Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Aminen der Formel

worin R1 einen Butyl- oder Pentylrest und R2 Methyl oder Äthyl oder einen Propyl- oder Butylrest bedeuten, und ihre Salze besitzen gute lokalanästhetische Eigenschaften und können dementsprechend als Heilmittel verwendet werden. In erster Linie sind hier zu erwähnen, das N-[ss-(m-n-Pentyloxyphenoxy)-äthyl]- methylamin, das N-[ss- (m-n-Pentyloxyphenoxy)-äthyl] isobutylamin, das N- [ss-(m-n-Butoxyphenoxy)-äthyl] - n-butylamin und das N-[ss-(m-n-Butoxyphenoxy)-äthyl]- äthylamin und ihre Salze.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man Verbindungen der Formel

worin R2 und n die oben gegebene Bedeutung haben, in der Phenolgruppe durch den Rest R, veräthert.
Die Verätherung wird zweckmässig unter Verwendung reaktionsfähiger Ester von Alkoholen der Formel R1OH, z. B. solchen von Halogenwasserstoffsäuren, wie Salzsäure, Brom- oder Jodwasserstoffsäure, oder Schwefelsäure, oder starken organischen Säuren, wie einer Sulfonsäure, z. B. einer Alkan- oder Arylsulfonsäure, durchgeführt, wobei man vorteilhaft in Anwesenheit alkalischer Kondensationsmittel, wie z. B. Alkali-alkoholaten oder -carbonaten, arbeitet.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form der freien Basen oder ihrer Salze. Aus den Salzen können in an sich bekannter Weise die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. der Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonäure, Weinsäure, Bernsteinsäure, Äpfelsäure, Methansulfonsäure, Äthansulfonsäure, Oxyäthansulfonsäure, Benzol- oder Toluolsulfonsäure, oder von therapeutisch wirksamen Säuren.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutisehen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
In dem folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
19,5 g (0,1 Mol) N-(m-Hydroxyphenoxy)-äthyln-propylamin werden zusammen mit 41,5 g (0,3 Mol) feingepulvertem wasserfreiem Kaliumcarbonat in 200 ml Methyläthylketon unter Rühren zum Rückfluss erhitzt. Nach Zugabe von 0,5 g Kaliumjodid tropft man langsam 13,6 g (0,1 Mol) n-Butylbromid zu. Nach zwölf Stunden lässt man abkühlen, nutscht von ungelöstem Material ab und dampft das Filtrat ein. Den Rückstand nimmt man in 200 ml Benzol auf und extrahiert mehrere Male mit verdünnter Natronlauge. Hierauf wäscht man die benzolische Phase mit Wasser neutral, trocknet sie über Natriumsulfat und dampft das Lösungsmittel ab. Der Rückstand wird im Hochvakuum im Claisenkolben destilliert. Das N-[ss-(m-n-Butoxyphenoxy)-äthyl]-n propylamin der Formel

wird als farbloses Ö1 erhalten.
Das Hydrochlorid schmilzt nach dem Umkristallisieren aus 2ithanol- Äther bei 147-148 .
Das als Ausgangsmaterial zur Verwendung gelangte N-(m-n-Hydroxyphenoxy)-äthyl-n-propylamirx kann wie folgt hergestellt werden:
In einer Lösung von 55 g (0,5 Mol) Resorcin in 133 g 1 Sgewichtsprozentiger Natronlauge lässt man in der Wärme 43 g (0,54 Mol) Äthylenchlorhydrin einlaufen und erwärmt anschliessend das Reaktionsgemisch unter energischem Rühren auf 90 bis 1000. Nach 15 Stunden kühlt man ab, stellt mit verdünnter Salzsäure leicht sauer und extrahiert mit Benzol. Aus den Benzolextrakten wird nach dem Verdampfen des Lösungsmittels das 2-(m-Hydroxyphenoxy)-äthanol durch fraktionierte Destillation als gelbes, hochviskoses Ö1 vom Kp.,,: 129-137 erhalten.
Man löst 60,0 g (0,39 Mol) 2-(m-Hydroxyphenoxy)-äthanol in 31 g (0,39 Mol) wasserfreiem Pyridin und tropft unter Wasserkühlung 49 g (0,41 Mol) Thionylchlorid zu. Anschliessend wird das Gemisch 1 Stunde auf 1000 erhitzt. Nach dem Abkühlen versetzt man mit 100 ml Wasser und verkocht 2 Stunden unter Rückfluss. Zugabe von weiteren 200 ml Wasser und Extraktion mit Benzol liefern ein dunkelgefärbtes Ö1, aus welchem durch fraktionierte Destillation das 2-(m-Hydroxyphenoxy) äthylchlorid als farblose Flüssigkeit vom Kp. 0 77-80 gewonnen werden kann.
36 g (0;21 Mol) 2-(m-Hydroxyphenoxy)-äthylchlorid und 59 g (1,0 Mol) n-Propylamin werden zusammen mit 100 ml Isopropanol 15 Stunden unter Rückfluss gekocht. Nach dem Abdampfen der leichtflüchtigen Anteile wird der Rückstand in 1000 ml in Salzsäure gegossen und die Neutralprodukte durch Ausschütteln mit wenig Benzol entfernt. Die saure wässerige Lösung wird mit Aktivkohle behandelt, filtriert, mit konz. wässerigem Ammoniak alkalisch gemacht und mit Methylenchlorid ausgezogen. Destillation des nach dem Entfernen des Lösungsmittels verbleibenden Rückstandes ergibt das N-[ss-(m-Hy- droxyphenoxyfäthyl]-n-propylamin der Formel

19.5 g (0.1 mol) of N- (m-hydroxyphenoxy) ethyln-propylamine are heated to reflux with stirring together with 41.5 g (0.3 mol) of finely powdered anhydrous potassium carbonate in 200 ml of methyl ethyl ketone. After adding 0.5 g of potassium iodide, 13.6 g (0.1 mol) of n-butyl bromide are slowly added dropwise. After twelve hours, the mixture is allowed to cool, undissolved material is filtered off with suction and the filtrate is evaporated. The residue is taken up in 200 ml of benzene and extracted several times with dilute sodium hydroxide solution. The benzene phase is then washed neutral with water, dried over sodium sulfate and the solvent is evaporated off. The residue is distilled in a Clais flask in a high vacuum. In a solution of 55 g (0.5 mol) of resorcinol in 133 g of 1% strength by weight sodium hydroxide solution, 43 g (0.54 mol) of ethylene chlorohydrin are allowed to run in while warm and the reaction mixture is then heated to 90 to 1000 with vigorous stirring. After 15 hours it is cooled, made slightly acidic with dilute hydrochloric acid and extracted with benzene. After evaporation of the solvent, 2- (m-hydroxyphenoxy) ethanol is obtained from the benzene extracts by fractional distillation as a yellow, highly viscous oil of bp. ,,: 129-137.
60.0 g (0.39 mol) of 2- (m-hydroxyphenoxy) ethanol are dissolved in 31 g (0.39 mol) of anhydrous pyridine and 49 g (0.41 mol) of thionyl chloride are added dropwise while cooling with water. The mixture is then heated to 1000 for 1 hour. After cooling, 100 ml of water are added and the mixture is refluxed for 2 hours. Addition of a further 200 ml of water and extraction with benzene yield a dark-colored oil, from which 2- (m-hydroxyphenoxy) ethyl chloride can be obtained as a colorless liquid with bp 0 77-80 by fractional distillation.
36 g (0; 21 mol) 2- (m-hydroxyphenoxy) ethyl chloride and 59 g (1.0 mol) n-propylamine are refluxed for 15 hours together with 100 ml isopropanol. After the volatile components have evaporated, the residue is poured into 1000 ml of hydrochloric acid and the neutral products are removed by shaking with a little benzene. The acidic aqueous solution is treated with activated charcoal, filtered, acidified with conc. made alkaline with aqueous ammonia and extracted with methylene chloride. Distillation of the residue remaining after removal of the solvent gives the N- [ss- (m-hydroxyphenoxyethyl] -n-propylamine of the formula
