Verfahren zur Herstellung von bisquaternisierten Aminen
Gegenstand der vorliegenden Erfindung ist ein Verfahren, zur Herstellung von bisquaternisierten Aminen, welches dadurch gekennzeichnet ist, dass man eine Carbonylverbindung der Formel

worin R, den 2, 6, 6-Trimethyl-cyclohexyl- (l)-rest oder dessen 1, 2-Dehydroform bezeichnet und wobei die C=C-Doppelbindung in der Seitenkette hydriert sein kann, mit einem a, c,-Diaminoalkan reduktiv kondensiert, das erhaltene bisekundäre Diamin alkyliert und das enstandene bitertiäre Diamin mit einem den Rest R abgebenden Quaternisierungsmittel umsetzt. (Bedeutung von R siehe weiter unten.)
Die Alkylreste der als Alkylierungs-bzw.
Quaternisierungsmittel verwendeten Verbindungen können geradkettig oder verzweigt sein. Bevorzugt sind die niederen Alkylreste, wie z. B. Methyl, Athyl, Propyl, Isopropyl, Butyl, Isobutyl, Amyl, Hexyl und Heptyl.
Es wird vorzugsweise ein a, ov-Diaminoalkan mit 2 bis 12 Kohlenstoffatomen verwendet. Die reduktive Kondensation wird zweckmässigerweise mittels Wasserstoff in Gegenwart eines Hydrierungskatalysators, wie z. B. Raney-Nickel, in einem inerten Lösungsmittel, z. B. in einem niederen Alkanol, wie Methanol oder Athanol, bei erhöhter Temperatur und unter erhöhtem Druck durchgeführt. Die Carbonylverbindung wird zweckmässigerweise mit etwa 1, Z2 Squivalent des a, cs-Diaminoalkans kondensiert.
Das erhaltene bisekundäre Diamin wird dann, wie gesagt, zum entsprechenden bitertiären Diamin alkyliert. Wenn der einzuführende Alkylrest der Methylrest ist, kann man die Alkylierung z. B. mittels Ameisensäure und Formaldehyd bei erhöhter Temperatur durchführen. Die Quaternisierung erfolgt zweckmässigerweise durch Behandlung des bitertiären Diamins mit einem Alkylhalogenid, wie Methylchlorid oder Methylbromid, einem Alkylester einer anorganischen oder organischen Säure, wie z. B. p-Toluol sulfonsäuremethylester, Benzolsulfonsäuremethylester, oder einem andern zweckentsprechenden Quaternisierungsmittel in einem Lösungsmittel, wie z. B. Methanol oder Athanol, bei Raumtemperatur oder er höhter Temperatur, nötigenfalls unter erhöhtem Druck.

in welcher m die Zahl 0 oder 1, R untereinander gleiche Alkylreste, vorzugsweise niedere Alkylreste, bedeuten und Ri den 2, 6, 6-Trimethyl-cyclohexyl-(l)- rest oder dessen 1, 2-Dehydroform, wobei die beiden Reste Ri untereinander gleich sind, R2 einen Alkylenrest, vorzugsweise einen solchen mit 2 bis 12 Kohlenstoffatomen, und X das Anion einer anorganischen Säure, wie z. Die Verbindungen der Formel II besitzen eine symmetrische Struktur, da die durch R bezeichneten Alkylreste untereinander gleich sind, beide Molekülhälften das gleiche Anion aufweisen und m in beiden Molekül- hälften die gleiche Zahl ist. Die symmetrischen bisquaternisierten Amine können allerdings in bezug auf die räumliche Konfiguration in verschiedenen Formen auftreten.
Die gemäss der Erfindung hergestellten bisquaternisierten Amine sind als antimikrobielle Mittel, beispielsweise zur Bekämpfung von Infektionen, die durch Bakterien, wie z. B. Streptokokken und Staphylokokken, Protozoen, wie z. B. Trichomonas vaginalis, oder Pilze, wie z. B. Trichophyton mentagrophytes und Microsporon lanosum, verursacht werden, verwendbar. Die Verfahrensprodukte sind insbesondere auch gegen Staphylokokkenstämme, die gegen Antibiotika resistent sind, wirksam. Sie können äusserlich, z. B. in Form von Cremen, Lösungen, Salben und dergleichen, oder parenteral, z. B. in einem sterilen Medium, wie sterilem Wasser, verabreicht werden.
Die Verfahrensprodukte können in üblicher Weise in therapeutisch wirksamer Dosierung Trägersubstanzen, Exzipientien und dergleichen einverleibt werden.
Im folgenden Ausführungsbeispiel sind alle Temperaturen in Celsiusgraden angegeben.
Beispiel
Zu einer Lösung von 49 g (0, 25 Mol) cis-Tetrahydrojonon und 14, 1 g (0, 12 Mol) 1, 6-Hexandiamin in 150 cm3 Athanol wird ein Teelöffel Raney-Nickel zugefügt. Durch Zugabe von Athanol wird das Volumen auf 300 cm3 gebracht. Das Gemisch wird bei 50 und einem Druck von 14 kgcms hydriert. Der Katalysator wird abfiltriert, das Filtrat eingeengt und das zurückbleibende 01 im Vakuum fraktioniert, wobei man
N, N'-Bis- (l-methyl-3- [2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-hexandiamin-(l, 6) vom Sdp. 192-202 (0, 02 mm) erhält.
Wenn man eine Probe mit alkoholischem Chlorwasserstoff behandelt, so erhält man
N, N'-Bis-(1-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-hexandiamin- (l, 6) dihydrochlorid vom Smp. 197-199 (nach Umlösen aus einem Gemisch von Athanol und Ather).
217 g (0, 456 Mol)
N, N'-Bis- (l-methyl-3- [2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-hexandiamin- (1, 6) werden mit 182 cm3 (3, 04 Mol) Ameisensäure (90 /o) versetzt. Die so erhaltene farblose Lösung wird gekühlt, worauf 91, 3 cm3 (1, 043 Mol) Formaldehyd (370/o) zugesetzt werden. Die Lösung wird während 2 Stunden bei Wasserdampftemperatur unter gelegentlichem Schütteln erhitzt und dann während 8 Stunden am Rückfluss erhitzt. Die flüchtigen Bestandteile werden bei Wasserdampftemperatur und im Wasserstrahlvakuum abdestilliert, und das zurückbleibende bl wird durch Zugabe von 50 /eigem Kaliumhydroxyd stark alkalisch gestellt. Das Reaktionsprodukt wird mit Ather ausgezogen.
Der Atherextrakt wird mit Wasser gewaschen, getrocknet und im Vakuum eingeengt. Das zurückbleibende 01 wird im Vakuum fraktioniert, wobei man
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-N, N'-dimethyl-hexan diamin-(1, 6) erhält ; Sdp. 230-2400/'0, 4 mm ; n 26¯ 1, 4833. Wenn man eine Probe dieser Verbindung mit einer äthano- lischen Chlorwasserstofflösung behandelt, so erhält man das kristalline Dihydrochlorid vom Smp. 183 bis 185 (nach Umkristallisieren aus einem Gemisch von Athanol und Acetonitril).
Eine Lösung von 5 g
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-N, N'-dimethyl-hexan diamin- (1, 6) in 150 cm3 Aceton wird mit 50 cm3 einer 40/oigen Lösung von Methylbromid in Aceton versetzt. Die Lösung wird über Nacht bei Raumtemperatur stehen- gelasse. Die sich dabei bildenden Kristalle werden abfiltriert, mit Aceton gewaschen und getrocknet, wobei man
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-N, N'-dimethyl-hexan diamin- (1, 6)-bis- (methobromid) vom Smp. 265-266 erhält.
Die im ersten Absatz dieses Beispiels beschriebene reduktive Kondensation wird wiederholt, doch werden anstelle von Raney-Nickel (1) 1, 5 g Platinoxyd und in einem weiteren Versuch (2) 1, 5 g Palladium-auf-Calciumcarbonat (Pd-Gehalt 1011/o) verwendet, wobei man dasselbe Produkt,
N, N'-Bis- (l-methyl-3- [2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-hexandiamin- (1, 6), erhält.
Zu einer Lösung von 5 g
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan diamin-(1, 6) in 100 cm3 Methanol und 4 werden 100 cm3 Methanol, die 10 g Methylchlorid enthalten, zugefügt.
Die Lösung wird während 15 Stunden bei 60 in einem geschlossenen Gefäss erhitzt. Die farblose Lösung wird eingeengt und der so erhaltene weisse Fest stoff aus einem Gemisch von Äthanol, Acetonitril und Äther kristallisiert, wobei man
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan diamin- (1, 6)-bis- (methochlorid)-hemihydrat erhält.
Zu einer Lösung von 5 g
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan diamin-(1, 6) in 100 cm3 Methanol werden 10 g p-Toluolsulfon- säuremethylester, gelöst in 100 cm3 Aceton, zugefügt. Die so erhaltene Lösung wird während 18 Stunden unter Rückfluss erhitzt und eingeengt, und der entstandene Sirup aus einem Gemisch von Acetonitril, Aceton und Ather kristallisiert, wobei man
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan diamin- (1, 6)-bis- (methyl-p-toluolsulfonat) vom Smp. 191-193 erhält.

Der Niederschlag wird abfiltriert, mit Wasser gewaschen und aus 25/oigem Athanol umkristallisiert, wobei man
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo hexyl]-propyl)-N, N'-dimethyl-hexan diamin- (1, 6)-bis- (methylnitrat), Smp. 287-288 unter Zersetzung, erhält.
Zu einer Lösung von 20 g
N, N'-Bis-(l-methyl-3-[2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan- diamin-(1, 6) in 250 cm3 Athanol werden 21, 8 g Athylbromid zugesetzt. Die Lösung wird während 15 Stunden bei 800 erhitzt. Die flüchtigen Bestandteile werden abdestilliert, und der Rückstand wird aus einem Gemisch von Aceton und Ather kristallisiert, wobei man
N, N'-Bis- (l-methyl-3- [2', 6', 6'-trimethyl-cyclo- hexyl]-propyl)-N, N'-dimethyl-hexan- diamin-(1, 6)-bis-(äthobromid) vom Smp. 196-198 erhält.

Compounds used in quaternizing agents can be straight-chain or branched. The lower alkyl radicals, such as. B. methyl, ethyl, propyl, isopropyl, butyl, isobutyl, amyl, hexyl and heptyl.
