EMI0001.0002
B. können durch Dehydrierung von 7-Methyl-1 lss-oxytestosteron oder 7,17-Dimethyl-11 ss-oxytestosteron oder deren 17 Acylaten 7-Methyl-9(11)-dehydrotestosteron oder 7,17-Dimethyl-9(11)-dehydrotestosteron bzw. deren 17-Acylate (1I1) erhalten werden;
7 .Methyl-9(11)-dehydrotestosteron, 7,17-Dimethyl-9(11)-dehydrotestosteron oder entsprechende 17-Acylate lassen sich durch Be handlung mit einer unterhalogenigen Säure zu einem 7-Methyl-9a halogen-1lss-oxytestosteron oder einem 7,17-Dimethyl-9a-halogen-11 ss-oxytestosteron oder 17-Ester (IV) umwandeln;
behandelt man das derart erhaltene 7-Methyl-9a-halogen-llss-oxytestosteron oder 7,17-Dimethyl-9a-halogen-11 ss-oxytestosteron oder deren 17-Ester mit einer schwachen Base, z. B.
Kalium- oder Natriumhydroxyd in geringem über schuss bei pH 8 bis 10 oder Kalium- oder Natrium acetat, so erhält man 7-Methyl-9ss,11ss-epoxytestosteron oder 7,17-Dimethyl-9ss,11 ss-epoxytestosteron oder 17-Ester (V);
Die reinen 7-Stereoisomeren können erhalten werden durch fraktionierte Kristallisation oder Chro- matographie, wobei eines der Isomeren rein gewon- neu wird, und Behandlung des restlichen Isomeren- Gemisches mit Chloranil, welches nur mit einem Isomer reagiert und das andere unverändert lässt;

Process for the production of new steroid compounds The present invention relates to a process for the production of new 7-methyl-testosterone of the formula
EMI0001.0002
in which R ss-oxy-, ss-oxy-a-methyl-,, ss-acyloxy- a-methyl- or ss-acyloxy- and R 'oxy-, a-acyloxy-, hydrogen, keto- or a 9 ( 11) double bond, which is characterized
that you can get a 6-dehydrosteroid of the formula
EMI0001.0014
with a methyl Grignard reagent in the presence of a 1,6-addition promoting agent.
The process according to the invention and some subsequent stages of the process are shown in the following reaction scheme:
EMI0002.0001
in which R ss-oxy-, ss-oxy-a-methyl-, ss-acyloxy- a-methyl or ss-acyloxy-, R 'oxy- (a and ss), a-acyloxy-, hydrogen, a 9 ( 11) double bond or keto, the acyl radical of the acyloxy group being that of an organic carboxylic acid,
is preferably a hydrocarbon carboxylic acid with 1 to 12 carbon atoms inclusive; X is a halogen of atomic weight 35 to 127 inclusive and X 'is a halogen of atomic weight 19 to 127 inclusive. In the above formulas, the 7-methyl groups can be in the a- and / 'position. The 7-methyltestosterones obtainable according to the invention can be converted into other 7-methyltestosterones; z.
B. by dehydrating 7-methyl-1 lss-oxytestosterone or 7,17-dimethyl-11 ss-oxytestosterone or their 17 acylates 7-methyl-9 (11) -dehydrotestosterone or 7,17-dimethyl-9 (11) -dehydrotestosterone or their 17-acylates (1I1) are obtained;
7 .Methyl-9 (11) -dehydrotestosterone, 7,17-dimethyl-9 (11) -dehydrotestosterone or corresponding 17-acylates can be treated with a hypohalous acid to a 7-methyl-9a halogen-1lss-oxytestosterone or a 7,17-dimethyl-9a-halogen-11 ß-oxytestosterone or 17-ester (IV) convert;
treat the 7-methyl-9a-halogen-llss-oxytestosterone or 7,17-dimethyl-9a-halogen-11ss-oxytestosterone or its 17-ester thereof with a weak base, e.g. B.
Potassium or sodium hydroxide in a slight excess at pH 8 to 10 or potassium or sodium acetate, this gives 7-methyl-9ss, 11ss-epoxytestosterone or 7,17-dimethyl-9ss, 11ss-epoxytestosterone or 17-ester ( V);
the 7-methyl-9ss, 1ss-epoxytestosterone, 7,17-dimethyl-9r, ', 11ss-epoxytestosterone or their 17 esters obtained in this way can be converted into 7-methyl-9a-fluoro-11ss-oxytestosterone or 7,17-dimethyl-9a-fluoro-1 lss-oxytestosterone and corresponding esters (V1) are implemented.
After each of these process steps, the 17 ester group, if present, can be hydrolyzed, specifically with acid in the case of compounds IV or otherwise with bases, in order to obtain the corresponding free 17-oxy compound.
The oxidation of a 7-methyl-9a-halogen-1 lss-oxytestosterone (IV or VI) with chromic acid leads to the corresponding 7-methyl-9a-halogen-11-ketotestosterone (VII). The 17-acylate group, if present, can here
hydrolyzed with alkali to form the corresponding 7-methyl-9a-halogen-11-ketotestosterone. 7-methyl-9a-halo-11-ketotestosterone-17-acylates can be hydrolyzed in alcoholic acid solution to the corresponding 7-methyl-9a-halo-11-ketotestosterone.
A 17-position ester group can be introduced into the above compounds having 17-oxy group by known methods. If a 17-methyl group is also present, stronger esterification conditions must be selected, as they are also known per se.
The 7-methyltestosterones (1I) prepared according to the invention and their secondary products have anabolic and anabolic effectiveness to an improved degree. They also have antiestrogenic, gonadotropin-preventing, progestational, growth-promoting, central nervous system regulating and tumor (especially breast tumors) preventing effectiveness.
The compounds prepared according to the invention can be processed into a large number of oral or parenteral dosage forms alone or in conjunction with other active ingredients and administered in this way. They can be administered with a pharmaceutical carrier which can be a solid or liquid material in which the compound can be dissolved, dispersed or suspended.
The solid preparations may be in the form of tablets, powders, capsules, pills or the like, preferably in unit dosage forms for ease of administration and accurate dosage. The liquid preparations can be in the form of solutions, emulsions, suspensions, syrups or elixirs. The oral forms are preferably used for the 17a-methyl-17ss-oxy compounds, which exert particularly advantageous oral effects.
The method according to the invention can be carried out in such a way that the starting 6-dehydrotestosterone (I) is treated with an excess of methyl Grignard reagent, e.g. B. methylmagnesium iodide or methylmagnesium bromide, is reacted in the presence of copper chloride or an equivalent catalyst.
Regarding other catalysts, reference is made to Grignard Reactions, Kharasch and Reinmuth, Prentice Hall, Inc. Publishers (1954), p.219. Copper chloride is preferred. Numerous inert solvents can be used, e.g. B. benzene, toluene, ethylene glycol dimethyl ether, ether, tetrahydrofuran or mixtures thereof.
The reaction temperature can vary between 0 C and the boiling point of the reaction mixture. A temperature between about 60 C and room temperature is usually preferred. Since the 17-oxygen function and, to a certain extent, the 11-oxy group also react with the Grignard reagent, it is advisable to use more than one molar equivalent of Grignard reagent in order to ensure a complete reaction with the 3-keto-d4. 6-diene system to ensure.
A ratio of at least 5 moles of Grignard per mole of steroid is preferred. Methyl magnesium bromide produces the highest yield of the desired product and is therefore the preferred reagent.
The hydrolysis of a 7-methyltestosterone-17-acylate (II, R - acyloxy) with alkali in an aqueous alcoholic solution or a dilute mineral acid gives the corresponding 7-methyltestosterone as free 17-alcohol.
The pure 7-stereoisomers can be obtained by fractional crystallization or chromatography, whereby one of the isomers is obtained in pure form, and treatment of the remaining isomer mixture with chloranil, which only reacts with one isomer and leaves the other unchanged;
this isomer can then be separated from the 6-dehydro compound formed, as described in Example 2.
The starting 6-dehydrotestosterones can be obtained as described in US Pat. No. 2,739,974, or by reacting testosterone with chloranil as described in Example 2, or by reacting a 3fl-oxy-5-androstene, the substituent R 'in the 11 position and R in the 17 position, with aluminum tert-butoxide and p-quinone (as described in preparation 2).
The starting 3f oxy-5-androstenes can be obtained by sodium borohydride reduction of a 3-enol ester of a testosterone (as described in preparation 1). Preparation 1: 17a-methyl-5-androstene-3ss, l li3,17 / 3-triol.
A solution of 5 g of 11f-oxy-17a-methyltestosterone (USA Patent No. 2,735,854), 25 ml of acetic anhydride, 100 mg of p-toluenesulfonic acid and 100 ml of toluene are refluxed in a nitrogen atmosphere for 41/2 hours heated.
The solvent was then removed by distillation in vacuo. The residue could not be crystallized. It was dissolved in 100 ml of 95 A / Aigem alcohol and 3 ml of 10% sodium hydroxide solution and cooled to 0.degree.
A solution of 5 g of sodium borohydride in 100 ml of 70% alcohol was added with stirring and cooling. After one hour, a further 2.5 g of sodium borohydride in 50 ml of 70 A / 0 alcohol were added. This solution was stored for three days at 5 ° C., then 15 ml of 10A /% sodium hydroxide solution were added and the mixture was heated to almost the boiling point. The alcohol was distilled off in vacuo. Ice and 3N hydrochloric acid were added to the concentrate with stirring.
The product precipitated and was collected. It was washed with water, dilute hydrochloric acid and again with water and dried. The yield was 5.7 g. This Mate rial was dissolved in 50 ml of tetrahydrofuran and slowly mixed with 1.5 g of lithium aluminum hydride with stirring. After about 3 minutes the reaction mixture hardened to a hard gel. 15 ml of ether were then added and the mixture was stirred for one hour.
Ethyl acetate and water were then added. The aqueous phase was separated off and extracted with ether and then with methylene chloride. The non-aqueous phase and the extracts were combined, dried over magnesium sulfate and filtered. On standing overnight, 1.6 g of crystals with a melting point of 223-230 ° C. separated out from the filtrate.
The mother liquor was evaporated to dryness and triturated with methylene chloride, a further 1.3 g of crystals with a melting point of 203-210 ° C. being obtained. The two portions were combined and recrystallized from 15 ml of alcohol which contained 0.5 ml of water. The yield of 17a-methyl-5-androstene-3ss, ll, ss, 17,8-triol was 1.7 g and the melting point was 227-230 C.
Recrystallization from ethyl acetate gave 1.2 g of the product melting at 230-235 C and having a rotation [a] D of -68 (dioxane).

Similarly, 6-dehydrotestosterone and 6-dehydrotestosterone acetate are converted into 7-methyltestosterone, 6-dehydro-11 a-oxytestosterone-17-acetate and 6-dehydro-11 a-oxytestosterone in 7-methyl-lla-oxytestosterone, 6- Dehydro-11 a-oxy-17-methyltestosterone in 7,17-dimethyl-lla-oxytestosterone, 6-dehydro-11-ketotestosterone acetate and 6-dehydro-11-ketotestosterone in 7-methyl-11-ketotestosterone,
6,9 (11) -bisdehydrotestosterone acetate and 6,9 (11) -bisdehydrotestosterone converted into 7-methyl-9 (11) -dehydrotestosterone and 6-dehydro-11-keto-17-methyltestosterone 7,17-dimethyl-11-ketotestosterone . The 7-isomers of each of the 7-methyltestosterones described in Examples 1, 2, 3 and 4 can be obtained by fractional crystallization of the mixture of isomers to remove the more insoluble isomer,
and reaction of the mixture formed from the mother liquor with chloranil as described in Example 2 to obtain the corresponding 7-methyl-6-dehydrotestosterone (which compound also has androgenic and anabolic activity), and finally separation of the remaining 7-methyltestosterone epimer from the 6-dehydro compound can be obtained.
7-Methyl-llss-oxytestosterone-17-acetate can be obtained from the product of Example 4 as follows: A solution of 1.0 g of 7-methyl-llss-oxytestosterone in 6 ml of dry and distilled pyridine was mixed with 6 ml of acetic anhydride treated. After standing at room temperature for 17 hours, the solution was poured into ice water. After standing for two hours, the mixture was filtered and the precipitate was washed with water and dried in vacuo.
After recrystallization from an acetone-hexane mixture, practically pure 7-methyl-11ss-oxytestosterone-17-acetate was obtained.
This compound can be oxidized as follows: A solution of 7-methyl-llss-oxytestosterone-17-acetate in acetic acid was shaken with an aqueous solution of chromic anhydride for 12 hours. The steroid product was well extracted with benzene and the extract washed with a sodium bicarbonate solution and then with water, dried over anhydrous sodium sulfate and evaporated. 7-methyl-11-ketotestosterone-17-acetate was obtained.
The corresponding 7-methyl-llss-oxytestosterone-17-benzoate can be prepared as follows: 0.3 g of 7-methyl-llss-oxytestosterone, suspended in 12 ml of dry benzene, were mixed with 0.3 ml of distilled benzoyl chloride and 0.3 g ml of dry pyridine treated. The mixture was stirred at room temperature for 17 hours. After cooling, the product was collected on a filter, washed with benzene and ether and dried. Recrystallization from ethyl acetate gave practically pure 7-methyl-llss-oxytestosterone 17-benzoate.
The oxidation to 7-methyl-11-ketotestosterone-17-benzoate can take place in the following way: A solution of 1.5 g of 7-methyl-11ss-oxytestosterone-17-benzoate in 80 ml of acetic acid was mixed with a solution of 0 , 74 g of chromic anhydride in 4 ml of water and 80 ml of acetic acid and left to stand for 5 hours at room temperature. The excess of chromic acid was destroyed by adding 10 ml of methanol and the solution was then concentrated in vacuo on the water bath.
The residue was extracted with ether after trituration with water and the ether solution was washed with dilute sodium hydroxide solution and water and then dried over anhydrous sodium sulfate. After evaporation of the ether, 7-methyl-11-ketotestosterone-17-benzoate was obtained.
7-Methyl-11-ketotestosterone can be obtained as follows: A solution of 1.0 g of 7-methyl-11-ketotestosterone-17-propionate in 50 ml of 1N methanolic potassium hydroxide solution containing 3 ml of water was added for 30 minutes Heated to reflux. The solution was then poured onto ice and the mixture was neutralized with dilute sulfuric acid. After standing, the formed precipitate was collected, washed with water and dried. The aqueous filtrate was extracted with methylene chloride to obtain an additional portion of the material.
After recrystallizing this residue from methylene chloride, practically pure 7-methyl-11-ketotestosterone is obtained.
