Verfahren zur Herstellung von Purinderivaten Es sind bisher nur sehr wenige Purinderivate be kannt, die zwei substituierte Aminogruppen im Kern enthalten. Sie werden teils durch Halogenaustausch (z. B. 2,6-Dialkylaminoalkylamino-dialkylamino-pu- rine, Journ. Amer. Chem. Soc., Band 67, 1945, Seite 1271) teils durch Chlorierung in Gegenwart tertiärer Amine (z.
B. 2,6-bis-Dialkylamino-purine, Journ. Amer. Chem. Soc., Band 74, 1952, Seite 3624) er halten.
Die vorliegende Erfindung betrifft nun ein Verfah ren zur Herstellung von Purinderivaten der Formel

Als Beispiele für bei dem erfindungsgemässen Ver fahren als Ausgangsstoffe verwendbare halogenhaltige Purine seien u. a. folgende genannt: 2,6,8-Trichlor-purin 2,6-Dichlor-7-methyl-purin, 2,6,8-Tribrom-7-methyl-purin, 2-Chlor-6,8-dijodid-7-methyl-purin, 2,6, 8-Trichlor-9-phenyl-purin, 2,6-Dichlor-8-oxy-9-phenyl-purin, 2-Chlor-6, 8-dipiperidino-7-methyl-purin, 2,6-Dichlor-8-anilino-7-methyl-purin,
2-Chlor-6-hydrazino-8-morpholino-7- methyl-purin.
Zu den für die Umsetzung mit den halogenhalti- gen Purinen geeigneten Verbindungen der Formel H-R gehören gegebenenfalls ein weiteres Heteroatom ent haltende cyclische Amine.
Es wird unter Zusatz eines säurebindenden Mit tels, z. B. eines Alkalihydroxyds, Alkalicarbonats, Al- kalialkoholats oder tertiären Amins, gegebenenfalls unter Verwendung eines Überschusses der Verbin dung H-R, wenn diese sich als säurebindendes Mittel eignet, gearbeitet.
Die Umsetzung kann in Ab- oder Anwesenheit von Lösungs- bzw. Verdünnungsmittel durchgeführt werden. Vorzugsweise wird sie jedoch in an der Um- setzung nicht teilnehmenden Lösungsmitteln, z. B. Aceton, Dioxan, Benzol und Dimethylformamid, durchgeführt. Wenn der Siedepunkt des verwendeten Lösungsmittels den durch einfachen Vorversuch zu ermittelnden, für den Ablauf der Umsetzung gün stigsten Temperaturbereich nicht erreichen lässt, kann unter Druck gearbeitet werden.
Auch Wasser und Alkohole können, vor allem in Abwesenheit von Alkalien und bei niederen Reak tionstemperaturen, als Lösungs- und Verdünnungs mittel dienen, da sie unter den Reaktionsbedingungen mit den halogenhaltigen Purinen praktisch nicht rea gieren. Ferner kann die Verbindung der Formel H-R, falls sie unter den Reaktionsbedingungen flüssig ist, im überschuss als Lösungs- oder Verdünnungsmittel Verwendung finden.
Als Reaktionsbeschleuniger für das erfindungs gemässe Verfahren haben sich z. B. Kupfer und Kup- fersalze sowie Kaliumjodid als besonders geeignet er wiesen.
Wenn das als Ausgangsstoff verwendete halogen- haltige Purin mindestens 2 Halogenatome als Substi- tuenten enthält, kann der Austausch dieser zwei oder mehr Halogenatome gegen den Rest R der Verbin dung H-R auch stufenweise durchgeführt werden. Es wurde nämlich gefunden, dass bei niederen oder mitt leren Temperaturen praktisch nur ein oder zwei Halogenatome ausgetauscht werden und sich erst bei höheren Temperaturen alle vorhandenen Halogen atome durch andere Gruppen ersetzen lassen.
So wird beispielsweise bei der Umsetzung von 2,6,8-Trichlor- 7-methyl-purin mit Morpholin in Dioxan oder Di- methylformamid unter Kühlung, das heisst bis etwa 5 C, nur ein Halogenatom gegen den Morpholino- rest ausgetauscht, der sehr wahrscheinlich in die 8- Stellung eintritt.
Wird bei der gleichen Umsetzung un ter Verwendung von Dioxan als Lösungsmittel kurz zeitig erwärmt, dann lassen sich zwei Halogenatome, sehr wahrscheinlich die in 6- und 8-Stellung stehen den, austauschen, während schliesslich durch 1/2stün- diges Sieden unter Verwendung von Morpholin im Überschuss ohne weiteres Lösungsmittel unter Rück fluss alle drei Halogenatome durch Morpholinogrup- pen ersetzt werden.
Das erfindungsgemässe Verfahren kann über raschenderweise unter besonders milden Bedingungen durchgeführt werden. So erfordert beispielsweise die Herstellung von 2-(,-Diäthylaminopropylamino)-6-di- äthylamino-7-methyl-purin sehr kräftige Reaktions bedingungen. Das als Ausgangsstoff verwendete 2- Chlor-6-diäthylamino-7-methyl-purin muss mit dem y-Diäthylaminopropylamin 17 Stunden auf 195 C unter Druck erhitzt werden (vgl. Journ. Amer. Chem. Soc., Band 67, 1945, Seite 1271).
Ausgehend von 2,6,8-Trichlor-7 - methyl - purin kann dagegen das 2,6,8-Tripiperidino-7-methyl-purin nach dem erfindungsgemässen Verfahren bereits durch zweistündiges Rückflusssieden (bei etwa 106 C) der Ausgangsstoffe erhalten werden.
Die nach dem erfindungsgemässen Verfahren er hältlichen neuen Purinderivate, die mindestens eine cyclische, gegebenenfalls ein weiteres Heteroatom auf weisende Aminogruppe und mindestens eine weitere substituierte Aminogruppe enthalten, sowie ihre in Wasser leicht löslichen Salze zeigen sehr wertvolle, grösstenteils bei Purinderivaten völlig unerwartete pharmakologische Eigenschaften, beispielsweise wir ken sie coronarerweiternd, analgetisch und sedativ.
So übertrifft beispielsweise das 2,6-Dimorpholino-7- methyl-purin bezüglich seiner coronarerweiternden Wirkung das Theophyllin um etwa das Fünffache.
Ausserdem zeigt es blutdrucksenkende und atem anregende Wirkungen. Das 2-Morpholino-6,8-dipipe- ridino-7-methyl-purin weist sehr gute analgetische und sedative Wirkungen auf.
Einige der nach dem erfindungsgemässen Verfah ren hergestellten Verbindungen zeigen ferner gute antipyretische Wirkungen, z. B. das 2,6-Dimorpho- lino-7-methyl-purin, 2-Morpholino-6,8-dipiperidino- 7-methyl-purin und das 2,6,8-Tripiperidino-7-methyl- purin.
<I>Beispiel 1</I> Herstellung verschiedener 2-Chlor-6,8-diamino- 7-methyl-purine aus 2,6,8 - Trichlor - 7-methyl-purin und den entsprechenden Aminen a) 2-Chlor-6,8-dipiperidino-7-mehyl-purin Zu einer Lösung von 9,5 g (0,04 Mol) 2,6,8-Tri- chlor-7-methyl-purin in 100 cm3 Dioxan werden un ter Umrühren 20 em3 Piperidin gegeben (Ausfallen von Piperidin-hydrochlorid), und anschliessend wird zum Sieden erhitzt.
Nach dem Erkalten wird das er haltene Gemisch in etwa 350 cm3 Wasser gegossen, wobei sich (unter Auflösen des Hydrochlorids) das Reaktionsprodukt zunächst als schmieriger, jedoch bald erstarrender Niederschlag abscheidet. Es wird abgesaugt, gewaschen und bei Zimmertemperatur ge trocknet. Ausbeute 10,2 g (76% d. Th.). Zur Reini gung wird die Verbindung einmal aus Methanol um kristallisiert: farblose, glänzende, längliche Blättchen; F = 140-1420 C.
b) 2-Chlor-6,8-dimorpholino-7-methyl-purin Die Herstellung erfolgt analog derjenigen der Piperidino-Verbindung. Ausbeute 75%. Nach Um-. fällen aus heisser 0,5n Salzsäure: schwach elfenbein- farbenes, mikrokristallines Pulver;
F=284-286"C. <I>Beispiel 2</I> 2-Chlor-6-morpholino-8-benzylamino-7- methyl-purin 3,1 g (0,01 Mol) 2,6-Dichlor-8-benzylamino-7- methylpurin (F<B>=</B> 226-228 C) werden mit 5 cm3 Morpholin in 30 cm3 Dimethylformamid etwa 1 Stunde lang auf etwa 125 C erhitzt.
Beim Eingiessen der erhaltenen Lösung in 150 cm3 Wasser fällt das Purinderivat zunächst als schmieriger, jedoch nach einigem Stehen erstarrender Niederschlag aus. Er wird abgesaugt, gewaschen und getrocknet: 3,1 g (86% d. Th.). Nach dem Umkristallisieren aus Me thanol: fast farblose, glänzende Nadeln, F = 211 bis 213 C.
<I>Beispiel 3</I> 2,6,8-Trimorpholino-purin 1,5 g (etwa 0,055 Mol) Trichlor-purin werden mit 20 cm3 Morpholin und 0,1g Kupfersulfat im Bom benrohr 2 Stunden lang auf 200-210 C erwärmt.
Das beim Aufnehmen des Reaktionsgemisches in etwa 100 cm-' Wasser ungelöst bleibende rohe Purinderivat wird abgesaugt: 0,9 g (48% d. Th.). Es wird zur Ana lyse dreimal aus Methanol umkristallisiert (farblose Nädelchen, F = 247-248 C) und bei 60 C und 0,1 Torr getrocknet.

d) 2-Morpholino-6,8-dipiperidino-7-methyl-purin Ausbeute 95%. Nach Umfällen aus 0,5n Salz säure-Alkohol (2: 1) mit Ammoniak: farbloses, mikrokristallines Pulver; F = 189-190 C.
e) 2-Pyrrolidino-6,8-dimorpholino-7-methyl-purin Ausbeute 89%. Nach Umfällen aus 0,1n Salzsäure: kleine farblose Nädelchen; F = 197-199 C (nach Trocknen bei 110 ' C).
2-Morpholino-6-hydrazino-8-morpholino-7- methyl-purin Ausbeute 42%. Aus Methanol fast farbloses, mi- krokristallines Pulver (kurze Prismen, F = 221 bis 223 C).
g) 2-Morpholino-6-diäthylamino-8-piperidino-7- methyl-purin Ausbeute 78%. a) 2,6-Dimorpholino-8-piperidino-7-methyl-purin 2,9 g (0,01 Mol) 2,6-Dichlor-8-piperidino-7-me- thyl-purin (F = 143-l45 C, erhalten aus 2,6,8-Tri- chlor-7-methyl-purin und Piperidin in Dimethylform- amid unter Kühlung) werden mit 10 cm3 (0,1 Mol) Morpholin im Bombenrohr etwa 2 Stunden lang auf 200 C erhitzt.
Das Reaktionsprodukt wird sofort ein mal aus etwa 1n Salzsäure umgefällt, abgesaugt, mit Wasser gewaschen und bei 110 C getrocknet (nahezu farbloses, mikrokristallines Pulver, F = 206-208 C). Ausbeute (praktisch reine Substanz) 3,6 g (93 d. Th.). Zur Analyse wird die Verbindung noch ein mal aus sehr verdünnter Schwefelsäure umgefällt und einmal aus Äthanol-Wasser (1 :2) umkristallisiert: farblose, mikrokristalline Prismen; F = 209-211 C.

Ausbeute 3,5 g (76% d. Th.). Zur Reinigung wird die Substanz einmal aus verdünnter Natronlauge mit Salzsäure und einmal aus heisser verdünnter Salzsäure mit Am moniak umgefällt.
Das so erhaltene elfenbeinfarbene Pulver zeigt bis 350 C keinen Schmelzpunkt.

b) Die Umsetzung des 2,6,8-Tribrom-7-methyl-pu- rins (aus Äthanol-Wasser [1 : 1]: blassgelbes, mikro kristallines Pulver [Prismen], F = 194-196 C) mit Morpholin verläuft in analoger Weise wie Versuch a.
<I>Beispiel 9</I> 2,6,8-Trimorpholino-9-phenyl-purin aus 2,6,8-Tri- chlor-9-phenyl-purin und Morpholin 2,7 g (0,009 Mol) 2,6,8-Trichlor-9-phenyl-purin werden mit 27 cm3 Morpholin im Bombenrohr 1112 Stunden lang auf 160 C erhitzt.
Beim Herausspülen des Reaktionsgemisches mit Wasser scheidet sich (un ter gleichzeitigem Auflösen des entstandenen Morpho- linhydrochlorids) das 2,6,8-Trimorpholino-9-phenyl- purin als farbloser, amorpher Niederschlag ab. Aus beute 2,5 g (63 % d. Th.). Zur Analyse wird die Ver bindung zweimal aus warmer 1n Salzsäure umgefällt und einmal aus Methanol umkristallisiert: fast farb loses, mikrokristallines Pulver; F = 223-224 .

b) 2,6,8-Tripiperidino-7-methyl-purin Die Herstellung dieser Verbindung erfolgt analog der der Morpholin-Verbindung durch zweistündiges Kochen am Rückfluss (bei etwa 106 C). Ausbeute 91 %. Nach Absaugen, Waschen und Trocknen: 1,0 g (55% d. Th.). Zur Reinigung wird die Verbindung einmal aus 0,5n Salzsäure umgefällt und einmal aus Methanol umkristallisiert: farblose, feine, mikrokristalline Nädel- chen; F = 244-245 C.

Beim Eingiessen des noch warmen Reaktionsgemisches (klare, gelb braune Lösung) in etwa 100 cm3 Wasser scheidet sich nach kurzer Zeit das 2,8-Dimorpholino-6-piperidino- 7-methyl-purin als gelber, kristalliner Niederschlag ab. Nach Absaugen, Waschen und Trocknen erhält man 3,4 g Substanz (58% d. Th.). Zur Analyse wird ein mal aus kalter 0,1n Salzsäure umgefällt und einmal aus Methanol-Wasser (1 :3) umkristallisiert: elfen beinfarbene, mikrokristalline Prismen; F = 207 bis 209 C.
