Verfahren zur Herstellung von O, O-Dialkyl-S-alkylthiolphosphaten
In der deutschen Patentschrift Nr. 835 145 wird ein Verfahren zur Herstellung von O, O-Dialkylthiono- phosphorsauren Ammoniumsalzen aus symmetrischen Dialkylphosphiten beschrieben, wobei an die intermediär entstehenden Dialkylphosphite in Gegenwart von Ammoniak Schwefel angelagert wird.
Diese Reaktion spielt sich folgendermassen ab :
Unter Addition von Schwefel an Phosphor bilden sich zunächst in praktisch quantitativer Ausbeute die Ammoniumsalze von O, O-Dialkylthionophosphorsäuren (I). Die Substanzen I stehen im Gleichgewicht mit der isomeren Thiolform (II)

Beim Alkylieren von (I) entstehen 0, O-Dialkyl S-alkylthiolphosphate, also Verbindungen, die sich von II ableiten.
Nach den Angaben der deutschen Patentschrift Nr. 835145 werden nur solche Dialkylphosphite erhalten, in denen die Alkoxygruppen der obigen Formeln primär Alkylreste darstellen. Auch in der weiteren Literatur wurden bisher nur solche Verbindun- gen beschrieben, in denen die Alkoxygruppen der sogenannten Formeln primärer Natur sind.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 0, 0-Dialkyl-S-alkylthiolphos- phaten der Formel

mit Schwefel und einer alkalisch reagierenden Verbindung in die entsprechenden 0, 0-Dialkylthiono- bzw.-thiolphosphorsauren Salze übergeführt werden und diese darauf mit Alkylierungsmitteln unter Alkylierung der Thiolgruppe weiter umgesetzt werden.
Als alkalisch reagierende Verbindung wird be vorzugt Ammoniak verwendet. Wie aus der oben gegebenen Definition von R2 ersichtlich ist, sind unter dem Ausdruck Alkylierungsmitteln auch solche Mittel zu verstehen, die einen substituierten Alkylrest oder einen Aralkylrest einführen. (R10 steht in diesen Formeln für den Rest eines sekundären Alkohols), wobei die Verbindungen der Stoffklasse III steht, wie aus den Formeln ersicht- lich, mit den Verbindungen der Stoffklasse IV im Gleichgewicht stehen.
Beim Alkylieren von IV entstehen dann die entsprechenden 0, 0-Dialkyl-S-alkylthiolphosphate, die sehr gute insektizide und teilweise auch nematizide Eigenschaften aufweisen. Sie sollen deshalb als Pflan zenschutzmittel Verwendung finden.
Zur Alkylierung von IV sollen bevorzugt solche Alkylverbindungen Verwendung finden, die zur Herstellung wirksamer Insektizide auf Basis von Phosphorsäureestern, in denen beide Esterreste primärer Natur sind, bekannt sind. Das sind vor allem :
Alkyl-bzw. Arylmercaptoalkylhalogenide,
Alkyl-bzw. Dialkylaminoalkylhalogenide,
Cyanalkylhalogenide, Alkoxycarbonyl- alkylhalogenide, Alkylmercaptocarbonylalkylhalogenide,
Alkyl-bzw. Dialkylaminocarbonylalkylhalogenide,
Aralkylhalogenide bzw. die entsprechenden Tosylate und andere mehr.
Die folgenden Beispiele erläutern das Verfahren : Beispiel 1

152 g (1 Mol) O-Methyl-O-sec.-butylphosphit (Kp. 2 mm/68 ) werden in 200 cms Methanol gelöst. Zu der Lösung gibt man 32 g feingepulverten Schwefel und leitet unter Rühren wasserfreies Ammoniak bei einer Innentemperatur von 30-40 bis zur vollständigen Lösung des Schwefels ein. Anschlie ssend wird das Lösungsmittel abgedampft. Es bleiben 105 g O-Methyl-O-sec.-butylthionophosphorsaures Ammonium zurück, das für weitere Umsetzungen rein genug ist. Ausbeute 52% d. Th.
In analoger Weise wie oben bereits beschrieben erhält man aus O-Methyl-O-isopropyl-phosphit durch Umsetzung mit Schwefel und Ammoniak in einer Ausbeute von 60% der Theorie das entsprechende O-Methyl-O-isoprapyl-thiolphosphonsaure Ammonium.

61 g (0, 3 Mol) O-Methyl-O-sec.-butylthiono- phosphorsaures Ammonium werden in 200 cm3 Acetonitril gelöst. Unter Rühren gibt man bei 75 bis s 80 37 g ss-Äthylmercaptoäthylchlorid hinzu. Man rührt noch 1 Stunde bei 80 und gibt dann das Reak tionsprodukt in 300 cm3 Eiswasser. Das ausgeschiedene 01 wird in 200 cm3 Benzol aufgenommen. Die Benzollösung wird mit Wasser gewaschen und anschliessend mit Natriumsulfat getrocknet. Beim Fraktionieren erhält man 64 g O-Methyl-O-sec.-butyl-S- (2-äthylmercaptoäthyl)-phosphorsäureester vom Kp.
0, 01 mm/94 . Ausbeute 79 % d. Th. Der neue Ester ist ein farbloses, wasserunlösliches O1.

80 g (0, 4 Mol) O-Methyl-O-sec.-butylthiono- phosphorisaures Ammonium werden in 250 cm3 Acetonitril gelöst. Bei 50-60 gibt man unter Rühren 45 g a-Chlormethylthioäthyläther zu. Man lässt eine Stunde bei angegebener Temperatur rüh- ren und arbeitet dann im Beispiel 1 auf. Man erhält auf diese Weise 72 g O-Methyl-O-sec.- butyl-S (äthylthiomethyl)-phosphorsäureester vom Kp.
0, 01 mm/81 . Ausbeute 71 % d. Th.
Der neue Ester ist ein farbloses, wasserunlös- liches 01.

Man schlämmt 94 g (0, 5 Mol) O-methyl-Oisopropylthiolphosphorsaures Ammonium in 250 cm3 Acetonitril an und fügt zu dieser Anschlämmung unter Rühren bei 50 bis 60 C 36 g (0, 5 Mol) Chlor-acetonitril. Anschliessend wird das REaktionsgemisch 3 Stunden bei 60 C gerührt, dann in 300 cm3 Eiswasser gegossen und das ausgeschiedene 01 in 300 cm3 Benzol aufgenommen. Die Benzol- lösung wäscht man mit Wasser und trocknet sie dann über Natriumsulfat.
Nach Abdestillieren des Lösungs- mittels werden 75 g (67 % der Theorie) des O-Methyl Oisopropylthiolphosphorsäure-S-cyanomethylesters in Form eines farblosen, wasserunlöslichen Gles vom Brechungsindex nD21 = 1,4774 erhalten.
Die mittlere Toxizität der Verbindung (DL5o) beträgt an der Ratte per os 5 mg je kg Tier.

Zu dieser Amchlämmung fügt man unter kräftigem Rühren bei 75 C 67 g (0, 5 Mol) ss-Chlorpropionnitril, rührt die Mischung 3 Stunden bei 75 bis 80 C und giesst sie dann in 300 cm3 Eiswasser. Das ausgeschiedene Öl wird in 300 cm3 Benzol aufgenommen, die Benzollösung mit Wasser gewaschen und über Natriumsulfat getrocknet. Nach Abdestillieren des Lösungsmittels erhält man 62 g (52% der Theorie) des O-Methyl-O-isoproplthiolphosphorsäure-S-(ss-cyanoäthyl)-esters als wasserunlösliches, farbloses 01 vom Brechungsindex nD21 = 1, 4787.
An der Ratte per os besitzb die Verbindung eine mittlere Toxizität (DL50 von 50 mg je kg Tier.

Zu einer Lösung von 94 g (0, 5 Mol) O-methyl O-isopropylthiolphosphonsaurem Ammonium in 300 cm3 Acetonitril fügt man bei 75 C 54 g (0, 5 Mol) Chloressigsäure-N-methylamid, gelöst in 50 cm3 Acetonitril, rührt das Reaktionsgemisch anschliessend noch 2 Stunden bei der angegebenen Temperatur und giesst es dann in 400 cm3 Eiswasser. Das ausgeschiedene 01 wird in 300 cm3 Chloroform aufgenommen, die Chloroformlösung über Natriumsulfat getrocknet und anschliessend das Lösungsmittel durch Abdestil- lieren entfernt.
Man erhält als Rückstand 71 g (59 % der Theorie) des O-Methyl-O-isopropylthiolphosphorsäure-S-(N-methyl-carbamyl-methyl-)esters in Form eines viskosen, wasserunlöslichen Mes.

Zu dieser Lösung fügt man unter Rühren bei 75 C 38 g (0, 5 Mol) p-Chloräthylthioäthyläther, erhitzt das Reaktionsgemisch noch 1 Stunde auf 80 C und giesst es dann in 300 cm3 Eiswasser. Die Ausbeute beträgt 64 g (78% d. Th.). Die mittlere Giftigkeit (DL5o) beträgt an der Ratte per os 5 mg je kg Tier.

When IV is alkylated, the corresponding 0, 0-dialkyl-S-alkylthiol phosphates are formed, which have very good insecticidal and in some cases also nematicidal properties. They should therefore be used as pesticides.
For the alkylation of IV, preference should be given to using those alkyl compounds which are known for the production of effective insecticides based on phosphoric acid esters, in which both ester residues are of a primary nature. These are mainly:

61 g (0.3 mol) of O-methyl-O-sec.-butylthionophosphoric acid ammonium are dissolved in 200 cm3 of acetonitrile. With stirring, 37 g of β-ethyl mercaptoethyl chloride are added at 75 to 80. The mixture is stirred for a further 1 hour at 80 and then the reaction product is added to 300 cm3 of ice water. The precipitated oil is taken up in 200 cm3 of benzene. The benzene solution is washed with water and then dried with sodium sulfate. Fractionation gives 64 g of O-methyl-O-sec.-butyl-S- (2-ethylmercaptoethyl) phosphoric acid ester of bp.
0.01 mm / 94. Yield 79% of theory Th. The new ester is a colorless, water-insoluble O1.

80 g (0.4 mol) of O-methyl-O-sec.-butylthionophosphoric acid ammonium are dissolved in 250 cm3 of acetonitrile. At 50-60, 45 g of α-chloromethylthioethyl ether are added with stirring.
