Verfahren zur HersteRung eines Antibiotikums Die vorliegende Erfindung betrifft die Herstellung <B>C</B> eines neuen Antibiotikums.
Bekanntlich besitzen Antibiotika grosse Büdeu- tung. Während sie früher im wesentlichen für thera peutische Zwecke verwendet wurden, benützt man sie heute mit grossem Erfolg auch auf andem Gebieten.
Es wurde gefunden, dassein Strahlenpilz (Aktino- mycete), welcher in einem geeigneten Nährmedium, gezüchtet wird, eine antibiotische Substanz zu liefern vermag, welche sich mittelsgeeigneter Methoden aus dem Züchtungsinedium extrahieren, lässt. Gegenstand der Erfindung ist nun ein Verfahren zur Herstellung eines neuen Antibiotikums, das dadurch gekenn zeichnet ist, dass man Streptomyces Cavourensis, Stamm<B>829.8.52,</B> in Kontakt mit einem flüssigen Medium züchtet,
welches Quellten für assimilierbaren Kohlenstoff und Stickstoff sowie Mineralsalze enthält, und nach dem Fermentieren das Mycelium aus der Brühe entfernt, aus derselben mit einem organischen Lösungsmittel das gebildete Flavensomycin in roher Form extrahiert und aus demerhaltenen Auszug das reine Antibiotikum isoliert. Die neue aktive Substanz, welche beim Züchten dieses Strahlenpilzes gebildet wird, soll Flavenso- mycin genannt werden. Die physikalischen und che mischen Eigenschaften von Flavensomycin sind von denjenigen der bereits bekannten Antibiotika ver schieden.
Der neue Strahlenpilz der Gattung Strepto- myces wurde aus landwirtschaftlichem Boden in Ca- vour, Piemont, isoliert und gehört zur Gruppe der chromogenen Arten.
Er wurde in den Versuchslaboratorien der Anmel- derin gezüchtet. Dieser Strahlenpilz kann, zur Zeit auch aus dem Universitäts,Institut des, Nahrungs- mittelinspektorats in Mailand bezogen werden.
Das vegetative Mycelium wird durch lange, dünne, verzweigte oder gehäufte Hyphen gebildet; dIas. Lu ft- myc,elium wird durch lange, gerade und wellenför mige Hyphen, gebildet, welche sich nur langsam in lange Konidiophoren mit rundlichen, nicht buschartig angeordneten Konidien umwandeln. Spiralen wurden nicht beobachtet.

Streptomyces-Art kann in üblicher Weise gezüchtet werden unter Benützung, von stationären Kulturen oder Tauchkulturen, wobei sich die, letzteren am besten eignen. Das Kultunriedium muss Kohle hydrate, eine geeignete Stickstoffquelle und Mineral salze enthalten. Die Züchtun.g oder Fermentierung kann bei einer Temperatur zwischen 24 und<B>32,0</B> erfolgen,<B>je</B> nach Belüftung, Rührung und Zusammen setzung des Nährmediums; maximale Ausbeuten er zielt man praktisch vom vierten bis zum siebenten Fermentierungstag.
Während der Fermentierung steigt der pH-Wert auf<B>7,8-8,</B> doch soll er diese Höhe zweckmässig nicht überschreiten.
Nach der Züchtung der das Fl-avensomycin erzeu genden Streptomyces-Pilze enthält die zurückblei bende Züchtungshrühe zahlreiche Substanzen, wie Reste der Nährstoffe, aufgelöstes Mycelium sowie die aktive Substanz, welche,<B>je</B> nach der Art des Kultur mediums und nach den Fermentierungsbedingungen, eine Konzentration von<B>1600</B> Einheiten/cm3 und mehr erreichen kann.
Das Flavensomycin lässt sich aus der Züchtungs brühe mit verschiedenen mit Wasser nicht misch baren Lösungsmitteln bei einem pH zwischen<B>7,5</B> und<B>8,5</B> extrahieren, beispielsweise mit Äther, Chloro form, Butanol, Äthylacetat, Petroläther, Benzol usw. Der das rohe Antibiotikum enthaltende Auszug wird im Vakuum bei Temperaturen zwischen 20 und<B>3 00<I>C</I></B> eingeengt.<B>Die</B> Reinigung des z.
B. in konzentrierter benzolischer Lösung vorliegenden Antibiotikums (Röhextrakt) kann auf zwei verschiedene Arten erfol gen: <B>1.</B> durch Chroniotographieren des Rohextraktes auf einer Aluminiumoxyd-Säule, wobei man in benzo- lischer Lösung eingeht, sorgfältig mit Ben-zol zur Entfernung von fetti <B>'</B> -en, gelbroten Fraktionen, welche inaktiv sind, und dann mit Äthylawtat zur Entfer nung einer weiteren,
inaktiven gelben Fraktion wäscht und schliesslich das Antibiotikum mit absolutem Methariol eluiert.
2. durch Ausfällen des Rohextraktes mit Petrol- äth#er, Auflösen des Niederschlages in Aceton, Wieder ausfällen mit Petroläther, Lösen in Benzol und Chromotographieren auf Aluminiumoxyd, wobei die Säule mit Benzol und Äthylacetat gewaschen und das Antibiotikum mit absolutem Methanol eluiert wird.
Aus der alkoholischen Lösung erhält man das Flavensomycin durch Abdampfen des Lösungsmittels. Flavensomycin, ist ein kristallines, zitronengelbes Pulver.
<I>Beispiel</I> Der genannte Pilzstamm wird in einem Agar- Asparagin-Glycerol-Medium bei 2711 <B>C</B> gezüchtet. In diesem Medium wird ein sehr gutes Sporenwachstum erzielt. Zur Beimpfung des Herstellungsmediums kann man entweder direkt von Sporen ausgehen oder aber von einer 24 Stunden alten, bei<B>270 C</B> im glei chen Medium gezüchteten Streptomyces-Tauchkultur.
Mit Sporen oder einem solchen Inokulum. beimpft man Fermentierungskolben, welche 20<B>g</B> Maltose, <B>5 g</B> Pepton, <B>2,5 g</B> Getreideweiche und<B>1</B> Liter Lei- tunaswasser enthalten. Der pH-Wert wird mit Na- triumhydroxyd auf<B>7</B> eingestellt. Nach beendeter Fer- mentierung wird der pH-Wert auf<B>7,5-8,5</B> erhöht, wobei man bei -einer Temperatur von<B>27-280 C</B> arbeitet.
Nach Beendigung der Fermentieiung entfernt man das Mycellum und extrahiert das Antibiotikum aus der Brühe, 3mal mit Benzol im Verhältnis<B>3: 1,</B> wobei dafür gesorgt wird, dass der pH im Bereiche von<B>7,5-8,5</B> bleibt. Unter diesen Bedingungen löst sich Flavensomycin in Benzol. Das Volumen des Lösungsmittelauszugs wird durch Konzentrieren im Vakuum in einem Temperaturbereich von 20-401> <B>C</B> vermindert; bei dieser Operation werden auch die letzten Spuren von Wasser entfernt.
Die Reinigung erfolgt durch Chromotographieren des Konzentrates auf einer mit Benzol beschickten Al.o.-Säule in folgender Weise: Das Antibiotikum wird zusammen mit einigen gelbroten Fraktionen adsorbiert, während die fettigen Fraktionen und rotviolette Pigmente nicht adsorbiert werden. Die Säule wird mehrmals mit Benzol und dann mit Äthylacetat gewaschen, welches eine gelbe, inaktive Fraktion cluiert; schliesslich wird mit abso lutem Mathanol eluiert, wobei die aktive Fraktion (Havensomycin) herauskommt.
Einige rote, inaktive Fraktionen, welche mit Aceton und Wasser eluierbar sind, bleiben in, der Säule.
Eine weitere Menge an obgenannteni Konzentrat wird wie folgt gereinigt: Man fällt den benzolischen Auszug des Anti biotikums mit Petroläther aus, löst den Niederschlag (welcher den aktiven Teil darstellt) in Aceton und fällt wiederum mit Petroläther aus. Die fettigen, gelbrot pi-Mentierten Fraktionen, welche auf der <B>C</B> Aluminiumoxyd-Säule nicht adsorbiert werden und deshalb ein l#ängeres Waschen der Säule mit Benzol nötig machen, werden auf diese Weise fast vollständig entfernt. Der Niederschlag wird in.
Benzol gelöst und auf einer Aluminiumoxyd-Säule chromotographiert. Nach dem Waschen mit Benzol und Äthylacetat cluiert man die Säule mit absolutem Methanel, wobei die aktive Fraktion anfällt.
Aus der alkoholischen, Lösung gewinnt man Flavensomycin durch Abdampfen des Lösungsmittels in Form eines zitronengelben kristallinen Pulvers, wel ches genügend hygroskopisch undgeruchlos ist.
Das erfindungsgemäss, hergestellte neue Anti biotikum zeigt die folgenden chemischen -.und physi kalischen Eigenschaften: Die hellzitronengelbe. Farbe von Fl-avens,omycin verändert sich weder in alka lischem noch in saurem Medium. Das Produkt ent hält 2,11 11/o Stickstoff; es enthält keinen Schwefel und keine Halogene. Ein Ultraviol-ett-Absorption-s- spektrum zeigt nur bei<B>251</B> y ein Maximum.
Das In-frarot-Absorptionsspektrum zeigt Haupt- Infrarot-Absorptionsmaxima bei<B>2,90, 3,26,</B> 3,45, <B>5,86,</B> 5,94,<B>6,17, 6,51, 6,92, 8,02, 9,05,</B> 10,04, <B>10,32, 10,91, 13,02</B> und<B>13,13</B> y (siehe Zeichnung). Die ersten, sechs gezeigten Maxima entsprechen den folgenden funktionellen Gruppen: OH,<B>CH, CO,</B> konjugiertes<B>CO,</B> konjugie#,rte Doppelbindung und Phenyl; die Anwesenheit der letztgenannten Gruppe und von konjugiertem<B>CO</B> ist nicht sicher.
In der Zeichnung wird das Infrarot-Absorptionsspektruni von Flavensomycin in Kaliumbromid mit einer Ken- zentration von<B>3</B> 1/o (untere Kurve B) dargestellt im Vergleich zu derjenigen einer Kaliumbromidi-Scheibe (obere Kurve<B>A),</B> aufgenommen mit einem Beck- man,n-IR/2-Spektrophatometer.
Das Antibiotikum löst sich sehr gut in einigen Lösungsmitteln, wie Methanol, Äthanel, Propylen- glykol. und Pyridm; gute Löslichkeit besteht in einigen weitern Alkohol-en, wie n-Propatiol- n-Butanol und Amylalkohol; ferner ist das Antibiotikum löslich in Eisessig und seinem Methyl-, Äthy;
lr, Propyl- oder Butylester, sowie in Aceton, Chloroform, Dioxan, und, Benzol. Auch in Wasser löst sich das Antibiotikum. Unlöslich ist das Antibiotikum in Glycerol, Tetra- chlorkehlenstoff, Schwefelkohlenstoff, Schwefeläth#cr, Hexan, Petroläther und Paraffinöl. Eine wässe,-rige Lösung von Flaverisornycin erhält man z. B. durch Zufügen von Wasser zu einer konzentrierten Lösung des Antibiotikums, z.
B. zu einer äthanolischen Lö sung mit -einer Konzentration von 0,2 g/cm3. Die Lösung schäumt.
Havensomycin gibt Farbreaktionrn mit Milisch-, Fehling- und Ehrlich-Reageris; keine, Farbreaktionen erfolgen mit Seliwanoff-, Tollens-, Millon-, Lieber- mann- und Sakaguchi-R.eagens und mit Biuret. Mit FeCl <B>3</B> ergibt sich ein Niederschlag ohne Farbreaktion.
Flavensoraycin ist ziemlich beständig, wenn man ,es im Vakuum als trockenes Pulver oder als Lösung in organischen Lösungsmitteln lagert. In, wässeriger Lösung, insbesondere bei geringen Konzentrationen, neig gt es dazu, seine Aktivität teilweise zu verlieren, und es wurde festgesellt, dass das Antibiotikum bei pH <B>8-10</B> rascher inaktiviert wird als. bei pH 4-6.
Eine wässerige Lösung von<B>100</B> y g/cms bei neu tralem pH und<B>180 C</B> verliert innerhalb von,<B>3</B> Tagen 40'% ihrer Aktivität. Eine wässerio",e Lösung miteiner Konzentration von<B>1</B> mg/cm3 mit neutralem pH, welche bei 40<B>C</B> gelagert wird, ist nach<B>7</B> Tagen un verändert;
die Aktivität der gleichen Lösung ist nach 6stündio,elm Erhitzen auf 7011 <B>C</B> oder nach 30minü- tigem Erhitzen auf 10011 <B>C</B> hundertfach verkleinert. Das Licht scheint den, Inaktivierungsvorgang kaum züi bceM, ussen. Die bes-te Stabilität in wässenger Lösungerzielt man bei pH <B>6,3-7.</B>
Flavensomycin hat eine bedeutende antifungale Wirkung und ist besonders wirksam gegenüber Hefe pilzen (Saccharomyces) und gegenüber Fadrnpflzeii von der Art Penicillium. Gegenüber Spaltpflzen (Schizomyce,t#en) ist es in Konzentrationen von wem - ger als<B>100</B> u g/cm3 unwirksam.
In der folgenden Tabelle wird das antibiotische Spektrum von Flavensomycin angegeben, welches die jeweilige Minimalkonzentration des Antibiotikums wieder-gibt, die, das Wachstum von bestimmten, Test organismen 48 Stunden lang verhindert. Die Ver dünnungsreihen wurden durchgeführt durch Zugabe von wässerigen Lösungen des Antibiotikums, welche aus<B>10</B> 0/eigen äthanolischen Lösungen hergestellt wurden.
Als Züchtungsmedium für die Testorganismen diente Agar-Malz. Wachstumstemperatur<B>270 C.</B><I>Er-</I> gebnisse nach 48 Stunden.
Neben, seiner starken pilzwachstumshemmenden Wirkung zeigt das erfindungsgemäss hergestellte Anti biotikum auch bemerkenswerte insekticide Wirkung

METHOD FOR PRODUCING AN ANTIBIOTIC The present invention relates to the production of a new antibiotic.
It is well known that antibiotics have a lot of potential. While in the past they were mainly used for therapeutic purposes, today they are also used with great success in other areas.
It has been found that a radiation fungus (actinomycete) which is cultivated in a suitable nutrient medium is able to deliver an antibiotic substance which can be extracted from the cultivation medium using suitable methods. The invention now relates to a method for producing a new antibiotic which is characterized in that Streptomyces cavourensis, strain <B> 829.8.52, </B> is grown in contact with a liquid medium,
which contains sources for assimilable carbon and nitrogen as well as mineral salts, and after fermentation the mycelium is removed from the broth, the flavensomycin formed is extracted from the same with an organic solvent and the pure antibiotic is isolated from the extract obtained. The new active substance which is formed during the cultivation of this radiation fungus is to be called flavensomycin. The physical and chemical properties of flavensomycin are different from those of the already known antibiotics.
The new ray fungus of the genus Streptomyces was isolated from agricultural soil in Cavour, Piedmont and belongs to the group of chromogenic species.
It was bred in the applicant's test laboratories. This ray fungus can currently also be obtained from the University, Institute of the Food Inspectorate in Milan.
