Verfahren zur Herstellung von 3-Phenyl-3-alkyl-2-pyrrolidonen Es wurde gefunden, dass sich die bisher unbe kannten 3-Phenyl-3-alkyl-2-pyrrolidone durch hyp notische und antikonvulsive Wirksamkeit auszeich nen.
Die Erfindung betrifft ein Verfahren zur Herstel lung von 3-Phenyl-3-Alkyl-2-pyrrolidonen, das da durch gekennzeichnet ist, dass man Phenylalkylaceto- nitrile, deren Alkylrest 1-6 C-Atome enthält, in ihre Alkalisalze überführt und mit Athylenoxyd zu a-Phe- nyl-a-alkyl-y-hydroxy-butyronitrilen umsetzt, diese durch Zusatz von Säuren und Erwärmen zu a-Phenyl- a-alkyl-y-hydroxy-butyrolactonen cyclisiert und letz tere mit Ammoniak behandelt.
Nach einer beispielsweisen Ausführungsform des Verfahrens werden Phenyl-alkyl-acetonitrile, deren Alkylrest 1-6 C-Atome enthält, in die Natriumsalze übergeführt und diese mit Äthylenoxyd, vorzugsweise in Gegenwart eines indifferenten Lösungsmittels, zu a-Phenyl-a-alkyl-y-hydroxy-butyronitrilen umgesetzt.
Diese werden durch Einwirkung einer Säure zu den 2-Imino-3-phenyl-3-alkyl-tetrahydrofuranen cyclisiert, welche, ohne isoliert zu werden, durch Erwärmung zu den a-Phenyl-a-alkyl-y-hydroxy-butyrolactonen hydrolysiert werden. Beim Behandeln mit Amrno- niak im Autoklaven bei 180-200 gehen diese in die 3-Phenyl-3-alkyl-2-pyrrolidone über.
Die neuen, so dargestellten 3-Phenyl-3-alkyl-2- pyrrolidone sind farblose, im Vakuum destillierbare, kristallisierbare und in Wasser schwerlösliche, neu tral reagierende Substanzen, die in den gebräuchlichen organischen Lösungsmitteln leicht löslich sind.
Sie sollen als Heilmittel Verwendung finden. <I>Beispiel 1</I> 3-Phenyl-3-äthyl-2-pyrrolidon: a-Phenyl-a-äthyl-y-hydroxy-butyronitril. 145 g Phenyl-äthyl-acetonitril werden in einem mit Rührwerk, Rückflusskühler, Tropftrichter und Thermometer versehenen 2-Liter-Rundkolben mit 39 g feinpulverisiertem Natriumamid in 400 cm3 ab solutem Äther vermischt. Unter lebhafter Ammoniak entwicklung beginnt die Mischung zu sieden.
Nach beendeter Reaktion wird 1/.1 Stunde am Rückfluss ge kocht und durch Durchblasen von Stickstoff die Hauptmenge des Ammoniaks entfernt. Die entstan dene, in Äther lösliche Natriumverbindung des Phe- nyl-äthyl-acetonitrils setzt sich unter Wärmeentwick lung mit einer ätherischen Lösung von 45g Athylen- oxyd zum a-Phenyl-a-äthyl-y-hydroxy-butyronitril um, das nach 1/Gstündigem Sieden am Rückfluss entsteht.
Der Kolbeninhalt wird gekühlt, mit Eis und Wasser versetzt, die ätherische Phase über Natriumsulfat ge trocknet und nach dem Abdampfen des Lösungsmit tels im Wasserstrahlvakuum destilliert. Der Siede punkt des farblosen öls liegt bei 13 mm Quecksilber druck zwischen 174-175 . a-Phenyl-a-äthyl-y-hydroxy-butyrolacton. a) 189,2 g a-Phenyl-a-äthyl-y-hydroxy-butyro- nitril werden unter intensivem Rühren zu 500 cm3 auf -10 abgekühlter 15 0/aiger Salzsäure gegeben.
Nach 5 Minuten Rühren werden 200 cm3 Eiswasser und 200 cm3 Äther zugefügt, die wässerige Phase abgetrennt und in Gegenwart von Eis mit 201/migem Ammoniak lakmusalkalisch gestellt, wobei ein öl aus fällt, welches mit Äther ausgezogen, getrocknet und destilliert wird. Bei 14 mm Druck gehen zwischen 154-158 120 g eines klaren farblosen öls über.
Das 2-Imino-3-phenyl-3-äthyl-tetrahydrofuran ist in Säuren in der Kälte klar löslich, aber aus den sauren wässerigen Lösungen fällt nach kurzer Zeit das a-Phe- nyl-a-äthyl-y-hydroxy-butyrolacton als farbloses öl aus. Durch Erwärmen auf 90 wird die Fällung ver vollständigt.
100 g 2-Imino - 3 - phenyl-3-äthyl-tetrahydrofuran werden zu 300 cm3 einer auf 90 erwärmten 3n Schwefelsäure unter Rühren zugegeben, wobei nach anfänglicher Lösung in kurzer Zeit das a-Phe- nyl-a-äthyl-y-hydroxy-butyrolacton ausfällt, welches mit Methylenchlorid ausgezogen, über Natriumsulfat getrocknet und bei 13 mm Druck destilliert wird. Zwischen 168-168,5 geht das Lacton über.
b) 189,2 g a-Phenyl-a-äthyl-y-hydroxy-butyronitril werden unter Rühren zu 400 cm3 15 /aiger Salzsäure gegeben und auf 90 erwärmt. Nach anfänglichem Lösen fällt a-Phenyl-a-äthyl-y-hydroxy-butyrolacton in öligen Tropfen aus, welche mit Äther extrahiert, über Natriumsulfat getrocknet und im Wasserstrahl vakuum destilliert werden. Das Lacton geht zwischen 168 und 168,5 bei 13 mm Druck über.
3-Phenyl-3-äthyl-2-pyrrolidon. 190 g a-Phenyl-a-äthyl-y-hydroxy-butyrolacton werden in 300 cm3 Methanol gelöst und in einem Autoklaven bei 180-200 mit 200 g 1001/oigem Ammoniak umgesetzt, im Vakuum zur Trockne ein geengt und aus Methanol-Wasser umkristallisiert. Das erhaltene 3-Phenyl-3-äthyl-2-pyrrolidon schmilzt bei 86-87 .
<I>Beispiel 2</I> 3-Phenyl-3-methyl-2-pyrrolidon. Ausgehend von Phenyl-methyl-acetonitril, erhält man analog der Umsetzung des oben beschriebenen Phenyl-äthyl-acetonitrils das a-Phenyl-a-methyl-y-hy- droxy-butyronitril (Kp. 10 mm/161-168 ), welches sich mit Salzsäure beim Erwärmen zum a-Phenyl-a- methyl-y-hydroxy-butyrolacton (Kp.
10 mm/160 bis 165 ) umsetzt. Das Erwärmen mit Ammoniak im Autoklaven führt zum 3-Phenyl-3-methyl-2-pyrroli- don (Smp. 79,5 ). Kp. 11 mm/198-200 .
<I>Beispiel 3</I> 3-Phetayl-3-n-propyl-2-pyrrolidon. Analog entsteht aus Phenyl-n-propyl-acetonitril über a-Phenyl-a-n-propyl-y-hydroxy-butyronitril (Kp. 10 mm/176 ) und a-Phenyl-a-n-propyl-y-hydroxy- butyrolacton (Kp. 12 mm/172-179 ) das 3-Phenyl- 3-n-propyl-2-pyrrolidon, das zwischen 208 und 209 bei 11 mm Druck siedet und bei 78,5 schmilzt.
<I>Beispiel 4</I> 3-Phenyl-3-isopropyl-2-pyrrolidon. Ausgehend von Phenyl-isopropyl-acetonitril wird in analoger Weise über a-Phenyl-a-isopropyl-y-hy- droxy-butyronitril (Kp. l0mm/173-178 ) und a-Phe- nyl-a-isopropyl-y-hydroxy-butyrolacton (Kp. 12 mm/ 170-176 ) das 3-Phenyl-3-isopropyl-2-pyrrolidon (Smp. 1l4 ) erhalten.

The invention relates to a process for the produc- tion of 3-phenyl-3-alkyl-2-pyrrolidones, which is characterized in that phenylalkylacetonitriles, the alkyl radicals of which contain 1-6 carbon atoms, are converted into their alkali metal salts and with ethylene oxide converts to a-phenyl-a-alkyl-y-hydroxy-butyronitriles, cyclizes them by adding acids and heating to give a-phenyl-a-alkyl-y-hydroxy-butyrolactones and treats the latter with ammonia.
According to an exemplary embodiment of the process, phenyl-alkyl-acetonitriles, the alkyl radicals of which contain 1-6 carbon atoms, are converted into the sodium salts and these are converted with ethylene oxide, preferably in the presence of an inert solvent, to a-phenyl-a-alkyl-y- hydroxy-butyronitrile implemented.
These are cyclized by the action of an acid to the 2-imino-3-phenyl-3-alkyl-tetrahydrofurans, which, without being isolated, are hydrolyzed to the a-phenyl-a-alkyl-γ-hydroxy-butyrolactones by heating. When treated with ammonia in an autoclave at 180-200, these change into the 3-phenyl-3-alkyl-2-pyrrolidones.
The new 3-phenyl-3-alkyl-2-pyrrolidones presented in this way are colorless, vacuum-distillable, crystallizable and sparingly water-soluble, neutrally reacting substances which are easily soluble in common organic solvents.
They are said to be used as remedies. <I> Example 1 </I> 3-phenyl-3-ethyl-2-pyrrolidone: a-phenyl-a-ethyl-y-hydroxy-butyronitrile. 145 g of phenylethyl acetonitrile are mixed with 39 g of finely powdered sodium amide in 400 cm3 of absolute ether in a 2 liter round bottom flask equipped with a stirrer, reflux condenser, dropping funnel and thermometer. The mixture begins to boil with a vigorous evolution of ammonia.
After the reaction has ended, the mixture is refluxed for 1/1 hour and most of the ammonia is removed by blowing nitrogen through it. The resulting, ether-soluble sodium compound of phenyl-ethyl-acetonitrile is converted under heat development with an ethereal solution of 45 g of ethylene oxide to a-phenyl-a-ethyl-y-hydroxy-butyronitrile, which after 1 / Ghouring boiling at reflux arises.
The contents of the flask are cooled, ice and water are added, the ethereal phase is dried over sodium sulfate and, after the solvent has evaporated, it is distilled in a water-jet vacuum. After stirring for 5 minutes, 200 cm3 of ice water and 200 cm3 of ether are added, the aqueous phase is separated off and made lactic-alkaline in the presence of ice with 201% ammonia, an oil precipitating out, which is extracted with ether, dried and distilled. At 14 mm pressure, between 154-158 120 g of a clear, colorless oil pass over.
The 2-imino-3-phenyl-3-ethyl-tetrahydrofuran is clearly soluble in acids in the cold, but the a-phenyl-a-ethyl-y-hydroxy-butyrolactone falls out of the acidic aqueous solutions after a short time as colorless oil. The precipitation is completed by heating to 90.
100 g of 2-imino-3-phenyl-3-ethyl-tetrahydrofuran are added with stirring to 300 cm3 of 3N sulfuric acid heated to 90, with the a-phenyl-a-ethyl-γ-hydroxy in a short time after the initial solution -butyrolactone precipitates, which is extracted with methylene chloride, dried over sodium sulfate and distilled at 13 mm pressure. Between 168-168.5 the lactone passes over.
b) 189.2 g of a-phenyl-a-ethyl-y-hydroxy-butyronitrile are added to 400 cm3 of 15% hydrochloric acid with stirring and heated to 90 °. After initial dissolution, a-phenyl-a-ethyl-γ-hydroxy-butyrolactone precipitates in oily drops, which are extracted with ether, dried over sodium sulfate and vacuum distilled in a water jet. The lactone passes between 168 and 168.5 at 13mm pressure.
3-phenyl-3-ethyl-2-pyrrolidone. 190 g of a-phenyl-a-ethyl-y-hydroxy-butyrolactone are dissolved in 300 cm3 of methanol and reacted in an autoclave at 180-200 with 200 g of 100% ammonia, evaporated to dryness in vacuo and recrystallized from methanol-water . The 3-phenyl-3-ethyl-2-pyrrolidone obtained melts at 86-87.
<I> Example 2 </I> 3-Phenyl-3-methyl-2-pyrrolidone. Starting from phenyl-methyl-acetonitrile, a-phenyl-a-methyl-y-hydroxy-butyronitrile (boiling point 10 mm / 161-168) is obtained analogously to the reaction of the phenyl-ethyl-acetonitrile described above with hydrochloric acid when heated to a-phenyl-a-methyl-y-hydroxy-butyrolactone (bp.
10 mm / 160 to 165). Heating with ammonia in the autoclave leads to 3-phenyl-3-methyl-2-pyrrolidone (melting point 79.5). Kp. 11 mm / 198-200.
<I> Example 3 </I> 3-Phetayl-3-n-propyl-2-pyrrolidone. Analogously, phenyl-n-propyl-acetonitrile is formed via a-phenyl-an-propyl-y-hydroxy-butyronitrile (b.p. 10 mm / 176) and a-phenyl-an-propyl-y-hydroxyl butyrolactone (b.p. 12 mm / 172-179) the 3-phenyl-3-n-propyl-2-pyrrolidone, which boils between 208 and 209 at 11 mm pressure and melts at 78.5.
<I> Example 4 </I> 3-Phenyl-3-isopropyl-2-pyrrolidone. Starting from phenyl-isopropyl-acetonitrile, a-phenyl-a-isopropyl-y-hydroxy-butyronitrile (b.p. 10 mm / 173-178) and a-phenyl-a-isopropyl-y-hydroxy are carried out in an analogous manner -butyrolactone (b.p. 12 mm / 170-176) obtained 3-phenyl-3-isopropyl-2-pyrrolidone (mp. 114).
