Verfahren zur Herstellung eines Dialdehydes Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines Dialdehydes, nämlich des be kannten 2,7-Dimethyl-octadien-(2,6)-in-(4)-dials- (1,8). Das neue erfindungsgemässe Verfahren besteht darin, dass man ein Acetylendimagnesiumhalogenid beidseitig mit einem 3-Alkoxy- bzw.
3-Acyloxy-2- methyl-propen-(2)-al-(1) kondensiert, hydrolysiert und das erhaltene Acetylendiol mit Säure behandelt. Das neue Verfahren kann ohne Isolierung von Zwischen produkten ausgeführt werden und ergibt gute Aus beuten an reinem Endprodukt, wobei die technische Durchführung sehr einfach ist.
Die benötigten Ausgangsmaterialien werden wie folgt hergestellt: 3-Alkoxy-2-methyl-propen-(2)-al-(1) Man kondensiert Orthoameisensäureäthylester mit Propenyl-äthyläther bei Gegenwart von Bortrifluorid- ätherat zu 1,1,3,3-Tetraäthoxy-2-methyl-propan (Sdp. 93 110 mm, nD = 1,4132).
Durch saure Hydrolyse mittels wässriger Schwefelsäure gewinnt man den freien Methylmalondialdehyd in wässriger Lösung und veräthert ihn durch azeotrope Destillation mit Benzol und Isopropylalkohol bei Gegenwart von p-Toluol- sulfonsäure als Katalysator. 3-Iospropoxy-2-methyl- propen-(2)-al-(1) siedet bei 83 /13 mm, nD = 1,4745.
Analog kann man 3-Äthoxy-2-methyl-propen-(2)- al-(1), Sdp. 78-80 113 mm, nD = 1,4755 gewinnen. 3-Acyloxy-2-methyl-propen-(2)-al-(1) Durch Neutralisieren einer wässrigen Lösung des freien Methylmalondialdehyds mit einem Äquivalent Natronlauge und Einengen erhält man das Natrium salz als weisses Pulver. Dieses wird in einem inerten Lösungsmittel, z.
B. Methylenchlorid, suspendiert und mit einem Säurechlorid in der Wärme umgesetzt. Die vom gebildeten NaCl filtrierte Lösung wird im Va kuum eingeengt und der Rückstand durch Destilla tion oder Kristallisation gereinigt.
Man erhält auf diese Weise z. B. farblose Nadeln des 3-Benzoyloxy-2-methyl-propen-(2)-al-(1), Smp. 79-80 , oder 3-Acetoxy-2-methyl-propen-(2)-al-(1), welches bei 75-77 /10 mm siedet und bei tiefer Tem peratur fest wird.
Die Durchführung des erfindungsgemässen Ver fahrens erfolgt in zwei Stufen; das Zwischenprodukt braucht indessen weder isoliert noch gereinigt zu wer den. Zweckmässigerweise stellt man zunächst eine Suspension eines Acetylendimagnesiumhalogenids her, indem man gereinigtes Acetylen etwa 12 Stunden durch eine Lösung (vorzugsweise in Äther) eines Alkyhnagnesiumhalogenids, z. B. Äthylmagnesium- bromid oder Hexyhnagnesiumbromid, leitet.
Man sorgt durch energisches Rühren, dass das in dem Lösungsmittel unlösliche Acetylendimagnesiumbromid fein suspendiert bleibt und lässt dann zur Durchfüh rung des erfindungsgemässen Verfahrens zweckmässig zwei Äquivalente eines 3-Alkoxy- bzw. 3-Acyloxy-2- methylpropen-(2)-als-(1) rasch zutropfen, worauf man anschliessend noch einige Zeit zum Kochen erwärmen kann.
Die Hydrolyse des gebildeten Kondensations produktes erfolgt vorzugsweise mit wässrigen Lösun gen von Ammoniumchlorid oder Ammoniumacetat. Die entstandenen Acetylendiole werden in der Regel als Öle erhalten und durch Zerewitinoffbestimmung charakterisiert.
Sie können ohne weitere Reinigung der Behandlung mit einer Säure, vorzugsweise einer starken, wässrigen Mineralsäure oder Sulfonsäure, wie Schwefelsäure, Phosphorsäure, p-Toluolsulfonsäure, oder einer starken organischen Säure, wie Essigsäure, Oxalsäure, unterworfen werden. Zu diesem Zweck kann man z.
B. bei Verwendung eines 3-Alkoxy- methylpropenals als Ausgangsmaterial eine ätherische Lösung des gebildeten Acetylendiols bei Raumtem peratur mit wässriger Schwefelsäure während etwa 10-15 Stunden schütteln. Bei Verwendung eines 3-Acyloxy-methylpropenals als Ausgangsmaterial empfiehlt es sich, das gebildete Acetylendiol mit Schwefelsäure 1-5 Stunden in Acetonlösung zu kochen.
Durch diese Säurebehandlung erfolgt Allyl- umlagerung und Verseifung der Alkoxy- bzw. Acyl- oxygruppen, wobei 2,7-Dimethyl-octadien-(2,6)-in- (4)-dial-(1,8) entsteht.
Gemäss einer andern Aus führungsform der Erfindung kann man das gebildete Kondensationsprodukt direkt mit Säure behandeln, ohne zuerst mittels Ammoniumsalzen zu hydroly- sieren. Auf diese Weise erfolgen die Hydrolyse der Metallsalze, die Allylumlagerung und die Verseifung vorhandener Alkoxy- bzw. Acyloxygruppen gleich zeitig unter Bildung des 2,7-Dimethyl-octadien-(2,6)- in-(4)-dials-(1,8).
Die Isolierung des Endproduktes aus dem Reak tionsgemisch kann durch Kristallisation, Destillation oder Sublimation der nach Entfernen des Lösungs mittels erhaltenen Rohprodukte erfolgen.
<I>Beispiel 1</I> Durch eine Lösung von Äthyhnagnesiumbromid in Äther (hergestellt aus 25 Gewichtsteilen Magnesium, 145 Gewichtsteilen Äthylbromid und 500 Raumteilen absolutem Äther) leitet man während 20 Stunden gereinigtes Acetylen bei 0 . Dann lässt man unter raschem Rühren eine Lösung von 64 Gewichtsteilen 3-Isopropoxy-2-methyl-propen-(2)-al-(1) in 50 Raum teilen absolutem Äther innert 5 Minuten zulaufen.
Anschliessend kocht man das Reaktionsgemisch wäh rend 1 Stunde unter Rückfluss, kühlt dann ab und hydrolysiert mit wässriger Ammoniumacetatlösung. Die Ätherschicht wird abgetrennt, einmal mit Na- triumbicarbonatlösung, dann mit Wasser gewaschen, mit Natriumsulfat getrocknet, filtriert und im Vakuum eingeengt. Man erhält etwa 70 Gewichtsteile 1,8- Di-isopropoxy - 3,6 - dihydroxy-2,7-dimethyl-octadien- (1,7)-in-(4) als zähes öl.
Die Zerewitinoffbestimmung zeigt zwei aktive Wasserstoffatome. Das Produkt wird in 200 Raumteilen gewöhnlichem Äther gelöst, eine Spur Hydrochinon und dann 50 Raumteile 3n Schwefelsäure zugegeben und unter Kohlendioxyd während 12 Stunden bei Raumtemperatur geschüttelt. Dann wird die Ätherschicht abgetrennt, neutral ge waschen, getrocknet und eingeengt. Der Rückstand, etwa 40 Gewichtsteile, kristallisiert beim Kühlen.
Auf Grund des Absorptionsspektrums besteht er zu mindestens 50% aus dem gewünschten 2,7-Dimethyl- octadien-(2,6)-in-(4)-dial-(1,8). Die Isolierung erfolgt durch Umkristallisieren aus Petroläther (Siedebereich 80-100 ), durch Sublimation im Hochvakuum bei 50 oder durch Destillation im Hochvakuum, Sdp. 95 bis 97 /0,05 mm. Smp. 68 , Absorptionsspektrum: Maxima 310 und 330 m,u., Ell% 2054 und 2180 (in Petroläther).
Verwendet man anstelle von 3 - Isopropoxy - 2 - methyl-propen-(2)-al-(1) ein 3-Acyloxy-2-methyl-pro- pen-(2)-al-(1), so wird die Hydrolyse mit Schwefel säure bei erhöhter Temperatur z. B. in kochendem Aceton durchgeführt.
<I>Beispiel 2</I> Zu der in gleicher Weise wie in Beispiel 1 herge stellten Suspension von Acetylendimagnesiumbromid in Äther gibt man eine Lösung von 170 Gewichts teilen 3 - Benzoyloxy- 2 -methyl-propen - (2)- a1 -(1) in 750 Raumteilen Methylenchlorid und verfährt dann gleich wie in Beispiel 1.
Das rohe 1,8-Dibenzoyloxy- 3,6 -dihydroxy - 2,7 - dimethyl-octadien- (1,7) -in- (4) (2aktive Wasserstoffatome) wird in 350 Raumteilen Aceton und 20 Raumteilen Schwefelsäure (20%ig) in einer Stickstoffatmosphäre während 2 Stunden unter Rückfluss gekocht. Man giesst das Reaktionsgemisch auf Wasser, neutralisiert mit Soda und isoliert das gebildete 2,7-Dimethyl-octadien-(2,6)-in-(4)-dial-(1,8) in gleicher Weise wie in Beispiel 1.
Process for the preparation of a dialdehyde The present invention relates to a process for the preparation of a dialdehyde, namely the known 2,7-dimethyl-octadiene- (2,6) -in- (4) -dials- (1,8). The new process according to the invention consists in that an acetylenedimagnesium halide is treated on both sides with a 3-alkoxy or
3-Acyloxy-2-methyl-propen- (2) -al- (1) condensed, hydrolyzed and the acetylenediol obtained treated with acid. The new process can be carried out without isolating intermediate products and yields good yields of pure end product, with the technical implementation being very simple.
The required starting materials are prepared as follows: 3-alkoxy-2-methyl-propen- (2) -al- (1) Ethyl orthoformate is condensed with propenyl ethyl ether in the presence of boron trifluoride etherate to give 1,1,3,3-tetraethoxy -2-methyl-propane (bp 93 110 mm, nD = 1.4132).
The free methylmalondialdehyde is obtained in aqueous solution by acid hydrolysis using aqueous sulfuric acid and is etherified by azeotropic distillation with benzene and isopropyl alcohol in the presence of p-toluenesulfonic acid as a catalyst. 3-Iospropoxy-2-methyl-propen- (2) -al- (1) boils at 83/13 mm, nD = 1.4745.
3-Ethoxy-2-methyl-propen- (2) -al- (1), bp 78-80 113 mm, nD = 1.4755 can be obtained analogously. 3-Acyloxy-2-methyl-propen- (2) -al- (1) By neutralizing an aqueous solution of the free methylmalondialdehyde with one equivalent of sodium hydroxide solution and concentrating, the sodium salt is obtained as a white powder. This is in an inert solvent, e.g.
B. methylene chloride, suspended and reacted with an acid chloride in the heat. The solution filtered from the NaCl formed is concentrated in vacuo and the residue is purified by distillation or crystallization.
In this way, for. B. colorless needles of 3-benzoyloxy-2-methyl-propen- (2) -al- (1), m.p. 79-80, or 3-acetoxy-2-methyl-propen- (2) -al- (1 ), which boils at 75-77 / 10 mm and solidifies at low temperatures.
The process according to the invention is carried out in two stages; the intermediate product, however, does not need to be isolated or purified. It is expedient to first prepare a suspension of an acetylenedimagnesium halide by passing purified acetylene for about 12 hours through a solution (preferably in ether) of an alkyl magnesium halide, e.g. B. Ethylmagnesiumbromid or Hexyhnagnesiumbromid, conducts.
It is ensured by vigorous stirring that the acetylenedimagnesium bromide, which is insoluble in the solvent, remains finely suspended and two equivalents of a 3-alkoxy or 3-acyloxy-2-methylpropene (2) -als- ( 1) drip in quickly, after which you can then heat to boil for some time.
The hydrolysis of the condensation product formed is preferably carried out with aqueous solutions of ammonium chloride or ammonium acetate. The acetylenediols formed are usually obtained as oils and characterized by determination of Zerewitinoff.
They can be subjected to treatment with an acid, preferably a strong, aqueous mineral acid or sulfonic acid, such as sulfuric acid, phosphoric acid, p-toluenesulfonic acid, or a strong organic acid, such as acetic acid or oxalic acid, without further purification. For this purpose you can z.
B. when using a 3-alkoxymethylpropenal as the starting material, shake an ethereal solution of the acetylenediol formed at room temperature with aqueous sulfuric acid for about 10-15 hours. When using a 3-acyloxymethylpropenal as the starting material, it is advisable to boil the acetylenediol formed with sulfuric acid in acetone solution for 1-5 hours.
This acid treatment results in allyl rearrangement and saponification of the alkoxy or acyloxy groups, with 2,7-dimethyl-octadiene- (2,6) -yne- (4) -dial- (1,8) being formed.
According to another embodiment of the invention, the condensation product formed can be treated directly with acid without first hydrolyzing using ammonium salts. In this way, the hydrolysis of the metal salts, the allyl rearrangement and the saponification of existing alkoxy or acyloxy groups take place simultaneously with the formation of 2,7-dimethyl-octadiene- (2,6) -in (4) -dials- (1, 8th).
The end product can be isolated from the reaction mixture by crystallization, distillation or sublimation of the crude products obtained after removal of the solution.
<I> Example 1 </I> Through a solution of ethyl magnesium bromide in ether (made from 25 parts by weight of magnesium, 145 parts by weight of ethyl bromide and 500 parts by volume of absolute ether), purified acetylene is passed at 0 for 20 hours. Then a solution of 64 parts by weight of 3-isopropoxy-2-methyl-propen (2) -al- (1) in 50 parts by volume of absolute ether is allowed to run in within 5 minutes with rapid stirring.
The reaction mixture is then refluxed for 1 hour, then cooled and hydrolyzed with aqueous ammonium acetate solution. The ether layer is separated off, washed once with sodium bicarbonate solution, then with water, dried with sodium sulfate, filtered and concentrated in vacuo. About 70 parts by weight of 1,8-di-isopropoxy-3,6-dihydroxy-2,7-dimethyl-octadiene- (1,7) -in- (4) are obtained as a viscous oil.
The Zerewitinoff determination shows two active hydrogen atoms. The product is dissolved in 200 parts by volume of ordinary ether, a trace of hydroquinone and then 50 parts by volume of 3N sulfuric acid are added and the mixture is shaken under carbon dioxide for 12 hours at room temperature. Then the ether layer is separated, washed neutral ge, dried and concentrated. The residue, about 40 parts by weight, crystallizes on cooling.
Based on the absorption spectrum, it consists of at least 50% of the desired 2,7-dimethyl-octadiene- (2,6) -in- (4) -dial- (1,8). The isolation takes place by recrystallization from petroleum ether (boiling range 80-100), by sublimation in a high vacuum at 50 or by distillation in a high vacuum, bp 95 to 97 / 0.05 mm. Mp. 68, absorption spectrum: maxima 310 and 330 m, u., Ell% 2054 and 2180 (in petroleum ether).
If a 3-acyloxy-2-methyl-propene- (2) -al- (1) is used instead of 3-isopropoxy-2-methyl-propen- (2) -al- (1), the hydrolysis takes place with sulfuric acid at elevated temperature z. B. carried out in boiling acetone.
<I> Example 2 </I> To the suspension of acetylenedimagnesium bromide in ether prepared in the same way as in Example 1 is added a solution of 170 parts by weight of 3-benzoyloxy-2-methyl-propene - (2) - a1 - ( 1) in 750 parts by volume of methylene chloride and then proceed in the same way as in Example 1.
The crude 1,8-dibenzoyloxy-3,6-dihydroxy-2,7-dimethyl-octadiene- (1,7) -in- (4) (2active hydrogen atoms) is dissolved in 350 parts by volume of acetone and 20 parts by volume of sulfuric acid (20% strength ) refluxed in a nitrogen atmosphere for 2 hours. The reaction mixture is poured into water, neutralized with soda and the 2,7-dimethyl-octadiene- (2,6) -in- (4) -dial- (1,8) formed is isolated in the same way as in Example 1.
