Verfahren zur Herstellung von neuen Derivaten des Pyrimido- [5, 4-d]-pyrimidins
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Derivaten des Pyrimido [5, 4-d]-pyrimidins der Formel :

worin zwei bis vier der Substituenten R1R4, die untereinander gleich oder verschieden sind, Aminogruppen, durch Alkyl-, Aryl-oder Aralkylreste, die ihrerseits substituiert oder unsubstituiert sein kön- nen, mono-od'er disubstituierte Aminogruppen, deren Stickstoffatom auch Glied eines substituierten oder unsubstituierten heterocyclischen Ringes sein kann, und der seinerseits durch ein weiteres Heteroatom unterbrochen sein kann, Acylaminogruppen, oder substituierte oder unsubstituierte Hydrazinooder Guanidinogruppen darstellen,
während die restlichen null bis zwei der Reste R1R4, die untereinander ebenfalls gleich oder verschieden sind, Wasserstoffatome, Halogenatome, Alkyl-, Aryl-oder Aralkylreste oder freie oder substituierte Hydroxyl oder Thiogruppen bedeuten.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Pyrimido- [5, 4-d]-pyrimidine der Formel I ist dadurch gekennzeichnet, dass in einem Pyrimido [5, 4-d]-pyrimidin der Formel II :

worin einer bis 3 der Reste Z !--Z4 gleiche oder verschiedene Halogenatome bedeuten, einer bis 3 der Reste eine freie oder substituierte Hydroxy- gruppe, eine freie oder substituierte Mercaptogruppe, eine freie Aminogruppe, eine durch Alkyl, Aryl- oder Aralkylreste, die ihrerseits substituiert oder unsubstituiert sein können, mono-oder disubstituierte Aminogruppe, deren Stickstoffatom auch Glied eines substituierten oder unsubstituierten Ringes sein kann, der seinerseits durch ein weiteres Heteroatom unterbrochen sein kann,
eine Acylaminogruppe oder eine substituierte oder unsubstituierte Hydrazino-oder Guanidinogruppe bedeuten und 0 bis 2 der Reste Z1Z4 den eben definierten Resten R1R4 entsprechen, mindestens 1 Halogenatom und mindestens einer der Reste Z1Z4, der in diesem Fall weder Wasserstoff noch Halogen, Alky, Aralkyl oder Aryl ist, durch Umsetzung mit einer Verbindung der Formel HR, in der R eine e freie Aminogruppe, eine durch Alkyl-, Aryl-oder Aralkylreste, die ihrerseits substituiert oder unsubstituiert sein können, mono-oder disubstituierte Aminogruppe, deren Stickstoffatom auch Glied eines substituierten oder unsubstituierten Ringes sein kann, der seinerseits durch ein weiteres Heteroatom unterbrochen sein kann,
eine Acylaminogruppe oder eine substituierte oder unsubstituierte Hydrazino-oder Guanidinogruppe bedeutet, durch den Rest R ersetzt wird, und dass die Umsetzung in Gegenwart säurebindender Mittel und bei Temperaturen zwischen-20 und 250 C erfolgt.
Im weiteren Verlauf der Beschreibung werden die Pyrimido- [5, 4-d]-pyrimidine der Einfachheit halber als Pyrimido-pyrimidine bezeichnet.
Bei dem erfindungsgemässen Verfahren werden in den Pyrimido-pyrimidinen der Formel II nicht nur Halogen, sondern auch weitere Substituenten, wie z. B. Hydroxyl-, substituierte Hydroxyl-, Aminooder substituierte Aminogruppen gegen den Rest R des Reaktionspartners HR ausgetauscht. So gelingt es beispielsweise
2, 6-Dichlor-4, 8-dioxy-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diamino-pyrimido-pyrimidin und
2, 6-Dichlor-4, 8-dipiperidino-pyrimido-pyrimidin durch Umsatz mit Anilin in 2, 4, 6, 8-Tetra-anilinopyrimido-pyrimidin umzuwandeln.
Als Ausgangssubstanzen der Formel II seien beispielsweise weiter genannt :
6-Chlor-4, 8-dioxy-pyrimido-pyrimidin,
6-Chlor-2-thio-4, 8-dimorpholino-pyrimido- pyrimidin,
2, 6-Dichlor-4, 8-diphenyloxy-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diphenylthio-pyrimido-pyrimidin,
4, 6, 8-Trichlor-2-thio-pyrimido-pyrimidin und
2, 6-Dichlor-4, 8-diäthyl-thio-pyrimido-pyrimidin.
Die als Ausgangsstoffe verwendeten Pyrimidopyrimidine der Formel II können beispielsweise durch Halogenierung von durch Ringschluss erhal tenen Oxypyrimido-pyrimidinen und durch anschlie ssenden teilweisen Austausch der Halogenatome hergestellt werden.
Als Beispiele für die Verbindungen HR, die für die Umsetzung mit den Pyrimido-pyrimidinen der Formel II geeignet sind, seien folgende genannt : Ammoniak, primaire oder sekundäre Amine, Guani- dine, Hydrazine, Aminoalkohole, Morpholine und Piperidine. Bei dem erfindungsgemässen Verfahren wird unter Zusatz eines säurebindenden Mittels, wie Alkalihydroxyd, Alkalicarbonat oder tertiären Aminen, oder gegebenenfalls mit einem Überschuss der Reaktionskomponente HR, sofern diese sich als säurebindendes Mittel eignet, gearbeitet.
Die Umsetzung kann in Ab-oder Anwesenheit von für die Reaktion inerten Lösungs-bzw. Ver dünnungsmitteln, z. B. Aceton, Dioxan, Benzol, Xylol oder Dimethylformamid, und gegebenenfalls unter Anwendung von Druck erfolgen. Wasser und Alkohole können vor allem in Abwesenheit von Alkalien und bei niederen Temperaturen ebenfalls als Lösungs-oder Verdünnungsmittel benützt werden, da sie unter diesen Bedingungen praktisch nicht mit den halogenhaltigen Pyrimidopyrimidinen reagieren. Auch die zweite Reaktionskomponente HR kann, falls sie unter den Reaktionsbedingungen flüssig ist, im Überschuss als Lösungs-oder Verdün- nungsmittel Verwendung finden.
Gewünschtenfalls können bei der Umsetzung Re aktionsbeschleuniger, z. B. Kupfer oder Kupfersalze, zugesetzt werden.
Die erfindungsgemäss erhaltenen Verbindungen können mit therapeutisch indifferenten Säuren in ihre Säureadditionssalze überführt werden. Die nach dem erfindungsgemässen Verfahren hergestellten Verbindungen besitzen ausgezeichnete therapeutische Eigen- schaften. Sie sind insbesondere cardiovaskulär wirksam. Während schon bei sehr niedrigen Dosen eine ausgezeichnete coronarerweiternde Wirkung, ohne wesentliche Beeinflussung des Blutdruckes, festzustellen ist, zeigt sich bei höherer Dosierung (ab etwa 0, 5-1 mg/kg) auch eine gute blutdrucksenkende Wirkung, die auf einer allgemeinen Gefässerweiterung und Verminderung des peripheren Widerstandes beruht.
Ausser den Coronarien werden besonders auch die Gehirngefässe erweitert, was sich in einer deutlichen und lang andauernden Mehrdurchblutung äussert. Dass die erwähnten Wirkungen nicht mit einer Schädigung des Herzens verbunden sind, wurde an 2, 6-Bis-(Diäthanolamino)-4, 8-dipiperidino-pyrimi do-pyrimidin bewiesen.
Beispiel 1
2, 4, 6, 8-Tetraanilino pyrimido-pyrimidin
Je 0, 01 Mol 2, 6-Dichlor-4, 8-diamino-pyrimidopyrimidin, 2, 6-Dichlor-4, 8-dioxy-pyrimido-pyrimidin oder 2, 6-Dichlor-4, 8-dipiperidino-pyrimido-pyrimidin wurden mit 45 g Anilin 25 Minuten lang am Rück- fluss gekocht. Beim Eingiessen der erhaltenen dunkelbraunen Lösung in 500 cm3 ln Salzsäure fiel das rohe Tetraanilino-pyrimido-pyrimidin als bräun- licher, amorpher Niederschlag aus. Nach dreimaligem Umkristallisieren aus Dioxan : kräftig kanariengelb gefärbte Nädelchen vom F. = 300-302 .
Beispiel 2
Verschiedene 2, 4, 6, 8-Tetraamino-pyrimido pyrimidine
Aus den entsprechenden 2, 6-Dichlor-4, 8-di- (Rsubstituierten)-pyrimido-pyrimidinen und den entsprechenden Aminen bei höherer Temperatur (gegebenenfalls unter Rückfluss oder Druck), wobei R eine gegebenenfalls substituierte Oxy-, Thio-oder Aminogruppe (einschliesslich quaternäre Aminogruppe) sein kann.
Folgende Verbindungen wurden beispielsweise als Ausgangsstoffe benutzt :
2, 6-Dichlor-4, 8-dithio-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diäthylthio-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diphenylthio-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-dioxy-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diäthoxy-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diphenoxy-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-diamino-pyrimido-pyrimidin,
2, 6-Dichlor-4, 8-bis- (diäthanolamino)-pyrimido- pyrimidin,
2, 6-Dichlor-4, 8-dimorpholino-pyrimido pyrimidin,
2, 6-Dichlor-4, 8-dipyridyl-pyrimido-pyrimidin chlorid. a) 2, 4, 6, 8-Tetra-{ > oxyäthylamino)-pyrimido- pyrimidin.
3, 9 g (0, 01 Mol) 2, 6-Dichlor-4, 8-diphenoxy-pyrimido-pyrimidin (F. 294-295 ) wurden mit 20 cm3 ss-Oxyäthylamin (Kp. 171 ) etwa 1 Stunde lang am Rückfluss erhitzt. Nach Eingiessen der erhaltenen Lösung in etwa 40 cm3 Wasser fiel nach mehrstündigem Stehen das Reaktionsprodukt als kristalli- ner Niederschlag aus. Nach Absaugen, Waschen und Trocknen betrug die Aubeute 2, 5 g (68 /o der Theorie). Aus Wasser : leicht bräunliche Prismen, F. 181 bis 183 .
Ähnlich der Verbindung a) wurden neben andern auch folgende Tetraamino-pyrimido-pyrimidine hergestellt b) 2, 4, 6, 8-Tetra- (methylamino)-pyrimido- pyrimidin, F. 227-228 . c) 2, 4, 6, 8-Tetra- (allylamino)-pyrimido- pyrimidin, F. 201-202 . d) 2, 4, 6, 8-Tetra- (benzylamino)-pyrimido- pyrimidin, F. 176-178 . e) 2, 4, 6, 8-Tetra-anilino-pyrimido-pyrimidin,
F. 300-302 .
2, 4, 6, 8-Tetra-(methyläthanolamino)-pyrimido- pyrimidin, F. 155-156 .
Beispiel 3 a) 2, 6-Bis-tdiäthanolamino) 4, 8-dipiperidino- pyrimido-pyrimidin.
12, 8 g (0, 04 Mol) 2, 6-Dichlor-4, 8-diäthylmer- capto-pyrimido-pyrimidin, F. = 190-192 , wurden mit 63 g (0, 6 Mol) Diäthanolamin etwa 1 Stunde auf 160 erhitzt. Beim Eingiessen der klaren, honigfarbenen Schmelze in 300 cm3 Wasser fiel das rohe Reaktionsprodukt als orangefarbener Niederschlag aus. Nach Absaugen, Waschen und Trocknen betrug die Ausbeute 9, 2 g (50, llo der Theorie). Zur Ent fernung geringer Mengen von 2, 4, 6-Tris- (diäthanol- amino)-8-äthylmercapto-pyrimido-pyrimidin wurde einmal aus 15 /oiger Essigsäure und einmal aus Methanol umkristallisiert : glänzende, leuchtend orangefarbene Prismen, F. = 182-183 .
4, 6 g (0, 01 Mol) 2, 6-Bis-diäthanolamino)-4, 8 diäthylmercapto-pyrimido-pyrimidin wurden mit 40 cm3 Piperidin im Bombenrohr etwa 3 Stunden lang auf 180 erhitzt. Bei Herausspülen des erhaltenen Gemisches mit etwa 300 cm3 Wasser schied sich das Reaktionsprodukt als schmieriger gelbbrauner Niederschlag ab. Es wurde sofort einmal aus verdünnter Salzsäure mittels Ammoniak umgefällt. Ausbeute 3, 0 g (630/o der Theorie).
Zur Analyse wurde die Verbindung einmal aus 10 /oiger Essigsäure mittels 10 /aiger Natriumacetatlösung umgefällt und zweimal aus Äthylenchlorid umkristal- lisiert : gelbe mikrokristalline Nädelchen, F. 143 bis 144 .
4, 8 g (0, 01 Mol) 2, 6-Bis-(diäthanolamino)-4- piperidino-8-äthylmercapto-pyrimido-pyrimidin bzw.
4, 6 g (0, 01 Mol) 2, 6-Bis- (diäthanolamino)-4, 8-di äthylmercapto-pyrimido-pyrimidin wurden mit 150 cm3 Piperidin im Einschlussrohr etwa 4 Stunden lang auf 200 erhitzt. Die erhaltene klare gelbe Lösung wurde im Vakuum weitgehend eingeengt. Beim Versetzen des verbleibenden Rückstandes mit etwa 150 cm3 Wasser schied sich das 2, 6-Bis-(diÏthanolamino)-4, 8-dipiperidino-pyrimid'o-pyrimidin alsbald erstarrende bräunlichgelbe Masse ab. Ausbeute 3, 4 g (670/o der Theorie).
Aus Äthylenchlorid : gelbe Prismen, F. 163-165 . b) 2, 4, 6-Tris- (diäthanolamino)-8-piperidino pyrimido-pyrimidin.
4, 6 g (0, 01 Mol) 2, 6-Bis-(diÏthanolamino-4, 8 diäthylmercapto-pyrimido-pyrimidin bzw. 3, 2g (0, 01 Mol) 2, 6-Dichlor-4, 8-diäthyl'mercapto-pyrimido- [5, 4-d]-pyrimidin wurden mit 15, 7 g (0, 15 Mol) Di äthanolamin etwa 3 Stunden lang auf 180 erhitzt.
Die erhaltene Schmelze wurde in 150 cm3 Wasser aufgenommen. Nach Neutralisieren mit Eisessig fiel nach mehrstündigem Stehen das Reaktionsprodukt als feiner gelber Niederschlag aus. Nach Absaugen, Waschen und Trocknen : 3, 0 g (60 /o der Theorie).
Zur Reinigung wurde einmal mit wenig Athylen- chlorid ausgekocht und einmal aus Äthylenchlorid- Aceton (1 : 1) umkristallisiert : gelbes mikrokristallines Pulver, F. = 157-159 .
Darstellung aus dem 2, 4, 6-Tris-(diÏthanolamin) 8-äthylmercapto-pyrimido-pyrimidin, analog dem 2, 6-Bis-(diäthanolamino)-4, 8-dipiperidino-pyrimido- pyrimidin (Versuch a). Ausbeute 57 /o der Theorie.
Aus Dioxan : hellgelbes mikrokristallines Pulver (Nädelchen), F. = 174-176 . c) 2, 6-Bis-(diäthanolamino)-4, 8-di-(ssoxy- äthylamino)-pyrimido-pyrimidin.
Darstellung aus dem 2, 6-Dichlor-4, 8-diäthyl- mercapto-pyrimido-pyrimidin über das 2, 6-Bis- (di äthanolamino)-4, 8-diäthylmercaptoipyrimido-pyrimi- din analog dem 2, 6-Bis-(diÏthanolamino)-4, 8-dipipe ridino-pyrimido-pyrimidin (Versuch a), jedoch ohne Druck (etwa 4 Stunden am Rückfluss). Ausbeute 73 zozo der Theorie. Aus Wasser : blassgelbe, glänzende Schuppen, F. = 238-241.
Beispiel 4
Verschiedene 4, 6, 8-Triamino-pyrimido-pyrimidine
Aus verschiedenen 6-Chlor-4, 8-di- (R-substituier- ten)-pyrimido-pyrimidinen (z. B. 6-Chlor-4, 8-di (carboxymethylthio)-pyrimido-pyrimidin und 6-Chlor 4, 8-diamino-pyrimido-pyrimidin) und den entsprechenden Aminen durch Erhitzen auf höhere Temperatur (gegebenenfalls am Rückfluss oder unter Druck). a) 4, 6, 8-Trianilino-pyrimido-pyrimidin.
3, 5 g (0, 01 Mol) 6-ChIor-4, 8-di- (carboxymethyl thio)-pyrimido-pyrimidin wurden mit 20 cm3 Anilin etwa 1 Stunde lang am Rückfluss erhitzt. Nach Aufnehmen des Reaktionsgemisches in etwa 100 cm3 Wasser und schwachem Ansäuern mit Salzsäure schied sich das 4, 6, 8-Trianilino-pyrimido-pyrimidin als fast farbloser Niederschlag ab. Nach Absaugen, Waschen und Trocknen : 3, 3 g (82 ;"/ der Theorie).
Aus Äthanol : schwach gelbliche glänzende Schüpp- chen, F. 203-204 .
Analog der Verbindung a) wurden (neben an dern) auch die folgenden 4, 6, 8-Triamino-pyrimido- pyrimidine hergestellt : b) 4, 6, 8-Tribenzylamino-pyrimido-pyrimidin,
F. 128-130 . c) 4, 6, 8-Tri- (o-methoxyanilino)-pyrimido- pyrimidin, F. 214-215 . d) 4, 6, 8-Tri- (ss-oxyäthylamino)-pyrimido- pyrimidin, F. 113115 . e) 4, 6, 8-Trimorpholino-pyrimido-pyrimidin,
F. 182-184 .
Beispiel 5 a) 2, 4, 6, 8-Tetraanilino-pyrimido-pyrimidin.
Aus Anilin und 2, 6-Dichlor-4, 8-dithio-pyrimidopyrimidin, 2, 6-Dichlor-4, 8-diäthylthio-pyrimido- pyrimidin und 2, 6-Dichlor-4, 8-däthoxy-pyrimido- pyrimidin. Jeweils 0, 1 Mol des entsprechenden Dichlor-pyrimido-pyrimidins wurden mit 40 cm3 Anilin etwa 1 Stunde lang am Rückfluss gekocht.
Beim Eingiessen der erhaltenen, dunkelbraunen Lösung in 500 cm3 In Salzsäure fiel das rohe Tetraanilino-pyrimido-pyrimidin als bräunlicher, amorpher Niederschlag aus. Ausbeute über 9oa/o. Nach mehrmaligem Umkristallisieren aus Dioxan erhielt man kräftig gelbe Nädelchen vom F. 300-302 . b) 2, 4, 6, 8-Tetraanilino-pyrimido-pyrimidin.
Aus 6-Chlor-2-thio4, 8-dimorpholino-pyrimido- pyrimidin und Anilin. Es wurde wie unter a) beschrieben gearbeitet, ausser dass das Reaktionsgemisch etwa 12 Stunden unter Rückfluss gekocht wurde. Die Ausbeute liegt ebenfalls in der Grössen- ordnung von goa/0.

The pyrimidopyrimidines of the formula II used as starting materials can be prepared, for example, by halogenation of oxypyrimidopyrimidines obtained by ring closure and by subsequent partial exchange of the halogen atoms.
The following are examples of the compounds HR which are suitable for reaction with the pyrimidopyrimidines of the formula II: ammonia, primary or secondary amines, guanidines, hydrazines, amino alcohols, morpholines and piperidines. The process according to the invention is carried out with the addition of an acid-binding agent, such as alkali hydroxide, alkali metal carbonate or tertiary amines, or optionally with an excess of the reaction component HR, if this is suitable as an acid-binding agent.
The reaction can be carried out in the absence or presence of solution or solution which are inert for the reaction. Ver thinners, z. B. acetone, dioxane, benzene, xylene or dimethylformamide, and optionally with the application of pressure. Water and alcohols can also be used as solvents or diluents, especially in the absence of alkalis and at low temperatures, since they practically do not react with the halogen-containing pyrimidopyrimidines under these conditions. The second reaction component HR, if it is liquid under the reaction conditions, can also be used in excess as a solvent or diluent.

The compounds obtained according to the invention can be converted into their acid addition salts with therapeutically inert acids. The compounds prepared by the process according to the invention have excellent therapeutic properties. They are particularly effective cardiovascularly. 3.9 g (0.01 mol) 2, 6-dichloro-4, 8-diphenoxypyrimidopyrimidine (F. 294-295) were refluxed with 20 cm 3 of β-oxyethylamine (boiling point 171) for about 1 hour heated. Example 3 a) 2,6-Bis-t-diethanolamino) 4,8-dipiperidino-pyrimido-pyrimidine.
12.8 g (0.04 mol) of 2,6-dichloro-4, 8-diethylmer-capto-pyrimido-pyrimidine, F. = 190-192, were added to 63 g (0.6 mol) of diethanolamine for about 1 hour 160 heated. When the clear, honey-colored melt was poured into 300 cm3 of water, the crude reaction product precipitated out as an orange-colored precipitate. After filtering off with suction, washing and drying, the yield was 9.2 g (50.100 of theory). 4.6 g (0.01 mol) of 2,6-bis-diethanolamino) -4, 8 diethylmercapto-pyrimido-pyrimidine were heated to 180 for about 3 hours with 40 cm3 of piperidine in a sealed tube. When the resulting mixture was rinsed out with about 300 cm3 of water, the reaction product separated out as a greasy yellow-brown precipitate. It was immediately reprecipitated once from dilute hydrochloric acid using ammonia. Yield 3.0 g (630 / o of theory).
For analysis, the compound was reprecipitated once from 10% acetic acid using 10% sodium acetate solution and recrystallized twice from ethylene chloride: yellow microcrystalline needles, F. 143 to 144.
4.8 g (0.01 mol) 2, 6-bis (diethanolamino) -4- piperidino-8-ethylmercapto-pyrimido-pyrimidine or
4.6 g (0.01 mol) of 2,6-bis (diethanolamino) -4, 8-diethylmercapto-pyrimidopyrimidine were heated to 200 with 150 cm3 of piperidine in the containment tube for about 4 hours. The clear yellow solution obtained was largely concentrated in vacuo. When the remaining residue was mixed with about 150 cm3 of water, the 2,6-bis- (diÏthanolamino) -4,8-dipiperidino-pyrimid'o-pyrimidine, which soon solidified, brownish-yellow material separated out. 4.6 g (0.01 mol) 2, 6-bis (diÏthanolamino-4, 8 diethylmercapto-pyrimido-pyrimidine or 3.2g (0.01 mol) 2, 6-dichloro-4, 8-diethyl ' mercapto-pyrimido- [5, 4-d] -pyrimidine were heated to 180 for about 3 hours with 15.7 g (0.15 mol) diethanolamine.

From aniline and 2,6-dichloro-4, 8-dithio-pyrimidopyrimidine, 2,6-dichloro-4, 8-diethylthio-pyrimidopyrimidine and 2,6-dichloro-4,8-dethoxypyrimidopyrimidine. In each case 0.1 mol of the corresponding dichloropyrimido-pyrimidine was refluxed with 40 cm3 of aniline for about 1 hour.
