Verfahren zur Herstellung eines Polyenaldehydes
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 13- [2', 6', 6'-Trimethyl-cyclohexen (1')-yl]-2, 7, 11-trimethyl-tridecapentaen-(2, 6, 8, 10, 12) in- (4)-al- (1), im folgenden 4, 5-Dehydro--apo-4- carotinal genannt.
Das Verfahren ist dadurch gekennzeichnet, da¯ man über die entsprechenden metallorganischen Acetylenverbindungen zuerst Acetylen mit 8- [2', 6', 6'-Tri methyl-cyclohexen- (1')-yl]-2, 6-dimethyl-octatrien (2, 4, 6)-al- (l)-im folgenden 4-Ctg-Aldehyd ge nannt-und anschliessend das erhaltene 10-[2', 6', 6' Trimethyl-cyclohexen-(1')-yl]-4,8-dimethyl- decatrien (4, 6, 8)-in-(1)-ol-(3) - im folgenden ¯-C21-Acetylencarbinol genannt-mit einem 3-Acyloxy-oder 3 Alkoxy-2-methyl-propen- (2)-al- (1) oder zuerst Acetylen mit einem 3-Alkoxy-2-methyl-propen- (2)-al- (1) und anschliessend das erhaltene 1-Alkoxy-2-methyl- 3-hydroxy-penten-(1)-in-(4) mit 8-[2',6',
6'-Trimethylcyclohexen- (1')-yl]-2, 6-dimethyl-octatrien- (2, 4, 6)al-(1) kondensiert und das erhaltene 1-Alkoxy-bzw.
1-Acyloxy-13- [2', 6', 6-trimethyl-cyclohexen- (10-yl]- 2, 7, 11-trimethyl-tridecatetraen-(l, 7, 9, 11)-in- (4)-diol- (3, 6)-mit Säure behandelt.
Die benötigten Ausgangsmaterialien können wie folgt hergestellt werden : 3-Alkoxy-2-methyl-propen-(2)-al-(1)
Man kondensiert Orthoameisensäureäthylester mit Propenyläthyläther in Gegenwart von Bortrifluorid ätherat zu 1, 1, 3, 3-Tetraäthoxy-2-methyl-propan (Siedepunkt : 93¯ C/10 mm; n20¯= 1, 4132). Durch saure Hydrolyse gewinnt man den freien Methylmalondial dehyd in wässeriger Lösung und veräthert ihn durch azeotrope Destillation mit Benzol und Isopropylalkohol in Gegenwart von p-Toluolsulfonsäure als Kataly- sator. Das gebildete 3-Isopropoxy-2-methyl-propen (2)-al-(1) siedet bei 83 C/13 mm ; n2rl = 1, 4745.
Analog kann man 3-Athoxy-2-methyl-propen- (2)-al- (1) (Siedepunkt : 78-80¯C/13 mm ; n2r4 = 1, 4755) gewinnen.
3-Acyloxy-2-methyl-propen-(2)-al-(1)
Durch Neutralisieren einer wässerigen Lösung des freien Methylmalonaldehyds mit 1 Aquivalent Natronlauge und Einengen erhält man das Natriumsalz als weisses Pulver. Dieses wird in einem inerten Lösungsmittel, z. B. Methylenchlorid, suspendiert und mit einem Säurechlorid in der Wärme umgesetzt. Die vom gebildeten NaCl filtrierte Lösung wird im Vakuum eingeengt, und der Rückstand durch Destillation oder Kristallisation gereinigt.
Man erhält z. B. mittels Benzoylchlorid farblose Nadeln des 3-Benzoyloxy-2-methyl-propen- (2)-al- (l) (Schmelzpunkt : 79-80 C) ; mittels Acetylchlorid gewinnt man das 3-Acetoxy-2-methyl-propen- (2)-al- (l), das bei 75-77¯ C/10 mm siedet und bei tieferer Temperatur fest wird.
In der ersten Stufe des erfindungsgemässen Verfahrens lässt man in einer Ausführungsform ¯-C19 Aldehyd in flüssigem Ammoniak mit einem Alkalioder Erdalkaliacetylid reagieren und setzt das gebildete Kondensationsprodukt, vorzugsweise nach Hydrolyse zum freien 4-C2l-Acetylencarbinol, mittels einer metallorganischen Reaktion mit einem 3-Acyloxy-bzw. 3-Alkoxy-2-methyl-propen- (2)-al- (l) um.
Als 3-Acyloxy-2-methyl-propen-(2)-al-(1) verwendet man mit Vorteil eine entsprechende niederaliphatische bzw. aromatische Acylverbindung, beispielsweise 3 Acetoxy-bzw. 3-Benzoyloxy-2-methyl-propen-(2)al-(1), und als 3-Alkoxy-2-methyl-propen- (2)-al- (l) ein entsprechendes niederaliphatisches Alkylderivat, beispielsweise 3-Athoxy-oder 3-Isopropoxy-2-methyl- propen- (2)-al- (1). Die Kondensation in flüssigem Ammoniak kann man unter erhöhtem Druck bei Raumtemperatur oder unter normalem Druck bei der Siedetemperatur des Ammoniaks ausführen.
Man kondensiert den ss-C-Aldehyd vorzugsweise mit Lithiumacetylid, das man vorgängig der Reaktion im gleichen Gefäss und im gleichen Ammoniak, das zur Kondensation verwendet wird, aus Lithiummethyl und Acetylen herstellen kann. Der ss-Cl9-Aldehyd kann in einem inerten Lösungsmittel, wie beispielsweise Ather, zugeführt werden. Die Hydrolyse des Kondensationsproduktes gelingt z. B. in flüssigem Ammoniak durch Zufügen eines Ammoniumsalzes, oder nach Entfernen des Ammoniaks wird durch Behandeln mit Säure hydrolysiert. Das -C2i-Acetylen- carbinol ist ein dickflüssiges Ol mit U.
V.-Absorptionsmaxima bei 280-281 und 291 m, in Petrol ätherlösung. Es zeigt bei der Zerewitinoff-Bestimmung in der Kälte 1 Mol und in der Wärme 2 Mol aktive Wasserstoffatome an. Die Kondensation des p-C2l- Acetylencarbinols mit dem 3-Acyloxy-bzw. 3-Alkoxy 2-methyl-propen- (2)-al- (l) erfolgt durch eine metallorganische Reaktion. Man lässt z. B. auf das ss-C2l- Acetylencarbinol in einem inerten Lösungsmittel 2 Mol Alkylmagnesiumhalogenid oder 2 Mol Phenyllithium einwirken. Das erste Mol wird von der Hydroxylgruppe gebunden, während das zweite Mol mit der Acetylenbindung reagiert und das endständige Kohlenstoffatom reaktionsfähig macht. Die gebildete Dimagnesiumhalogenid-Verbindung bzw.
Dilithium Verbindung setzt man dann im gleichen Lösungsmittel mit dem 3-Acyloxy-bzw. 3-Alkoxy-2-methyl-pro- pen- (2)-al- (l) um. Vorzugsweise behandelt man das , B-C2l-Acetylencarbinol in einem Lösungsmittel, wie Äther, mit 2 Mol Alkylmagnesiumhalogenid und kondensiert die gebildete Dimagnesiumhalogenid-Verbindung ohne Isolierung und Reinigung mit 1 Mol 3-Acyloxy-bzw. 3-Alkoxy-2-methyl-propen- (2)-al- (l).
Das Kondensationsprodukt wird am besten ohne Reinigung in üblicher Weise hydrolysiert, beispielsweise durch Eingiessen in ein Gemisch von Eis und verdünnter Essigsäure oder verdünnter Ammoniumchlo- ridlösung, wobei das entsprechende Acetylendiol als zähes 0l erhalten wird, das durch U. V.-Absorptionsspektrum und Zerewitinoff-Bestimmung charakterisiert wird. Es wird am besten ohne Reinigung direkt weiter umgesetzt.
In einer weiteren Ausführungsform der ersten Stufe des Verfahrens wird zuerst ein 3-Alkoxy-2-me thyl-propen- (2)-al- (1) in flüssigem Ammoniak mit einem Alkali-bzw. Erdalkaliacetylid kondensiert und das gebildete l-Alkoxy-2-methyl-3-hydroxy-penten- (l)-in- (4) wie bereits für das,-C2l-Acetylencarbinol beschrieben durch eine metallorganische Reaktion mit / ?-C-Aldehyd umgesetzt.
In der zweiten Stufe wird das erhaltene Acetylendiol wie gesagt einer Säurebehandlung unterworfen, wobei unter doppelter Allylumlagerung und gleichzeitiger Wasser-und Säure-bzw. Alkoholabspaltung 4, 5-Dehydro--apo-4-carotinal entsteht. Die Reaktion wird zweckmässig in einem inerten Lösungsmittel, z. B. Aceton, Alkohol oder Ather, in Gegenwart einer wässerigen oder wasserfreien Säure, z. B. Essigsäure, p-Toluolsulfonsäure, Salzsäure usw., durchgef hrt.
Das 4, 5-Dehydro--apo-4-carotinal ist eine kristalline Verbindung, die im U. V.-Spektrum ein charakteristisches Absorptionsmaximum bei 402-404 mÁ in Petroläther aufweist.
Das erfindungsgemäss erhältliche 4, 5-Dehydro ¯-apo-4-carotinal stellt einen wertvollen, gelbfärben- den Farbstoff dar und kann als Lebensmittelfarbstoff verwendet werden. Ferner ist er ein wichtiges Zwischenprodukt zur Synthese anderer wertvoller Carotinoidpigmente.
Beispiel 1
Man leitet in die Lösung von 1, 9 g Lithium in 900 ml flüssigem Ammoniak bis zur vollständigen Umsetzung trockenes, acetonfreies Acetylen ein. Dann gibt man unter energischem Rühren innert 20 Minuten eine Lösung von 61, 2 g 8- [2', 6', 6'Trimethyl- cyclohexen- (1')-yl]-2, 6-dimethyl-octatrien-(2, 4, 6)-al (1) in 200 ml trockenem Ather zu und rührt die Reaktionsmischung unter Feuchtigkeitsausschluss intensiv während 20 Stunden. Darauf gibt man in kleinen Portionen 35 g Ammoniumchlorid zu und lässt das Ammoniak verdampfen. Nach Zugabe von 260 ml Wasser wird die Atherschicht abgetrennt, mit Wasser gewaschen, mit Natriumsulfat getrocknet und im Vakuum eingeengt.
Das zurückbleibende rötliche 61 wird im Vakuum scharf getrocknet. Man erhält 66 g
10- [2', 6', 6'- Trimethyl-cyclohexen- (1')-yl]-4, 8-dime thyl-decatrien- (4, 6, 8)-in- (1)-0l- (3) ; U. V.-Absorptionsmaxima bei 280, 5 und 291 mli. (in Petroläther). Dieses wird in 200 ml absolutem Ather gelöst und zu einer aus 12, 3 g Magnesium und 67 g Athylbromid in 200 ml absolutem Ather hergestellten Athylmagne siumbromidlösung unter Rühren bei 15-20 C all mählich zugegeben.
Anschliessend erhitzt man in einer
Stickstoffatmosphäre 1 Stunde unter Rückfluss, kühlt mit Eiswasser ab, verdünnt mit 200 ml trockenem Methylenchlorid und lässt eine Lösung von 42 g 3 Benzoyloxy-2-methyl-propen- (2)-al- (l) in 200 ml Methylenchlorid rasch zuflie¯en. Man rührt weitere 2
Stunden unter Eiskühlung, giesst das Reaktionsgemisch auf eiskalte verdünnte Essigsäure, trennt die organische Schicht ab, wäscht sie mit Wasser, trocknet mit Natriumsulfat und dampft das Lösungsmittel im Vakuum bei 30 C ab. Man erhält 108 g rohes
13- [2', 6', 6'-Trimethyl-cyclohexen-(l')-yl]-2, 7, 11-tri methyl-1-benzoyloxy-tridecatetraen- (1, 7, 9, 1 l)-in- (4)- diol- (3, 6) als gelbes, zähes 01, das ohne Reinigung direkt weiter umgesetzt wird.
Zu diesem Zweck wird es in 700 ml Isopropanol gelöst, mit 5 ml Eisessig versetzt und 3 Stunden in einer Stickstoffatmosphäre gekocht. Nach dem Verdünnen mit Wasser wird das
Reaktionsprodukt mit Ather extrahiert, mit verdünn- ter Natriumbicarbonatlösung gewaschen und mit Na triumsulfat getrocknet. Der Ather wird im Vakuum abgedampft, und man erhält 80 g rohes 13-[2', 6', 6' Trimethyl-cyclohexen-(l')-yll-2, 7, 11-trimethyl-trideca- pentaen- (2, 6, 8, 10, 12)-in- (4)-al- (l), das durch Chromatographie, L¯sungsmittelverteilung oder Kristallisation gereinigt werden kann.
Das Produkt bildet aus Petroläther (Siedebereich 80-100¯ C) oder aus Athanol gelbe Blättchen (Schmelzpunkt 82-83 C) und weist im U. V.-Spektrum ein Absorptionsmaximum bei 402-404 mu (E"/-= 1790) in Petroläther- lösung auf.
Beispiel 2
Man gibt zu einer Lösung von Lithiumacetylid in flüssigem Ammoniak (hergestellt nach den Angaben des Beispiels 1 aus 1, 5 g Lithium) eine Lösung von 25 g 3-Isopropoxy-2-methyl-propen- (2)-al- (l) in 50 ml absolutem Ather und rührt das Gemisch energisch während 18 Stunden bei der Siedetemperatur des Ammoniaks. Man fügt portionenweise 35 g Ammoniumchlorid zu und lässt das Ammoniak verdampfen. Nach Zugabe von 200 ml Wasser wird mit Ather extrahiert. Man wäscht die ätherische Lösung mit Wasser, trocknet mit Natriumsulfat und dampft im Vakuum ein. Man erhält 29 g 1-Isopropoxy-2-methyl 3-hydroxy-penten- (l)-in- (4) als gelbliches 01, das ohne Reinigung weiter umgesetzt wird.
Die Zerewiti- noff-Bestimmung zeigt in der Kälte 0, 96 Mol und in der Wärme 1, 91 Mol aktive Wasserstoffatome an.
Das Produkt wird in 200 ml absolutem Ather gelöst und zu einer Athylmagnesiumbromidlösung- (hergestellt aus 9, 5 g Magnesium und 50 g Athylbromid in 200 ml absolutem Ather) unter Rühren bei 15 bis 20 C allmählich zugegeben. Anschliessend erhitzt man in Stickstoffatmosphäre 1 Stunde unter R ckfluss, kühlt mit Eiswasser ab, gibt eine Lösung von 51 g 8-[2', 6', 6'-Trimethyl-cyclohexen-(l')-yl]-2, 6-di methyl-octatrien- (2, 4, 6)-al- (1) in 400 ml absolutem Ather zu und rührt 2 Stunden in Stickstoffatmosphäre.
Man giesst die Reaktionslösung auf eiskalte Ammoniumchloridlösung, trennt die Atherschicht ab, wäscht mit gesättigter Ammoniumchloridlösung und trocknet mit Natriumsulfat. Die so erhaltene Lösung des 13 [2', 6', 6'-Trimethyl-cyclohexen- (1')-yl]-2, 7, 11-trime thyl-1-isopropoxy-tridecatetraen- (1, 7, 9, 11)-in- (4)-diol- (3, 6) wird mit 160 ml 12 /oiger alkoholischer Salzsäure versetzt und 16 Stunden bei Raumtemperatur stehengelassen. Dann wird die Lösung mit Wasser und mit 5 /o iger Natriumbikarbonatlösung gewaschen, mit Natriumsulfat getrocknet und im Vakuum abgedampft. Man erhält 62 g Rohprodukt, das durch Chromatographie, L¯sungsmittelverteilung oder Kri stallisation gereinigt werden kann.
Es bildet aus Alkohol gelbe Blättchen (Schmelzpunkt 82-83 C) und hat ein U. V.-Absorptionsmaximum bei 402-404 m, u (E 1% = 1770) in Petroläther.
Beispiel 3
40 g 10- [2', 6', 6'-Trimethyl-cyclohexen-(l')-yl]- 4, 8-dimethyl-decatrien- (4, 6, 8)-in- (l)-ol- (3) werden, wie im Beispiel 1 angegeben, mit einer Athylmagnesiumbromidlösung umgesetzt. Zur abgekühlten Gri gnardlösung gibt man dann eine Lösung von 18 g 3-Isopropoxy-2-methyl-propen- (2)-al- (1) in 10 ml absolutem Ather und rührt in Stickstoffatmosphäre 2 Stunden bei 05O C. Hierauf giesst man auf kalte gesättigte Ammoniumchloridlösung und gewinnt das Reaktionsprodukt auf übliche Weise.
Man erhält 62 g rohes 13- [2', 6', 6'-Trimethyl- cyclohexen- (1')-yl]-2, 7, 11-trimethyl-1-isopropoxytridecatetraen- (1, 7, 9, 11)-in- (4)-diol- (3, 6), das in 500 ml Aceton gelöst und nach Zugabe einer Lösung von 6 g p-Toluolsulfonsäure in 60 ml Wasser 11/2 Stunden in StickstofEatmosphäre gekocht wird. Das entstandene 13-[2', 6', 6'-Trimethyl-cyclohexen-(l')-yl]- 2, 7, 11-trimethyl-tridecapentaen-(2, 6, 8, 10,12)-in-(4)al-(1) wird wie in den vorangehenden Beispielen ge wonnen und gereinigt. Schmelzpunkt 82-83 C ; U. V.-Absorptionsmaximum bei 402-404 m, in Pe troläther.

The free methylmalondialdehyde is obtained in aqueous solution by acid hydrolysis and is etherified by azeotropic distillation with benzene and isopropyl alcohol in the presence of p-toluenesulfonic acid as a catalyst. 3-Athoxy-2-methyl-propen- (2) -al- (1) (boiling point: 78-80¯C / 13 mm; n2r4 = 1.4755) can be obtained analogously.
3-acyloxy-2-methyl-propen- (2) -al- (1)
Neutralizing an aqueous solution of the free methylmalonaldehyde with 1 equivalent of sodium hydroxide solution and concentrating it gives the sodium salt as a white powder. The 4,5-dehydro-apo-4-carotinal is a crystalline compound which has a characteristic absorption maximum in the U.V. spectrum at 402-404 mA in petroleum ether.
The 4,5-dehydro-apo-4-carotinal which can be obtained according to the invention is a valuable, yellow coloring material and can be used as a food coloring material. It is also an important intermediate in the synthesis of other valuable carotenoid pigments.

In the solution of 1.9 g of lithium in 900 ml of liquid ammonia, dry, acetone-free acetylene is introduced until the reaction is complete. Example 2

The resulting solution of 13 [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') - yl] -2, 7, 11-trimethyl-1-isopropoxy-tridecatetraen- (1, 7, 9, 11) -in- (4) -diol- (3, 6) is mixed with 160 ml of 12% alcoholic hydrochloric acid and left to stand for 16 hours at room temperature. The solution is then washed with water and with 5% sodium bicarbonate solution, dried with sodium sulfate and evaporated in vacuo. 62 g of crude product are obtained which can be purified by chromatography, solvent distribution or crystallization.
It forms yellow leaflets from alcohol (melting point 82-83 C) and has an U.V. absorption maximum at 402-404 m, u (E 1% = 1770) in petroleum ether.
Example 3
40 g of 10- [2 ', 6', 6'-trimethyl-cyclohexen- (l ') - yl] - 4, 8-dimethyl-decatrien- (4, 6, 8) -in- (l) -ol- (3) are, as indicated in Example 1, reacted with an ethylmagnesium bromide solution. A solution of 18 g of 3-isopropoxy-2-methyl-propen- (2) -al- (1) in 10 ml of absolute ether is then added to the cooled Gri gnard solution and the mixture is stirred in a nitrogen atmosphere for 2 hours at 05O C. The mixture is then poured on cold saturated ammonium chloride solution and recover the reaction product in the usual way.
62 g of crude 13- [2 ', 6', 6'-trimethylcyclohexen- (1 ') - yl] -2, 7, 11-trimethyl-1-isopropoxytridecatetraene (1, 7, 9, 11) are obtained -in- (4) -diol- (3, 6), which is dissolved in 500 ml of acetone and, after adding a solution of 6 g of p-toluenesulfonic acid in 60 ml of water, is boiled for 11/2 hours in a nitrogen atmosphere. The resulting 13- [2 ', 6', 6'-trimethyl-cyclohexen- (l ') - yl] - 2, 7, 11-trimethyl-tridecapentaen- (2, 6, 8, 10,12) -in- (4) al- (1) is obtained and purified as in the previous examples. Melting point 82-83 C; U.V. absorption maximum at 402-404 m, in petroleum ether.
