Verfahren zur Herstellung von Pimariein Die Erfindung bezieht sich auf ein Verfahren zur Herstellung eines neuen Antibiotikums, nämlich von Pimaricin, das dadurch gekennzeichnet ist, dass man Streptomyces natalensis nova species unter aeroben Bedingungen in submerser oder Oberflächenkultur züchtet. Das Antibiotikum kann aus der flüssigen Kultur und/oder dem Mycel mit Vorteil durch Extrahieren mittels eines mit Wasser begrenzt mischbaren Alko hols, z.
B. Butanol, oder durch Behandeln mit Metha nol oder einer Lösung von Calciumchlorid in Metha nol mit nachfolgendem Konzentrieren des Extraktes und Reinigen mit Hilfe von Lösungsmitteln, isoliert werden.
Der im folgenden eingehend beschriebene Mikro organismen-Stamm wurde aus einer Bodenprobe aus Pieter-Maritzburg (Provinz Natal, Südafrika) gewon nen. Es handelt sich um eine Streptomyces-Art, welcher der Name Streptomyces natalensis gegeben wurde.
Die morphologischen und physiologischen Eigen schaften des Mikroorganismus sind folgende: Morphologische Eigenschaften: Hyphen verzweigt, unregelmässig verdreht, in der Wachstumszone oft praktisch gerade und zueinander parallel, 0,3-0,7,a dick, gewöhnlich von gleichförmiger Dicke, jedoch manchmal mit örtlichen Verdickungen.
Die sporen- bildend:en Hyphen bilden einfüssige, seitliche Zweige am Lufihnycel. Die Sporen liegen in unregelmässig gekräuselten, selten in lose spiralig gewundenen Ket ten vor und sind durch kurze Zwischenstücke ohne Protoplasma voneinander getrennt, oval, rund oder orangenförmig 0,7-1,2 X 0,7-0,8,a.
Die Kultur weist beim Züchten auf verschiedenen Substraten nach 7 Tagen bei 25 (wenn nicht anders angegeben) die in folgender Tabelle 1 angegebenen Eigenschaften auf:

Streptomyces natalensis nov. spec. kann auch in die Reihe der Neo-Ingri von Baldacci (vgl. Sym posium Actinomycetales. VIth Congr. Intern. Microb. Roma, <B>1953,</B> Seite 31) eingereiht werden.
Zu be merken ist, dass ausser dem beschriebenen Stamm von Streptomyces natälensis nov. spec. auch andere ver wandte Stämme verwendet werden können, die im grossen und :ganzen die gleichen Eigenschaften besitzen und das Antibiotikum Pimaricin erzeugen; sie können betrachtet werden als Subspezies, Varietäten, Rassen, Formen, Gruppen serologischer oder anderer Typen, Varianten, Phasen, spontane Mutanten, Modifikatio nen oder dergleichen dieser Spezies. Ferner kommen auch z. B. durch Bestrahlen oder Behandeln mit Sub stanzen mit toxischer Wirkung erhältliche Mutanten in Betracht.
Das neue Antibiotikum Pimaricin ist in der Hauptsache aktiv gegenüber saprophytischen und parasitären Pilzen und Hefen.
Tabelle 11 gibt einen Überblick über die anti biotischen Eigenschaften von Pimaricin im Verhältnis zu Hefen und Pilzen, die bezüglich des menschlichen Körpers nicht pathogen sind. Für jede Spezies ist die Menge des Antibiotikums in Mikrogramm/ml des Nähr mittels angegeben, die gerade das Wachstum hemmt.
Tabelle IlI zeigt die entsprechende Hemmung einer Anzahl von Hefen und Pilzen, die bezüglich des menschlichen Körpers pathogen sind.

Ein wesentlicher Vorteil des Pimaricius ist der sehr geringe phytotoxische Effekt im Gegensatz zu den meisten bisher gefundenen fungiciden Antibiotika. Infolgedessen ist das neue Antibiotikum auch in der Landwirtschaft und im Gartenbau sehr geeignet als Mittel zur Bekämpfung von Pflanzenkrankheiten, um so mehr als es sich leicht ausbreitet und systematisch wirkt. So dringt Pimaricin z. B. in Erbsen (Pisum sativum) bei dem Eintauchen in eine verdünnte wäss- rige Lösung ein.
Dies ergibt sich aus der Möglichkeit der Extraktion von Pimaricin aus den Keimblättern und Keimen von behandelter Saat.
Die Tabellen IVa und IVb zeigen die f ungizide Wirkung von Pimaricin. Das Pilzmycel (u. a. Asco- chyta pisi) wird in der Saat vernichtet.

Versuche mit Ratten und Mäusen zeigten. eine sehr niedrige Toxizität. Zur Bestimmung der akuten Albionratten-Toxizität von Pimaricin wurde eine Probe entsprechend 985 Mikrogramm/mg Ratten ge geben, die dann sieben Tage gehalten und untersucht wurden.
Mit der Pimaricin-Suspension in Wasser wurden die folgenden Resultate (Durchschnittswerte von Versuchen mit 40 Ratten) erhalten: LDSo oral 1,500 mg/kg LD5o intramuskulär 2,000 mg/kg LDSO subkutan 5,000 mg/kg LDSO intraperitoneal 250 mg/kg Pimaricin hat keine Reizwirkung auf die Haut und Schleimhäute.
Im reinen Zustand ist Pimaricin eine weisse, kristalline Verbindung von amphoterem Charakter, deren Salze auf übliche Weise erhalten werden kön nen. Es gibt mit FeCl. keine Farbreaktion, mit kon zentrierter Phosphorsäure eine rosa, unstabile Fär bung. Konzentrierte Salzsäure und Schwefelsäure er geben eine blaue bzw. olivgrüne Verfärbung. Das Antibiotikum entfärbt Bromwasser. Diese Tatsache und das Infrarot- sowie das Ultraviolett-Spektrum (über die weiter unten berichtet wird) zeigten, dass Pimaricin ein sogenanntes Polyen-Antibiotikum ist.
Die Löslichkeit in Wasser ist sehr gering und be trägt 8 mg in 100 ml bei 2011. In organischen Lösungs mitteln, z. B. Alkoholen, Glykolen und Ketonen, ist das Antibiotikum besser löslich, insbesondere in Alko holen mit 1-6 Kohlenstoffatomen, z.
B. in Methyl alkohol und n-Butylalkohol und in Cellosolve. Weiter ist es löslich in Pyridin, Dimethylformamid, Di- methylacetamid, Eisessig und Alkalihydroxyd. In ali- phatischen Kohlenwasserstoffen wie Pentan, Hexan, Cyclohexan und dergleichen ist die Substanz prak tisch unlöslich.
Bei Raumtemperatur ist Pimaricin eine sehr stabile Verbindung. Eine Lösung in Wasser (5 mg in 100 ml) behält ihre volle Aktivität bei während 7 Tagen bei einem pH-Wert von 7 bei 25 . Bei pH 2 verliert die Lösung in drei Tagen bei dieser Temperatur 5011/o ihrer Aktivität. Bei pH 10 ist die Halbwertzeit bei 25 sechs Tage. Bei erhöhten Tem peraturen ist die Lösung weniger stabil.
Eine Lösung der angegebenen Konzentration von Pimaricin in Wasser vom pH 2 verliert bei 90 in 15 Minuten 9011/o ihrer Aktivität;
beim pH 6,5 15% und beim pH 9 50% der Aktivität. In methylalkoholischer Lösung ist Pimaricin bei höheren Temperaturen (25 bis 601) beständig.
Die antibiotische Aktivität des Pimaricins wurde mikrobiologisch mit Saccharomyces cerevisiae-Holland-Stamm als Testorganismen im Vergleich zu einem reinen Standardpräparat geprüft.
Pimaricin weist keinen Schmelzpunkt auf, son dern beginnt sich bei etwa 150 zu zersetzen. Die spezifische Drehung beträgt

(c = 0,083 % in 100 % Methanol). Das Molekular- gewicht der Substanz ist etwa 727.
Die vermutliche empirische Formel ist C;sH.0N014. Die Elementar analyse ergab folgende Werte: C 57,770/c, H 7,270/0, N 1,95% und O 33,01%.
Das Ultraviolett-Absorptionsspektrum von Pima- ricin weist Maxima bei 290, 304 .und 318 mjc mit einer Spitze bei etwa 280 und ein Minimum bei etwa 250 m,cc (C = 3,93 Mikrogramm/ml in Metha nol) auf.
Eine Probe von Pimaricin, in eine Kalium bromidplatte C = 0,5 0/a) eingepresst, zeigt folgende Infrarot-Absorption (in cm-'): <U>3460</U>,<U>2985</U>,<U>1721,</U> <B>1</B>637, 1577, 1441,<U>1401</U>,<U>1381</U>,<U>1275</U>, 1269, 1238, <U>1192</U>,<U>1</U><B>1</B><U>76</U>,<U>1136</U>,<U>1109</U>,<U>1066</U>,<B>1</B><U>006</U>, _988, 948, 887, 85-5-,844, -803,-1-9-4. (Bei den unterstrichenen Wellenlängen ist die Absorption stark bis sehr stark.) Die Figur zeigt eine graphische Darstellung des Infrarotspektrums von Pimaricin,
aufgenommen mit Hilfe einer Kaliumbromidpiatte.
Das erfindungsgemässe Verfahren zur Herstellung von Pimaricin wird beispielsweise wie folgt ausge führt: Streptomyces natalensis wird aerob in stationären Kulturen oder eingetaucht in eine Nährflüssigkeit unter sterilen Bedingungen in geschlossenen Gefässen, ausgerüstet mit Rühreinrichtungen, denen zur Be lüftung steriler Sauerstoff oder sterile Luft während des Züchtens zugeführt werden kann, gezüchtet.
Das Nährmedium kann aus den üblichen Stoffen bestehen; es enthält vorteilhaft eine Kohlenstoffquelle und eine Quelle von fermentierbarem organischem und/oder anorganischem Stickstoff, wobei vorzugs weise Mineralsalze, wie Phosphate, Kaliumsalze und Natriumsalze, und Spuren verschiedener Metalle, zu gegen sind. Die in der Praxis verwendeten Ausgangs stoffe sind im übrigen oft hinreichend mit diesen Mineralsalzen verunreinigt, so dass sich eine beson dere Zugabe erübrigt.
Als Kohlenstoffquelle können hierbei lösliche und unlösliche Kohlehydrate, z. B. Glukose, Saccharose, Laktose oder Stärke, verwendet werden. Zucker alkohole, wie z. B. Glycerin, sind geeignet. Die Menge der Kohlenstoffquelle in dem Medium kann in weiten Grenzen variieren, abhängig von der Natur des ver wendeten Kohlehydrats und von der übrigen Zusam mensetzung des Mediums; im allgemeinen beträgt sie zwischen 0,5 und 5 Gew.O/o des Mediums.
Als Stickstoffquelle kann eine grosse Anzahl von Substanzen in Frage kommen, z. B. hydrolysiertes oder mchthydrolysiertes Kasein, Maische bzw. Ge- treidequellflüssigkeit, Pepton, Fleischextrakt, ent fettetes oder nichtentfettetes Sojamehl, Erdnussmehl, Fischmehl, Nitrate. Die Wahl der Stickstoffquelle hängt für gewöhnlich von der übrigen Zusammen setzung des Mediums ab, die ihrerseits derart gewählt wird, dass das Antibiotikum auf möglichst wirtschaft liche Weise hergestellt werden kann. In dieser Ver bindung mag erwähnt werden, dass geringe Mengen von stickstoffhaltigen Materialien, wie z. B.
Hefe extrakt, Destillationslösungen, lösliche Fischsubstan zen usw., die Ausbeute an Pimaricin in bestimmten Medien beträchtlich erhöhen. Auch Lipoid-Substan- zen wie z. B. fette Öle vegetabilischen und tierischen Ursprungs (z. B. Sojaöl und Fischöl) vermögen die Ausbeute in einem wesentlichen Masse zu steigern.
Die Dauer der Fermentation hängt in hohem Masse von der Zusammensetzung des Nährmediums ab. Für gewöhnlich variiert sie zwischen 48 und 120 Stunden, jedoch kann mit der Fermentierung auch während einer längeren Zeit fortgefahren wer den, z. B. bis zu 14 Tagen, wenn die erhöhte Aus beute an dem so erhaltenen Antibiotikum die grösseren Kosten einer längeren Fermentationsdauer recht fertigt.
Die Temperatur der Fermentation kann zwischen 15 und 30 liegen; bevorzugt sind Temperaturen von etwa 26-28a.
Mit dem optimalen Wachstum der Mikroorga nismen und der Ausbeute von Pimaricin kann der pH-Wert variieren, insbesondere während der ersten Phase der Fermentation, z. B. zwischen 5 und B. Das Nährmedium wird, z. B. nach dem Sterilisieren, vorzugsweise auf einen pH-Wert zwischen 6 und 7 eingestellt. Die Fermentation wird vorzugsweise aus- geführt bei einem pH-Wert zwischen 6,5 und 8, da in diesem Falle die höchsten Ausbeuten erhalten werden.
Der pH-Wert kann während der Fermen tation durch Zusetzen von Alkaiihydroxyd oder Säure in regelmässigen Intervallen unter sterilen Bedin gungen konstant gehalten werden. üblicherweise jedoch wird Calciumcarbonat als eine Art Puffer substanz in Mengen von 0,2-1 Gew.O/o des Mediums verwendet. Die Menge der in das Medium während der Fermentierung unter sterilen Bedingungen ein geführten Luft hängt in hohem Masse von der Gestalt des Gefässes, der Geschwindigkeit und der Form des Rührers ab. Im allgemeinen variiert diese Menge zwischen 0,1 und 4 Liter pro Liter Nährmedium pro Minute.
Als Impfmaterial werden vorzugsweise 48 bis 72 Stunden alte Vorkulturen von Streptomyces natalensis verwendet. Zur Erzielung guter Ausbeuten ,und zur Verhinderung von Schwankungen der Ergeb nisse wird das Animpfen vorzugsweise mit Kultur mengen von 1-7 Vol /o des Nährmediums. im Hauptfexmentationsgefäss durchgeführt. Es leuchtet ein, dass bei grossen Fermentationsgefässen einige Por tionen von Vorkulturen verwendet werden müssen.
Es ist jedoch auch möglich, die Vorkultur als eine einzige Portion, z. B. in einer Menge von 10 % in das Hauptfermentierungsgefäss zwecks Erzeugen einer neuen Kultur einzugeben. Die gesamte Fermentierung kann auch kontinuierlich durchgeführt werden.
Das Pimaricin kann aus der flüssigen Kultur auf verschiedene Weise gewonnen werden. Gemäss der Erfindung wird zur Gewinnung die Löslichkeit des Antibiotikums in organischen Lösungsmitteln ausge nutzt, die mit Wasser in einem beschränkten Masse mischbar sind, z. B. Butanol. Das Verfahren kann z. B. folgendermassen durchgeführt werden: Nach Erhalten einer hinreichend aktiven Flüssigkeit wird diese auf einen pH-Wert von etwa 10 eingestellt, um die aktive Substanz aus dem sogenannten Mycel freizumachen, wonach es abfiltriert wird. Die Flüssig keit kann dann z.
B. mit n-Butanol extrahiert werden bei einem pH zwischen 3 und 10. Wird die Flüssig keit angesäuert, so wird vorzugsweise mit Phosphor säure gearbeitet; etwa gebildete Ausfällungen können entfernt und nötigenfalls ebenfalls .extrahiert werden. Der n-Butanolextrakt wird durch azeotrope Destilla tion konzentriert; die rohe aktive Substanz kristalli siert dabei. Sie kann durch selektive Fällung, z. B.
aus Eisessig, Pyridin, Dimethylformamid und derglei chen, gereinigt werden. Bei Fermentierungen mit Aus beuten über 700 Mikrogramm/ml kann die Ausbeute an Pimaricin sehr erheblich durch Extrahieren des Mycels erhöht werden.
Vorteilhaft ist die Verwendung von Methylalkohol, dessen Lösefähigkeit durch Zu- gabe von 1-3 % Calciumchlorid erhöht werden kann, als Extraktionsmittel.
<I>Beispiel 1</I> a) Herstellung der Impfkultur.
Aus einem Röhrchen mit einer Kultur von Strepto- myces natalensis nov. spec. auf z. B. Hafermehl Agar, bei der eine gute Sporenbildung stattgefunden hat, werden kleine Mengen Conidium unter sterilen Be dingungen in Schüttelflaschen von etwa 2 Liter Fassungsvermögen, in die 500 cms eines wässrigen Nährmediums eingefüllt sind, übertragen. Das Nähr medium besteht aus:
Pepton 0,5% Konzentrierte Maischflüssigkeit (50 % Trockensubstanz) 0,61/o Glukose 1,01/0 Kochsalz (eingestellt auf pH = 7,0 mit Alkalihydroxyd) 1,011/0 Nach der Inkubation unter dauerndem Schütteln bei 26 während 48 Stunden ist die Kultur für das Animpfen in dem Hauptfermentationsmedium fertig. b) Herstellung der Kultur.
Ein Liter der oben beschriebenen Impfkultur wird unter sterilen Bedingungen in ein mit Rührer und Einblasvorrichtung für sterile Luft ausgerüstete Fer- mentierungsgefäss, das 15 Liter eines wässrigen Kul turmediums folgender Zusammensetzung enthält, ein gefüllt:

1/0 Dieses Kulturmedium wird mit Aikalihydroxyd auf einen pH-Wert von 6-9 eingestellt. Nach einer Inkubationszeit von 48 Stunden bei 27 unter dauern der Belüftung und dauerndem Rühren enthält die Kultur 610 Mikrogramm Pimaricin pro ml.
Andere wässrige Kulturmedien, mit denen Aus beuten derselben Grössenordnung erhalten werden können, können die folgende Zusammensetzung haben: A. Erdnussmehl 20/a Kartoffelstärke 11/0 Konzentrierte Maischflüssigkeit (50 0/a Trockensubstanz) 2% Kochsalz 0,5()10 Magnesiumsulfat 0,10/0 Primäres Kaliumphosphat 0,
05110 Calciumcarbonat 0,611/0 Kaliumhydroxyd bis pH 6,5 Mit diesem Medium wurden nach dem Animpfen mit 4 % Impfkultur nach 72 Stunden Inkubation und Belüftung bei 26 590 Mikrogramm Pimaricin pro ml Fermentationsflüssigkeit erhalten. B.
Zuckerrübenmelasse (Zuckergehalt etwa 50 %) 4% Laktose 1 0/0 Konzentrierte Marschflüssigkeit 2 % Natriumsulfat 10 aq. 0,10/0 Calciumcarbonat 0,5 % Kaliumhydroxyd bis pH 7,
1 Mit diesem Medium wurden nach dem Animpfen mit 5% Impfkultur und 120 Stunden Inkubation und Belüftung bei 27 640 Mikrogramm Pimaricin pro ml Fermentationsflüssigkeit erhalten.

51/o In diesem Medium bildeten sich nach Animpf.en mit 3 % Impfkultur und 148 Stunden Inkubation und Belüftung bei 26 535 Mikrogramm Pimaricin pro ml Fermentationsflüssigkeit. D.
Sojamehl (grob) 511/o Sojaöl 0,5% Konzentrierte Marschflüssigkeit (50 % Feststoffe) 0,21/o Glukose 10/0.
Primäres Kaliumphosphat 0,021/o Ammoniumsulfat 0,5010 Caleiumcarbonat 10/0 Mit diesem Medium wurden nach dem Animpfen mit 3 0/a Impfkultur in :einem 15-Liter-Fermentie- rungsgefäss und Inkubation und Belüftung während 168 Stunden bei 28 690 Mikrogramm Pimaricin pro ml Ferrnentierungsflüssigkeit gebildet.
Nach dem Behandeln des Mycels, das ausgepresst und in einer reichlichen Menge Wasser mit verdünntem Alkali hydroxyd vom pH-Wert 10 gerührt worden war, konnte in dem Filtrat des Mycels 20,4 g Pimaricin festgestellt werden.
<I>Beispiel 2</I> (Isolierung des rohen Pimaricins) 4 Liter der gemäss Beispiel 1 erhaltenen Kultur flüssigkeit wurden nach vollendeter Fermentierung - Gehalt an Pimaricin 590 Mikrogramm/ml - reit 10 % igem Natriumhydroxyd auf pH 10 eingestellt,
dann von dem Mycel durch Filtrieren mittels Kiesel- gur als Flltriermittel (2 %) befreit. Das Filtrat der Kultur betrug 3,7 Liter; die Aktivität betrug 570 Mikrogramm/m1; das Filtrat wurde mit Phosphor säure zu pH 3 angesäuert. Dieses angesäuerte Filtrat wurde nacheinander mit 750, 350 bzw. 350 ml n-Butanol extrahiert.
Der Butanol-Extrakt wurde dann dreimal mit je 120 ml einer 4 %igen Borax- lösung und dann zweimal mit je 120 ml Wasser ge waschen. Der Butanolextrakt enthielt dann 1400 Mikrogramm/ml. Nach dem azeotropischen Ein dampfen auf 100 ml trennten sich 0,64 g von nicht völlig reinem Pimaricin in kristallinem Zustand ab.
(Aktivität 900 Mikrogramm/ml). Aus dem Rest des Extraktes konnte eine Gesamtmenge von 1,42 g un reines Pimariein durch weiteres Eindampfen (Akti vität 395 Mikrogramm/mg) erhalten werden.
Die Ausbeute, bezogen auf die Gesamtaktivität der voll- kommen fermentierten Flüssigkeit, betrug 48,3 %. <I>Beispiel 3</I> (Isolierung von rohem Pimaricin) 15 Liter einer gemäss der Arbeitsweise von Bei spiel 1 :
erhaltenen Kulturflüssigkeit nach vollendeter Fermentierung, die 610 Mikrogramm/ml Pimaricin enthielt, wurde auf pH 10,3 mit 35 %.igem Kalium- hydroxyd eingestellt und dann von dem Mycel mit tels 50 g Hyflo (eingetragene Marke) als Filtrier mittel befreit.
Das Filtrat wurde dann mit Phosphor säure auf pH 2,8 angesäuert.. Die gebildete Fällung wurde abfiltriert, und zwar mittels 10 g Hyflo . Die Fällung wurde dann eine halbe Stunde mit 100 ml n-Butanol gerührt. Nach dem Absetzen wurde der Butanol'extrakt abgezogen und der Rückstand mit 100 ml Wasser gewaschen.
Der gesamte Butanol, extrakt wurde dreimal mit je 10 ml einer 4 % igen Boraxlösung und danach dreimal mit je 10 ml Wasser gewaschen. Er wurde dann im Vakuum bei 44' auf 40 m1 eingedampft.
Nach dem Kühlen auf 0 , Ab saugen und Trocknen, wurden 0;5l g mattgelbes kristallines Pimaricin mit einer Aktivität von 850 Mikrogramm/mg erhalten. Das Kulturfiltrat wurde in der in Beispiel 1 beschriebenen Weise weiterbehan delt. Erhalten wurden zwei Fraktionen.: 3,21 g (Akt.
890 Mikrogramm/mg) und 6,8 g (Akt. 223 Mikro- gramm/mg). Die Gesamtausbeute betrug 51,5 %, <I>Beispiel 4</I> (Isolierung von rohem Pimaricin) 10 Liter eines gemäss Beispiel 1 erhaltenen voll ständig fermentierten Kulturmediums (Aktivität 640 Mikrogramm/ml) wurden auf pH 10 mit 15 0/aigem Kaliumhydroxyd eingestellt.
Danach wurde das Mycel zentrifugiert; das klare Zentrifugat wurde mit 10n Salzsäure auf pH 6,9 eingestellt. Danach wurde nacheinander mit 2000, 1000 und 1000 ml Isoamyl- alkohol extrahiert. Die vereinigten Isoamylalkohol- extrakte wurden nacheinander dreimal mit je 300 ml einer 4 0/aigen Natriumcarbonatlösung und dann zweimal- mit je 300 ml Wasser gewaschen.
Der so behandelte Extrakt wurde azeotrop auf 100 ml ein gedampft; dabei trennten sich 2,62g eines mattgelben kristallinen Pimaricins mit einer Aktivität von 900 Mikrogramm/mg ab. Ausbeute 36,8%. <I>Beispiel 5</I> (Isolierung von rohem Pimaricin) 20 Liter einer gemäss Beispiel 1 erhaltenen, voll ständig fermentierten Kultur von Akt.
700 Mikro- gramm/ml wurden mit 20 /o igem Natriumhydroxyd auf pH 10 eingestellt. Das Mycel wurde danach mittels Hyflo (200 g) abfiltriert. Das klare Kultur- filtrat wurde nacheinander mit 4000, 2000 und <B>1000</B> ml n Butanol extrahiert.
Die vereinigten Buta- nolextrakte wurden zweimal mit je 500 ml 4 /o iger Boraxlösung und dann zweimal mit je 500 ml Wasser gewaschen. Danach wurde der pH-Wert des Butanol- extraktes mit 10n Salzsäure auf 6,8 eingestellt; der Extrakt wurde azeotrop im Vakuum auf 250 ml ein gedampft.
Dabei trennten sich 9,85 g mattgelben, kristallinen Pimaricins (Aktivität 913 Mikrogramm/mg ab. Ausbeute 60,8 %.
<I>Beispiel 6</I> (Isolierung von rohem Pimaricin aus Mycel) 15 Liter einer gemäss Beispiel 1 erhaltenen, voll ständig fermentierten Kultur von Streptomyces nata- lensis nov. spec. wurden mit Eisessig auf einen pH- Wert von 4,1 eingestellt. Danach wurden 200 g Hyflo als Filtriermittel zugegeben, mit dem die Lösung gerührt wurde.
Das Mycel wurde dann so trocken als möglich ausgepresst bis zu einem Gesamt gewicht von<B>1700</B> g (1 g dieses Mycels wurde in diesem Zustand während einer Stunde mit 40 ml Methylalkohol extrahiert, um die Zahl der Mikro gramm Pimaricin zu bestimmen. Der Methanolextrakt wurde auf 100 ml aufgefüllt und .spektrophotometrisch gemessen. Der Gehalt der Methanollösung an Pima- ricin betrug 120 Mikrogramm/ml).
Der Pimaricingehalt des gesamten Mycels betrug 20,4g. Das abgepresste Mycel wurde während einer halben Stunde bei Raumtemperatur mit 8 Liter Methanol, in dem 2 % Calciumchlorid gelöst waren, extrahiert.
Der so erhaltene Extrakt wurde mit 1 Liter Wasser verdünnt und von Methanol im Vakuum be freit; dabei bildete sich ein kristalliner Niederschlag von Pimaricin, der nach dem Absaugen, Waschen mit Wasser und Trocknen im Vakuum 14,73g wog und eine Aktivität von 9 Mikrogramm/mg besass.
(65 /a der theoretischen Ausbeute.) <I>Beispiel 7</I> (Reinigung von Pimaricin) 10 g gemäss den Beispielen 1-6 erhaltenes, un reines Pimaricin (Aktivität 890 Mikrogramm/mg) wurden unter gelindem Erwärmen in 80 ml Eisessig gelöst und dann so rasch als möglich durch Filtrieren von ungelösten Verunreinigungen befreit.
Das klare, gelbbraune Filtrat wurde mit 1500 ml Wasser ver dünnt; der pH-Wert dieser Lösung wurde mit 33 /oigem Natriumhydroxyd auf 6,3 eingestellt. Nach dem Kühlen wurde die gebildete kristalline Fällung zentrifugiert und zweimal mit einer Gesamtmenge von 500 ml Wasser gewaschen und abgesaugt. Die kristal line Masse auf dem Filter wurde wieder mit 200 ml Wasser gewaschen, danach im Vakuum über Phos- phorpentoxyd getrocknet.
Das Gewicht des so erhal tenen sehr blassgelben Produktes betrug 7,07 g; seine Aktivität betrug 960 Mikrogramm/mg.
Die oben mitgeteilte Behandlung wurde mit 60 ml Eisessig und 1000 ml Wasser wiederholt. Die Sub stanz wurde dreimal mit je 200 ml Wasser ge- waschen. Das so erhaltene Produkt wog 5,35 g und besass eine Aktivität von 995 Mikrogramm/mg. Ge samtausbeute: 59,8 %.
<I>Beispiel 8</I> (Reinigung von Pimaricin) 10 g gemäss den Beispielen 1-6 erhaltenes, un reines Pimaricin (Aktivität 890 Mikrogramm/mg) wurde unter gelindem Erwärmen in<B>100</B> ml Dimethyl- formamid gelöst. Diese Lösung wurde abfiltriert zur Entfernung ungelöster Verunreinigungen, das Filter wurde wieder mit 30 ml Dimethylformamid ge waschen. Zu dem braungefärbten klaren Filtrat wur den 250 ml Wasser zugegeben, wonach das Anti- bio:tikum im kristallinen Zustand ausfiel.
Das Produkt wurde abgesaugt, das Filter wieder mit 100 ml Was ser gewaschen. Die kristalline Masse wurde danach im Vakuum getrocknet. Das so erhaltene Produkt wog 9,24 g. Nach Wiederholung der oben beschrie benen Behandlung wurde schliesslich ein mattgelbes kristallines Produkt von einem Gewicht von 7,9 g mit einer Aktivität von 985 Mikrogramm/mg erhal- ten. Gesamtausbeute: 87,5%.
<I>Beispiel 9</I> (Herstellung des Natriumsalzes) 2,5 g gemäss der Arbeitsweise nach Beispielen 1 bis 6 erhaltenes Pimaricin (Aktivität 985 Mikro gramm/mg) wurden unter mechanischem Rühren in 20 ml Methanol suspendiert. Danach wurden 0,145 g Natriumhydroxyd, gelöst in 0,3 ml Wasser, zu dieser Suspension zugegeben. Das sich zunächst lösende Pimaricin kristallisierte nach wenigen Minuten als Natriumsalz aus.
Nach weiteren<B>30</B> Minuten Rühren und 24stündigem Stehenlassen bei 0 wurde die kristalline Masse abgesaugt und mit 5 ml Äthyl- alkohol und 10 ml Diäthyläther gewaschen. Nach dem Trocknen im Vakuum betrug das Gewicht des weissen, nadelförmig kristallinen Natriumsalzes 2,16 g (Gehalt 995 Mikrogramm/mg = 87,3 % der berechneten Aktivität). Andere Salze können in entsprechender Weise hergestellt werden.

Experiments with rats and mice showed. very low toxicity. To determine the acute toxicity of pimaricin to albion rats, a sample corresponding to 985 micrograms / mg was given to rats, which were then kept and examined for seven days.

In the pure state, pimaricin is a white, crystalline compound of amphoteric character, the salts of which can be obtained in the usual way. There is with FeCl. no color reaction, with concentrated phosphoric acid a pink, unstable color. Concentrated hydrochloric acid and sulfuric acid give a blue or olive green discoloration. The antibiotic decolorizes bromine water. This fact and the infrared and ultraviolet spectrum (which will be reported below) indicated that pimaricin is a so-called polyene antibiotic.

The antibiotic activity of pimaricin was tested microbiologically with Saccharomyces cerevisiae-Holland strain as test organisms in comparison with a pure standard preparation.

The inventive method for the production of pimaricin is carried out, for example, as follows: Streptomyces natalensis is aerobically in stationary cultures or immersed in a nutrient liquid under sterile conditions in closed vessels, equipped with stirring devices, which are supplied with sterile oxygen or sterile air for ventilation during cultivation can be bred.
The nutrient medium can consist of the usual substances; it advantageously contains a carbon source and a source of fermentable organic and / or inorganic nitrogen, with preference being given to mineral salts such as phosphates, potassium salts and sodium salts, and traces of various metals. In addition, the starting materials used in practice are often sufficiently contaminated with these mineral salts, so that special addition is not necessary.
As a carbon source, soluble and insoluble carbohydrates, e.g. B. glucose, sucrose, lactose or starch can be used. Sugar alcohols, such as B. glycerin are suitable. The amount of carbon source in the medium can vary within wide limits, depending on the nature of the carbohydrate used and on the rest of the composition of the medium; in general it is between 0.5 and 5% by weight of the medium.
A large number of substances can be used as the nitrogen source, e.g. B. hydrolyzed or mchthydrolyzed casein, mash or grain swelling liquid, peptone, meat extract, defatted or not defatted soy meal, peanut meal, fish meal, nitrates. The choice of nitrogen source usually depends on the rest of the composition of the medium, which in turn is chosen so that the antibiotic can be produced in the most economical way possible. The duration of the fermentation depends to a large extent on the composition of the nutrient medium. Usually it varies between 48 and 120 hours, but fermentation can be continued for a longer period of time, e.g. B. up to 14 days if the increased yield of the antibiotic obtained in this way justifies the greater costs of a longer fermentation period.
The fermentation temperature can be between 15 and 30; temperatures of about 26-28a are preferred.
With the optimal growth of the microorganisms and the yield of pimaricin, the pH can vary, especially during the first phase of fermentation, e.g. B. between 5 and B. The nutrient medium is, for. B. after sterilization, preferably adjusted to a pH between 6 and 7. The fermentation is preferably carried out at a pH between 6.5 and 8, since in this case the highest yields are obtained.
The pH value can be kept constant during fermentation by adding alkali hydroxide or acid at regular intervals under sterile conditions. Usually, however, calcium carbonate is used as a kind of buffer substance in amounts of 0.2-1% by weight of the medium. The amount of air introduced into the medium during fermentation under sterile conditions depends to a large extent on the shape of the vessel, the speed and the shape of the stirrer. In general, this amount varies between 0.1 and 4 liters per liter of nutrient medium per minute.
Pre-cultures of Streptomyces natalensis that are 48 to 72 hours old are preferably used as inoculation material. In order to achieve good yields and to prevent fluctuations in the results, the inoculation is preferably carried out with culture quantities of 1-7 vol / o of the nutrient medium. carried out in the main fermentation vessel. The product was filtered off with suction and the filter was washed again with 100 ml of water. The crystalline mass was then dried in vacuo. The product thus obtained weighed 9.24 g. After repeating the treatment described above, a pale yellow crystalline product weighing 7.9 g with an activity of 985 micrograms / mg was finally obtained. Overall yield: 87.5%.
