Verfahren zur Herstellung von 6,7-Dehydro-ss-apo-3-carodnal
Gegenstand der vorliegenden Erfindung ist ein neues Verfahren zur Herstellung von 15- [2, 6,6-Tri methyl-cyclohexen- (l)-yl]-4,9,13-trimethyl-pentadeca- hexaen- (2, 4,8,10,12,14)-in- (6)-al- (1), welches dadurch gekennzeichnet ist, dass man 1, 1-Dialkoxy-2-methyl- penten- (2)-in- (4) mittels einer metallorganischen Reaktion mit 8- [2, 6,6-Trimethyl-cyclohexen-(l)-yl]2,6-dimethyl-octatrien- (2,4,6)-al- (l) kondensiert, das Kondensationsprodukt mit Säure behandelt und das erhaltene 13- [2,6,6-Trimethyl-cyclohexen-(l)-yl]2,7,11-trimethyl-tridecapentaen- (2,6,8,10,12)-in- (4) al- (l) acetalisiert,
das Acetal mit einem Vinyläther kondensiert und das Kondensationsprodukt mit Säure behandelt. Das Reaktionsprodukt kann an der Dreifachbindung partiell hydriert und isomerisiert werden.
Man kann z. B. die 1, 1-Diäthylverbindung der als Ausgangsmaterial genannten C6-Acetale wie folgt herstellen : Man kondensiert Orthoameisensäureäthyl- ester mit Propenyläther in Gegenwart von Bortrifluoridätherat zu 1, 1, 3,3-Tetraäthoxy-2-methyl-propan (Siedepunkt 930110mm ; nD = 1,4132). Durch saure Hydrolyse gewinnt man den freien Methylmalondialdehyd in wässeriger Lösung ; man ver äthert ihn durch azeotrope Destillation mit Benzol und Äthylalkohol in Gegenwart von p-Toluolsulfosäure als Katalysator.
Den gebildeten 1-Athoxy-2- methyl-propen- (l)-al- (3) (Siedepunkt 78-80 113 mm ; nid = 1,4755) wird mit einer Alkalimetall-oder Erdalkalimetallverbindung von Acetylen in flüssigem Ammoniak kondensiert und anschliessend mit Orthoameisensäureäthylester und einem Gemisch von p-Toluolsulfosäure und Phosphorsäure behandelt, wobei man das gewünschte l, l-Diäthoxy-2-methyl penten- (2)-in- (4) erhält. Siedepunkt 45-46 l0, 1 mm ; nid = 1,4520-1,4540 ; UV-Absorptionsmaximumbei 225 m, (in Alkohol).
Geeignete Ausgangsmaterialien sind insbesondere Verbindungen, in welchen die 1, 1-Dialkoxyreste niederaliphatische Radikale, wie z. B. die Athoxy-oder Isopropoxygruppe, darstellen.
Die Kondensation des, e-Ct9-Aldehyds erfolgt, wie gesagt, mit einem metallorganischen Derivat des C6-Acetals, zweckmässig in Gegenwart eines inerten Lösungsmittels, wie z. B. Benzol oder Ather. Vorzugsweise kondensiert man den ss-Ctg-Aldehyd mit einer Grignard-oder Lithiumverbindung der genannten Acetale, die man vorgängig der Reaktion im gleichen Gefäss und im gleichen Lösungsmittel, die zur Kondensation verwendet werden, z. B. aus Magnesium, Athylbromid und dem entsprechenden Acetal herstellen kann.
Das erhaltene metallorganische Kondensationsprodukt wird am besten ohne weitere Reinigung in üblicher Weise hydrolytisch zersetzt, beispielsweise durch Eingiessen in ein Gemisch von Eis und verdünnter Ammoniumchloridlösung, wobei die gebildete Acetylenverbindung in Form eines zähen Ols erhalten wird. Diese Acetylenverbindung wird, wie gesagt, danach einer Säurebehandlung unterworfen, wobei unter Wasser-und Alkoholabspaltung ein Aldehyd entsteht. Diese Reaktion wird zweckmässig in einem inerten Lösungsmittel, wie z. B.
Aceton, Alkohol oder Ather, in Gegenwart einer wässerigen oder wasserfreien Säure, wie z. B. Essigsaure, p-Toluolsulfosäure, Salzsäure usw., durchgeführt. Die Acetalisierung des gebildeten Aldehyds erfolgt zweckmässig durch Umsetzung mit einem Gemisch von Alkohol und Orthoameisensäureester in Gegenwart eines sauren Katalysators, wie z. B.
Phosphorsäure oder p-Toluolsulfosäure.
Zur Vornahme der Kondensation mit einem Vinyläther, z. B. dem Vinyläthyläther, arbeitet man zweckmässig in Gegenwart eines inerten Lösungsmit tels, wie z. B. Benzol, Ather oder Petroläther. Als Kondensationsmittel verwendet man saure Katalysatoren, wie z. B. Zinkchlorid, Bortrifluoridätherat usw.
Die erhaltenen Ätheracetale werden anschliessend, gleich wie die Kondensationsprodukte des (3-Ci- Aldehyds, der Säurebehandlung unterworfen. Die Säurebehandlung wird besonders vorteilhaft durch Erwärmen auf 100 mit einem Gemisch von Natriumacetat in wässerigem Eisessig vorgenommen. Die vorhandene dreifach ungesättigte Bindung des erfindungsgemäss erhaltenen 6,7-Dehydro-, 8-apo-3-carotinals kann partiell hydriert werden. Diese Hydrierung erfolgt zweckmässig mit Wasserstoff in Gegenwart von Metallkatalysatoren in einem inerten Lösungsmittel, wie z. B. Petroläther. Ein geeigneter Katalysator ist ein durch Zugabe von Blei und Chinolin desaktivierter Palladium-Calciumcarbonat-Katalysator. Die Isomerisierung findet schon unter milden Bedingungen statt, z.
B. durch Wärme-oder Lichteinfluss.
Der erfindungsgemäss erhaltene Polyenaldehyd stellt einen wertvollen Farbstoff dar. Er kann als Lebensmittelfarbstoff sowie als Geflügelfutterzusatz für die Eidotterfärbung verwendet werden. Weiter stellt er ein Zwischenprodukt für die Synthese wichtiger Carotinoidpigmente dar.
Beispiel
Man stellt zunächst unter Eiskühlung aus 2,7 g Magnesium, 14 g Äthylbromid und 30 cm3 absolutem Äther eine Lösung von Äthylmagnesiumbromid her. Dazu tropft man innert 30 Minuten unter kräfti- gem Rühren ohne Kühlung eine Lösung von 16,8 g 1, 1-Diäthoxy-2-methyl-penten-(2)-in-(4) in 30 cm3 absolutem Benzol, wobei die Reaktionslösung stets Ieicht siedet. Die Grignardverbindung scheidet sich zum Teil körnig, zum Teil in zähflüssiger Form aus.
Man kocht noch während 30 Minuten am Rückfluss und tropft dann innert 30 Minuten unter Kühlung mit Wasser eine Lösung von 22 g ff-Ct9-Aldehyd in 50 cm3 absolutem Äther zu, wobei bis auf wenig ungelöste Anteile eine klare gelbe Lösung entsteht.
Man rührt noch während 3 Stunden bei Raumtemperatur weiter und giesst dann die Reaktionslösung auf eine Mischung von 20 g Ammoniumchlorid und Eis.
Nach Extraktion mit Ather, Waschen mit Wasser, Trocknen über Natriumsulfat und Eindampfen im Vakuum bei 45 , erhält man 39 g 13- [2, 6,6-Trimethyl-cyclohexen- (1)-yl]-2, 7,11-trimethyl-1, l-di äthoxy-6-hydroxy-tridecatetraen-(2, 7,9,11)-in- (4) als zähflüssiges Kondensationsprodukt. UV-Absorptionsmaximum bei 281 m, (in Petroläther).
Das erhaltene Kondensationsprodukt wird in 200 cm3 Äther gelöst und nach Zugabe von 20 cm3 alkoholischer Salzsäure über Nacht bei Raumtemperatur stehengelassen. Die entstandene Lösung wird vorerst mit Natriumbicarbonatlösung und anschlie ssend mit Wasser gewaschen, mit Natriumsulfat getrocknet und im Vakuum bei 45 eingedampft. Man erhält dabei etwa 29 g eines zähflüssigen stark ge färbten Ols. Dieses wird in 150 cm3 Petroläther (Siedeintervall 30-45 ) gelöst, angeimpft und wäh- rend einigen Stunden bei-10 stehengelassen. Der ausgeschiedene Kristallbrei wird abgenutscht und mehrmals mit kaltem Petroläther gewaschen.
Man trocknet im Vakuum bei 30 und erhält 17 g 4,5-De hydro--apo-4-carotinal als ockerfarbiges Pulver.
Schmelzpunkt 82-83 ; UV-Absorptionsmaximum bei 400 m, (in Petroläther).
Diese Verbindung kann auch wie folgt gewonnen werden : Zu einer ausgehend von 8 g Lithium bereiteten Lösung von Lithiumamid in 2000 cm3 flüssi- gem Ammoniak tropft man innert einer Stunde 168 g 1, 1-Diäthoxy-2-methyl-penten-(2)-in-(4) zu und spült mit 20 cm3 Äther nach. Man rührt noch während 4-5 Stunden und fügt dann innerhalb einer Stunde eine Lösung von 220 g B-C, 9-Aldehyd in 750 cm3 absolutem Äther zu. Das Reaktionsgemisch wird über Nacht weitergeruhrt und anschliessend, wie weiter oben beschrieben, unter Verwendung von 100 g Ammoniumchlorid hydrolysiert und aufgearbeitet, wobei man 4,5-Dehydro-, 6-apo-4-carotinal erhält.
18 g 4,5-Dehydro-ss-apo-4-carotinal werden in 17 cm3 Orthoameisensäureäthylester suspendiert, mit einer Lösung von 0,24 cm3 Phosphorsäure und einer Spur p-Toluolsulfosäure in 8 cm3 absolutem Alkohol versetzt und einige Stunden bei Raumtemperatur verrührt. Wenn alles gelöst ist, lässt man noch 15 Stunden stehen, gibt unter Eiskühlung 1,5 cm3 Pyridin und hierauf 50 cm3 5 /0, ige natlösung zu und extrahiert mit Petroläther. Man wäscht den Extrakt mit Wasser, trocknet mit Natriumsulfat und dampft im Vakuum ein.
Der Rück- stand wird im Hochvakuum bei 60 getrocknet und man erhält 21,7 g 13- [2, 6,6-Trimethyl-cyclohexen (1)-yl]-2, 7,11-trimethyl-1, 1-diäthoxy-tridecapentaen (2,6,8,10,12)-in- (4) als gelbes 01. UV-Absorptionsmaxima bei 366 und 380 m, lt (in Petroläther). Dieses wird mit 4 cm3 10"/oiger Zinkchloridlösung in Essigester und hierauf tropfenweise mit 4,27 g Vinyläther unter Rühren bei 30-35 versetzt.
Man lässt das Reaktionsgemisch 15 Stunden stehen, gibt zum entstandenen rohen 15- [2, 6,6-Trimethyl-cyclohexen- (1)-yl]4,9,13-trimethyl-1,1,3-triäthoxy-pentadecapentaen (4,8,10,12,14)-in- (6) eine Lösung von 5 g Natriumacetat in 50 cm3 954/piger Essigsäure zu und erhitzt unter energischem Rühren in Stickstoffatmosphäre 4 Stunden auf 95-100 , wobei der gebildete Essigester langsam abdestilliert. Man lässt die Reaktionsmischung unter Rühren erkalten, wobei das Reaktionsprodukt auskristallisiert. Es wird abfiltriert, mit Wasser gewaschen und aus Petroläther oder Athanol umkristallisiert. Man erhält 13 g 6,7-Dehydro-fl-apo3-carotinal als orangerote Plättchen.
Schmelzpunkt 122 ; UV-Absorptionsmaximum bei 417-418 m, (in Petroläther).

B. produce the 1,1-diethyl compound of the C6-acetals mentioned as starting material as follows: Ethyl orthoformate is condensed with propenyl ether in the presence of boron trifluoride ether to give 1,1,3,3-tetraethoxy-2-methylpropane (boiling point 930110mm; nD = 1.4132). The free methylmalondialdehyde is obtained in aqueous solution by acid hydrolysis; it is etherified by azeotropic distillation with benzene and ethyl alcohol in the presence of p-toluenesulfonic acid as a catalyst.
The 1-ethoxy-2-methyl-propen- (1) -al- (3) formed (boiling point 78-80 113 mm; nid = 1.4755) is condensed with an alkali metal or alkaline earth metal compound of acetylene in liquid ammonia and then condensed treated with ethyl orthoformate and a mixture of p-toluenesulfonic acid and phosphoric acid, the desired l, l-diethoxy-2-methyl-pentene- (2) -yne- (4) being obtained. Boiling point 45-46 l0.1 mm; nid = 1.4520-1.4540; UV absorption maximum at 225 m, (in alcohol).
Suitable starting materials are in particular compounds in which the 1, 1-dialkoxy radicals are lower aliphatic radicals, such as. B. the ethoxy or isopropoxy group represent.
The condensation of the, e-Ct9-aldehyde takes place, as said, with an organometallic derivative of C6-acetal, conveniently in the presence of an inert solvent, such as. B. benzene or ether. Preferably, the ß-Ctg-aldehyde is condensed with a Grignard or lithium compound of the acetals mentioned, which are used prior to the reaction in the same vessel and in the same solvent that are used for the condensation, e.g. B. from magnesium, ethyl bromide and the corresponding acetal.

The polyenaldehyde obtained according to the invention is a valuable coloring agent. It can be used as a food coloring agent and as a poultry feed additive for coloring egg yolks. It is also an intermediate for the synthesis of important carotenoid pigments.

A solution of ethylmagnesium bromide is first prepared from 2.7 g of magnesium, 14 g of ethyl bromide and 30 cm3 of absolute ether while cooling with ice. A solution of 16.8 g of 1,1-diethoxy-2-methyl-pentene- (2) -in- (4) in 30 cm3 of absolute benzene is added dropwise to this over a period of 30 minutes with vigorous stirring without cooling, with the reaction solution always simmering. The Grignard compound is partly granular and partly in a viscous form.
The mixture is refluxed for a further 30 minutes and a solution of 22 g of ff-Ct9-aldehyde in 50 cm3 of absolute ether is then added dropwise over a period of 30 minutes while cooling with water, a clear yellow solution being formed except for a few undissolved portions.
The mixture is stirred for a further 3 hours at room temperature and the reaction solution is then poured onto a mixture of 20 g of ammonium chloride and ice.
After extraction with ether, washing with water, drying over sodium sulfate and evaporation in vacuo at 45, 39 g of 13- [2,6,6-trimethyl-cyclohexen (1) -yl] -2,7,11-trimethyl are obtained -1, l-diethoxy-6-hydroxy-tridecatetraen- (2, 7,9,11) -in- (4) as a viscous condensation product. UV absorption maximum at 281 m, (in petroleum ether).
The condensation product obtained is dissolved in 200 cm3 of ether and, after the addition of 20 cm3 of alcoholic hydrochloric acid, left to stand overnight at room temperature. The resulting solution is first washed with sodium bicarbonate solution and then with water, dried with sodium sulfate and evaporated at 45 ° in vacuo. 18 g of 4,5-dehydro-ss-apo-4-carotinal are suspended in 17 cm3 of ethyl orthoformate, mixed with a solution of 0.24 cm3 of phosphoric acid and a trace of p-toluenesulfonic acid in 8 cm3 of absolute alcohol and stirred for a few hours at room temperature. When everything is dissolved, it is left to stand for another 15 hours, 1.5 cm3 of pyridine and then 50 cm3 of 5/0 sodium solution are added while cooling with ice, and the mixture is extracted with petroleum ether. The extract is washed with water, dried with sodium sulfate and evaporated in vacuo.
The residue is dried in a high vacuum at 60 and 21.7 g of 13- [2,6,6-trimethyl-cyclohexen (1) -yl] -2, 7,11-trimethyl-1, 1-diethoxy are obtained tridecapentaen (2,6,8,10,12) -in- (4) as yellow 01. UV absorption maxima at 366 and 380 m, lt (in petroleum ether). The reaction mixture is allowed to cool while stirring, the reaction product crystallizing out. It is filtered off, washed with water and recrystallized from petroleum ether or ethanol. 13 g of 6,7-dehydro-fl-apo3-carotinal are obtained as orange-red platelets.
