Verfahren zur Herstellung von Pteridinderivaten
Es sind bisher nur sehr wenige Pteridinderivate bekannt, die mindestens drei freie oder substituierte Aminogruppen im Kern enthalten. Sie werden im all gemeinen durch Halogenaustausch dargestellt (vgl. z. B. C. Schenker, Ph. D. Thesis, Cornell University, 1949).
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Pteridinderivaten der Formel

worin mindestens einer der Substituenten R-R4 ein Stickstoffatom, das Glied eines gegebenenfaLls durch ein weiteres Heteroatom unterbrochenen Rin- ges ist, mindestens zwei weitere der Reste Rt-R4 eine freie oder substituierte Aminogruppe oder ein Stickstoffatom, das Glied eines gegebenenfalls durch ein weiteres Heteroatom unterbrochenen Ringes ist, und der vierte der Reste R-R4 eine freie oder substituierte Hydroxyl-oder Thiogruppe bedeutet.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Pteridinderivate ist dadurch gekennzeich- net, dass man ein balogenhaltiges Pteridin der Formel

worin einer der Reste Z1-Z4 ein Halogenatom, 1-3 der Reste Z1-Z4 ein Stickstoffatom, das Glied eines gegebenenfallls durch ein weiteres Heteroatom unterbrochenen Ringes ist und 0-2 der Reste Z1-Z4 eine freie oder substituierte Aminogruppe bedeuten, mit einer Verbindung der Formel
H-R, worin R eine freie oder substituierte Hydroxyl-oder Mercaptogruppe bedeutet, in Anwesenheit eines sÏurebindenden Mittels bei Temperaturen von-20 bis +2500C, gegebenenfalls unter Druck, umsetzt.
Ein Beispiel für bei dem erfindungsgemässen Verfahren als Ausgangsstoffe verwendbare halogenhaltige Pteridine ist 4-Chlor-2, 6, 7-tripiperidino-pteridin.
Zu den f r die Umsetzung mit den halogenhaltigen Pteridinen geeigneten Verbindungen der Formel H-R gehören beispielsweise folgende : gegebenenfalls substituierte Alkohole, Phenole oder gegebenenfalls substituierte Mercaptane und Thiophenole.
Als säurebindende Mittel kommen z. B. Alkali- hydroxyde, Alkalicarbonate, Alkalialkoholate oder tertiäre Amine, gegebenenfalls ein Uberschuss der Verbindung H-R, wenn diese sich als säurebindendes Mittel eignet, in Frage.
Die Umsetzung kann in Ab-oder Anwesenheit von Lösungs-bzw. Verdünnungsmitteln, durchgefuhrt werden. Vorzugsweise wird sie jedoch in an der Umsetzung g nicht teilnehmenden L¯sungsmitteln, z. B.
Aceton, Dioxan, Benzol und Dimethylformamid, durchgeführt. Wenn sich der Siedepunkt des verwendeten Lösungsmittels den durch einfachen Vorversuch zu ermittelnden, für den Ablauf der Umsetzung günstigsten Temperaturbereich nicht erreichen lässt, kann unter Druck gearbeitet werden. Die Verbindung der Formel H-R kann, falls sie unter den Reaktions- bedingungen flüssig ist, im Vberschuf3 als Lösungsoder Verdünnungsmittel Verwendung finden.
Die nach dem erfindungsgemässen Verfahren er hältlichen neuen Pteridinderivate sowie ihre in Wasser meist leicht löslichen Salze zeigen sehr wertvolle neue, bei Ptcridinderivaten völlig unerwartete, phar makologische Eigenschaften.
Beispiel 1
4-Athoxy-2, 6, 7-trimorpholino-pteridin
8, 4 g (0, 02 Mol) 4-Chlor-2, 6, 7-trimorpholino pteridin wurden mit einer Lösung von 0, 5 g (etwa 0, 02 Mol) Natrium in 300 cm3 absolutem Alkohol 2 Stunden lang am Rückfluss erhitzt. Nach Absaugen einer geringen Menge ungelöster Substanz begann sich beim Erkalten das Reaktionsprodukt in nadelförmigen Kristallen abzuscheiden. Zur Vervollständigung der Fällung wurde mit der dreifachen Menge Wasser versetzt. Nach Absaugen, Waschen und Trocknen betrug die Ausbeute 7, 3 g (= 85"/o d. Th.). Zur Analyse wurde die Verbindung zweimal aus Methanol umkri stallisiert. Fast farblose, mikrokristalline Nädelchen, F. =19 8-200 C.

Beispiel 2
4-(¯-¯thoxy-Ïthoxy)-2, 6, 7-trimorpholino-pteridin
8, 4 g (0, 02 Mol) 4-Chlor-2, 6, 7-trimorpholino- pteridin wurden in eine Lösung von 0, 5 g (0, 022 Mol) Natrium in 40 cm3 Athylglykol und 60 cm3 Dioxan eingetragen und dann 30 Minuten lang auf ungefähr
100 C erhitzt. Nach Filtrieren der hei3en Reaktions- lösung und nach Zugabe von 200 cm3 Wasser schied sich das Umsetzungsprodukt in orangefarbenen Nadeln ab. Ausbeute 7, 6 g (= 80 11/o d. Th.).
Zur Reinigung wurde die Substanz einmal aus Methanol umkristallisiert, einmal aus verdünnter Salzsäure umgefällt und einmal aus Benzol umkristallisiert ; sehr feine watteartig verfilzte, elfenbeinfarbene Nädelchen ; nach Trocknen bei 110 C : F. =153-154 C.

Beispiel 3
4-Phenoxy-2, 6, 7-trimorpholino-pteridin
In eine auf etwa 60 C erwärmte Schmelze aus
10 g Phenol und I g Natriumhydroxyd wurden 4, 2 g (0, 01 Mol) 4-Chlor-2, 6, 7-trimorpholino-pteridin eingetragen und anschliessend 10 Minuten lang auf 180 bis 200 C erwärmt. Beim Aufnehmen des Reaktions- gemisches in etwa 200 cm3 verdünntem Ammoniak schied sich das 4-Phenoxy-2,6,7-trimorpholino-pteridin als gelblicher Niederschlag ab.
Es wurde nach einigem Stehenlassen abgesaugt, gewaschen und getrocknet. Nach Umkristallisieren aus Methanol erhielt man schwach gelbliche, sehr kleine Nädelchen, F. =239-240"C. Ausbeute (reine Substanz) 1, 2 g (=25 /o d. Th.).

Beispiel 4 4-Athylthio-2, 6, 7-trimorpholino-pteridin
Zu einer Lösung von 6, 0 g (etwa 0, 014 Mol) 4-Chlor-2, 6, 7-trimorpholino-pteridin in 200 cm3 sie- dendem Dimethylformamid wurden langsam 5 cm3 Athylmercaptan (gelöst in 20 cm3 4n Natronlauge) zutropfen gelassen und anschliessend das Ganze un gefähr 30 Minuten lang am Rückfluss erhitzt.
Nach Abfiltrieren einer geringen Menge abgeschiedener Substanz wurde die erhaltene Lösung mit Wasser versetzt und dann im Vakuum bis auf etwa 50 cm3 ein gedampft. Das abgeschiedene Reaktionsprodukt wurde abgesaugt, gewaschen und getrocknet. Ausbeute 4, 0 g (= 64 ID/o d. Th.). Zur Analyse wurde die e Verbindung zweimal aus Methanol umkristallisiert.
Kleine, leuchtend gelbe Prismen (kristallmethanolhaltig !) ; nach vierstündigem Trocknen bei 110 C : F. = 193-195 C.

Beispiel 5
4-Phenylthio-2, 6, 7-trimorpholino-pteridin
4, 2 g (0, 01 Mol) 4-Chlor-2, 6, 7-trimorpholino- pteridin wurden mit 5 cm3 Thiophen : ol und 2 cm3 Pyridin in 50 cm3 Dimethylformamid 1 1/2 Stunden lang unter Rückfluss erhitzt. Das nach Abdampfen des Lösungsmittels (im Vakuum) zurückbleibende rohe, ölige Reaktionsprodukt konnte nach Digerieren mit verdünntem Ammoniak abgesaugt werden. Nach einmaligem Umkristallisieren aus Methanol betrug die Ausbeute 3, 5 g (=71 /o d. Th.). Zur Analyse wurde nochmals aus Methanol umkristallisiert. KrÏftig gelbes, mikrokristallines Pulver, F. =186-187 C.

Process for the preparation of pteridine derivatives
So far, very few pteridine derivatives are known which contain at least three free or substituted amino groups in the core. They are generally represented by halogen exchange (see, for example, C. Schenker, Ph. D. Thesis, Cornell University, 1949).
The present invention relates to a process for the preparation of pteridine derivatives of the formula

8.4 g (0.02 mol) of 4-chloro-2,6,7-trimorpholino pteridine were refluxed for 2 hours with a solution of 0.5 g (about 0.02 mol) of sodium in 300 cm3 of absolute alcohol . 5 cm3 of ethyl mercaptan (dissolved in 20 cm3 of 4N sodium hydroxide solution) were slowly added dropwise to a solution of 6.0 g (about 0.014 mol) of 4-chloro-2,6,7-trimorpholino-pteridine in 200 cm3 of boiling dimethylformamide and then refluxed the whole thing for about 30 minutes.
