Verfahren zur Herstellung von Phenylalaninderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Phenylalanin-Derivaten der allgemeinen Formel:

worin R ein Radikal aus einem niedrigen Alkyl mit gerader Kette bedeutet. Diese Verbindungen sind bei der Behandlung von Hypertension nützlich.
Eine dieser Verbindungen, das a-Methyl-ss-(3,4- dioxyphenyl)alanin ist schon in der Literatur beschrieben. Diese Verbindung wird so hergestellt, dass man das 5-Methyl-5-(3,4-dimethoxybenzyl)hydan- toin mit Bariumhydroxyd zum a-Methyl-ss-(3 ,4-di- methoxyphenyl)alanin in Form des Bariumsalzes, reagieren lässt, anschliessend die Bariumionen durch Ausfällen mit Schwefelsäure entfernt, das Filtrat mit einer Base neutralisiert und die freie Aminosäure erhält, danach die freie Aminosäure mit 48 % iger Bromwasserstoffsäure bei Rückfluss unter Bildung des a-Methyl-B-(3,4-dioxyphenyl)alanin-hy- drobromids behandelt und schliesslich diese Verbindung neutralisiert und die freie Aminosäure a-Methyl p-(3,4-dioxyphenyl)alanin erhält.
Es ist entsprechend der vorliegenden Erfindung gefunden worden, dass ein Hydantoin der Formel:

worin R dasselbe bedeutet wie oben angegeben, direkt zum Hydrobromid des gewünschten Endproduktes a-(niedrig Alkyl)-ss - (3,4 - dioxyphenyl)alanin, unter Benützung einer wässrigen Bromwasserstoffsäure-Lösung von 35-55 Gew. % an HBr, hydrolysiert werden kann. Diese Hydrolyse wird bei einer Temperatur im Bereiche von etwa 900 C bis zur Rückflusstemperatur durchgeführt. Es ist von Vorteil, die Hydrolyse mit einer eigen Bromwasserstoffsäure bei einer Rückflusstemperatur von etwa 1260 C vorzunehmen.
Das so erhaltene Aminosäurehydrobromid wird in die freie Aminosäure, mit einer Base oder einem andern Protonakzeptor, wie Natriumhydroxyd, Diäthylamin, einem basischen Ionenaustauschharz, wie Amberlite IR-45 , oder Athylenoxyd, neutralisiert. Durch die Ausschaltung der Hydrolyse mit Bariumhydroxyd erzielt man eine höhere Gesamtausbeute bei der Konversion des Hydantoins in die gewünschte Aminosäure und gleichzeitig wird das Verfahren, durch die Herabsetzung der Anzahl der Reaktionsstufen und durch die Ausschaltung des oben angeführten Verfahrens zur notwendigen Entfernung der Bariumionen, vereinfacht.
Beispiel 1 a-Methyl-B-(3 4-dioxyphenyl) alanin
17 g von 5- Methyl - 5- (3,4 - dimethoxybenzyl)- hydantoin und 90 ml einer wässrigen Lösung, enthaltend 485S HBr wurde in eine masche gefüllt und bei Rückfluss, bei etwa 1260-1280 C, während 18 Stunden erhitzt. Nach fünfminutigem Erhitzen bei dieser Temperatur löste sich der Körper auf. Danach wurde der Überschuss an Bromwasserstoffsäure durch Erhitzen im Vakuum, bei 20 mm Hg auf einem Wasserbad bei 900 C entfernt. Das Erhitzen wurde fortgesetzt, bis der grösste Teil des Wassers ab destillierte. Dies erkannte man an der Bildung von Kristallen. Die Kristalle wurden in einem Lösungsmittelgemisch von 50 ml Methanol und 5 ml Wasser bei 300 C gelöst.
Diese Lösung wurde danach auf 50 C gekühlt und mit wässrigem 12 N Ammoniumhydroxyd auf pH 6 neutralisiert. Danach verdünnte man diese Lösung mit 300 ml Aceton, kühlte auf 0 C und hielt diese Temperatur während 3 Stunden aufrecht. Das rohe a-Methyl-A-(3, 4-dioxyphenyl)- alanin, welches ausfiel, wurde filtriert und dreimal mit 10 ml Portionen Aceton gewaschen. Die Farbe dieses Produktes war grau. Der Ertrag betrug 13,2 g mit 97%iger Ausbeute; F = 275-2800 C.
Das rohe α-Methyl-ss-(3,4-dioxyphenyl)alanin wurde zu 140 ml Wasser, welches 150 mg Natriumbisulfit enthielt, bei 90-950 C hinzugefügt. Nachdem sich der feste Körper löste, fügte man 500 mg Entfärbungskohle und 150 mg Weinsäure hinzu. Das Gemisch wurde in einer Stickstoffatmosphäre während 10 Minuten bei 90-950 C gerührt und filtriert.
Anschliessend kühlte man das Filtrat auf 0 C und liess es während 12 Stunden bei 0 C stehen. Das gereinigte Produkt wurde filtriert und dreimal mit 10-ml-Portionen Aceton gewaschen. Der Ertrag betrug 7,1 g, mit 52,5% iger Ausbeute; F = 316 bis 3170 C (Zers.). Diese Zersetzungstemperatur wurde auf diese Weise bestimmt, dass man eine Probe mit 20 pro Minute von 2800 C an bis zur Zersetzung erhitzte. Die Mutterlauge wurde auf 15 ml konzentriert, was eine Ausscheidung von weitern 2,45 g zur Folge hatte; F = 280-2820 C.
Beispiel 2 α-Äthyl-ss-(3,4-dioxyphenyl)alanin
Ein Gemisch von 5,0 g (0,018 Mole) von 5 Äthyl-5-(,4-dimethoxybenzyl)hydantoin und 30 ml einer 48% eigen wässrigen Bromwasserstoffsäure wurde während 46,5 Stunden unter Rückfluss behandelt.
Die Lösung wurde danach im Vakuum konzentriert.
Den Rückstand löste man in tert. Butanol auf und dampfte ihn im Vakuum ab; dies wurde wiederholt.
Danach wurde der Rückstand in Wasser gelöst und die Lösung filtriert. Um den pH auf 6 zu bringen, gab man konzentrierten Ammoniumhydroxyd hinzu.
Die Lösung wurde hernach durch Erhitzen mit 200 mg von Darco -Aktivkohle entfärbt und filtriert. Zwecks weiterer Entfärbung führte man eine kleine Menge Schwefeldioxyd in das Filtrat ein. Die Lösung wurde anschliessend im Vakuum, bis sich schwere Kristalle bildeten, konzentriert und dann gekühlt und bei etwa 5-100 C während etwa 16 Stunden gehalten. Die so gebildeten Kristalle von α-Äthyl-ss-(3,4-dioxyphenylalanin wurden durch Filtrieren gewonnen, nacheinander mit kaltem Wasser und Äther gewaschen und bei etwa 75-800 C im Vakuum getrocknet. Der Ertrag betrug 1,87 g, das ist mit einer 46% eigen Ausbeute; F = 308-3090 C.

One of these compounds, α-methyl-ss- (3,4-dioxyphenyl) alanine, has already been described in the literature. This compound is produced in such a way that the 5-methyl-5- (3,4-dimethoxybenzyl) hydantoin reacts with barium hydroxide to give a-methyl-ss- (3,4-dimethoxyphenyl) alanine in the form of the barium salt then the barium ions are removed by precipitation with sulfuric acid, the filtrate is neutralized with a base and the free amino acid is obtained, then the free amino acid with 48% hydrobromic acid at reflux to form a-methyl-B- (3,4-dioxyphenyl) treated alanine-hydrobromids and finally this compound neutralized and the free amino acid a-methyl p- (3,4-dioxyphenyl) alanine obtained.
It has been found in accordance with the present invention that a hydantoin of the formula:

where R is the same as stated above, hydrolyzed directly to the hydrobromide of the desired end product a- (lower alkyl) -ss - (3,4-dioxyphenyl) alanine, using an aqueous hydrobromic acid solution of 35-55% by weight of HBr can be. This hydrolysis is carried out at a temperature in the range from about 900 C to the reflux temperature. Example 1 a-Methyl-B- (3 4-dioxyphenyl) alanine
17 g of 5-methyl-5- (3,4-dimethoxybenzyl) -hydantoin and 90 ml of an aqueous solution containing 485S HBr were placed in a mesh and heated at reflux, at about 1260-1280 ° C., for 18 hours. After heating for five minutes at this temperature, the body dissolved. Thereafter, the excess hydrobromic acid was removed by heating in vacuo at 20 mm Hg on a water bath at 900 ° C. This solution was then cooled to 50 ° C. and neutralized to pH 6 with aqueous 12 N ammonium hydroxide. This solution was then diluted with 300 ml of acetone, cooled to 0 ° C. and this temperature was maintained for 3 hours. The crude α-methyl-A- (3, 4-dioxyphenyl) alanine which precipitated was filtered and washed three times with 10 ml portions of acetone. The color of this product was gray. The yield was 13.2 g with a 97% yield; F = 275-2800 C.
The crude α-methyl-ss- (3,4-dioxyphenyl) alanine was added to 140 ml of water containing 150 mg of sodium bisulfite at 90-950 ° C. After the solid was dissolved, 500 mg of decolorizing charcoal and 150 mg of tartaric acid were added. The mixture was stirred in a nitrogen atmosphere at 90-950 ° C. for 10 minutes and filtered.
The filtrate was then cooled to 0 C and left to stand at 0 C for 12 hours. The purified product was filtered and washed three times with 10 ml portions of acetone. The yield was 7.1 g, with a 52.5% yield; F = 316 to 3170 C (dec.). This decomposition temperature was determined in this way by heating a sample at 20 per minute from 2800 ° C. until decomposition. The mother liquor was concentrated to 15 ml, which resulted in a further excretion of 2.45 g; F = 280-2820 C.
Example 2 α-Ethyl-ss- (3,4-dioxyphenyl) alanine
A mixture of 5.0 g (0.018 moles) of 5 ethyl 5- (, 4-dimethoxybenzyl) hydantoin and 30 ml of 48% aqueous hydrobromic acid was refluxed for 46.5 hours.
The solution was then concentrated in vacuo.
The residue was dissolved in tert. Butanol on and evaporated in vacuo; this was repeated.
The residue was then dissolved in water and the solution was filtered. Concentrated ammonium hydroxide was added to bring the pH to 6.
The solution was then decolorized by heating with 200 mg of Darco® activated charcoal and filtered. A small amount of sulfur dioxide was introduced into the filtrate for further discoloration. The solution was then concentrated in vacuo until heavy crystals formed and then cooled and kept at about 5-100 ° C. for about 16 hours. The crystals of α-ethyl-ss- (3,4-dioxyphenylalanine thus formed were collected by filtration, washed successively with cold water and ether, and dried in vacuo at about 75-800 ° C. The yield was 1.87 g, the is with a 46% own yield; F = 308-3090 C.
