Verfahren zur Herstellung von Alkaloiden Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Isolierung von Reserpin aus einem Reserpin im Gemisch mit anderen schwach basischen Alkaloiden enthaltendem Material durch Umsetzung mit zur Salzbildung mit schwachen Basen geeigneten Säuren oder Salzen solcher Säuren.
Verschiedene Verfahren zur Isolierung von Re- serpin sind bereits bekannt, u. a. auch die Reinigung von Reserpin durch Umsetzung mit zur Salzbildung mit schwachen Basen geeigneten Säuren oder Salzen. Diese Verfahren führen gut zu den genannten Alka loiden. Zur Isolierung allerdings der reinen Alkaloide bedurfte es meist noch besonderer Reinigungsmetho den, wie Chromatographie oder wiederholter frak tionierter Kristallisation.
Es wurde nun gefunden, dass man auf einfache Weise Reserpin in hoher Reinheit gewinnen kann und somit eine gute Trennung von Reserpin und Deserpidin erreicht, wenn man das die Alkaloide enthaltende Material mit Rhodanwasserstoffsäure oder deren Salzen behandelt, aus der erhaltenen Mischung das rhodanwasserstoffsaure Salz des Reser- pins abscheidet und das entstandene Reserpinrhodanid in das freie Reserpin überführt.
Das Ausgangsmaterial kann z. B. ein Reserpin und Deserpidin enthaltender Extrakt aus Pflanzen der Rauwolfia- oder Alstonia-Arten sein. Besonders zu nennen sind aus folgenden Pflanzen gewonnene Extrakte:
Rauwolfia vomitoria Afz., Rauwolfia ca- nescens, Rauwolfia serpentina Benth., Rauwolfia inebrians, Rauwolfia indecora und Alstonia constricta F. Muell., deren Gewinnung bekannt ist. So ist es möglich, z.
B. einen Benzol- oder Toluolextrakt von diesen Pflanzen, der vorteilhafterweise von ange feuchtetem Pflanzenmaterial, insbesondere Wurzel material, hergestellt ist, als Ausgangsmaterial zu verwenden.
Als solches dienen aber auch Chloro form-, Äthylendichlor- oder Trichloräthylen- oder Acetonextrakte. Es ist aber auch möglich, von einem unreinen Reserpin auszugehen oder einen Rauwolfia- oder Alstoniaextrakt zu benützen, aus dem bereits die Hauptmenge Reserpin .entfernt ist, und der ein Gemisch von restlichem Reserpin und Deserpidin enthält.
Insbesondere kann man auch von Mutter laugen ausgehen, die nach einer ersten Kristallisa tion von Reserpin anfallen, vorzugsweise von sol chen, welche Deserpidin und restliches Reserpin im ungefähren Verhältnis 1:l enthalten.
Je nach der Wahl des Ausgangsmaterials kann das neue Verfahren, z. B. zur Gewinnung von Reser- pin und Deserpidin, zur Reinigung von Reserpin, zur Aufarbeitung von Reserpin-Mutterlaugen auf Re- serpin und Deserpidin oder zur Auftrennung von Reserpin-Deserpidin-Gemischen dienen.
Eine besondere Ausführungsform des Verfahrens besteht darin, dass man ein Reserpin und Deserpidin enthaltendes Produkt mit Rhodanwasserstoffsäure behandelt, so dass ein überwiegender Teil des Re- serpins in ein Rhodanwasserstoffsäuresalz übergeführt und das Deserpidin mindestens in einem überwie genden Teil nicht umgesetzt wird,
das heisst durch Umsetzung mit ungefähr der für die überführung des vorhandenen Reserpins in ein rhodanwasserstoff- saures Salz berechneten Menge Rhodanwasserstoff- säure, und dieses Salz abtrennt.
Zur erfindungsgemässen Umsetzung mit Rhodan- wasserstoffsäure verwendet man zweckmässig eine Lösung des Ausgangsmaterials in einem organischen Lösungsmittel, in welchem das Reserpinrhodanid schwer löslich ist, gegebenenfalls mit einem Gemisch mit Wasser. So eignen sich z. B.
Lösungen in halo- genierten aliphatischen Kohlenwasserstoffen, wie Chloroform, Methylenchlorid, Äthylenchlorid, Tri- chloräthan und dergleichen, niederen Alkoholen, wie Äthanol oder Methanol, Äther, wie Dimethyläther, oder Fettsäuren, wie Essigsäure, oder in erster Linie deren Gemischen, wie Chloroform-Äther, Chloro- form-Methanol,
Methanol-Wasser oder Methanol- Essigsäure. Die Rhodanwasserstoffsäure kann als solche verwendet oder aber auch erst in der Reak tionslösung gebildet werden. So kann sie z. B. in Form eines ihrer Salze, wie ein Alkali- oder Erd- alkalisalz, z. B. Kalium-, Calcium- oder Ammonium rhodanid, Verwendung finden. In diesem Fall soll die Reaktionslösung eine Säure enthalten, wie eine Fettsäure, vorzugsweise Essigsäure, oder Phosphor säure, die zur Freisetzung der Rhodanwasserstoff- säure dient.
Es ist aber auch möglich, von einem Material auszugehen, das Reserpin in Form eines Salzes enthält, wobei Rhodanwasserstoffsäure durch doppelte Umsetzung erhalten wird, und sogleich das Reserpinrhodanid bildet.
Aus der Reaktionslösung kann das Rhodanid in einfacher Weise, z. B. durch Filtrieren, abgetrennt werden. Es lässt sich in üblicher Weise, z. B. durch Behandeln mit alkalischen Mitteln, wie Ammoniak oder Natronlauge, in das freie basische Material umwandeln, aus dem Reserpin durch übliche Kristal lisation in reiner Form gewonnen werden kann. Besonders vorteilhaft ist es, das Salz in wässrig- methanolischer Lösung mit Ammoniak zu behandeln, wobei reines Reserpin kristallisiert.
Je nach der im Ausgangsmaterial vorhandenen Menge Deserpidin und der verwendeten Menge Rho- danwasserstoffsäure kann das abgetrennte Salz auch Deserpidinrhodanid enthalten. Bei der Freisetzung des basischen Materials und der nachfolgenden Kri stallisation bleibt Des.erpidin als leichter lösliches Alkaloid in Lösung und kann daraus in bekannter Weise isoliert werden.
In den nachstehenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Tem peraturen sind in Celsiusgraden angegeben. <I>Beispiel 1</I> 1 Gewichtsteil pulverisierter Extrakt aus Rau- wolfia vomitoria Afz. oder Rauwolfia serpentina Benth. Wurzeln wird in einem Gemisch von 2 Vo- lumteilen Eisessig und 8 Volumteilen Methanol warm gelöst,
unter Rühren mit 6 Volumteilen Wasser ver setzt und der Niederschlag abfiltriert. Das Filtrat wird bei 60 mit Reserpinrhodanid geimpft. Unter Rühren wird 1 Volumteil 50%ige wässrige Kalium- rhodanidlösung zugegeben. Das Gemisch lässt man einige Stunden bei Raumtemperatur stehen und nutscht dann das gebildete Reserpinrhodanid ab. Der Rückstand wird mit Methanol gewaschen.
1 Gewichtsteil Reserpinrhodanid wird mit 8 Vo- lumteilen Methanol und 4 Volumteilen 3,5o/oigem Ammoniak aufgeschlämmt und bei 60 Innentempe ratur während 30 Minuten gerührt. Nach dem Ab- kühlen lässt man einige Stunden bei Raumtemperatur stehen und nutscht dann das freigesetzte Reserpin ab. Es wird mit Wasser, Methanol und Methanol äther gewaschen. Man erhält so reines Reserpin.
Der als Ausgangsmaterial verwendete Extrakt kann nach den im USA-Patent Nr. 2 752 351 enthal tenen Angaben erhalten werden. Man kann z. B. 1 Gewichtsteil gemahlene Wurzeln von Rauwolfia ser- pentina Benth. oder Rauwolfia vomitoria Afz. mit 1 Volumteil Wasser gleichmässig anfeuchten.
Das Wurzelmaterial wird dann mit 5-l0 Volumteilen Toluol während 5-7 Stunden am Rückfluss extra hiert, wobei mit entsprechendem Vakuum die Siede temperatur des Toluols auf etwa 65 einreguliert wird. Der hell bräunliche Toluolextrakt wird von eventuell vorhandenem Wasser abgetrennt und im Vakuum zur Trockene eingedampft.
<I>Beispiel 2</I> 1 Gewichtsteil rohes Reserpin wird in einem Gemisch von 2 Volumteilen Methanol und 2 Volum- teilen Eisessig warm gelöst. Bei 60 Innentemperatur und gutem Rühren werden 6 Volumteile Wasser zu gefügt und entstandene Abscheidungen abfiltriert. Das klare Filtrat wird mit Reserpinrhodanid ge impft und bei 60 unter gutem Rühren mit 0,4 Vo- lumteilen 5011/oiger wässriger Kaliumrhodanidlösung versetzt.
Nach dem Abkühlen wird das Gemisch einige Stunden stehen gelassen und anschliessend fil triert. Das abgenutschte Reserpinrhodanid wird mit Methanol gewaschen.
1 Gewichtsteil Reserpinrhodanid wird in 8 Vo- lumteilen Methanol und 4 Volumteilen 3,5 /o igem Ammoniak aufgeschlämmt und bei 60 Innentempe ratur während 30 Minuten gerührt. Nach dem Ab kühlen lässt man einige Stunden bei Raumtemperatur stehen und nutscht dann das freigesetzte Reserpin ab. Der Rückstand wird mit Wasser, Methanol und Methanoläther gewaschen. Man erhält so reines Reserpin.
<I>Beispiel 3</I> Man löst eine Mischung von 0,05 Gewichtsteilen Reserpin und 0,05 Gewichtsteilen Deserpidin in 2 Volumteilen Chloroform und 50 Volumteilen Äther und tropft 10 Volumteile einer Rhodanwasserstoff- säurelösung, die durch Versetzen von 10 Volum- teilen 0,1-n.
methanolischer Ammoniumrhodanid- lösung mit 10 Volumteilen Methanol und 2 Volum- teilen Eisessig und Auffüllen mit Äther auf ein Volumen von 100 Volumteilen erhalten wird, unter mechanischem Rühren zu. Diese :Mischung wird mit 25 Volumteilen Petroläther versetzt. Nach 15 Minu ten wird das ausgeschiedene Reserpinrhodanid ab filtriert und mit Äther nachgewaschen.
Man erhält 0,066 Gewichtsteile Reserpinrhodanid vom F. 239 bis 240 (Zerr.). Das Filtrat, das im wesentlichen Deserpinbase enthält, wird 7mal mit je 25 Volum- teilen 2-n. Essigsäure ausgeschüttelt, die saure Lö sung mit Ammoniak unter Kühlung alkalisch gestellt und das erhaltene Produkt durch Filtrieren abge- trennt. Man erhält so 0,037 Gewichtsteile eines Basengemisches, das zu 85 % aus Deserpidin und 15 0/0 Reserpin besteht.
<I>Beispiel 4</I> Nimmt man anstelle der 10 Volumteile Rhodan- wasserstoffsäurelösung nur 9 Volumteile derselben und verfährt im übrigen wie in Beispiel 3 beschrie ben, so erhält man 0,062 Gewichtsteile Reserpin- rhodanid und 0,041 Gewichtsteile Deserpidinbase, die noch 221/o Reserpin enthält.
<I>Beispiel 5</I> 0,065 Gewichtsteile eines nach Beispiel 4 er haltenen Deserpidin-Reserpin-Gemisches, enthaltend 22% Reserpin, werden in 2 Volumteilen Chloroform gelöst und mit 100 Volumteilen Äther versetzt.
Dazu gibt man 2,5 Volumteile der in Beispiel 1 beschriebenen methanolischen Rhodanwasserstoff- säurelösung, fügt 5 Volumteile Petroläther zu und filtriert nach 15minutigem mechanischem Rühren ab.
Man erhält so 0,016 Gewichtsteile Reserpin- rhodanid und 0,045 Gewichtsteile eines Basen- gemisches, das aus 87% Deserpidin und 13% Re- serpin besteht.
Das in den Beispielen 3-5 erhaltene Reserpin- rhodanid kann, wie in den Beispielen 1 und 2 gezeigt, in reines Reserpin umgewandelt werden.
<I>Beispiel 6</I> 1 Gewichtsteil Extrakt aus Wurzeln von Rau- woifia vomitoria Afz. oder Rauwolfia serpentina Benth. wird in einem Gemisch von 4 Volumteilen Methanol und 1 Volumteil Eisessig warm gelöst,
unter Rühren mit 6 Volumteilen Wasser versetzt und der Niederschlag aus dem noch warmen Ge misch abfiltriert. Man impft das Filtrat bei etwa 60 mit Reserpinrhodanid und versetzt es unter Rühren mit 0,5 Volumteilen 50%iger wässriger Kalium- rhodanidlösung. Nach einigen Stunden Stehen bei Raumtemperatur wird das Gemisch genutscht und das auf dem Filter zurückbleibende,
rohe Reserpin- rhodanid mit Methanol gewaschen. Das erhaltene Salz lässt sich, wie in Beispiel 1 beschrieben, in das basische Reserpin überführen.
Der als Ausgangsmaterial verwendete Extrakt (Alkaloidgehalt etwa 0,2 bis 0,3 Gewichtsteile) kann, wie in Beispiel 1 beschrieben, hergestellt werden.
<I>Beispiel 7</I> 1 Gewichtsteil Äthylenchloridextrakt aus Wur zelmaterial von Rauwolfia vomitoria Afz. oder Rau- wolfia serpentina Benth. wird in einem Gemisch von 8 Volumteilen Methanol und 2 Volumteilen Eisessig warm gelöst. Bei etwa 40 Innentemperatur und gutem Rühren fügt man 6 Volumteile Wasser zu und filtriert den Niederschlag aus dem noch warmen Gemisch ab.
Das klare Filtrat wird mit Reserpin- rhodanid geimpft und bei etwa 60 Innentemperatur unter gutem Rühren mit 2 Volumteilen 50%iger wässriger Kaliumrhodanidlösung versetzt. Nach dem Abkühlen wird das Gemisch etliche Stunden stehen gelassen und anschliessend filtriert. Das abgenutschte, rohe Reserpinrhodanid wird mit - Methanol ge waschen.
Es lässt sich, wie in Beispiel 1 beschrieben, in das freie Reserpin überführen.
Zur Herstellung des als Ausgangsmaterial ver wendeten Äthylenchloridextraktes (Alkaloidgehalt etwa 0,1 Gewichtsteile) kann man z. B. 1 Gewichts teil gemahlene Wurzeln von Rauwolfia serpentina Benth. oder Rauwolfia vomitoria Afz. mit etwa 0,8 Volumteilen Wasser gleichmässig anfeuchten und dann mit 5-10 Volumteilen Äthylenchlorid wäh rend 5-7 Stunden am Rückfluss extrahieren.
Der braune Äthylenchloridextrakt wird von eventuell vor handenem Wasser abgetrennt und im Vakuum zur Trockne eingedampft.
Process for the preparation of alkaloids The present invention relates to a process for the isolation of reserpine from a reserpine in a mixture with other weakly basic alkaloids-containing material by reaction with acids or salts of such acids suitable for salt formation with weak bases.
Various methods for isolating reserpin are already known, including a. also the purification of reserpine by reaction with acids or salts suitable for salt formation with weak bases. These processes lead well to the alkaloids mentioned. To isolate the pure alkaloids, however, special purification methods such as chromatography or repeated fractional crystallization were usually required.
It has now been found that reserpine can be obtained in a simple manner in high purity and thus a good separation of reserpine and deserpidine can be achieved if the material containing the alkaloids is treated with hydrofluoric acid or its salts, from the resulting mixture the rhodanic acid salt of the reser - pins deposited and the reserpine rhodanide formed is converted into the free reserpine.
The starting material can e.g. B. be an extract containing reserpine and deserpidine from plants of the Rauwolfia or Alstonia species. Particular mention should be made of extracts obtained from the following plants:
Rauwolfia vomitoria Afz., Rauwolfia canescens, Rauwolfia serpentina Benth., Rauwolfia inebrians, Rauwolfia indecora and Alstonia constricta F. Muell., Whose extraction is known. So it is possible, for.
B. a benzene or toluene extract from these plants, which is advantageously made of moist plant material, especially root material, is used as the starting material.
Chloro form, ethylene dichloro or trichlorethylene or acetone extracts also serve as such. However, it is also possible to start from an impure reserpine or to use a Rauwolfia or Alstonia extract from which the main amount of reserpine has already been removed and which contains a mixture of residual reserpine and deserpidine.
In particular, one can also start from mother liquors which are obtained after a first crystallization of reserpine, preferably from those which contain deserpidine and residual reserpine in an approximate ratio of 1: 1.
Depending on the choice of starting material, the new process, e.g. They are used, for example, to obtain reserpine and deserpidine, to purify reserpine, to work up reserpine mother liquors to produce reserpine and deserpidine or to separate reserpine-deserpidine mixtures.
A particular embodiment of the process consists in treating a product containing reserpine and deserpidine with hydrofluoric acid so that a predominant part of the reserpine is converted into a hydrofluoric acid salt and at least a predominant part of the deserpidine is not converted,
that is, by reacting with approximately the amount calculated for converting the reserpine present into a salt of rhodanic acid, and separating this salt.
A solution of the starting material in an organic solvent in which the reserpine rhodanide is sparingly soluble, optionally with a mixture with water, is expediently used for the reaction according to the invention with hydrofluoric acid. So are z. B.
Solutions in halogenated aliphatic hydrocarbons such as chloroform, methylene chloride, ethylene chloride, trichloroethane and the like, lower alcohols such as ethanol or methanol, ethers such as dimethyl ether, or fatty acids such as acetic acid, or primarily their mixtures such as chloroform Ether, chloroform-methanol,
Methanol-water or methanol-acetic acid. The hydrofluoric acid can be used as such or it can only be formed in the reaction solution. So she can z. B. in the form of one of their salts, such as an alkali or alkaline earth salt, z. B. potassium, calcium or ammonium rhodanide, use. In this case, the reaction solution should contain an acid, such as a fatty acid, preferably acetic acid, or phosphoric acid, which is used to release the thiocyanate acid.
However, it is also possible to start from a material which contains reserpine in the form of a salt, with hydrofluoric acid being obtained by double conversion and immediately forming the reserpine rhodanide.
The rhodanide can be extracted from the reaction solution in a simple manner, e.g. B. by filtration, are separated. It can be in the usual way, for. B. by treating with alkaline agents, such as ammonia or sodium hydroxide solution, convert into the free basic material, from which reserpine can be obtained in pure form by conventional crystallization. It is particularly advantageous to treat the salt in an aqueous methanolic solution with ammonia, with pure reserpine crystallizing.
Depending on the amount of deserpidine present in the starting material and the amount of hydrofluoric acid used, the separated salt can also contain deserpidine rhodanide. When the basic material is released and the subsequent crystallization occurs, Des.erpidin remains in solution as a more readily soluble alkaloid and can be isolated therefrom in a known manner.
In the examples below, parts by weight and parts by volume have the same relationship as the relationship between grams and cubic centimeters. The temperatures are given in degrees Celsius. <I> Example 1 </I> 1 part by weight of powdered extract from Rauwolfia vomitoria Afz. or Rauwolfia serpentina Benth. Roots are dissolved warm in a mixture of 2 parts by volume of glacial acetic acid and 8 parts by volume of methanol,
while stirring with 6 parts by volume of water ver sets and the precipitate is filtered off. The filtrate is inoculated with reserpine rhodanide at 60. 1 part by volume of 50% strength aqueous potassium rhodanide solution is added with stirring. The mixture is left to stand for a few hours at room temperature and then the reserpine rhodanide formed is filtered off with suction. The residue is washed with methanol.
1 part by weight reserpine rhodanide is slurried with 8 parts by volume of methanol and 4 parts by volume of 3.5% ammonia and stirred at an internal temperature of 60 minutes for 30 minutes. After cooling, the mixture is left to stand for a few hours at room temperature and then the released reserpine is sucked off. It is washed with water, methanol and methanol ether. This gives pure reserpine.
The extract used as the starting material can be obtained according to the information contained in U.S. Patent No. 2,752,351. You can z. B. 1 part by weight of ground roots of Rauwolfia ser- pentina Benth. or Rauwolfia vomitoria Afz. Moisten evenly with 1 volume part of water.
The root material is then extracted with 5-10 parts by volume of toluene for 5-7 hours under reflux, the boiling temperature of the toluene being regulated to about 65 with an appropriate vacuum. The light brownish toluene extract is separated from any water that may be present and evaporated to dryness in vacuo.
<I> Example 2 </I> 1 part by weight of crude reserpine is dissolved warm in a mixture of 2 parts by volume of methanol and 2 parts by volume of glacial acetic acid. At an internal temperature of 60 ° and stirring thoroughly, 6 parts by volume of water are added and any deposits formed are filtered off. The clear filtrate is inoculated with reserpine rhodanide, and 0.4 parts by volume of 5011 / o aqueous potassium rhodanide solution are added at 60 ° with thorough stirring.
After cooling, the mixture is left to stand for a few hours and then filtered. The suction filtered reserpine rhodanide is washed with methanol.
1 part by weight of reserpine rhodanide is slurried in 8 parts by volume of methanol and 4 parts by volume of 3.5% ammonia and stirred at an internal temperature of 60 minutes for 30 minutes. After cooling, the mixture is left to stand for a few hours at room temperature and the released reserpine is sucked off. The residue is washed with water, methanol and methanol ether. This gives pure reserpine.
<I> Example 3 </I> A mixture of 0.05 parts by weight of reserpine and 0.05 parts by weight of deserpidine is dissolved in 2 parts by volume of chloroform and 50 parts by volume of ether and 10 parts by volume of a rhodanic acid solution are added dropwise and 10 parts by volume are added 0.1-n.
methanolic ammonium thiocyanate solution with 10 parts by volume of methanol and 2 parts by volume of glacial acetic acid and filling up with ether to a volume of 100 parts by volume is obtained, with mechanical stirring. This: mixture is mixed with 25 parts by volume of petroleum ether. After 15 minutes the excreted reserpine rhodanide is filtered off and washed with ether.
0.066 part by weight of reserpine rhodanide with a melting point of 239 to 240 (Zerr.) Is obtained. The filtrate, which essentially contains deserpine base, becomes 2-n times with 25 parts by volume each time. Shaken out acetic acid, the acidic solution was made alkaline with ammonia while cooling and the product obtained was separated off by filtration. This gives 0.037 part by weight of a base mixture which consists of 85% deserpidine and 15% reserpine.
<I> Example 4 </I> If, instead of the 10 parts by volume of the rhodanic acid solution, only 9 parts by volume of the same and the rest of the procedure is as described in Example 3, 0.062 part by weight of reserpine rhodanide and 0.041 part by weight of deserpidine base are obtained, which are still 221 / o Contains reserpine.
<I> Example 5 </I> 0.065 part by weight of a deserpidine-reserpine mixture obtained according to Example 4, containing 22% reserpine, is dissolved in 2 parts by volume of chloroform and 100 parts by volume of ether are added.
To this are added 2.5 parts by volume of the methanolic hydrofluoric acid solution described in Example 1, 5 parts by volume of petroleum ether are added and, after mechanical stirring for 15 minutes, the mixture is filtered off.
This gives 0.016 part by weight of reserpine rhodanide and 0.045 part by weight of a base mixture which consists of 87% deserpidine and 13% reserpine.
The reserpine rhodanide obtained in Examples 3-5 can, as shown in Examples 1 and 2, be converted into pure reserpine.
<I> Example 6 </I> 1 part by weight of extract from roots of Rauwoifia vomitoria Afz. or Rauwolfia serpentina Benth. is dissolved warm in a mixture of 4 parts by volume of methanol and 1 part by volume of glacial acetic acid,
6 parts by volume of water are added while stirring and the precipitate is filtered off from the still warm mixture. The filtrate is inoculated with reserpine rhodanide at about 60 and mixed with 0.5 parts by volume of 50% aqueous potassium rhodanide solution while stirring. After standing for a few hours at room temperature, the mixture is suction filtered and the remaining on the filter,
crude reserpine rhodanide washed with methanol. As described in Example 1, the salt obtained can be converted into the basic reserpine.
The extract used as the starting material (alkaloid content about 0.2 to 0.3 parts by weight) can be prepared as described in Example 1.
<I> Example 7 </I> 1 part by weight of ethylene chloride extract from root material from Rauwolfia vomitoria Afz. or Rau- wolfia serpentina Benth. is dissolved warm in a mixture of 8 parts by volume of methanol and 2 parts by volume of glacial acetic acid. At about 40 internal temperature and stirring well, 6 parts by volume of water are added and the precipitate is filtered off from the still warm mixture.
The clear filtrate is inoculated with reserpine rhodanide and 2 parts by volume of 50% aqueous potassium rhodanide solution are added at an internal temperature of about 60 degrees with thorough stirring. After cooling, the mixture is left to stand for several hours and then filtered. The sucked off, crude reserpine rhodanide is washed with - methanol ge.
As described in Example 1, it can be converted into free reserpine.
To prepare the ethylene chloride extract used as the starting material (alkaloid content about 0.1 parts by weight) you can, for. B. 1 part by weight of ground roots of Rauwolfia serpentina Benth. or Rauwolfia vomitoria Afz. Evenly moisten with about 0.8 parts by volume of water and then extract with 5-10 parts by volume of ethylene chloride for 5-7 hours under reflux.
The brown ethylene chloride extract is separated from any water present and evaporated to dryness in vacuo.
