Verfahren zur Herstellung von Pteridinderivaten
Es sind bisher nur sehr wenige Pteridinderivate bekannt, die mindestens drei freie oder substituierte Aminogruppen im Kern enthalten. Sie werden im allgemeinen durch Halogenaustausch dargestellt (vgl. z. B. C. Schenker, Ph. D. Thesis, Cornell University, 1949).
Die vorliegende Erfindung betrifft nun ein Verfahren zur Herstellung von Pteridinderivaten der Formel

Als Beispiele für bei dem erfindungsgemässen Verfahren als Ausgangsstoffe verwendbare halogenhaltige Pteridine seien unter anderem folgende genannt :

2, 4, 6, 7-Tetrabrom-pteridin,
2, 4, 7-Trichlor-6-phenyl-pteridin,
2, 4, 7-Trichlor-6-carboxymethyl-pteridin,
2-Benzyl-4, 6, 7-trichlor-pteridin,
2-Methylamino-4, 6, 7-trichlor-pteridin,
2, 4-Dichlor-6, 7-bis-(diäthylamino)-pteridin,
2-Athylthio-4-chlor-6, 7-dimorpholino-pteridin,
2-Phenoxy-4-chlor-6, 7-dimorpholino-pteridin.
Als säurebindende Mittel kommen in Frage z. B.
Alkalihydroxyde, Alkalicarbonate, Alkalialkoholate oder tertiäre Amine, gegebenenfalls ein Überschuss der Verbindung H-R, wenn diese sich als säurebindendes Mittel eignet.
Die Umsetzung kann in Ab-oder Anwesenheit von Lösungs-bzw. Verdünnungsmitteln durchgeführt werden. Vorzugsweise wird sie jedoch in an der Umsetzung nicht teilnehmenden Lösungsmitteln, z. B.
Aceton, Dioxan, Benzol und Dimethylformamid, durchgeführt. Wenn der Siedepunkt des verwendeten Lösungsmittels den durch einfachen Vorversuch zu ermittelnden, für den Ablauf der Umsetzung günstig- sten Temperaturbereich nicht erreichen lässt, kann unter Druck gearbeitet werden. Auch Wasser und Alkohole können, vor allem in Abwesenheit von Alkalien und bei niederen Reaktionstemperaturen, als Lösungs-und Verdünnungsmittel dienen, da sie unter den Reaktionsbedingungen mit den halogenhaltigen Pteridinen praktisch nicht reagieren. Ferner kann die Verbindung der Formel H-R, falls sie unter den Reaktionsbedingungen flüssig ist, im Uberschuss als Lösungs-oder Verdünnungsmittel Verwendung finden.
Zur Durchführung des erfindungsgemässen Verfahrens kann ein Reaktionsbeschleuniger, z. B.
Kupfer und Kupfersalze sowie Alkalijodide, verwendet werden.
Wenn das als Ausgangsstoff verwendete halogenhaltige Pteridin mindestens 2 Halogenatome als Substituenten enthält, kann der Austausch dieser zwei oder mehr Halogenatome gegen den Rest R der Verbindung H-R auch stufenweise ausgeführt werden. So werden beispielsweise bei der Umsetzung des 2, 4, 6, 7-Tetrachlor-pteridins mit Aminen bei niedrigen Temperaturen, vorteilhaft in einem Lö sungsmittel unter Kühlung, nur die beiden Halogenatome in 6-und 7-Stellung gegen Aminogruppen ausgetauscht. Wird bei der gleichen Umsetzung auf mittlere Temperaturen (um 100 ) erwärmt, dann lässt sich ausser den beiden in 6-und 7-Stellung stehenden Halogenatomen noch ein drittes, sehr wahrscheinlich in 2-Stellung, durch eine Aminogruppe ersetzen.
Bei Erhitzen von 2, 4, 6, 7-Tetrachlor-pteridin mit Aminen auf höhere Temperaturen, oberhalb etwa 170 , gegebenenfalls unter Druck und in Anwesenheit eines Reaktionsbeschleunigers, werden schliesslich alle vorhandenen Halogenatome gegen Aminogruppen ausgetauscht.
Die Halogenatome der als Ausgangsstoffe verwendeten halogenhaltigen Pteridine, auch die verhältnismässig schwer austauschbaren in 2-und vor allem in 4-Stellung, lassen sich, wie sich überraschenderweise gezeigt hat, besonders leicht durch den Rest eines cyclischen, gegebenenfalls ein weiteres Heteroatom enthaltenden Amins ersetzen.
Die nach dem erfindungsgemässen Verfahren er hältlichen neuen Pteridinderivate sowie ihre in Wasser meist leicht löslichen Salze, zeigen sehr wertvolle neue, bei Pteridinderivaten völlig uner wartete, pharmakologische Eigenschaften.
Beispielsweise zeigen das 2- B-Oxyäthylamino)-
4-chlor-6, 7-dipiperidino-pteridin und besonders das 4-Athoxy-2, 6, 7-trimorpholino-pteridin neben einer geringen Blutdruckwirkung eine sehr gute corona erweiternde Wirkung.
Das 2, 4-Dimorpholino-6, 7-bis- (dimethylamino)- pteridin ist nicht nur sehr gut coronaerweiternd, sondern auch ausgezeichnet sedativ und antipyretisch wirksam. Das 2, 4-Dimorpholino-6-7-dipiperidino- pteridin schliesslich zeigt sowohl sedative wie vor allem auch eine sehr gute analgetische Wirkung.
Beispiel 1 2-Morpholino-4-chlor-6, 7-dipiperidino-pteridin
7, 4 g (0, 02 Mol) 2, 4-Dichlor-6, 7-dipiperidino- pteridin (erhalten aus 2, 4, 6, 7-Tetrachlor-pteridin und Piperidin in Dioxan unter Kühlung, F. = 186-187 ) wurden mit 5 cm3 Morpholin in 120 cm3 Dioxan etwa 1 Stunde lang am Rückfluss erhitzt, wobei sich Morpholin-hydrochlorid abschied. Bei Zugabe von etwa 250 cm3 Wasser zu dem erhaltenen Reaktionsgemisch schied sich das gebildete Pteridin-Derivat unter Auflösung des Morpholin-hydrochlorids als gelber, zunächst schmieriger Niederschlag ab.
Das nach kurzem Stehenlassen fest gewordene Reaktionsprodukt wurde abgesaugt, mit Wasser gewaschen und bei 110 getrocknet. Ausbeute 8, 1 g (-97"/ der Theorie). Zur Analyse wurde die Verbindung einmal aus sehr verdünnter Salzsäure umgefällt und einmal aus Athanol umkristallisiert. Sehr feine, hellgelbe Nädelchen, F. = 158-159 .

Beispiel 2
2, 4-Dimorpholino-6, 7-bis- (dimethylamino)-pteridin
5, 7 g (0, 02 Mol) 2, 4-Dichlor-6, 7-bis- (dimethyl- amino)-pteridin (erhalten aus 2, 4, 6, 7-Tetrachlorpteridin und absoluter alkoholischer Dimethylaminlösung unter Kühlung, F. = 247-248 ) wurden mit 17, 2 g (0, 2 Mol) Morpholin im Einschlussrohr etwa 2 Stunden lang auf 200 erhitzt. Das Reaktionsprodukt wurde mit Wasser aus dem Rohr gespült, abgesaugt, gewaschen und getrocknet. Die Ausbeute betrug 7, 7 g (= 99 /o der Theorie).
Zur Reinigung wurde die Verbindung zweimal aus sehr stark ver dünnter Salzsäure umgefällt und einmal aus Methanol umkristallisiert. Gelbe, mikrokristalline, meist flache Prismen, F. = 191-192 .

Beispiel 3
4-Chlor-2, 6, 7-tripiperidino-pteridin
Eine Lösung von 10, 8 g (0, 04 Mol) 2, 4, 6, 7 Tetrachlorpteridin in 150 cm3 Dioxan wurde nach Zugabe von 25, 5 g (0, 3 Mol) Piperidin etwa 1 Stunde lang am Rückfluss erhitzt. Piperidinhydrochlorid schied sich als gelber, kristalliner Niederschlag ab.
Bei Zugabe von etwa 500 cm3 Wasser zum Reaktionsgemisch fiel das Umsetzungsprodukt unter Auflösung des Hydrochlorids als brauner, schmieriger Niederschlag aus, der nach mehrstündigem Stehen erstarrte. Nach Absaugen, Waschen und Trocknen erhielt man 16, 0 g (= 96"/o der Theorie) des Produktes. Zur Reinigung wurde die Verbindung einmal aus 0, ln Salzsäure umgefällt und einmal aus Methanol umkristallisiert. Kleine, gelblichbraune Prismen, F. = 147-148 .
Beispiel 4
2, 4, 6, 7-Tetramorpholino-pteridin
4, 5 g (0, 01 Mol) 2, 4, 6, 7-Tetrabrom-pteridin wurden mit 25 cm3 Morpholin im Bombenrohr etwa 2 Stunden lang auf 200-220 erhitzt. Beim Aufnehmen des Rohrinhaltes in verdünnte Salzsäure wurde eine klare Lösung erhalten, aus der auch nach Versetzen mit Ammoniak das Reaktionsprodukt nicht ausfiel.
Es schied sich erst nach teilweisem Eindampfen ab.
Zur Entfernung anorganischer Salze wurde das Rohprodukt mit warmem, trockenem Benzol digeriert, das Ungelöste abgesaugt und die Lösung im Vakuum zur Trockne eingedampft. Ausbeute 4, 0 g (= 85 /o der Theorie). Zur Analyse wurde das Tetramorpholino-pteridin zweimal aus etwa 0, 1n Salzsäure umgefällt. Kräftig gelbes, mikrokristallines Pulver (Prismen), F. = 187-188 .

Beispiel 5 2- (-0xyäthylamino)-4-morpholino-6, 7 dipiperidino-pteridin
7, 8 g (0, 2 Mol) 2-(B-Oxyäthylamino)-4-chlor- 6, 7-dipiperidino-pteridin wurden mit 15 cm3 Morpholin und 1 cm3 kaltgesättigter wässriger Kupfersulfatlösung 2 Stunden lang im Rohr auf 200 erhitzt. Mit 150 cm3 Wasser wurde das Reaktionsprodukt (braungelber, amorpher Niederschlag) herausgespült, abgesaugt, gewaschen und anschliessend sofort einmal aus n/10 Salzsäure umgefäüt. Ausbeute (nach Trocknen im Vakuum bei Zimmertemperatur) 6, 0 g (= 68"/o der Theorie).
Zur Analyse wurde das Tetraamino-pteridin einmal aus Dioxan-Wasser (1 : 1) und zweimal aus Dioxan umkristallisiert. Drüsen- förmig miteinander verwachsene, mikroskopisch kleine, gelbe Kristalle. F. = 168-1700 (nach länge- rem Trocknen bei 110 ).

Beispiel 6
2-Methylamino-4-chlor-6, 7-dipiperidino-pteridin
In eine Lösung von 5, 6 g (0, 02 Mol) 2-Methylamino-4, 6, 7-trichlor-pteridin in 150 cm3 Dioxan wurden unter Kühlen und Umrühren 10 cm3 Piperidin langsam eingetropft, wobei Piperidin-hydrochlorid ausfiel. Beim Eingiessen des Reaktionsgemisches in 500 cm3 Wasser fiel unter gleichzeitiger Auflösung des Hydrochlorids das 2-Methylamino4-chlor-6, 7-dipiperidino-pteridin als brauner, amorpher Niederschlag aus. Es wurde abgesaugt, mit Wasser gewaschen und bei 110 getrocknet. Ausbeute 2, 0 g (= 276/o der Theorie). Zur Analyse wurde die Substanz einmal aus kalter 0, ln Salzsäure umgefällt und zweimal aus Methanol umkristallisiert.
Gelbes mikrokristallines Pulver, F. = 240-2420.

2-Methylamino-4, 6, 7-trimorpholino-pteridin
5, 6 g (0, 02 Mol) 2-Methylamino-4, 6, 7-trichlorpteridin wurden mit 20 cm3 Morpholin im Druckrohr 2 Stunden lang auf 200 erhitzt. Der sich bildende braune, kristalline Niederschlag wurde mit etwa 300 cm3 Wasser herausgespült, abgesaugt, gewaschen und getrocknet. Ausbeute 6, 5 g (= 76 /o der Theorie). Zur Reinigung wurde einmal aus kalter 0, 1n Salzsäure umgefällt und zweimal aus Methanol umkristallisiert. Leuchtend gelbe,, mikrokfistalline tetragonale Prismen, F. = 254-2560.
Beispiel 8
2, 4, 7-Trimorpholino-6-phenyl-pteridin
3, 1 g (0, 01 Mol) 2, 4, 7-Trichlor-6-phenyl-pteridin (erhalten aus 2, 4, 7-Trioxy-6-phenyl-pteridin mit Phosphorpentachlorid und Phosphoroxychlorid am Rückfluss, F. = 157-158 ) wurden mit 20 cm3 Morpholin und 0, 5 g Natriumjodid im Bombenrohr 2 Stunden lang auf 200 erhitzt. Das Reaktionsgemisch wurde mit etwa 250 cm3 Wasser herausgespült und der gelbe Niederschlag des Triaminopteridins abgesaugt, gewaschen und getrocknet. Ausbeute 4, 5 g (97"/o. der Theorie). Zur Reinigung wurde die Substanz einmal aus kalter 0, 1n Salzsäure umgefällt und zweimal aus Methanol umkristallisiert. Kräftig gelbes, mikrokristallines Pulver (Prismen), F. = 201 bis 202 .

Beispiel 9
2-Phenyl-4, 6, 7-trimorpholino-pteridin
3, 1 g (0, 01 Mol) rohes 2-Phenyl-4, 6, 7-trichlor- pteridin (F. = 187-189 , erhalten durch Erhitzen des 2-Phenyl-4, 6, 7-trioxy-pteridins mit Phosphorpentachlorid und Phosphoroxychlorid unter Druck) wurden mit 60 cm3 Morpholin (Kpt28 ) 2 Stunden lang unter Rückfluss gekocht. Beim Eingiessen des noch warmen Reaktionsgemisches in etwa 300 cm3 Wasser schied sich das 2-Phenyl-4, 6, 7-trimorpho lino-pteridin als gelber Niederschlag ab. Nach ein maligem Umfallen aus 2n Schwefelsäure betrug die Ausbeute 4, 4 g (= 9511/o der Theorie).
Zur weiteren Reinigung wurde einmal aus Methanol umkristallisiert. Gelbes, mikrokristallines Pulver, F. = 209 bis 210 .

Beispiel 10
2-Athylthio-4-pyrrolidino-6, 7-dimorpholino- pteridin
4, 0 g (0, 01 Mol) 2-ÄthyIthio-4-cMor-6, 7-di- morpholino-pteridin wurden mit 20 cm3 Pyrrolidin im Bombenrohr 2 Stunden lang auf etwa 200 erhitzt. Das Reaktionsprodukt wurde mit Wasser aus dem Rohr gespült und sofort einem aus verdünnter Salzsäure durch Zugabe konzentrierten Ammoniaks umgefällt. Das rohe 2-Athylthio-4-pyrrolidino-6, 7dimorpholino-pteridin schied sich dabei als zähe, jedoch nach einigem Stehenlassen erstarrende Masse ab. Es wurde zur Reinigung noch dreimal aus etwa 0, 1n Salzsäure umgefällt.
Ausbeute 1, 7 g (= 39 lo der Theorie). Nach Umkristallisieren aus Methanol-Wasser (1 : 1) erhielt man leuchtend gelbe, unregelmässige Kristalle, F. = 118-120 .
Beispiel 11
2 (4)-Oxy-4 (2)-6, 7-trimorpholino-pteridin
1, 5 g (0, 0045 Mol) 2 (4)-Oxy-4 (2)-chlor-6, 7trimorpholino-pteridin wurden mit 15 cm3 Morpholin 2 Stunden lang im Bombenrohr auf etwa 200 erhitzt.
Aus der beim Ausspülen des Rohres mit Wasser erhaltenen alkalischen Lösung schied sich beim Neutralisieren mit 2n Schwefelsäure das Reaktionsprodukt als bräunlicher Niederschlag ab. Nach Absaugen, Waschen und Trocknen erhielt man 1, 0 g (= 59 fl/o der Theorie). Zur Analyse wurde einmal aus 2n Salzsäure umgefällt. Schmutzig gelbes Pulver, F. = 242-243 .

2, 4-Dimorpholino-7-dimethylamino-pteridin
2, 4 g (0, 01 Mol) 2, 4-Dichlor-7-dimethylamino- pteridin (F. des Rohproduktes = 172-175 , erhalten aus 2, 4, 7-Trichlorpteridin und absoluter alkoho- lischer Dimethylaminolösung in Dioxan unter Küh- lung) wurden mit 15 cm3 Morpholin im Druckrohr etwa 2 Stunden lang auf 2000 erhitzt. Beim Aufnehmen des braunen, geleeartigen Reaktionsgemisches in etwa 300 cm3 Wasser schied sich das rohe 2, 4-Dimorpholino-7-dimethylamino-pteridin als elfenbeinfarbener, amorpher Niederschlag ab.
Nach Absaugen, Waschen mit Wasser und Trocknen bei 110 betrug die Ausbeute 2, 4 g (= 70 /o der Theorie). Nach einmaligem Umfällen aus 0, ln Salzsäure und zweimaligem Umkristallisieren aus Methanol erhielt man elfenbeinfarbene, mikrokristalline, rautenförmige Blättchen, F. = 194-195 .

2, 7-Dimorpholino-4-pyrrolidino-6-carboxy methyl-pteridin
2, 0 g (0, 05 Mol) 2, 7-Dimorpholino-4-chlor-6carboxymethyl-pteridin (F. des Rohproduktes etwa 150 , erhalten aus 2, 4, 7-Trichlor-6-carboxymethyl- pteridin durch Kochen mit Morpholin in Dioxan am Rückfluss) wurden mit 15 cm3 Pyrrolidin im Bombenrohr 2 Stunden lang auf etwa 200 erhitzt. Aus der beim Herausspülen des Reaktionsgemisches mit etwa 200 cm3 Wasser erhaltenen dunkelbraunen Lösung fiel erst nach Neutralisieren mit Salzsäure und teilweisem Einengen das rohe Pteridinderivat als brauner, flockiger Niederschlag aus.
Nach Absaugen, Waschen und Trocknen im Vakuum bei Zimmertemperatur erhielt man 1, 2 g (= 56"/o der Theorie). Zur Reinigung wurde dreimal aus 0, 1n Salzsäure durch Neutralisieren mit 2n Ammoniak umgefällt. Hellolivfarbenes Pulver, F. = 115-117 (vorher Sintern).

Process for the preparation of pteridine derivatives
So far, very few pteridine derivatives are known which contain at least three free or substituted amino groups in the core. They are generally prepared by halogen exchange (see, e.g., C. Schenker, Ph. D. Thesis, Cornell University, 1949).
The present invention now relates to a process for the preparation of pteridine derivatives of the formula

Examples of halogen-containing pteridines which can be used as starting materials in the process according to the invention include the following:

As acid-binding agents come into question z. B.
Alkali hydroxides, alkali carbonates, alkali alcoholates or tertiary amines, optionally an excess of the compound H-R, if this is suitable as an acid-binding agent.
The implementation can be in the absence or presence of solution or. Diluents are carried out. However, it is preferably used in solvents which do not participate in the reaction, e.g. B.
Acetone, dioxane, benzene and dimethylformamide. If the boiling point of the solvent used does not allow the temperature range to be determined by a simple preliminary experiment to be reached and the most favorable temperature range for the course of the reaction to be reached, pressure can be used. Water and alcohols can also serve as solvents and diluents, especially in the absence of alkalis and at low reaction temperatures, since they practically do not react with the halogen-containing pteridines under the reaction conditions. Furthermore, the compound of the formula H-R, if it is liquid under the reaction conditions, can be used in excess as a solvent or diluent.
To carry out the process according to the invention, a reaction accelerator, e.g. B.
Copper and copper salts as well as alkali iodides can be used.
If the halogen-containing pteridine used as the starting material contains at least 2 halogen atoms as substituents, these two or more halogen atoms can also be exchanged for the radical R of the compound H-R in stages. For example, when converting 2,4,6,7-tetrachloropteridine with amines at low temperatures, advantageously in a solvent with cooling, only the two halogen atoms in the 6- and 7-positions are exchanged for amino groups. If the same reaction is heated to medium temperatures (around 100), in addition to the two halogen atoms in the 6- and 7-position, a third, very likely in the 2-position, can be replaced by an amino group.
When 2, 4, 6, 7-tetrachloropteridine is heated with amines to higher temperatures, above about 170, optionally under pressure and in the presence of a reaction accelerator, all halogen atoms present are finally exchanged for amino groups.
The halogen atoms of the halogen-containing pteridines used as starting materials, including those in the 2-position and especially in the 4-position, which are relatively difficult to replace, can, as has surprisingly been shown, be replaced particularly easily by the remainder of a cyclic amine, optionally containing a further heteroatom.
The new pteridine derivatives obtainable by the process according to the invention and their salts, which are usually readily soluble in water, show very valuable new pharmacological properties which are completely unexpected in the case of pteridine derivatives.
For example, show the 2- B-Oxyäthylamino) -
4-chloro-6, 7-dipiperidino-pteridine and especially 4-ethoxy-2, 6, 7-trimorpholino-pteridine have a very good corona-expanding effect in addition to a low blood pressure effect.
The 2,4-dimorpholino-6, 7-bis- (dimethylamino) - pteridine is not only very good at expanding the corona, but also has excellent sedative and antipyretic properties. Finally, the 2,4-dimorpholino-6-7-dipiperidino-pteridine shows both sedative and, above all, very good analgesic effects.
Example 1 2-Morpholino-4-chloro-6,7-dipiperidino-pteridine
7.4 g (0.02 mol) 2, 4-dichloro-6, 7-dipiperidino-pteridine (obtained from 2, 4, 6, 7-tetrachloro-pteridine and piperidine in dioxane with cooling, mp = 186-187 ) were refluxed with 5 cm3 of morpholine in 120 cm3 of dioxane for about 1 hour, during which morpholine hydrochloride separated out. When about 250 cm3 of water were added to the reaction mixture obtained, the pteridine derivative formed separated out as a yellow, initially greasy precipitate with dissolution of the morpholine hydrochloride.
The reaction product, which had solidified after standing for a short time, was filtered off with suction, washed with water and dried at 110. Yield 8.1 g (-97 "/ of theory). For analysis, the compound was reprecipitated once from very dilute hydrochloric acid and once recrystallized from ethanol. Very fine, pale yellow needles, F. = 158-159.

Example 2
2,4-Dimorpholino-6, 7-bis (dimethylamino) pteridine
5.7 g (0.02 mol) 2, 4-dichloro-6, 7-bis (dimethyl-amino) pteridine (obtained from 2, 4, 6, 7-tetrachloropteridine and absolute alcoholic dimethylamine solution with cooling, F. = 247-248) were heated with 17.2 g (0.2 mol) of morpholine in the containment tube to 200 for about 2 hours. The reaction product was rinsed from the tube with water, filtered off with suction, washed and dried. The yield was 7.7 g (= 99 / o of theory).
For purification, the compound was reprecipitated twice from very dilute hydrochloric acid and recrystallized once from methanol. Yellow, microcrystalline, mostly flat prisms, F. = 191-192.

Example 3
4-chloro-2, 6, 7-tripiperidino-pteridine
A solution of 10.8 g (0.04 mol) 2, 4, 6, 7 tetrachloropteridine in 150 cm 3 dioxane was refluxed for about 1 hour after addition of 25.5 g (0.3 mol) piperidine. Piperidine hydrochloride separated out as a yellow, crystalline precipitate.
When about 500 cm3 of water were added to the reaction mixture, the reaction product precipitated out as a brown, greasy precipitate with dissolution of the hydrochloride, which solidified after standing for several hours. After suctioning off, washing and drying, 16.0 g (= 96 "/ o of theory) of the product were obtained. For purification, the compound was reprecipitated once from 0.1N hydrochloric acid and once recrystallized from methanol. Small, yellowish-brown prisms, F. = 147-148.
Example 4
2, 4, 6, 7-tetramorpholino-pteridine
4.5 g (0.01 mol) 2, 4, 6, 7-tetrabromopteridine were heated to 200-220 for about 2 hours with 25 cm3 of morpholine in a sealed tube. When the tube contents were taken up in dilute hydrochloric acid, a clear solution was obtained, from which the reaction product did not precipitate even after addition of ammonia.
It separated out only after partial evaporation.
To remove inorganic salts, the crude product was digested with warm, dry benzene, the undissolved material was filtered off with suction and the solution was evaporated to dryness in a vacuum. Yield 4.0 g (= 85 / o of theory). For the analysis, the tetramorpholino-pteridine was reprecipitated twice from about 0.1N hydrochloric acid. Strong yellow, microcrystalline powder (prisms), F. = 187-188.

Example 5 2- (-0xyäthylamino) -4-morpholino-6,7 dipiperidino-pteridine
7.8 g (0.2 mol) of 2- (B-oxyethylamino) -4-chloro-6, 7-dipiperidino-pteridine were heated to 200 in a tube for 2 hours with 15 cm3 of morpholine and 1 cm3 of cold-saturated aqueous copper sulfate solution. The reaction product (brown-yellow, amorphous precipitate) was rinsed out with 150 cm3 of water, filtered off with suction, washed and then immediately converted once from n / 10 hydrochloric acid. Yield (after drying in vacuo at room temperature) 6.0 g (= 68 "/ o of theory).
For analysis, the tetraamino-pteridine was recrystallized once from dioxane-water (1: 1) and twice from dioxane. Microscopic, yellow crystals that have grown together like glands. F. = 168-1700 (after longer drying at 110).

Example 6
2-methylamino-4-chloro-6,7-dipiperidino-pteridine
10 cm3 of piperidine were slowly added dropwise to a solution of 5.6 g (0.02 mol) of 2-methylamino-4, 6, 7-trichloropteridine in 150 cm3 of dioxane, with cooling and stirring, piperidine hydrochloride precipitating. When the reaction mixture was poured into 500 cm3 of water, the 2-methylamino-4-chloro-6, 7-dipiperidino-pteridine precipitated as a brown, amorphous precipitate with simultaneous dissolution of the hydrochloride. It was filtered off with suction, washed with water and dried at 110. Yield 2.0 g (= 276 / o of theory). For analysis, the substance was reprecipitated once from cold 0.1N hydrochloric acid and recrystallized twice from methanol.
Yellow microcrystalline powder, m.p. = 240-2420.

Example 7
2-methylamino-4, 6, 7-trimorpholino-pteridine
5.6 g (0.02 mol) of 2-methylamino-4, 6, 7-trichloropteridine were heated with 20 cm 3 of morpholine in a pressure tube to 200 for 2 hours. The brown, crystalline precipitate that formed was rinsed out with about 300 cm3 of water, filtered off with suction, washed and dried. Yield 6.5 g (= 76 / o of theory). For purification, it was reprecipitated once from cold 0.1N hydrochloric acid and recrystallized twice from methanol. Bright yellow, microcrystalline tetragonal prisms, F. = 254-2560.
Example 8
2, 4, 7-trimorpholino-6-phenyl-pteridine
3.1 g (0.01 mol) 2, 4, 7-trichloro-6-phenyl-pteridine (obtained from 2, 4, 7-trioxy-6-phenyl-pteridine with phosphorus pentachloride and phosphorus oxychloride at reflux, m.p. = 157 -158) were heated to 200 in a sealed tube for 2 hours with 20 cm3 of morpholine and 0.5 g of sodium iodide. The reaction mixture was rinsed out with about 250 cm3 of water and the yellow precipitate of the triaminopteridine was filtered off with suction, washed and dried. Yield 4.5 g (97% of theory). For purification, the substance was reprecipitated once from cold 0.1N hydrochloric acid and recrystallized twice from methanol. Strong yellow, microcrystalline powder (prisms), mp = 201 to 202.

Example 9
2-phenyl-4, 6, 7-trimorpholino-pteridine
3.1 g (0.01 mol) of crude 2-phenyl-4, 6, 7-trichloropteridine (F. = 187-189, obtained by heating the 2-phenyl-4, 6, 7-trioxy-pteridine with Phosphorus pentachloride and phosphorus oxychloride under pressure) were refluxed with 60 cm3 of morpholine (Kpt28) for 2 hours. When the still warm reaction mixture was poured into about 300 cm3 of water, the 2-phenyl-4, 6, 7-trimorpholino-pteridine separated out as a yellow precipitate. After reprecipitating once from 2N sulfuric acid, the yield was 4.4 g (= 9511 / o of theory).
For further purification, it was recrystallized once from methanol. Yellow, microcrystalline powder, F. = 209 to 210.

Example 10
2-Ethylthio-4-pyrrolidino-6, 7-dimorpholino-pteridine
4.0 g (0.01 mol) of 2-EthyIthio-4-cMor-6, 7-dimorpholino-pteridine were heated with 20 cm3 of pyrrolidine in a sealed tube to about 200 for 2 hours. The reaction product was rinsed from the tube with water and immediately reprecipitated from dilute hydrochloric acid by adding concentrated ammonia. The crude 2-ethylthio-4-pyrrolidino-6, 7-dimorpholino-pteridine separated out as a tough, but solidified mass after standing for a while. For purification, it was reprecipitated three times from about 0.1N hydrochloric acid.
Yield 1.7 g (= 39 lo of theory). After recrystallization from methanol-water (1: 1), bright yellow, irregular crystals were obtained, mp = 118-120.
Example 11
2 (4) -Oxy-4 (2) -6,7-trimorpholino-pteridine
1.5 g (0.0045 mol) of 2 (4) -oxy-4 (2) -chloro-6, 7-trimorpholino-pteridine were heated with 15 cm3 of morpholine to about 200 in a sealed tube for 2 hours.
From the alkaline solution obtained when the tube was rinsed with water, the reaction product separated out as a brownish precipitate on neutralization with 2N sulfuric acid. After filtering off with suction, washing and drying, 1.0 g (= 59 fl / o of theory) was obtained. For analysis, it was reprecipitated once from 2N hydrochloric acid. Dirty yellow powder, F. = 242-243.

Example 12
2,4-Dimorpholino-7-dimethylamino-pteridine
2.4 g (0.01 mol) 2,4-dichloro-7-dimethylaminopteridine (F. of the crude product = 172-175, obtained from 2,4,7-trichloropteridine and absolute alcoholic dimethylamino solution in dioxane under cooling - lung) were heated with 15 cm3 of morpholine in a pressure tube to 2000 for about 2 hours. When the brown, jelly-like reaction mixture was taken up in about 300 cm3 of water, the crude 2,4-dimorpholino-7-dimethylamino-pteridine separated out as an ivory-colored, amorphous precipitate.
After filtering off with suction, washing with water and drying at 110, the yield was 2.4 g (= 70 / o of theory). After reprecipitation once from 0.1N hydrochloric acid and twice recrystallization from methanol, ivory-colored, microcrystalline, diamond-shaped flakes were obtained, m.p. 194-195.

Example 13
2,7-Dimorpholino-4-pyrrolidino-6-carboxy methyl-pteridine
2.0 g (0.05 mol) 2, 7-dimorpholino-4-chloro-6carboxymethyl-pteridine (F. of the crude product about 150, obtained from 2, 4, 7-trichloro-6-carboxymethyl-pteridine by boiling with morpholine in dioxane at reflux) were heated to about 200 for 2 hours with 15 cm3 of pyrrolidine in a sealed tube. The crude pteridine derivative precipitated as a brown, flaky precipitate from the dark brown solution obtained when the reaction mixture was rinsed out with about 200 cm3 of water after neutralization with hydrochloric acid and partial concentration.
After suctioning off, washing and drying in vacuo at room temperature, 1.2 g (= 56 "/ o of theory) were obtained. For cleaning, it was reprecipitated three times from 0.1N hydrochloric acid by neutralizing with 2N ammonia. Light olive-colored powder, m.p. = 115- 117 (previously sintered).
