Verfahren zur Herstellung eines p-Nitro-benzoesäureesters. Es wurde gefunden, dass man zu einer neuen therapeutisch wertvollen Verbindung gelangen kann, wenn man eine Verbindung der Formel

und eine Verbindung der Formel CH3 (CH@)la # CH, . R', worin R und R' reaktionsfähige, mit Aus nahmeeines in ihnen enthaltenen Sauerstoff atoms sich bei der Reaktion abspaltende Reste bedeuten, aufeinander einwirken lässt. Die so erhältliche neue Verbindung bildet weisse Kristalle vom Schmelzpunkt 44-45 .
Sie zeigt eine ausgesprochene Wirksamkeit gegen Wurmerkrankungen am Warmblüter und ist, da sie überraschenderweise auch gut verträglich ist, zur therapeutischen Behand lung solcher Erkrankungen geeignet. Ausser- dem zeigt sie auch Wirkung gegen bakte rielle, z. B. durch Pneumokokken oder Strepto- kokken verursachte Infektionskrankheiten der Warmblüter. Sie soll dementsprechend therapeutische Anwendung finden.
Die neue Verbindung ist in Wasser un-. löslich, wenig löslich in Alkohol, hingegen leicht löslich in indifferenten organischen Lösungsmitteln. Für ihre praktische Anwen dung ist ihre Löslichkeit in Ölen und Lipoi den von Vorteil.
Die Herstellung der neuen Verbindung kann nach den für die Darstellung von Estern aromatischer Carbonsäuren an sich üblichen Arbeitsweisen erfolgen. Man verfährt z. B. derart, dass man die Mischung der p-Nitro- benzoesäure mit Dodecylalkohol mit Chlor- oder Bromwasserstoff sättigt und einige Zeit erhitzt oder dass man unter Zusatz von Schwefelsäure oder einer andern Säure er hitzt;
oder dass man ein p-Nitro-benzoesäure- halogenid mit dem Dodecylalkohol umsetzt, wobei unter Halogenwasserstoffentwicklung der Ester entsteht; oder dass man das Säure halogenid mit dem Dodecylalkohol in Gegen wart eines säurebindenden Mittels, z.
B. orga nischer Basen, wie Pyridin, oder Alkalien, wie Alkalicarbonat oder -hydroxyd, im in differenten oder wässrigen Mittel umsetzt; oder dass man das Säureanhydrid mit dem Dodecylalkohol in an sich üblicher Weise umsetzt; oder dass man Salze der p-Nitro- benzoesäure mit einem reaktionsfähigen Dodecylester, wie z. B. Dodecylhalogenid oder Dodecylsulfat, umsetzt.
Man kann auch niedere Ester durch Umesterung mit dem Dodecylalkohol in den Ester verwandeln. <I>Beispiel:</I> 92,5 g p-Nitro-benzoylchlorid werden mit 93 g Dodecylalkohol 20 Stunden auf 120 bis 125 erhitzt. Nach dem Erkalten der Mi schung wird Methylalkohol hinzugegeben, abgesaugt und mit Methylalkohol gewaschen. Naeh dem Umkristallisieren des Reaktions produktes aus Methylalkohol erhält man den p-Nitro-benzoesäure-dodecylester in weissen Kristallen vom F. 44-45 .
Dieselbe Verbindung erhält man, wenn eine 3Sischung von 92,5 g p-Nitro-benzo-,#1- chlorid, 93 g Dodecylalkohol und 75 am' Xylol tropfenweise mit 41,5 g Pyridin unter Eiskühlung versetzt wird, so. dass die Tempe ratur innen, 70 nicht überschreitet. Dann wird die Mischung 3 Stunden auf 120-13,0" erhitzt. Das Xylol wird unter vermindertem Druck entfernt und der Rückstand mit. Äther und Wasser versetzt.
Die ätherische Lösung wird mit n-Natronlauge gewaschen, getrock net und der Äther verdampft. Beim Umkri- stallisieren des Rückstandes aus Aceton- Methylalkohol erhält man den p-Nitro- benzoesäure-dodecylester wiederum in weissen Kristallen vom F. 44-45 .
Man erhält die Verbindung auch, wenn man eine Mischung von 33,4 g p-Nitro- benzoesäure und 61 g Dodecy lalkohol mit Chlorwasserstoff :sättigt. Die Mischung wird dann noch 5 Stunden zum Sieden unter Durchleiten von Chlorwasserstoff erhitzt, wobei Lösung eintritt. Nach Aufgiessen der llischun.g auf Eis wird mit Äther versetzt. Die ätherische Lösung wird mit n-Natron- lauge gewaschen, getrocknet und der Äther entfernt.
Man erhält nach dein L mkristalli- sieren des Rückstandes den oben beschriebe nen Ester.
Man erhält. die Verbindung auch, wenn man 19,5 g p-Nitro-henzoesäure-äthylester mit 18,6 g Dodecvlalkohol und 0, ? g Na trium einige Tage verrührt. Nach dem Auf giessen der Mischung auf Eiund Ausziehen mit n-Natronlauge wird das Reaktionspro- dukt getrocknet und fraktioniert. @Zan er hält .den p-Nitro-lbenzoeeäure-dodecy.le:stei.- vom Kp. ":

It shows a pronounced effectiveness against worm diseases in warm-blooded animals and is, since it is surprisingly well tolerated, suitable for the therapeutic treatment of such diseases. The new compound is un- in water. soluble, sparingly soluble in alcohol, but easily soluble in inert organic solvents. Their solubility in oils and lipids is advantageous for their practical application.
The preparation of the new compound can be carried out according to the procedures customary per se for the preparation of esters of aromatic carboxylic acids. One proceeds z. B. in such a way that the mixture of p-nitrobenzoic acid with dodecyl alcohol is saturated with hydrogen chloride or hydrogen bromide and heated for some time or that it is heated with the addition of sulfuric acid or another acid;
or that a p-nitrobenzoic acid halide is reacted with the dodecyl alcohol, the ester being formed with evolution of hydrogen halide; or that the acid halide with the dodecyl alcohol in the presence of an acid-binding agent, for.
B. organic bases, such as pyridine, or alkalis, such as alkali carbonate or hydroxide, in different or aqueous agents; or that the acid anhydride is reacted with the dodecyl alcohol in a conventional manner; or that salts of p-nitrobenzoic acid with a reactive dodecyl ester, such as. B. dodecyl halide or dodecyl sulfate.
Lower esters can also be converted into the ester by transesterification with the dodecyl alcohol. <I> Example: </I> 92.5 g of p-nitrobenzoyl chloride are heated with 93 g of dodecyl alcohol to 120 to 125 hours for 20 hours. After the mixture has cooled down, methyl alcohol is added, suction filtered and washed with methyl alcohol. After recrystallization of the reaction product from methyl alcohol, the p-nitrobenzoic acid dodecyl ester is obtained in white crystals with a melting point of 44-45.
The same compound is obtained when 41.5 g of pyridine are added dropwise to a mixture of 92.5 g of p-nitro-benzo, # 1 chloride, 93 g of dodecyl alcohol and 75% of xylene while cooling with ice. that the temperature inside does not exceed 70. The mixture is then heated to 120-13.0 "for 3 hours. The xylene is removed under reduced pressure and the residue is treated with ether and water.
The ethereal solution is washed with sodium hydroxide solution, dried and the ether is evaporated. When the residue is recrystallized from acetone-methyl alcohol, the p-nitrobenzoic acid dodecyl ester is again obtained in white crystals of 44-45.
The compound is also obtained when a mixture of 33.4 g of p-nitrobenzoic acid and 61 g of dodecyl alcohol is saturated with hydrogen chloride. The mixture is then heated to boiling for a further 5 hours while passing through hydrogen chloride, during which time solution occurs. After the mixture has been poured onto ice, ether is added. The ethereal solution is washed with sodium hydroxide solution, dried and the ether removed.
After crystallizing the residue, the ester described above is obtained.
You get. the connection also, if 19.5 g of p-nitro-benzoic acid ethyl ester with 18.6 g of dodecyl alcohol and 0,? g sodium stirred for a few days.
