Isobutylamin, Cyclopentylamin,
Cyclohexylmethylamin, Morpholin,
S-Methyl-cysteamin, S-Benzyl-cysteamin,
3-Methoxy-propylamin, Furfurylamin, p-Chlor-benzylamin, p-Methoxy-anilin,
N-Methylanilin, ss-Phenäthylamin und
Dipropylamin.
Besonders leicht reagieren Dimethylamin.

Process for the preparation of benzene disulfonamides
It is already known that electron-withdrawing substituents, for example nitro or cyano groups, loosen aromatically bound halogen atoms to such an extent that they can be exchanged for amino groups. This effect was previously
EMI1.1
wherein X is a chlorine or bromine atom, R1, R2, R3 and R4 are hydrogen atoms, saturated or unsaturated aliphatic hydrocarbon radicals optionally substituted by hydroxyl groups, aryl or aralkyl radicals optionally substituted by halogen atoms, alkyl or alkoxy groups or, together with the nitrogen atom, members of a saturated heterocyclic ring mean and in which the saturated or unsaturated aliphatic hydrocarbon radicals or
the alkylene groups in the aralkyl radicals can be interrupted by oxygen or sulfur, can be obtained in good yield and in a simple manner if a dihalogenodisulfohalide of the general formula is used
EMI1.2
preferably evaluated preparatively for the production of nitroanilines (cf. Houben-Weyl, 4th edition, Volume XI, Part 1, pages 63 and 64, and Table 10).
It has now been found that benzene disulfonamides of the general formula can be combined with a compound of the formula
EMI1.3
brings to implementation and the resulting dihalo-disulfonamides of the formula
EMI1.4
a compound of the formula
EMI1.5
can act.
As starting materials for the method according to the invention, for example
4,6-dichlorobenzene-1,3-disulfonic acid dichloride,
4,6-Dibwmbenzot- 1, 3-disulfonic acid dichloride and the corresponding -dibromide into consideration.
The starting materials are advantageously obtained by heating 1,3-dichlorobenzene or 1,3-dibromobenzene with four to six times the amount by weight of chlorosulfonic acid at temperatures of about 1700 ° C.
Work-up is conveniently carried out by pouring the cooled reaction mixture into ice. The disulfochlorides formed in the reaction are generally obtained in crystalline form and are practically sufficiently pure that further recrystallization is unnecessary. The yields are generally 70-75% of theory.
The first reaction stage of the process according to the invention consists in that a dihalodisulfohalide with ammonia or an amine of the general formula
EMI2.1
brings to implementation. According to an advantageous embodiment of the process, the starting materials, if appropriate in the presence of a solvent or diluent, are reacted with ammonia, preferably liquid ammonia, or with the above-mentioned amine at moderately elevated temperatures, advantageously at temperatures below 400.degree Excess of the amine in question is used to bind the hydrohalic acid liberated during the reaction. However, other inorganic or organic bases or compounds with a basic reaction can also be used to trap the hydrohalic acid.
Examples are alkali bicarbonates, calcium oxide, triethylamine or dimethylaniline. As a solvent, for. B. alcohols, ethers or ketones such as methanol, ethanol, acetone, dioxane or tetrahydrofuran in question.
The work-up can, for. B. be done in such a way that the reaction mixture is introduced into dilute aqueous mineral acid, such as hydrochloric acid, the dihalodisulfonamide separating in most cases in crystalline form. After suctioning off, washing and drying, the crude product can be used directly for further conversion.
The dihalogenated benzene-disulfonamides obtained are in the second reaction stage with compounds of the general formula
EMI2.2
treated, whereby it is expedient to use a two to four-fold excess of the basic reaction component in order to capture the hydrogen halide released during the reaction. The excess amounts of amine favor the yield without affecting the course of the reaction, because even if an excess of amine is used, the second halogen atom in the molecule of the dihalogenated benzenedisulfonamides is surprisingly not exchanged for the remainder of an amine, unless the temperature required for the exchange of the first halogen atom is exceeded very much.
If the amines are technically difficult to obtain, it is advisable to reduce the amine excess and instead add a tertiary organic base such as triethylamine, pyridine or dimethylaniline to bind the hydrohalic acid.
In addition to ammonia, practically any desired primary or secondary aliphatic, aromatic, araliphatic or heterocyclic amine is suitable as a reaction component of the dihalogenodisulfonamides. For example, the following amines may be mentioned: allylamine, ethylamine, ss, ss-diethoxy-ethylamine,
Isobutylamine, cyclopentylamine,
Cyclohexylmethylamine, morpholine,
S-methyl-cysteamine, S-benzyl-cysteamine,
3-methoxy-propylamine, furfurylamine, p-chlorobenzylamine, p-methoxy-aniline,
N-methylaniline, ss-phenethylamine and
Dipropylamine.
Dimethylamine reacts particularly easily. Benzylamine, pyrrolidine or piperidine, but it is also possible to cause aniline to react, although higher reaction temperatures are generally required.
The reaction can be carried out either in the melt or using a solvent or diluent, for example water or water-miscible inert solvents such as methanol, propanol or glycols being suitable. The reaction mixture is heated either under reflux or in an autoclave, depending on the type of starting materials used. The duration of the reaction is generally about 1-2 hours.
After the reaction has ended, the reaction mixture is poured into acidified water, the desired end product generally separating out in crystalline form. The crude product can optionally be purified by recrystallization from a suitable solvent or solvent mixture, such as water / dimethylformamide or itthanol / water.
The temperatures required for the reaction depend to a large extent on the choice of amino component and are generally between +40 and 2000 C. For example, when using cyclic amines, such as pyrrolidine, morpholine or piperidine, the reaction is already successful at temperatures between 40 and 700 C.
Secondary aliphatic amines and benzylamine, for example, can generally be reacted at temperatures between 70 and 900.degree. C., while primary amines, such as ethylamine, allylamine and isobutylamine, require reaction temperatures between about 90 and 1100.degree. When using ammonia, it is advisable to work at about 110 to 130.degree. The highest reaction temperatures are required when using aromatic amines.
A particularly advantageous embodiment of the process according to the invention is given when it is a question of the preparation of benzene disulfonamides whose formula the substituents of the aromatic amino group correspond to those of the sulfonic acid amido groups. Such process products to which the general formula
EMI3.1
(in which R1, R2 and X have the meaning given) can be conveniently prepared in such a way that a disulfohalide of the formula
EMI3.2
initially at temperatures below 40 C in excess ammonia, optionally liquid ammonia,
enters or with a compound of the formula
EMI3.3
brings to reaction and the reaction mixture obtained is heated to moderately elevated temperatures or to temperatures between 80 and 1500 C without isolation of intermediate products. The reaction product is worked up in the manner indicated above.
Some of the products of the process are new compounds and have valuable therapeutic properties. In particular, they are suitable as diuretics and saluretics. In addition, they are valuable intermediates for the production of therapeutically interesting secondary compounds.
The fact that, according to the process according to the invention, only one halogen atom of the dihalogenodisulfohalides, even in the presence of an excess of base, against ammonia or
Amines exchanged was not to be expected without further ado. Furthermore, it was surprising that the process according to the invention succeeds in consistently good yields without the use of catalysts and often even at moderately elevated temperatures, without any significant side reactions, for example transamidations of the sulfonic acid amide groups, taking place.
For the preparation of benzene-disulfonamides, which are substituted in the aromatic nucleus by halogen atoms and amino groups, only the following methods have hitherto been known: 1. Treatment of m-haloanilines with excess chlorosulfonic acid [cf. Liebigs Ann. d.
Chem. 623 (1959), page 157] or 2nd reaction of m-haloanilines with chlorosulfonic acid in the presence of sodium chloride to give the corresponding aniline-2,4-disulfochloride and conversion of the intermediate product obtained into the corresponding aniline-disulfonamide [cf. z. B. Belgian Patent No. 564006 and monthly journals for chemistry 48 (1927), page 87].
The known processes give only moderate yields and work with considerable technical difficulties; In particular, when sodium chloride is used, the strong foaming of the reaction mixture which occurs is a problem. A further disadvantage is that the known processes in question are not very suitable when applied to the above-mentioned compounds substituted on the nitrogen atom by hydroxyalkyl, alkenyl, aryl, aralkyl or heterocyclic radicals, since there is a risk that the chlorosulfonic acid with these residues in reaction and inconsistent products arise. These disadvantages are avoided by the novel process according to the invention described above in that the remainder of the amine, which could give rise to side reactions, is first introduced into the halobenzene disulfonamide that has already been formed.
