Procédé de préparation d'un dérivé de la dichloracétanilide
La présente invention a pour objet un procédé de préparation d'un dérivé de la dichloracétanilide, pos sédant des propriétés amibicides utiles.
On sait que le composé dichloracét-4-hydroxy-Nméthylanilide, désigné ci-après composé G possède des propriétés qui le rendent intéressant pour le traitement clinique des amibiases chez les humains.
On a également établi que certains esters de ce composé, notamment le benzoate, présentent des propriétés amibicides. Des procédés permettant de préparer ces esters aussi économiquement que possible sont donc désirables.
Le benzoate et autres esters du composé G peuvent tre préparés par estérification directe de ce dernier composé, conformément aux procédés décrits dans le brevet britannique No 794762. L'économie de ces procédés est déterminée dans une grande mesure par la disponibilité et le prix de revient de la matière de départ, la p-hydroxy-N-méthylaniline, et des procédés utilisant des matières différentes et moins coûteuses sont désirables. En outre, il peut parfois tre plus avantageux économiquement de produire un ester du composé G sans devoir préparer ce dernier composé comme produit intermédiaire.
Le procédé selon l'invention, de préparation d'esters du composé G, présente les avantages cités cidessus.
Ce procédé a pour objet la préparation d'un composé de formule:

dans laquelle R1 est un radical acyle de formule
R2CO-, RO désignant un groupe alcoyle, alcényle ou phényle ou un groupe alcoyle substitué par au moins un atome de chlore, ou R1 est un radical de formule XSO2 -, dans laquelle X désigne un radical alcoyle, phényle ou phényle substitué. Ce procédé est caractérisé en ce que l'on dichloroacétyle du p-aminophénol en p-dichloroacétamido-phénol, en ce que l'on estérifie le groupe hydroxyle de ce dernier, obtenant ainsi un composé de formule:

et en ce que l'on méthyle ce dernier.
Le diagramme schématique ci-après (fig. 1) permettra de mieux comprendre le procédé selon l'invention. On peut voir que la matière première essentielle est le p-amino-phénol (A), et que pour convertir ce composé en les esters désirés du composé G, il est nécessaire d'introduire un groupe dichloracétyle et un groupe méthyle sur l'atome d'azote.

Le p-aminophénol (A) peut tre dichloroacétylé par l'une quelconque des méthodes suivantes:
a) Traitement du composé A avec de la cyanhy
drine de chloral en présence d'un agent de liaison
d'acide, la cyanhydrine de chloral étant ajoutée au
mélange réactionnel comme telle ou préparée in
situa; cette méthode est décrite dans le brevet
britannique No 786806. b) Traitement du composé A avec du chlorure de
dichloracétyle, si désiré en présence d'un agent
de liaison d'acide. c) Traitement du composé A avec de l'anhydride
dichloracétique, si désiré en présence d'un agent
de liaison d'acide. d) Traitement du composé A avec de la N :
N-bis-
dichloracétamide. e) Traitement d'un mélange du composé A et d'acide
dichloracétique en présence de trichlorure de
phosphore. f) Traitement du composé A avec du dichloraceto-
nitrile en solution dans un alcool aliphatique infé
rieur.
On obtient ainsi le composé M. Avant de méthyler le composé M, il est nécessaire de protéger le groupe hydroxyle par l'introduction du groupe Ri et la formation du composé N. Le composé N peut ensuite tre méthylé, par exemple par traitement avec un halogénure de méthyle, de préférence l'iodure, en présence d'un alcali, notamment d'hydroxyde de potassium, ou par traitement avec du sulfate de diméthyle, en présence, par exemple, de carbonate de potassium, ou par traitement avec du p-toluène-sulfo- nate de méthyle. Qn obtient ainsi le composé (a).
Exemple 1
Préparation du benzoate de 4-(dichloracét-N-méthyl
amido)-phényle (composé a avec Rt=C6H5CO-)
par la voie p-aminophénol (A) + composé M + composé N N composé a.
I) Le p-aminophénol (A) est converti en p-dichloracétamidophénol (M) par l'une des méthodes suivantes: a) On agite un mélange de 10,9 g de p-aminophénol,
22 g de cyanhydrine de chloral et 75 ml de pyri
dine anhydre, tout en ajoutant lentement et en
refroidissant 30ml de triéthylamine. Une réac
tion exothermique se produit et on règle la vitesse
d'addition de la triéthylamine de manière que la
température du mélange réactionnel se maintienne
entre 10 et 150 C. Lorsque l'addition est terminée,
on enlève le bain réfrigérant et on coule le mé
lange dans 200 mi d'un mélange de glace et d'eau.
On isole par filtration le produit solide qui s'est
séparé, on le lave à l'acide chlorhydrique dilué
jusqu'à ce qu'il soit exempt de pyridine et on le
sèche sous vide. On recristallise le produit résul
tant dans un mélange de 35 ml d'alcool et 50 ml
d'eau. On obtient ainsi le p-dichloracétamidophé
nol sous forme d'un solide cristallin incolore de
point de fusion égal à 137-1390 C. b) A une solution de 109 g de p-aminophénol dans
un mélange de 196 mi d'acide chlorhydrique
5,1 N et 1,4 litre d'eau, on ajoute en agitant une
solution de 295 g d'hydrate de chloral dans
525 ml d'eau, 3 g de sulfite de sodium heptahy
drate et 94 g de cyanure de sodium à 90%.
On
ajoute ensuite au mélange, en agitant, 535 mi
d'hydroxyde de sodium 5,04 N. La température
monte à 490 C et on agite le mélange pendant
encore 2 heures, durant lesquelles la température
tombe à 250 C. On ajoute de la glace au mélange
de manière à réduire la température à 130 C et
on lave le solide qui s'est séparé avec de l'eau,
puis avec de l'acide chlorhydrique N, et finale
ment avec de l'eau, avant de le recristalliser dans
un mélange de 200 mi d'alcool et 350 ml d'eau.
On obtient ainsi le p-dichloracétamidophénol sous
forme d'un solide cristallin fondant à 137-1390 C. c) On traite, en agitant et à une température de
S-10o C, un mélange de 16,5 g de p-aminophénol,
22g d'acétate de sodium trihydrate, 100ml de
benzène, 200 ml d'eau et 0,5 ml de solution satu
rée de bisulfite de sodium, avec une solution de
22,5 g de chlorure de dichloracétyle dans 50ml de
benzène. Après avoir terminé l'addition, on agite
le mélange pendant encore une heure à tempéra
ture ordinaire, on isole par filtration le produit
solide qui s'est séparé, on le lave à l'eau et on
le recristallise deux fois dans de l'éthanol aqueux.
On obtient ainsi le p-dichloracétamidophénol sous
forme d'un solide cristallin de point de fusion de 137-139 C. (Trouvé: C = 43,3 ; H = 3,2%.
C8H7O2NCl2 correspond à C=43,7; H=3,2%.)
II) Le p-dichloracétamidophénol (M) est converti en benzoate de 4-dichloracétamidophényle (composé
N avec R1 = CGH5CO-) de la manière suivante:
On chauffe à 1000 C, en agitant, un mélange de 44 g de p-dichloracétamidophénol et 70ml de pyridine sèche, jusqu'à obtention d'une solution limpide, que l'on refroidit à 130 C en agitant, et on obtient une suspension. On ajoute à cette suspension, lentement et en agitant, 24 ml de chlorure de benzoyle, en refroidissant à 14-160 C et, lorsque l'addition est terminée, on agite le mélange pendant encore 15 minutes, avant de le traiter avec 400 mi d'eau glacée.
Après avoir agité pendant encore 2 heures, on isole par filtration le produit solide qui s'est séparé, et on le lave à reau, puis avec de l'acide chlorhydrique N, et finalement à l'eau, après quoi on l'essore bien sur le filtre et on le recristallise dans du cellosolve. On obtient ainsi le benzoate de 4-dichloracétamidophé- nyle, sous forme d'un solide cristallin incolore de point de fusion égal à 207-2080 C.
(Trouvé C=55,5; H=3,5%. C10H1O8NC12 correspond à C=55,6; H=3,4%.)
III) Le benzoate de 4-dichloracétamidophényle obtenu comme décrit sous 11) est ensuite converti en benzoate de 4-(dichloracét-N-méthylamido) -phényle (composé a avec R' =C6H5CO -) par l'une ou l'autre des méthodes suivantes a) On chauffe à reflux en agitant un mélange de
16,2 g de benzoate de 4-dichloracétamidophényle
et 200 ml d'acétone et on traite avec 11 g d'hy
droxyde de potassium broyé, puis, 2 minutes plus
tard, avec 10,6 g d'iodure de méthyle et, après
encore 5 minutes, par 4,5 g d'iodure de méthyle.
Lorsque l'addition est terminée, on chauffe le
mélange à reflux pendant encore 10 minutes,
avant de le refroidir à la température ordinaire,
et on le filtre. On évapore le filtrat et les eaux de
lavage à sec et on dissout le résidu dans 100ml
de benzène. On filtre la solution benzénique, on
évapore le filtrat à sec, et chauffe le résidu à
1000 C sous une pression de 2 mm de mercure
pendant une brève durée.
On lave le résidu par
décantation avec 200 ml d'éther de pétrole (zone
d'ébullition 40-600 C) et on le recristallise dans
l'alcool. On obtient ainsi le benzoate de 4-(di
chloracét-N-méthylamido)-phényle sous forme
d'un solide cristallin de point de fusion de 118
1190 C. b) On chauffe à reflux pendant 4 heures, en agitant,
un mélange de 10g de benzoate de 4-dichlor
acétamidophényle, 10g de carbonate de potas
sium, 4 g de sulfate de diméthyle et 50 ml d'acé
tone, après quoi on refroidit à la température
ordinaire et on filtre. On évapore le filtrat à sec
et on recristallise le résidu deux fois dans l'alcool.
On obtient ainsi le benzoate de 4-(dichloracét-N
méthylamido)-phényle sous forme d'un solide
cristallin de point de fusion de 118-119o C.
Exemple 2
Préparation de l'acétate de 4-(dichloracét-N-méthyl-
amido)-phényle (composé a avec R' = CH5CO -
par la voie p-aminophénol (A) + composé M
composé N + composé a.
I) Le p-aminophénol (A) est converti en p-di chloracétamidophénol (M) par l'une des méthodes décrites dans l'exemple 1 (I).
II) Le p-dichloracétamidophénol (M) est converti en acétate de 4-dichloracétamidophényle (composé N avec = Rl=CH3CO-) de la manière suivante:
On prépare une solution en dissolvant 22 g de 4-dichloracétamidophénol dans 35 ml d'hydroxyde de sodium 5 N et 100 g d'eau glacée. On ajoute à la solution 70 g de glace et 14 ml d'anhydride acétique et on secoue vigoureusement le mélange pendant une brève durée. On isole par filtration le produit solide qui s'est séparé, on le lave à reau et on le recristallise dans de l'alcool. On obtient ainsi l'acétate de 4dichloracétamidophényle sous forme d'un solide cristallin blanc de point de fusion de 168-169oC.
(Trouvé: C=45,9 ; H=3,5%. 'C,0H9O3'NCl2 correspond à C=45,8; H=3,4%).
III) L'acétate de 4-dichloracétamidophényle est ensuite converti en acétate de 4-(dichloracét-N-méthylamido)-phényle (composé a avec Rl=CH3CO-) de la manière suivante:
On chauffe à reflux pendant 4,5 heures un mélange de 7,86 g d'acétate de 4-dichloracétamidophényle, 8 g de carbonate de potassium anhydre, 4 g de sulfate de diméthyle, et 40 ml d'acétone sèche, après quoi on refroidit à température ordinaire et on filtre.
On évapore le filtrat à sec et on recristallise le résidu dans de l'alcool. On obtient ainsi l'acétate de 4-dichloracét-N-méthylamido)-phényle sous forme d'un solide cristallin de point de fusion de 89,50 C.
Process for preparing a dichloroacetanilide derivative
The present invention relates to a process for preparing a derivative of dichloroacetanilide, possessing useful amoebicidal properties.
It is known that the compound dichloroacet-4-hydroxy-N-methylanilide, hereinafter referred to as compound G, has properties which make it useful for the clinical treatment of amoebiasis in humans.
It has also been established that certain esters of this compound, in particular benzoate, exhibit amebicidal properties. Methods of preparing these esters as economically as possible are therefore desirable.
The benzoate and other esters of compound G can be prepared by direct esterification of the latter compound, in accordance with the processes described in British Patent No. 794762. The economy of these processes is determined to a large extent by the availability and the cost price. starting material, p-hydroxy-N-methylaniline, and methods using different and less expensive materials are desirable. In addition, it can sometimes be more economically advantageous to produce an ester of compound G without having to prepare the latter compound as an intermediate product.
The process according to the invention for preparing esters of compound G has the advantages mentioned above.
The object of this process is the preparation of a compound of formula:

This process is characterized in that p-aminophenol is dichloroacetylated into p-dichloroacetamido-phenol, in that the hydroxyl group of the latter is esterified, thus obtaining a compound of formula:

and in that the latter is methylated.
The schematic diagram below (FIG. 1) will make it possible to better understand the process according to the invention. It can be seen that the essential raw material is p-amino-phenol (A), and that in order to convert this compound into the desired esters of compound G, it is necessary to introduce a dichloroacetyl group and a methyl group on the atom. nitrogen.

III) The 4-dichloracetamidophenyl benzoate obtained as described under 11) is then converted into 4- (dichloracet-N-methylamido) -phenyl benzoate (compound a with R '= C6H5CO -) by one or the other of following methods a) Heated to reflux with stirring a mixture of

and 200 ml of acetone and treated with 11 g of hy
crushed potassium droxide, then, 2 minutes more
late, with 10.6 g of methyl iodide and, after
another 5 minutes, with 4.5 g of methyl iodide.

mixture at reflux for a further 10 minutes,
before cooling it to room temperature,
and we filter it. The filtrate and the water from
wash dry and the residue is dissolved in 100ml
of benzene. The benzene solution is filtered,
evaporates the filtrate to dryness, and heats the residue to
1000 C at a pressure of 2 mm of mercury
for a short time.
The residue is washed with
decantation with 200 ml of petroleum ether (zone
40-600 C) and recrystallized from
the alcohol. We thus obtain the benzoate of 4- (di
chloracet-N-methylamido) -phenyl in the form
of a crystalline solid with a melting point of 118

Example 2
Preparation of 4- (dichloroacet-N-methyl- acetate
amido) -phenyl (compound a with R '= CH5CO -
via the p-aminophenol (A) + compound M route

I) p-aminophenol (A) is converted into p-di chloroacetamidophenol (M) by one of the methods described in Example 1 (I).
II) p-dichloroacetamidophenol (M) is converted to 4-dichloroacetamidophenyl acetate (compound N with = Rl = CH3CO-) in the following way:
A solution is prepared by dissolving 22 g of 4-dichloroacetamidophenol in 35 ml of 5N sodium hydroxide and 100 g of ice water. 70 g of ice and 14 ml of acetic anhydride are added to the solution and the mixture is shaken vigorously for a short time. The solid product which separated is isolated by filtration, washed with water and recrystallized from alcohol. 4dichloroacetamidophenyl acetate is thus obtained in the form of a white crystalline solid with a melting point of 168-169oC.
(Found: C = 45.9; H = 3.5%. C, 0H9O3'NCl2 corresponds to C = 45.8; H = 3.4%).
III) The 4-dichloracetamidophenyl acetate is then converted into 4- (dichloracet-N-methylamido) -phenyl acetate (compound a with Rl = CH3CO-) in the following way:
A mixture of 7.86 g of 4-dichloroacetamidophenyl acetate, 8 g of anhydrous potassium carbonate, 4 g of dimethyl sulfate, and 40 ml of dry acetone is heated under reflux for 4.5 hours, after which it is cooled to room temperature and filtered.
The filtrate is evaporated to dryness and the residue is recrystallized from alcohol. 4-Dichloracet-N-methylamido) -phenyl acetate is thus obtained in the form of a crystalline solid with a melting point of 89.50 C.
