Verfahren zur Herstellung neuer sekundärer Amine
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von sekundären Aminen der Formel

worin R einen gesättigten niederen Kohlenwasserstoffrest, der auch in der Kohlenstoffkette durch Sauerstoff unterbrochen sein kann, und R, einen niederen Kohlenwasserstoffrest bedeuten.
Der gegebenenfalls durch Sauerstoff unterbrochene gesättigte niedere Kohlenwasserstoffrest R ist vor allem aliphatischer Natur, z. B. ein niederer Oxaalkylrest, wie der 4-Oxa-pentyl-(l)-rest, oder vorzugsweise ein niederer, gerader oder verzweigter Alkylrest, z. B. Methyl, Äthyl, Isopropyl, Butyl, sek.-Butyl, Amyl, Isoamyl oder Hexyl, vor allem aber n-Propyl. Als niedere Kohlenwasserstoffreste Rt sind die eben erwähnten, vor allem Alkylreste, wie Methyl, oder Alkenylreste, wie Allyl, anzuführen.
Die neuen Verbindungen und ihre Salze besitzen ausgesprochen sympathicolytische Wirkung mit einer bradykarden und sedativen Wirkungskomponente.
Sie sollen dementsprechend als Medikamente verwendet werden.
Besonders wertvoll sind die Verbindungen der Formel

worin R4 für einen Sithyl-, Propyl-oder Butylrest steht, und ihre Salze.
Die sedative Wirkungskomponente ist besonders ausgeprägt beim N-(o-Methoxyphenoxy-äthyl)-N-n- propylamin und seinen Salzen, dessen Herstellung damit auch einen besonderen Gegenstand der Erfindung darstellt.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, dass man eine Verbindung der Formel

mit einer durch Reduktion den Alkohol HOR ergebenden Oxoverbindung unter reduzierenden Bedingungen umsetzt oder die Verbindung der vorstehenden Formel mit der genannten Oxoverbindung umsetzt und die so erhaltene Schiffsche Base der Reduktion z. B. mit Wasserstoff in Gegenwart von Katalysatoren, wie Nickelkatalysatoren, z. B. Raney Nickel, oder mit Di-Leichtmetallhydriden, wie Natriumborhydrid, unterwirft.
Die Reaktion kann in an sich bekannter Weise, in An- oder Abwesenheit von Verdünnungs- und/oder Kondensationsmitteln, bei gewöhnlicher oder erhöhter Temperatur im offenen oder im geschlossenen Gefäss unter Druck durchgeführt werden.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form der Basen oder ihrer Salze.
Aus den Salzen können in an sich bekannter Weise die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. der Halogenwasserstoffsäuren, Schwefelsäure, Salpeter- säure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Weinsäure, Bernsteinsäure, Apfelsäure, Methansulfonsäure, Äthansulfonsäure, Oxyäthansulfonsäure, Benzoloder Toluolsulfonsäure oder von therapeutisch wirksamen Säuren.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
16,7 g (0,1 Mol) ss-(o-Methoxyphenoxy)-äthyl- amin werden in 50 cm3 absolutem Benzol gelöst und bei Raumtemperatur mit 6,0 g (0,103 Mol) Propionaldehyd versetzt. Das sich sofort bildende Wasser wird in einem Wasserabscheider azeotrop entfernt. Hierauf dampft man das Lösungsmittel ab und destilliert den Rückstand im Hochvakuum. Die Schiffsche Base der Formel

ist eine gelbliche Flüssigkeit vom Kp. 005 : 79-820. a) 10,35 g (0,05 Mol) Schiffsche Base werden in 100 cm3 Methanol und 50 cm3 Wasser zum Sieden erhitzt. Unter kräftigem Rühren gibt man hierauf in 5-10 Portionen insgesamt 5 g (etwa 0,12 Mol) Natriumborhydrid zu.
Nachdem die Wasserstoffentwicklung abgeklungen ist, kühlt man auf 0", gibt 200 cm3 Wasser zu und extrahiert mit Äther. Das Produkt wird im Hochvakuum destilliert. Das N-(o Methoxy-phenoxyäthyl)-n-propylamin der Formel

destilliert als farbloses Öl bei 0,08 mml82-84t.
Das Hydrochlorid bildet feine farblose Nadeln aus Äthanol-Äther vom F. 141-1420 b) 10,35 g (0,05 Mol) Schiffsche Base werden in 150 cm3 Äthanol gelöst und in Gegenwart von 3-4 g Raney-Nickel in einem Druckautoklaven bei 100 atü/500 hydriert. Nach kurzer Zeit ist ein Mol Wasserstoff aufgenommen. Nach dem Abfiltrieren des Katalysators wird das Lösungsmittel verdampft und der Rückstand destilliert. Das Hydrierungprodukt ist nach Kp., Analyse und Schmelzpunkt des Hydrochlorids völlig identisch mit dem oben beschriebenen N-(o-Methoxy-phenoxyäthyl)-n-propylamin.
Das als Ausgangsstoff verwendete Amin wird z. B. wie folgt erhalten:
Zu 40 g (0,173 Mol) o-Methoxy-phenoxy-äthylbromid in 200 cm3 Methanol gibt man 40 g flüssigen Ammoniak und erhitzt im Schüttelautoklaven 10 Stunden lang auf 100 . Nach dem Abkühlen wird der überschüssige Ammoniak abgeblasen und das Methanol verdampft. Den Rückstand nimmt man in Chloroform auf, wäscht zweimal mit Wasser aus und trocknet über wasserfreiem Natriumsulfat. Nach dem Verdampfen des Lösungsmittels wird der Rückstand im Wasserstrahlvakuum destilliert. Das/3-(o-Methoxy- phenoxy)-äthylamin der Formel

Neutralisiert man die Base mit einer Lösung von Chlonvasserstoff in Essigester, so erhält man ein farbloses Hydrochlorid, welches nach Umkristallisieren aus Äthanol-Äther bei 116-117 schmilzt.

The invention relates to a process for the preparation of secondary amines of the formula

The new compounds and their salts have a pronounced sympathicolytic effect with a bradycardic and sedative component.

The inventive method for the preparation of the new compounds consists in that one compound of the formula

reacts with an oxo compound resulting from reduction to the alcohol HOR under reducing conditions or reacts the compound of the above formula with the oxo compound mentioned and the thus obtained Schiff base of the reduction z. B. with hydrogen in the presence of catalysts such as nickel catalysts, e.g. B. Raney nickel, or with di-light metal hydrides such as sodium borohydride, subjects.
The reaction can be carried out in a manner known per se, in the presence or absence of diluents and / or condensing agents, at normal or elevated temperature in an open or in a closed vessel under pressure.

The starting materials are known or can be obtained by methods known per se.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations, which contain them or their salts in a mixture with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral or parenteral administration.
In the following example the temperatures are given in degrees Celsius.

16.7 g (0.1 mol) of ss- (o-methoxyphenoxy) ethylamine are dissolved in 50 cm3 of absolute benzene and 6.0 g (0.103 mol) of propionaldehyde are added at room temperature. 40 g of liquid ammonia are added to 40 g (0.173 mol) of o-methoxy-phenoxy-ethyl bromide in 200 cm3 of methanol and the mixture is heated to 100 for 10 hours in a shaking autoclave. After cooling, the excess ammonia is blown off and the methanol is evaporated. The residue is taken up in chloroform, washed twice with water and dried over anhydrous sodium sulfate.
