worin R, Wasserstoff oder einen Methylrest, R2 und R3 niedere Alkylreste bedeuten, die sich von den bekannten Verbindungen durch.die Methylgruppe in 5-Stellung unter scheiden, tiefen und langdauernden Schlaf bewirken.
Gemäss vorliegender Erfindung werden die Verbindungen der angeführten allgemei nen Formel dadurch gewonnen, dass man ein 2,4-Dioxo-3,3-dialkyl-piperidin bzw. sein N- Methyl-Derivat durch Umsetzung mit einem Ameisensäureester in Gegenwart eines Alkali metalles in das 2,4-Dioxo-3,3-Dialkyl-5-oxy- methylen-piperidin bzw. sein N-hlethyl-Deri- vat überführt und dieses zum 2,4-Dioxo-3,3- dialkyl-5-methyl-piperidin bzw.
zu seinem N-hIethyl-Derivat reduziert. Dieselben Ver bindungen können auch dadurch gewonnen werden, dass die Oxymethylen-Verbindungen durch Umsetzung mit Ammoniak, Alkyl- oder Dialkylaminen in die Aminomethylen-,-Alkyl- oder Dialkylamino-methylen-Verbindungen übergeführt und diese der Reduktion unter worfen werden.
Die neuen Verbindungen sind farblos, fest oder flüssig, sie lösen sich leicht in Wasser und in den gebräuchlichen organischen Lö-. sungsmitteln und können unter vermindertem Druck unzersetzt destilliert werden. Sie sollen als Arzneimittel Verwendung finden.
Gegenstand des Patentes ist nun ein Ver fahren zur Herstellung von 2,4-Dioxo-3,3-di- äthyl-5-methyl-piperidin, das dadurch gekenn zeichnet ist, dass man 2,4-Dioxo-3,3-diäthyl- piperidin mit einem Ameisensäureester in Gegenwart eines Alkalimetalles in das 2,4 Dioxo-diäthyl-5-oxymethylen-piperidin über führt und dieses zum 2,4-Dioxo-3,3-diäthyl-5- methyl-piperidin reduziert.
<I>Beispiel:</I> In 100 Raumteilen absolutem Benzol wer den 24 Gewichtsteile Natriumpulver suspen diert und unter Rühren mit einer friseh her gestellten Lösung von 150 Gewichtsteilen Ameisensäuremethylester und 165 Gewichts teilen 2,4-Dioxo-3,3-diäthyl-piperidin in 900 Raumteilen absolutem Benzol versetzt. Die Temperatur wird durch Wasserkühlung zwi schen 25 und 28 C gehalten. Man rührt wäh rend 12 Stunden. Dann wird das Reaktions gemisch unter Kühlung mit 200 Raumteilen 0,6 n-1\Tatronlauge versetzt.
Die wässerige Schicht wird abgetrennt und mit 351/o Salz säure congosauer gestellt. Das 2,4-Dioxo-3,3- diäthyl-5-oxymethylen-piperidin fällt in guter Ausbeute fest aus. Nach Umlösen aus Chloro- form-Petroläther schmilzt es bei 140-141 C.
5 Gewichtsteile 2,4-Dioxo-3,3-diäthyl-5-oxy- methylen-piperidin werden mit etwa 2 Ge wichtsteilen alkoholfeuchtem Raney-Nickel in 25 Raumteilen Methanol bei 100 atü und 120 C mit Wasserstoff hydriert. Nach Auf nahme von 21M1 Wasserstoff wird die Hydrie rung unterbrochen, die Lösung vom Kataly sator getrennt und eingeengt und das Roh produkt im Vakuum destilliert. Das bei 74 bis 75 C schmelzende 2,4-Dioxo-3,3-diäthyl- 5-methyl-piperidin siedet unter 16 mm Druck zwischen 178-185 C.
