Als die 1,6-Addition beschleunigendes Reagenz eignet sich z. B. Kupfer(I)-chlorid.
Eine Estergruppe in 17-Stellung kann in die er findungsgemäss erhältlichen Verbindungen mit 17 ständiger OH-Gruppe in bekannter Weise eingeführt werden. Bei Anwesenheit einer Methylgruppe in 17- Stellung sind, wie bekannt, schärfere Veresterungs- bedingungen anzuwenden.
Zu den organischen Carbonsäuren, aus denen erfindungsgemässe Acylate erhalten werden, gehören gesättigte und ungesättigte aliphatische und aroma tische Säuren, wie Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, tert.-Butylessigsäure, Valeriansäure, Isovaleriansäure, Capronsäure, Caprinsäure, Decansäure, Dodecansäure, Acrylsäure,
Crotonsäure, Hexinsäure, Heptinsäure, Octinsäure, Cyclobutancarbonsäure, Cyclopentancarbonsäure, Cyclohexancarbonsäure, Dimethylcyclohexancarbonsäure, Benzoesäure, Zolylsäure, Naphthoesäure, Äthylbenzoesäure, Phenylessigsäure, Naphthylessigsäure, Phenylvaleriansäure, Zimtsäure, Phenylpropiolsäure,
Phenylpropionsäure, p-Hexyloxyphenylpropionsäure, p-Bütyloxyphenylpropionsäure und ähnliche p-Alkoxyphenylalkansäuren.
Die erfindungsgemäss erhältliche Verbindung mit R"=Y=H kann oxydiert werden, z. B. mit Chrom säure oder einem ähnlichen Oxydationsmittel, wobei auf bekannte Weise die entsprechende 17-Ketover- bindung erhalten wird.
Die erfindungsgemäss herstellbaren 7a-Methyl-19- nortestosterone besitzen erhöhte anabolische und an- drogene Wirkung bei verbessertem Verhältnis von anabolischem zu androgenem Effekt. Sie besitzen ferner antiöstrogene, gonadotropininhibierende,
pro gestationale uhdwachstumsfördernde sowie dieFrucht- barkeit herabsetzende und das Zentralnervensystem beruhigende Wirkung. Besonderes Interesse besitzt das 7a - Methyl -17a - äthinyl-19-nortestosteron, das stärker wirksam ist im Hinblick auf die Herabsetzung der Fruchtbarkeit, aber eine geringere progestatio- nale Wirkung als das nahe verwandte, bekannte 17a-Äthinyl-19-nortestosteron besitzt.
Die 7a-Methyl-19-nortestosterone sind ferner Zwischenprodukte bei der Herstellung der entspre chenden 7a-Methyl-östrone und 7a-Methyl-östradiole, die verbesserte gonadotropininhibierende, die Frucht barkeit herabsetzende und den Cholesteringehalt im Blut senkende Eigenschaften aufweisen. Die erfin- dungsgemäss erhältlichen 7a-Methyl-19-nortestoste- rone können z.
B. unter Verwendung von Septomyxa affinis oder ähnlichen, dehydrierend wirkenden Mikroorganismen unter Bildung der entsprechend den 7a-Methyföstrone und -östradiole dehydriert werden.
Die erfindungsgemäss erhältlichen Verbindungen können Säugetieren, Vögeln und Menschen in einer Vielfalt von oralen und parenteralen Veräbrei- chungsformen einzeln oder im Gemisch mit anderen, gleichzeitig wirksamen Verbindungen zugeführt wer den.
Sie können zusammen mit einem festen oder flüssigen Träger, in dem sie gelöst, dispergiert oder suspendiert vorliegen, angewendet werden. Festen Zu sammensetzungen kann die Form von Tabletten, Pul vern, Kapseln, Pillen oder dergleichen zwecks ein facher Verabreichung und genauer Dosierung vor zugsweise mit einheitlicher Steroidkonzentration, ge geben werden. An flüssigen Zusammensetzungen kommen Lösungen, Emulsionen, Suspensionen, Sirupe oder Elixiere in Frage.
Die in, 17a-Stellung alkylier- ten 17ss-Hydroxyverbindungen werden vorzugsweise oral verabreicht, da sie hierbei eine besonders günstige Wirkung entfalten.
Bei der Durchführung des erfindungsgemässen Verfahrens wird das 6 - Dehydro -19 - nortestosteron mit einem überschuss an Methyl-Grignardreagens, z.
B. Methylmagnesiumjodid, Methylmagnesiumbro- mid oder dergleichen, in Gegenwart von Kupfer(I)- chlorid oder einem entsprechenden Katalysator um gesetzt [bezüglich anderer Katalysatoren vergleiche Grignard Reactions, Kharasch und Reinmutz, Pren- tice Hall, Inc. Publishers (1954), S. 219].
Kupfer(1)- chlorid wird bevorzugt. An Lösungsmitteln kommen Benzol, Toluol, Äthylenglykoldimethyläther, Äther, Tetrahydrofuran und dergleichen oder ein Gemisch aus 2 der genannten Lösungsmittel in Frage. Die Reaktionstemperatur kann zwischen -40 und dem Siedepunkt des Reaktionsgemisches, vorzugsweise zwischen etwa 0 und 60 liegen.
Das Mengenverhält nis beträgt vorzugsweise mindestens 5 Mol Grignard- verbindung pro Mol Steroid. Methylmagnesiumbro- mid ergibt die höchsten Ausbeuten und wird daher bevorzugt.
Die als Ausgangsmaterialien verwendeten Ver bindungen können wie folgt erhalten werden: Die Herstellung von 6-Dehydro-19-nortestoste- ronacetat durch Umsetzung von 19-Nortestosteron mit Acetanhydrid und Acetydchlorid in Pyridinlösung, anschliessende Bromierung des so erhaltenen 3,
17ss- Diacetoxy-19-nor-43=5-androstadiens und Bromwas- serstoffabspaltung aus dem so gebildeten 6-Brom-19- nortestosteron-17-acetat ist in der britischen Patent- Schrift Nr. 833 183 beschrieben.
Durch Ersatz des in der ersten Verfahrensstufe verwendeten Acetanhydrids und Acetylchlorids durch entsprechende. Anhydride und Säurechloride erhält man andere 17-Acylate. Die freien 17-Hydroxyverbindungen können durch übliche Entacylierung erhalten werden.
Ersatz des 19-Nortestosterons durch ein 17-Alkyl- 19-nortestosteron ergibt bei Verwendung entspre chender Säureanhydride und Säurechloride 17-Alkyl- 6-dehydro-19-nortestosteron-17-acylate. Die 17-Hy- droxyverbindungen können durch eines der bekann ten Entacylierungsverfahren, z.
B. durch Behandeln mit einer Base wie Natrium- oder Kaliumhydroxyd in wässriger oder wässrig-alkoholischer Lösung, erhalten werden.
Das erfindungsgemäss erhältliche 7a-Methyl-17a- äthyl-19-nortestosteron und dessen 17-Acylate haben androgene und anabolische Wirksamkeit mit verbes sertem therapeutischem Verhältnis;
sie besitzen ferner antiöstrogene, gonadotropininhibierende, progestatio- nale,wachstumsfördernde uhddas Zentralnervensystem regulierende Wirkungen, die sie zur Verwendung in der Human- und Veterinärmedizin bei Säugetieren geeignet machen.
7a-Methyl-17a-äthyl-testosteron wird durch Um setzung von 6-Dehydro-17a-äthyl-testosteron (US Patentschrift Nr. 2 739 974) mit einem Methyl- Grignardseagens in Gegenwart eines die 1,6-Addition fördernden Mitteln, z. B. Kupfer(1)-Chlorid, erhalten.
<I>Beispiel</I> 7a-Methyl-19-nortestosteron-acetat 30 ml Tetrahydrofuran (erhalten durch Percolie- ren von techn. Tetrahydrofuran über eine Säule aus neutralem Aluminiumoxyd unter Verwerfen der ersten 15 ml des Percolats) wurden in einem Eisbad gekühlt und unter Rühren in einer Stickstoffatmosphäre mit 25 ml. einer 3molaren Methylmagnesiumbromicllösung und 0,4 g Kupfer(1)-bromid versetzt.
Diesem Gemisch wurde eine Lösung von 3 g 6-Dehydro-19-nortest- osteron-17-acetat und geringen Mengen CuBr in 50 ml wie vorstehend gereinigtem Tetrahydrofuran unter Rühren zugesetzt.
Das Gemisch wurde unter Kühlen noch 10 Minu ten gerührt und sodann in ein mit Stickstoff durch- spültes Gemisch aus mit Eis verdünnter Salzsäure, gesättigt mit Natriumchlorid-Äther, gegossen. Die Ätherschicht wurde abgetrennt und nacheinander mit Kochsalzlösug, verdünnter Natronlauge + Kochsalz und wiederum mit Kochsalzlösung gewaschen.
Die Ätherextrakte wurden über Magnesiumsulfat getrock net, filtriert und zur Trockne eingedampft. Der Rück stand wurde in 5 ml Pyridin + 5 ml Acetanhydrid gelöst.
Nach 18stündigem Stehen bei Raumtempera tur wurde Eis und Wasser zugegossen und das Pro dukt mit Äther extrahiert. Der Extrakt wurde mit ver- dünnter Säure, verdünnter Natronlauge und Wasser gewaschen, getrocknet und eingedampft. Der Rück stand wurde in Methylenchlorid gelöst und die Lö sung an Magnesiumsilikat (Florisil) chromatogra- phiert. Die Säule wurde mit Skellysolve-B-Hexanen mit steigendem Gehalt an Aceton el'uiert. Die Frak tionen,
in welchen durch IR-Analyse ein Gehalt an 7a - Methyl - 19 - nortestosteron - 17 - acetat ermittelt wurde, wurden vereinigt und zur Trockne einge dampft.
Der ölige Rückstand wurde in Aceton gelöst und über eine mit Aceton getränkte Säule von 20 g Celite und 10g Darco chromatographiert. Die Säule wurde mit Aceton eluiert. Die ersten 750 ml wurden zur Trockne eingedampft, der Rückstand aus 7a-Me- thyl-19-nortestosteron-17-acetat wurde aus Methanol umkristallisiert. Man erhielt 1 g kristallines Material vom F. 111-114 ; [ab + 48 (Chloroform).
Das UV-Spektrum dieser Verbindung (in Äthanol) besass ein Maximum bei 240 m,u (s = 17,350).
Analyse: Berechnet für C21Hgp02: C 76,32 H 9,15 Gefunden: C 76,28 H 9,44 Aus obiger Verbindung kann wie folgt 7a-Methyl- 19-nortestosteron hergestellt werden: Eine Lösung von 3 g 7a-Methyl-19-nortestoste- ron-17-acetat in 40 ml einer 5 % igen Lösung von Kaliumcarbonat in 80 % igem wässrigem Methanol wurde unter Stickstoff 2 Stunden am Rückfluss ge kocht.
Das Gemisch wurde mit Äther extrahiert, der Ätherextrakt mit Wasser gewaschen und über Magne- siumsulfat getrocknet, filtriert und zur Trockne ein gedampft. Der Rückstand wurde mit Äther verrieben, der dabei resultierende Feststoff wurde isoliert. Man erhielt so 7a-Methyl-19-nortestosteron als kristallinen Feststoff vom F.140-145,5 . Eine analysenreine Probe schmolz bei 145-146 ; [a]n + 55 (Chloro form) (nach Umkristallisieren aus einem Gemisch aus Aceton und Skellysolve B).
Das UV-Spektrum (in Äthanol) besass ein Maximum bei 241 mu (a = 17,150).
Analyse: Berechnet für <B>C19H2802:</B> C 79,12 H 9,78 Gefunden: C 79,13 H 10,19 Die Oxydation der 17-Hydroxyverbindung zum 7a-Methyl-19-norandrosten-3,17-dion lässt sich wie folgt durchführen: Eine Aufschlämmung von 1,4 g Chromtrioxyd in 15 ml Pyiidin wurde unter Rühren und Kühlen einer Lösung von 1,4 g 7a-Methyl-19-nortestosteron in 17 ml Pyridin zugegeben.
Nach beendeter Zugabe wurde das Gemisch bei Raumtemperatur (etwa 20 ) 20 Stunden gerührt, sodann mit einem Gemisch aus gleichen Teilen Benzol und Äther verdünnt und über eine Schicht aus Diatomeenerde (Celite) filtriert. Die Filterschicht wurde gut mit einem 1 : 1-Gemisch aus Äther und Benzol, dann Wasser und wiederum mit dem Lösungsmittelgemisch gewaschen. Das Filtrat und die organische Schicht der Waschflüssigkeit wur den vereinigt und mehrmals mit Wasser gewaschen.
Das Waschwasser wurde nochmals mit Äther + Ben zol (1 :1) extrahiert. Die vereinigten organischen Schichten wurden getrocknet und zur Trockne ein gedampft. Der Rückstand wurde mit Äther verrieben; der dabei erhaltene Feststoff wurde abfiltriert. Man erhielt 1,4 g kristallines 7a-Methyl-19-norandrosten- 3,17-dion vom F. 195-198 . Eine durch Umkristalli- sieren aus Aceton erhaltene Analysenprobe schmolz bei 201-204 .
Das UV-Absorptionsspektrum dieser Verbindung (in Äthanol) zeigte ein Maximum bei 239,5 m,u (s = 17,000).

17ss-diacetoxy-19-nor-43 = 5-androstadiene and splitting off of hydrogen bromine from the 6-bromo-19-nortestosterone-17-acetate thus formed are described in British Patent No. 833 183.
By replacing the acetic anhydride and acetyl chloride used in the first process stage with corresponding ones. Other 17-acylates are obtained from anhydrides and acid chlorides. The free 17-hydroxy compounds can be obtained by conventional deacylation.

<I> Example </I> 7a-methyl-19-nortestosterone acetate 30 ml of tetrahydrofuran (obtained by percolating technical grade tetrahydrofuran over a column of neutral aluminum oxide while discarding the first 15 ml of the percolate) were cooled in an ice bath and 25 ml of a 3 molar methylmagnesium bromicl solution and 0.4 g of copper (1) bromide are added while stirring in a nitrogen atmosphere.
A solution of 3 g of 6-dehydro-19-nortest-osterone-17-acetate and small amounts of CuBr in 50 ml of tetrahydrofuran purified as above was added to this mixture with stirring.
The mixture was stirred for a further 10 minutes while cooling and then poured into a mixture of hydrochloric acid diluted with ice and saturated with sodium chloride-ether, flushed with nitrogen. The ether layer was separated and washed successively with sodium chloride solution, dilute sodium hydroxide solution + sodium chloride solution and again with sodium chloride solution.
The ether extracts were getrock net over magnesium sulfate, filtered and evaporated to dryness. The residue was dissolved in 5 ml of pyridine + 5 ml of acetic anhydride.
After standing for 18 hours at room temperature, ice and water were poured in and the product was extracted with ether. The extract was washed with dilute acid, dilute sodium hydroxide solution and water, dried and evaporated. The residue was dissolved in methylene chloride and the solution was chromatographed on magnesium silicate (Florisil). The column was eluted with Skellysolve-B-hexanes with increasing acetone content. The fractions
in which a content of 7a - methyl - 19 - nortestosterone - 17 - acetate was determined by IR analysis, were combined and evaporated to dryness.
The oily residue was dissolved in acetone and chromatographed on an acetone-soaked column of 20 g of Celite and 10 g of Darco. The column was eluted with acetone. The first 750 ml were evaporated to dryness, the residue from 7α-methyl-19-nortestosterone-17-acetate was recrystallized from methanol. 1 g of crystalline material with a melting point of 111-114; [from + 48 (chloroform).
The UV spectrum of this compound (in ethanol) had a maximum at 240 m, u (s = 17.350).
Analysis: Calculated for C21Hgp02: C 76.32 H 9.15 Found: C 76.28 H 9.44 From the above compound, 7a-methyl-19-nortestosterone can be prepared as follows: A solution of 3 g of 7a-methyl-19 -nortestosterone-17-acetate in 40 ml of a 5% solution of potassium carbonate in 80% aqueous methanol was refluxed under nitrogen for 2 hours.
The mixture was extracted with ether, the ether extract was washed with water and dried over magnesium sulfate, filtered and evaporated to dryness. The residue was triturated with ether and the resulting solid was isolated. This gave 7a-methyl-19-nortestosterone as a crystalline solid of material 140-145.5. A reagent grade sample melted at 145-146; [a] n + 55 (chloro form) (after recrystallization from a mixture of acetone and Skellysolve B).
The UV spectrum (in ethanol) had a maximum at 241 mu (a = 17.150).

After the addition was complete, the mixture was stirred at room temperature (about 20) for 20 hours, then diluted with a mixture of equal parts benzene and ether and filtered through a pad of diatomaceous earth (Celite). The filter layer was washed well with a 1: 1 mixture of ether and benzene, then water, and again with the mixed solvent. The filtrate and the organic layer of the washing liquid were combined and washed several times with water.
The wash water was extracted again with ether + benzene (1: 1). The combined organic layers were dried and evaporated to dryness. The residue was triturated with ether; the solid thus obtained was filtered off. 1.4 g of crystalline 7a-methyl-19-norandrosten-3,17-dione with a melting point of 195-198 were obtained. An analytical sample obtained by recrystallization from acetone melted at 201-204.
The UV absorption spectrum of this compound (in ethanol) showed a maximum at 239.5 m, u (s = 17,000).
