mit Halogencarbonsäuren bzw. deren Estern der Formel Hal alkylen. COO R, wobei X und R die obige Bedeutung haben, zweckmässig in einem geeigneten Lösungsmittel in Gegenwart eines säurebindenden Mittels kondensiert. Falls mit einem Ester kondensiert wurde, können die Xanthon-oxyalkylen-carbonsäure- alkylester zu den entsprechenden Carbonsäuren verseift und umgekehrt freie Säuren entsprechend verestert werden.
Die so erhaltenen Oxyxanthonderivate besitzen starke gefässerweiternde Wirkung, insbesondere auf die Coronargefässe. Sie sind in dieser Beziehung z. B. dem Papaverin und dem Khellin bei der Behandlung von Coronarspasmen eindeutig überlegen.
Beispiel I
Eine Mischung aus 8,5 g l-Oxyxanthon, 7,5 g Bromessigsäureäthylester, 3 g wasserfreiem Kaliumcarbonat und 200 cm3 Aceton wird 12 Stunden unter Rühren am Rückfluss erhitzt. Nach dem Erkalten saugt man vom ausgeschiedenen Bromkalium ab und wäscht mit Aceton nach. Das Acetonfiltrat wird bis zur Trockene abgedunstet, der verbleibende Rückstand mehrmals mit Wasser verrieben, jeweils abgesaugt und schliesslich im Vakuumtrockenschrank getrocknet. Anschliessend wird aus Alkohol umkristallisiert.
Man erhält so Xanthon-l-oxyäthylacetat der Formel

Ausbeute: 9,5 g. F. = 124-1260.
In ganz analoger Weise können, ausgehend von 2-, 3- und 4-Oxyxanthon, die entsprechenden Xanthon-oxyäthylacetate hergestellt werden, und zwar
Xanthon-2-oxyäthylacetat vom F. = 97 980 (aus Alkohol),
Xanthon-3-oxyäthylacetat vom F. = 122-124 (aus Alkohol),
Xanthon-4-oxyäthylacetat vom F. = 160-161 (aus Alkohol).
Verseijung:
7 g Xanthon-l-oxyäthylacetat werden mit 40 cm3 alkoholischer Kalilauge 4 Stunden unter Rühren und Rückfluss erhitzt. Nach Abdestillieren des Lösungsmittels löst man den Rückstand in 150 cm3 Wasser, filtriert von Verunreinigungen und fällt das Filtrat mit verdünnter Salzsäure. Der abgeschiedene Kristallbrei wird abgesaugt, mit Wasser neutral gewaschen und getrocknet. Anschliessend wird aus Alkohol umkristallisiert.
Ausbeute: 5 g Xanthon-oxyessigsäure. F. = 190 bis 1920.
Aus den isomeren Xanthon-oxyäthylacetaten erhält man in gleicher Weise die entsprechenden Xanthon-oxyessigsäuren:
Xanthon-2-oxyessigsäure vom F. = 180-181 (aus Alkohol),
Xanthon-3-oxyessigsäure vom F. = 196-198 (aus Alkohol),
Xanthon-4-oxyessigsäure vom F. = 238-240 (aus Alkohol).
Die Xanthon-oxyessigsäuren können auch durch Kondensation der Oxyxanthone mit Monochloressigsäure in Gegenwart von säurebindenden Mitteln hergestellt werden.
Veresterung:
8 g Xanthon-2-oxyessigsäure werden in 60 cm3 Isopropanol unter Rühren zum Sieden erhitzt. Innerhalb von 10 Minuten tropft man sodann eine Lösung von 3,8 g N-y-Dimethylaminopropylchlorid in 10 cm3 Isopropanol zu, wobei die Suspension in eine klare Lösung übergeht. Es wird anschliessend noch etwa 4 Stunden am Rückfluss erhitzt. Nach dem Erkalten versetzt man die klare Lösung bis zur Trübung mit Äther und lässt sie einige Tage im Eisschrank stehen. Die abgeschiedenen Kristalle werden abgesaugt, mit Äther gewaschen und im Vakuumexsiccator getrocknet. Man erhält so das Xanthon-2-oxyessigsäure-dimethylamino-propylester-chlorhydrat.
Ausbeute: 9,5 g. F. = 119-122 .

alkylene with halocarboxylic acids or their esters of the formula Hal. COO R, where X and R have the above meaning, suitably condensed in a suitable solvent in the presence of an acid-binding agent. If condensation was carried out with an ester, the xanthone-oxyalkylene-carboxylic acid alkyl esters can be saponified to give the corresponding carboxylic acids and, conversely, free acids can be esterified accordingly.
The oxyxanthone derivatives obtained in this way have a strong vasodilating effect, in particular on the coronary vessels. You are z. B. clearly superior to papaverine and khelline in the treatment of coronary spasms.
Example I.
A mixture of 8.5 g of 1-oxyxanthone, 7.5 g of ethyl bromoacetate, 3 g of anhydrous potassium carbonate and 200 cm3 of acetone is refluxed for 12 hours while stirring. After cooling, the precipitated potassium bromide is suctioned off and washed with acetone. The acetone filtrate is evaporated to dryness, the remaining residue is triturated several times with water, in each case suctioned off and finally dried in a vacuum drying cabinet. It is then recrystallized from alcohol.
This gives xanthone-1-oxyethyl acetate of the formula

The corresponding xanthone oxyäthylacetate can be prepared in a completely analogous manner, starting from 2-, 3- and 4-oxyxanthone, namely

7 g of xanthone-1-oxyethyl acetate are heated with 40 cm3 of alcoholic potassium hydroxide solution for 4 hours with stirring and reflux. After the solvent has been distilled off, the residue is dissolved in 150 cm3 of water, impurities are filtered off and the filtrate is precipitated with dilute hydrochloric acid. The separated crystal slurry is filtered off with suction, washed neutral with water and dried. It is then recrystallized from alcohol.
Yield: 5 g of xanthone-oxyacetic acid. F. = 190 to 1920.
The corresponding xanthone oxyacetic acids are obtained in the same way from the isomeric xanthone oxyethyl acetates:
Xanthone-2-oxyacetic acid from F. = 180-181 (from alcohol),
Xanthone-3-oxyacetic acid from F. = 196-198 (from alcohol),
Xanthone-4-oxyacetic acid from F. = 238-240 (from alcohol).
The xanthone-oxyacetic acids can also be prepared by condensation of the oxyxanthones with monochloroacetic acid in the presence of acid-binding agents.
Esterification:
8 g of xanthone-2-oxyacetic acid are heated to the boil with stirring in 60 cm3 of isopropanol. A solution of 3.8 g of N-γ-dimethylaminopropyl chloride in 10 cm 3 of isopropanol is then added dropwise over the course of 10 minutes, the suspension turning into a clear solution. It is then refluxed for about 4 hours. After it has cooled down, ether is added to the clear solution until it becomes cloudy and it is left to stand in the refrigerator for a few days. The deposited crystals are filtered off with suction, washed with ether and dried in a vacuum desiccator. The xanthone-2-oxyacetic acid dimethylamino propyl ester chlorohydrate is obtained in this way.
Yield: 9.5 g. F. = 119-122.
