Die vorliegende Erfindung betrifft Verfahren zur Herstellung von neuen substituierten Diphenyläthern mit wertvollen biologischen Eigenschaften.

in welcher X und Y unabhängig voneinander Chlor oder Brom bedeuten und Z Wasserstoff, Chlor oder Brom bedeutet, sind bisher nicht bekanntgeworden. Wie nun gefunden wurde, besitzen diese Verbindungen antimikrobielle Eigenschaften von überraschender Intensität und Breite, zugleich ist ihre Toxizität relativ gering.
Sie lassen sich infolge dieser Eigenschaften wie auch ihre Stabilität, weitgehenden Farblosigkeit und fehlenden Reizwirkung auf Haut und Schleimhäute als antimikrobielle Wirkstoff auf den verschiedensten Anwendungsgebieten einsetzen, z. B. zum Schützen von organischen Materialien und Gegenständen vor Mikroorganismen, besonders vor Bakterienbefall, weiter als desinfizierende Zusätze zu Seifen und Waschmitteln sowie zu Hautsalben und andern Körperpflegemitteln.
Besonders wertvoll ist ihre Verwendbarkeit als Wirkstoffe zur Heilung von Krankheitszuständen der Haut, des Intestinalsystems und der Harnwege des Warmblüters, die sich aus der hervorragenden Wirksamkeit gegenüber den weiter unten genannten pathogenen Pilzen und Bakterien, der weitgehend in unveränderter, wirksamer Form erfolgten Ausscheidung aus dem Körper sowie der relat, geringen Toxizität ergibt.
Die Verbindung der allgemeinen Formel I zeigen beispielsweise gegenüber folgenden grampositiven und gramnegativen Bakterien sowie pathogenen Pilzen eine vorzügliche wachstumshemmende Wirkung: Staphylococcus aureus Smith, Staphylococcus lactis, Bacillus pumilus, Bacillus subtilis, Corynebacterium diphtheriae, Clostridium botulinum, Clostridium butyricum, Clostridium welchii, Clostridium tetani, Klebsiella pneumoniae, Alcaligenes faecalis, Salmonella pullorum, Salmonella typhi, Salmonella paratyphi A und B, Salmonella typhi murium, Salmonella enteritidis, Shigella dysenteriae, Shigella flexneri, Brucella abortus, Proteus mirabilis, Achromobacter spec., Serratia marcescens, Pasteurella pseudotuberculosis;
Trichophyton mentagrophytes, Trichophyton rubrum, Trichophyton tonsurans var. sabourandi, Trichophyton schonleini, Trichophyton quinckeanum, Microsporon canis, Microsporon gypseum, Blastomyces dermatidis, Sporotrichum schenkii, Epidermophyton floccosum. Die Verbindungen der allgemeinen Formel I können beispielsweise als Wirkstoffe von Händedesinfektionsmitteln, Hautpflegemitteln, Wundsalben, Augensalben und weiteren äusserlich anwendbaren Mitteln sowie auch als Wirkstoffe von Mitteln zur inneren Anwendung dienen. Von letzteren seien z. B. Lutschtabletten zur Mund- und Rachendesinfektion sowie Tabletten und Dragees zur Desinfektion des Intestinalsystems und der Harnwege genannt.
Man erhält die Verbindungen der allgemeinen Formel I, indem man eine Verbindung der allgemeinen Formel II,

in welcher X, Y und Z die oben angegebene Bedeutung haben, in ein Diazoniumsalz überführt und auf letzteres einen Alkohol, insbesondere Äthanol bei dessen Siedepunkt, oder unterphosphorige Säure einwirken lässt.
Die Ausgangsstoffe der allgemeinen Formel II kann man z. B. erhalten, indem man die entsprechenden, durch Halogen und eine Nitrogruppe substituierten 2-Alkoxydiphenyläther entalkyliert und dann zu den entsprechenden Aminoverbindungen mit freier Hydroxylgruppe reduziert.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel a) In die Schmelze von 317 g 2-Methoxy-4-chlorphenol (4-Chlor-guaiacol) und 384 g 2, 5-Dichlor-1- nitro-benzol werden unter gutem Rühren bei 115-1200 innerhalb etwa 4 Stunden 223 g 50,3 % ige Kaliauge eingetropft. Durch einen absteigenden Kühler lässt man Wasser und geringe Mengen organischer Stoffe abdestilieren. Dann wird die Temperatur während 12 Stunden auf 145-1500 gehalten. Nach dem Erkalten wird das Reaktionsgemisch in die Mischung von 3000 ml Wasser und 140 ml 30 %ige Natronlauge eingegossen, das Reaktionsprodukt in Äther aufgenommen, die Ätherlösung neutralgewaschen und eingedampft; zuletzt im Vakuum.
Der als Öl zurückbleibende 2-Methoxy-2'-ni tro4,4'-dichlor-diphenyläther wird roh weiterverarbeitet. b) 400 g Eisenpulver werden mit 1000 ml Wasser und 20 ml 80% Der Essigsäure unter gutem Rühren 15 Minuten zum Sieden erhitzt. Nun lässt man durch einen geheizten Tropftrichter den rohen 2-Methoxy 2'-nitro-4,4'-dichlordiphenyläther innerhalb etwa 2 Stunden zulaufen und beendigt die Reduktion durch 12stündiges Rückflusskochen des Reaktionsgemisches. Die Masse wird nun mit Natriumcarbonat phenolphthaleinalkalisch gestellt und nach Zugabe von 1000 ml Chlorbenzol noch einmal zum Sieden erhitzt. Das heisse Gemisch wird zur Entfernung von Eisenschlamm durch Tierkohle filtriert, das Filtrat mit Salzsäure kongosauer gestellt und das Chlorbenzol mit Wasserdampf abdestilliert.
Der Rückstand wird mit Natronlauge neutralisiert und der 2-Methoxy-2'-amino-4,4'-dichlor-diphenyl äther nach dem Erstarren abfiltriert, mit Wasser vermahlen, wieder filtriert, neutralgewaschen und getrocknet. Dieses bei 73-760 schmelzende Rohprodukt wird in der nächsten Reaktionsstufe ohne weitere Reinigung eingesetzt. c) 426 g fein gemahlener 2-Methoxy-2'-amino-4,4'dichlor-diphenyläther werden in eine Mischung aus 1500 ml Bromwasserstoffsäure 48 % und 500 ml Essigsäure eingetragen und 48 Stunden zum Sieden erhitzt.
Das Reaktionsgemisch wird nach dem Erkalten mit Natronlauge abgestumpft, bis es Kongopapier nur noch violett färbt, und mit Natriumacetat kongoneutral gestellt. Nun wird das Gemisch filtriert, das Filtergut neutralgewaschen und unter Zusatz von 250 ml Natronlauge 30 % in 2000 ml Wasser gelöst, die Lösung filtriert und das Reaktionsprodukt durch Zugabe von Salzsäure bis zur kongovioletten Reaktion ausgefällt, abfiltriert, neutral gewaschen und getrocknet. Nach dem Umkristallisieren aus Ligron unter Zusatz von Tierkohle schmilzt der 2-Hydroxy-2'-amino-4,4'-dichlor-diphenyl- äther bei 126-128 . d) In ein Gemisch von 65 ml konz. Salzsäure und 175 ml Wasser werden unter gutem Rühren 67,5 ml fein gemahlener 2-Hydroxy-2'-amino-4,4'-dichlor-diphenyläther eingetragen.
Bei 0-5 werden 55 g 33 % ige Natriumnitritlösung unter dem Flüssigkeitsniveau zulaufen gelassen und die entstandene Diazosuspension nach weiteren 15 Stunden Rühren in 2500 ml Äthyl- alkohol und 7 g Kupferpulver eingerührt. Wenn keine Diazoverbindung mehr nachweisbar ist, wird das Gemisch filtriert. Aus dem Filtrat wird der Alkohol abdestilliert, der Rückstand in Äther aufgenommen, die wässrige Phase abgetrennt, die ätherische Phase neutralgewaschen und eingedampft. Das zurückbleibende Rohprodukt wird unter Wasserstrahlvakuum destilliert und das bei 197-2040/12 Torr übergehende, beim Abkühlen erstarrende Destillat aus Petroläther umkristallisiert. der erhaltene 2- Hydroxy -4,4' - dichlordiphenyläther schmilzt bei 78-79 .

in which X and Y are independently chlorine or bromine and Z is hydrogen, chlorine or bromine, have not yet become known. As has now been found, these compounds have antimicrobial properties of surprising intensity and breadth, and at the same time their toxicity is relatively low.
As a result of these properties, as well as their stability, extensive colorlessness and lack of irritation on the skin and mucous membranes, they can be used as antimicrobial agents in a wide variety of fields of application, e.g. B. to protect organic materials and objects from microorganisms, especially from bacterial infestation, further as disinfectant additives to soaps and detergents as well as to skin ointments and other personal care products.
Their usability as active ingredients for the healing of disease states of the skin, the intestinal system and the urinary tract of warm-blooded animals is particularly valuable, which results from their excellent effectiveness against the pathogenic fungi and bacteria mentioned below, which are largely excreted from the body in an unchanged, effective form and the relatively low toxicity.

The compounds of general formula I can serve, for example, as active ingredients in hand disinfectants, skin care products, wound ointments, eye ointments and other externally applicable agents, and also as active ingredients in agents for internal use. The starting materials of the general formula II can be, for. B. obtained by dealkylating the corresponding 2-alkoxydiphenyl ethers substituted by halogen and a nitro group and then reducing to the corresponding amino compounds with free hydroxyl groups.

Example a) In the melt of 317 g of 2-methoxy-4-chlorophenol (4-chloro-guaiacol) and 384 g of 2,5-dichloro-1-nitro-benzene are 223 g with thorough stirring at 115-1200 within about 4 hours g 50.3% potassium hydroxide solution was added dropwise. The crude 2-methoxy 2'-nitro-4,4'-dichlorodiphenyl ether is now run in through a heated dropping funnel over the course of about 2 hours and the reduction is ended by refluxing the reaction mixture for 12 hours. The mass is then made alkaline to phenolphthalein with sodium carbonate and, after adding 1000 ml of chlorobenzene, heated again to the boil. The hot mixture is filtered through animal charcoal to remove iron sludge, the filtrate is acidified to Congo with hydrochloric acid and the chlorobenzene is distilled off with steam.
The residue is neutralized with sodium hydroxide solution and the 2-methoxy-2'-amino-4,4'-dichloro-diphenyl ether is filtered off after solidification, ground with water, filtered again, washed neutral and dried. This crude product, melting at 73-760, is used in the next reaction stage without further purification. c) 426 g of finely ground 2-methoxy-2'-amino-4,4'-dichloro-diphenyl ether are introduced into a mixture of 1500 ml of hydrobromic acid 48% and 500 ml of acetic acid and heated to boiling for 48 hours.
After cooling, the reaction mixture is blunted with sodium hydroxide solution until it only turns Congo paper purple, and made Congo-neutral with sodium acetate. At 0-5, 55 g of 33% sodium nitrite solution are allowed to run in below the liquid level and, after stirring for a further 15 hours, the resulting diazo suspension is stirred into 2500 ml of ethyl alcohol and 7 g of copper powder. When the diazo compound can no longer be detected, the mixture is filtered. The alcohol is distilled off from the filtrate, the residue is taken up in ether, the aqueous phase is separated off, the ethereal phase is washed neutral and evaporated. The remaining crude product is distilled under a water jet vacuum and the distillate which passes over at 197-2040 / 12 Torr and solidifies on cooling is recrystallized from petroleum ether. the 2-hydroxy -4,4'-dichlorodiphenyl ether obtained melts at 78-79.
