Verfahren zur Herstellung neuer 2u-Flnorsteroide der 19-Norandrostenreihe Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung gewisser neuer 2a-Fluor- steroide der 19-Norandrostenreihe. Das erfindungs- gemässe Verfahren sowie die Herstellung der Aus gangsstoffe und die Weiterverarbeitung der Endstoffe lassen sich durch folgende Formeln darstellen:

Der Ausdruck niedrige Alkoxygruppe bedeutet einen Alkoxyrest mit 1 bis 8 Kohlenstoffatomen.
Die erfindungsgemäss erhaltenen Verbindungen der Formel IV sowie die der Formeln V und VI sind anabolisch-androgene Mittel mit verbesserten thera peutischen Verhältnissen; sie eignen sich für die Be handlung von Schwächezuständen, bei welchen ein Gewebeaufbau erwünscht ist. Sie sind auch geeignet als Futterzusätze für Nutztiere, als progestationale Mittel und als Gonadotropin-Inhibitoren bei Schwan- gerschafts- und Menstruationsstörungen.
Ferner wei sen sie das Zentralnervensystem beruhigende, cardio- tonische und antiartheriosklerotische Wirksamkeit auf. Zur oralen oder parenteralen Verabreichung an den Organismus können sie in die üblichen pharmazeuti schen Träger einverleibt werden, z. B. in Tabletten, mit oder ohne enterischem Überzug, in Pillen oder Kapseln mit oder ohne verzögernder Wirkung, Gra nulate oder Kugeln, wässrige oder alkoholische Sus pensionen oder Lösungen mit oder ohne Zusatz von Lösungsvermittlern, in Salben, Gelees, Cremes, Sup positorien, Stäbchen usw.
Die erfindungsgemäss hergestellten Verbindungen können leicht zu pharmazeutischen Präparaten für orale und parenterale Verabreichung an Säugetiere, Vögel und dergleichen verarbeitet werden.
Die Herstellung der Ausgangsmaterialien für die vorliegende Erfindung kann aus den 19-Nortestoste- ronverbindungen gemäss Formel I, das heisst 19-Nortestosteron, 17a-Methyl-19-nortestosteron, 17a-Äthyl-19-nortestosteron, 17a-Äthynyl-19-nortestosteron, 17a-(1-Propynyl)-19-nortestosteron und 17a-Allyl-19-nortestosteron erfolgen.
Zur Herstellung der Ausgangsstoffe des erfin dungsgemässen Verfahrens wird die entsprechende 19- Nortestosteronverbindung der Formel I mit einem niederen Alkylformiat, einem niederen Alkyloxalat oder einem niederen Alkyl-trifluoracetat in Gegen wart eines basischen Alkalimetallkondensationsmittels nach den in den USA-Patentschriften Nrn. 2 767 198, 2 790 814 und 2 884 429 beschriebenen Arbeitswei sen kondensiert, um eine Verbindung der Formel II zu erhalten, worin M ein Alkalimetall ist.
Diese Ver bindung oder deren Enolform (M=H), die in bekann ter Weise durch Ansäuern des Alkalimetallenolates erhalten wird, wird sodann erfindungsgemäss unter Bildung einer Fluorverbindung der Formel III mit Perchlorylfluorid behandelt. Die Umsetzung kann bei Zimmertemperatur oder niedrigerer Temperatur, vor zugsweise bei Temperaturen um 0 oder weniger, in einem organischen Lösungsmittel, wie z. B. Methanol, durchgeführt werden. Das Perchlorylfluorid kann durch die Lösung hindurchgeleitet oder in einem Lö sungsmittel zugegeben werden.
Der am C.,-Atom be findliche, die Carbonylgruppe enthaltende Rest wird sodann mit einer Alkalimetallbase und einem Alkanol, z. B. Natriummethoxyd in Methanol, entfernt, unter Bildung der erfindungsgemässen 2,i-Fluorverbindung IV, die nach üblichen Methoden, z. B. durch Kristal lisation, Chromatographie oder dergleichen, gereinigt werden kann.
Beispiele für derart herstellbare Verbindungen sind: 2a-Fluor-19-nortestosteron, 2a-Fluor-17a-methyl-19-nortestosteron, 2a-Fluor-17a-äthyl-19-nortestosteron, 2a-Fluor- 17-äthyl-19-nortestosteron, 2a-Fluor- 17a-äthynyl-19-nortestosteron, 2a-Fluor-17a-(1-propynyl)-19-nortestosteron und 2a-Fluor-17a-allyl-19-nortestosteron. Das so hergestellte 2a-Fluor-19-nortestosteron (IV)
kann nach üblichen Methoden in 17-Acylate der Formel V übergeführt werden, z. B. durch Umset zung mit einem entsprechenden Säureanhydrid oder Säurehalogenid, einem Ester durch Umesterung, einer Säure in Gegenwart eines Veresterungskatalysators oder dergleichen.
Geeignete Acylierungsmittel sind Carbonsäuren, insbesondere solche mit 1 bis 12 C-Atomen, deren Anhydride oder Halogenide. Es können z.
B. ali- phatische Säuren, wie Ameisensäure, Essigsäure, Pro- pionsäure, Buttersäure, Valeriansäure, Hexansäure, Laurinsäure, Trimethylessigsäure, Isobuttersäure, Iso- valeriansäure, tert.-Butylessigsäure, cycloaliphatische Säuren, wie /3-Cyclopentyl-propiosäure, Cyclohexan- carbonsäure,
Cyclohexylessigsäure, alkylaromatische Säuren, wie Benzoesäure, Phenylessigsäure, /3-Phenyl- propionsäure, o-, m-, p-Tolylsäure, gesättigte Dicar- bonsäuren (die in wasserlösliche Salze, z. B. Natrium salze, überführbar sind), z. B. Bernsteinsäure, Adipin- säure, ungesättigte Monocarbonsäuren, z. B.
Acryl säure, Crotonsäure, Undecylensäure, Propiolsäure, 2- Butinsäure, Undecolsäure, Zimtsäure, ungesättigte Di- carbonsäuren (die in wasserlösliche Salze überführbar sind), z. B. Malein- und Citraconsäure, oder deren An hydride oder Halogenide, zur Veresterung des 2a- Fluor-19-nortestosterons verwendet werden.
Wird als Acylierungsmittel eine freie Säure verwendet, so führt man die Reaktion vorzugsweise in Gegenwart eines Veresterungskatalysators, z. B. p-Toluolsulfonsäure- chlorid, Trifluoressigsäureanhydrid, p-Toluolsulfon- säure, Trifluoressigsäure, Schwefelsäure oder derglei chen durch.
Das 2a-Fluor-19-nortestosteron kann gewünsch- tenfalls nach den üblichen Methoden zur Oxydation der 17-ständigen Hydroxylgruppe von Testosteron in eine Ketogruppe, z.
B. mit Aluminium-tert.-butylat und Aceton oder Cyclohexanon in Gegenwart eines trockenen, organischen Lösungsmittels, wie Toluol, Benzol, Petroläther, Dioxan oder dergleichen oder deren Gemischen, zum 2a-Fluor-19-nor-4-androsten- dion-(3,17) oxydiert werden. Vorzugsweise wird die Oxydation mit Chromsäure nach dem Verfahren der USA-Patentschrift Nr. 2 697 715 durchgeführt. Das so hergestellte 2a-Fluor-19-nor-4-androsten-dion-(3, 17) kann auf übliche Weise, z.
B. durch Chromato- graphie oder Umkristallisation, gereinigt werden. <I>Beispiel 1</I> 2a-Fluor- 19-nortestosteron Ein Gemisch aus 25 g 19-Nortestosteron, 80 cm3 tert.-Butylalkohol und 25 cm-- Diäthyloxalat wurde unter Rühren in einer Stickstoffatmosphäre erwärmt, bis das Steroid gelöst war. Das Gemisch wurde auf 45 abgekühlt, mit 25 % igem Natriummethylat in Methanol versetzt und ohne Temperaturregelung 30 Minuten lang gerührt.
Danach betrug die Temperatur des Reaktionsgemisches 28 . Das Gemisch wurde so dann in ein Gemisch aus 340 cm3 Skellysolve-B- Hexan und 170 ein- Äther gegossen und unter Stick stoff 10 Minuten lang gerührt. Das als feiner Nieder schlag ausfällende Natriumenolat des 2-Athoxyoxalyl- 19-nortestosterons wurde schnell abfiltriert, mit 200 cm ,3 eines Gemisches aus Skellysolve-B-Hexan und Äther (1 : 1) gewaschen und im Vakuum 18 Stunden getrocknet.
Das getrocknete Natriumenolat wurde in 680 cm3 Methanol gelöst und auf -10 gekühlt. Dann wurde unter Rühren langsam eine kalte Lösung von 10 g Perchlorylfluorid in 400 cm?, Methanol zugegeben. Während der Zugabe des Perchlorylfluorids wurde die Temperatur unterhalb -2 gehalten.
Das so her gestellte 2 - Fluor - 2 - äthoxyoxalyl-19-nortestosteron wurde mit 25 cm3 25 % iger Natriummethylatlösung umgesetzt, um die Äthoxyoxalylgruppe zu entfernen. Nach 30minutigem Rühren wurde mit Essigsäure ein pH-Wert von 6 eingestellt und bei vermindertem Druck auf 200 cm s eingeengt. Das Produkt wurde durch Zugabe von 150 cm3 Wasser ausgefällt und das Gemisch noch so lange gerührt, bis der Nieder schlag erhärtete.
Das 2a-Fluor-19-nortestosteron wurde abfiltriert und in einem Vakuumtrockenschrank bei 40 16 Stunden lang getrocknet. Es wurde an 700 g Florisil (synthet. Magnesiumsilicat) unter Verwendung von 650 cm3 Methylenchlorid chromato- graphiert. Die Säule wurde mit Skellysolve-B-Hexan plus 5, 10, 12, 14 und 16 % Aceton entwickelt.
Das 2a-Fluor-19-nortestosteron wurde dabei mit Skelly- solve-B-Hexan plus 10, 12 und 14 % Aceton eluiert; der Rückstand dieser Fraktionen wurde aus Skelly- solve-B-Hexan umkristallisiert und ergab 15,77 g 2.@-Fluor-19-nortestosteron vom F.160 bis 163 , + 102 (Alkohol), UVA mä,@ : 241 mg (E _ <B>15750).</B>

Zur Veresterung des erhaltenen 2a-Fluor-19-nor- testosterons wurde eine Lösung von 3 g 2a- Fluor - 19 - nortestosteron in 20 cms trockenem Pyridin mit 20 cm3 Acetanhydrid behandelt und 22 Stunden bei 26 stehengelassen. Das Reak tionsgemisch wurde dann in 250 cm3 Wasser gegos sen und 2 Stunden bei Zimmertemperatur gerührt.
Das kristalline Produkt wurde abfiltriert, mit Wasser gewaschen und getrocknet und ergab 3,1 g 2a-Fluor- 19-nortestosteron-acetat. Die Analysensubstanz wurde durch Umkristallisieren aus Methanol erhalten.

Ist das Acylierungsmittel fest, so kann ein inertes Lösungsmittel wie Toluol, Xylol oder Dioxan zur Herstellung eines flüssigen Veresterungsmediums zu gegeben werden.
Zur Oxydation zum 2a-Fluor-19-nor-4-androsten- dion-(3,17) wurde eine Lösung von 1,58 g 2a-Fluor- 19-nortestosteron in einem Gemisch aus 35 cm3 Toluol und 15 cm3 Cyclohexanon zwecks Trocknen der Lösung so lange erwärmt, bis etwa 10 cm3 über gegangen sind. Zur trockenen Lösung werden 1,5 g Aluminium-tert,-butylat gegeben; es wird so lange ge kocht, bis die Umsetzung vollständig ist.
Ein über schuss an gesättigter, wässriger Kaliumtartratlösung wird sodann zugegeben, und die Lösungsmittel wer den durch Wasserdampfdestillation entfernt. Der Rückstand wird mit Methylenchlorid extrahiert, ge trocknet und an 150 g Magnesiumsilikat ehromato- graphiert. Die Säule wird mit Skellysolve-B-Hexan, das steigende Mengen Aceton enthält, entwickelt. 2a- Fluor-19-nor-4-androsten-dion-(3,17) wird als kristal liner Feststoff erhalten.
1 g 2a-Fluor-19-nortestosteron wird bei Zimmer temperatur mit 20 % igem überschuss an Chromtri- oxyd in 25 cm-' Eisessig, der 1 cm3 Wasser enthält, bis zur Beendigung der Reaktion oxydiert. Das Reak tionsgemisch wird sodann in ein grosses Volumen Wasser gegossen und sorgfältig mit Methylenchlorid extrahiert. Die Methylenchloridlösung wird mit Na- triumbicarbonatlösung gewaschen und getrocknet.
Die getrocknete Lösung wird an 40 g Magnesiumsilikat chromatographiert. Die Säule wird mit Hexangemisch, das steigende Mengen Aceton enthält, entwickelt. Man eluiert im wesentlichen reines 2a-Fluor-19-nor- 4-androsten-dion-(3,17).

In the same way, 17a-ethyl-19-nortestosterone, 17a-ethynyl-19-nortestosterone, 17a- (1-propynyl) -19-nortestosterone and 17a-allyl-19-nortestosterone are converted into 2a-fluoro-17a-ethyl-19- nortestosterone, 2a-fluoro-17a-ethynyl-19-nortestosterone, 2a-fluoro-17a- (1-propynyl) -19-nortestosterone and 2a-fluoro-17a-allyl-19-nortestosterone.
To esterify the 2a-fluoro-19-nor- testosterone obtained, a solution of 3 g of 2a-fluoro-19-nortestosterone in 20 cms. The reaction mixture was then poured into 250 cm3 of water and stirred for 2 hours at room temperature.
The crystalline product was filtered off, washed with water and dried, giving 3.1 g of 2a-fluoro-19-nortestosterone acetate. The analyte was obtained by recrystallization from methanol.
It melted at 148; [a] n: + 82 (CHCl3).

If the acylating agent is solid, an inert solvent such as toluene, xylene or dioxane can be added to produce a liquid esterification medium.
A solution of 1.58 g of 2a-fluoro-19-nortestosterone in a mixture of 35 cm3 of toluene and 15 cm3 of cyclohexanone was used for drying purposes for the oxidation to 2a-fluoro-19-nor-4-androstenedione- (3.17) the solution is heated until about 10 cm3 has passed over. 1.5 g of aluminum tert-butoxide are added to the dry solution; it is cooked until the implementation is complete.

1 g of 2a-fluoro-19-nortestosterone is oxidized at room temperature with a 20% excess of chromium trioxide in 25 cm- 'glacial acetic acid containing 1 cm3 of water until the reaction has ended. The reaction mixture is then poured into a large volume of water and carefully extracted with methylene chloride. The methylene chloride solution is washed with sodium bicarbonate solution and dried.
The dried solution is chromatographed on 40 g of magnesium silicate. The column is developed with hexane mixture containing increasing amounts of acetone. Essentially pure 2a-fluoro-19-nor-4-androstenedione (3.17) is eluted.
