bei 159 bis 1610. b) 623 g 2-Methoxy-4, 2'-dichlor-4'-nitro-diphenyläther werden in 400 ml Dioxan in Gegenwart von 250 g Raney-Nickel bei Raumtemperatur und Normaldruck katalytisch hydriert. Nach Aufnahme der berechneten Menge Wasserstoff wird vom Raney-Nickel abfiltriert und der 2-Methoxy-4,2'-dichlor-4'-amino-diphenyläther durch Ausfällen mit Wasser abgeschieden, abfiltriert, gewaschen und getrocknet, Smp. 100-102 . c) In die Mischung von 254 mi konz. Salzsäure und 1600 ml Wasser werden unter gutem Rühren bei Raumtemperatur 204 g gut gemahlener 2-Methoxy-4,2'-dichlor- 4'-amino-diphenyläther eingetragen.
Die entstandene Suspension wird auf 0-5 gekühlt und bei dieser Temperatur 225 g 33 % ige Natriumnitritlösung unter das Flüssigkeitsniveau zulaufen gelassen. Das Gemisch wird noch 12 Stunden bei 0-5 verrührt.
In die Lösung von 400 g Kupfersulfat krist. und 106 g Natriumchlorid in 1280 ml Wasser lässt man bei 800 eine Lösung von 86 g Natriumsulfit und 60 g Natriumhydroxyd in 640 mi Wasser einlaufen. Man lässt das entstandene Kupferchlorür absitzen, giesst das überstehende Wasser ab und reinigt den Niederschlag durch dreimaliges Dekantieren mit Wasser.
Der Rückstand wird in 640 ml konz. Salzsäure gelöst, auf 65-700 erwärmt und unter Rühren die nach dem ersten Abschnitt von c) hergestellte Diazosuspension eingetragen. Nach dem Erkalten wird der wässrige Teil abgegossen, der harzartige organische Teil in Äther aufgenommen, die Ätherlösung mit verdünnter Natronlauge ausgeschüttelt, neutralgewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rück, stand wird unter Wasserstrahlvakuum destilliert. Der erhaltene 2 Methoxy - 4,2',4' - trichlordiphenyläther siedet bei 210 bis 2170.
Diazotiert man anstelle von 2-Methoxy-4,2'-dichlor4'-amino-diphenyläther 2-Methoxy-4, 4'-dichlor-2'-amino- diphenyläther in gleicher Weise und verrührt die entstandene Diazosuspension mit 10000 ml siedendem Äthyl alkohol und 28 g Kupferpulver, bis keine Diazoverbindung mehr nachweisbar ist (etwa 10 Minuten), so erhält man nach Filtration und Entfernung des überschüssigen Alkohols 2-Methoxy-4,4'-dichlor-diphenyl äther vom Kp. 12 = 197-2030. d) In die Lösung von 187,5 g 2-Methoxy-4,2',4'trichlordiphenyläther in 800 ml Benzol werden 243 g Aluminiumchlorid gegeben und das Reaktionsgemisch unter Rühren 30 Minuten zum Sieden erhitzt.
Nach dem Abkühlen wird es in EisSalzsäure gegossen, die benzolische Schicht abgetrennt und mit Wasser und Natronlauge durchgeschüttelt. Die mimosa-alkalische wässrige Schicht wird abgetrennt, durch Einblasen von Wasserdampf von den letzten Resten Benzol befreit, filtriert und mit Salzsäure angesäuert. Der ausgefallene 2-Hydroxy-4, 2', 4'-tri- chlordiphenyläther ist zunächst schmierig, erstarrt aber nach einiger Zeit. Er wird abfiltriert, gewaschen und getrocknet. Nach Umkristallisieren aus Petroläther schmilzt er bei 60-610.
In analoger Weise erhält man den 2-Hydroxy-4,4'- dichlor-diphenyläther vom Smp. 78-79 aus der am Ende von c) genannten Methoxyverbindung. e) Verwendet wan bei a) anstelle von 4-Chlor-2methoxyphenol 4Brom-2-methoxy-phenol und verfährt im übrigen wie in den Stufen a)-d) angegeben, so erhält man als Endprodukt den unter einem Druck von 12 Torr bei 223-2290 siedenden 2-Hydroxy-4-brom 2',4'-dichlor-diphenyläther.

Their usability as active ingredients for the healing of disease states of the skin, the intestinal system and the urinary tract of warm-blooded animals is particularly valuable, which results from their excellent effectiveness against the pathogenic fungi and bacteria mentioned below, which are largely excreted from the body in an unchanged, effective form as well as the relatively low toxicity.

The resulting suspension is cooled to 0-5 and, at this temperature, 225 g of 33% sodium nitrite solution are allowed to run below the liquid level. The mixture is stirred for a further 12 hours at 0-5.
In the solution of 400 g of copper sulfate crystall. and 106 g of sodium chloride in 1280 ml of water are allowed to run in a solution of 86 g of sodium sulfite and 60 g of sodium hydroxide in 640 ml of water at 800. The copper chloride formed is allowed to settle, the excess water is poured off and the precipitate is purified by decanting three times with water.
The residue is concentrated in 640 ml. Dissolved hydrochloric acid, heated to 65-700 and introduced the diazo suspension prepared according to the first section of c) with stirring. After cooling, the aqueous part is poured off, the resinous organic part is taken up in ether, the ether solution is shaken out with dilute sodium hydroxide solution, washed neutral, dried over sodium sulphate and evaporated. Instead of 2-methoxy-4,2'-dichloro-4'-amino-diphenyl ether, 2-methoxy-4, 4'-dichloro-2'-aminodiphenyl ether is diazotized in the same way and the resulting diazo suspension is stirred with 10,000 ml of boiling ethyl alcohol and 28 g of copper powder, until no more diazo compound can be detected (about 10 minutes), after filtration and removal of the excess alcohol, 2-methoxy-4,4'-dichloro-diphenyl ether of b.p. 12 = 197-2030 is obtained. d) 243 g of aluminum chloride are added to the solution of 187.5 g of 2-methoxy-4,2 ', 4'trichlorodiphenyl ether in 800 ml of benzene and the reaction mixture is heated to boiling for 30 minutes while stirring.
After cooling, it is poured into ice / hydrochloric acid, the benzene layer is separated off and shaken with water and sodium hydroxide solution. The mimosa-alkaline aqueous layer is separated off, freed from the last residues of benzene by blowing in steam, filtered and acidified with hydrochloric acid. The precipitated 2-hydroxy-4, 2 ', 4'-trichlorodiphenyl ether is initially greasy, but solidifies after some time. It is filtered off, washed and dried. After recrystallization from petroleum ether, it melts at 60-610.
The 2-hydroxy-4,4'-dichloro-diphenyl ether of melting point 78-79 is obtained in an analogous manner from the methoxy compound mentioned at the end of c). e) If 4-bromo-2-methoxyphenol is used instead of 4-chloro-2-methoxyphenol in a) and the procedure is otherwise as indicated in steps a) -d), the end product obtained under a pressure of 12 torr at 223 -2290 boiling 2-hydroxy-4-bromo 2 ', 4'-dichloro-diphenyl ether.
