Verfahren zur Herstellung von 14-Dehydro-steroiden Es ist bekannt, dass Steroid-14,16-diene bei der katalytischen Reduktion 14-Iso-17-iso-steroide lie fern, d. h. Steroide mit .einer Konfiguration in 14- und 17-Stellung, die der normalen der entsprechen den Naturstoffe nicht entspricht.
Es wurde nun gefunden, dass sich bei der Reduk tion von Steroid-14,16-dienen mit nascierendem Was serstoff 14-Dehydro-steroide mit der normalen Kon figuration in 17-Stellung bilden. Diese Verbindungen lassen sich, wie bekannt ist, zu Steroiden mit nor maler Konfiguration in 14-Stellung hydrieren. Gegen stand der Erfindung ist demgemäss ein Verfahren zur Herstellung von 14-Dehydro-steroiden, dadurch ge kennzeichnet, dass man Steroid-14,16-diene mit nas- cierendem Wasserstoff reduziert.
Man kann dabei nach den üblichen Methoden arbeiten. Besonders geeignet ist die Bildung des nas- cierenden Wasserstoffes durch Reaktion eines Alkali- oder Erdalkalimetalles, insbesondere von Lithium, ferner auch Natrium, Kalium, Calcium oder Alkali- metallamalgamen, mit Alkoholen allein und,.'oder Stickstoffbasen.
Von den Alkoholen sind die alipha- tischen zu nennen, wie Methanol, Äthanol, Propanol, Isopropanol, Butanol, Amylalkohol und dergleichen. Als Stickstoffbasen eignen sich in erster Linie Ammo niak, ferner auch aliphatische Amine, wie Methylamin, Äthylamin, Propylamin, Äthylendiamin, entweder allein oder in Kombination.
Wird die Reduktion mit oben genannten Metallen und einem Alkohol allein durchgeführt, so arbeitet man zweckmässig bei erhöhter Temperatur, beispiels weise zwischen 30 und 100 . Ist bei der Reduktion eine flüchtige Stickstoffbase zugegen, so sind tiefere Temperaturen einzuhalten, im Falle von Ammoniak etwa -40 , oder es kann unter Druck und höherer Temperatur, z. B. Raumtemperatur, gearbeitet wer den. Die verfahrensgemässe Reduktion lässt sich vor zugsweise in Gegenwart von organischen Lösungs mitteln, wie Äthern, z. B. Diäthyläther, Dimethoxy- äthan, Dioxan, Kohlenwasserstoffen, z.
B. Petrol- äther, Toluöl u. a., durchführen.
Als Ausgangsstoffe eignen sich aus natürlichen Steroiden oder durch Totalsynthese gewonnene, d. h. optisch aktive oder racemische Steroid-14,16-diene, ferner auch entsprechende Verbindungen der 19-Nor- und"oder D-Homo-Reihen. Besonders wichtig sind die Ausgangsstoffe mit einer freien oder funktionell abgewandelten Acetyl-, Oxyacetyl- oder Carboxyl- gruppe in 17-Stellung, die in 3-,
11- und/oder 18- Stellung eine freie oder funktionell abgewandelte Oxy- oder Oxogruppe, beispielsweise eine veresterte oder verätherte Oxygruppe oder acetalisierte Oxo- gruppe aufweisen und, oder weitere Doppelbindungen enthalten, wie ausgehend vom Kohlenstoffatom 5, d. h. in 4,5- bzw. 5,6-Stellung. Funktionell abge wandelte Oxyacetylresten sind z.
B. solche mit ver esterten oder verätherten Oxygruppen. Eine funk tionell abgewandelte Carboxylgruppe ist in erster Linie eine veresterte Carboxylgruppe, ferner z. B. die Säureamid- oder Nitrilgruppe. Solche Ausgangs stoffe sind bekannt oder können nach den Verfahren der schweizerischen Patente Nrn.343392, 341495, 335386 und 343393 hergestellt werden.
Unter den Verfahrensbedingungen reduzierbare Gruppen, z. B. Oxogruppen, können intermediär ge schützt werden, insbesondere durch überführung in acetalisierte Oxogruppen, andernfalls werden sie gleichzeitig reduziert, z. B. Oxo- zu Oxygruppen. Oxygruppen lassen sich nach bekannten Methoden durch Dehydrierung in Oxogruppen zurückverwan deln.
Die Verfahrensprodukte können als Heilmittel oder als Zwischenprodukte zu ihrer Herstellung Ver- wendung finden. Das nach den Angaben in Beispiel 1 erhältliche 14-Dehydro-progesteron ist progestativ wirksam.
In den folgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Tempe raturen sind in Celsiusgraden angegeben. <I>Beispiel 1</I> 1 Gewichtsteil reines d5;14;
1e-3ss_Oxy-20-oxo- pregnatrien vom F. = 186-187 wird in einem Ge misch von 40 Volumteilen Dioxan und 40 Volum- teilen Äther gelöst und unter Feuchtigkeitsausschluss bei -40 bis -50 unter heftigem Durchmischen in eine Lösung von 1,25 Gewichtsteilen Lithium in 600 Volumteilen flüssigem Ammoniak eingetropft. Nach 1 Stunde werden dem Reaktionsgemisch trop fenweise 55 Volumteile n-Propanöl zugefügt.
Nach Zusatz von 1 Gewichtsteil Lithium wird das Reak tionsgemisch noch 1/2 Stunde gerührt und dann auf Eis gegossen.
Die Aufarbeitung durch Extraktion mit Äther liefert 1 Gewichtsteil rohes Reduktionsprodukt, das in bekannter Weise in Toluollösung mit Aluminium- tert.-butylat und Cyclohexanon oxydiert wird. Da es bekannt ist, dass 20-Oxy-pregnan-Derivate bei der Oxydation nach Oppenauer oftmals uneinheitliche Reaktionsprodukte liefern, ist es zu empfehlen, das Rohprodukt einer Nachoxydation mit Chromtrioxyd (0,85 Gewichtsteile) in Pyridin (25 Volumteile) bei 20 zu unterwerfen.
Die normale Aufarbeitung lie fert dann ein Rohprodukt, das nach chrematographi- scher Reinigung 14-Dehydro-progesteron liefert (0,3 Gewichtsteile), das aus Hexan in groben Prismen vom F. = 127-128 kristallisiert. Durch weiteres Umkristallisieren steigt der Schmelzpunkt auf 134 .
Die Konstitution dieser Verbindung lässt sich durch die nachstehenden Überführungen beweisen: Durch Reduktion des 14-Dehydro-progesterons nach den obigen Angaben und Nachoxydation, z. B. mittels des Pyridin-Chromsäure-Komplexes erhält man das d14-3,20-Dioxo-allopregnen vom F. = 186 bis 187 ; [a]D = -i-78 (in Chloroform), das sich auch aus dem bekannten dl-1-3ss-Acetoxy-20-oxo-pregnen durch Hydrolyse und Oxydation mit Pyridin-Chrom- säure-Komplex oder nach Oppenauer gewinnen lässt.
<I>Beispiel 2</I> 1 Gewichtsteil reines d5;14;1a-3ss-Oxy-20-oxo- pregnatrien vom F. = 186-187 , 310 my (log a = 4,14), wird in einem Gemisch von 20 Vo- lumteilen Dioxan und 20 Volumteilen Äther gelöst und unter Feuchtigkeitsausschluss und intensivem Durchmischen bei -45 in .eine Lösung von 0,13 Gewichtsteilen Lithium in 300 Volumteilen flüssigem Ammoniak eingetropft.
Nach 1 Stunde wird dem Reaktionsgemisch tropfenweise n-Propanol bis zur Entfärbung zugefügt. Nach Zugabe von 0,03 Ge- wichtsteilen Lithium wird das Reaktionsgemisch wei tere 30 Minuten gerührt, dann wiederum mit n-Pro- panol bis zur Entf'ärbung versetzt, worauf die beiden letzten Operationen (Zugabe von Lithium und Ent- färbung mit n-Propanol) nochmals wiederholt wer den.
Die Aufarbeitung geschieht, indem durch Weg nahme der Kühlung das Ammoniak abgedampft wird, worauf das Reaktionsgemisch mit 2n Schwefelsäure neutralisiert wird. Die Aufarbeitung durch Extraktion mit Äther liefert 1 Gewichtsteil Rohprodukt, aus welchem sich durch chromatographische Reinigung an Aluminiumoxyd und fraktioniertes Kristallisieren aus Methanol das d5:14-3ss,20a-Dioxy-pregnadien vom F. = 210-213 und das d5;14_3ss,20ss-Dioxy-preg- nadien vom F.<B>=</B> 218-220 isolieren lassen. -Das 3,20-Diacetat der ersteren Verbindung schmilzt bei 178-179 [a]D = -32 (CHCIs);
auch das isomere Diacetat schmilzt bei 178-179 ; [a]D = -10 (CHCl,1). Beide Verbindungen geben in der Misch probe eine Depression des Schmelzpunktes von etwa 20 .
Die Oxydation der an C-20 epimeren d5;14-3ss,20- Dioxy-pregnadiene nach Oppenauer in Benzol mit Aluminium-tert.-butylat und Aceton, gefolgt von einer Nachoxydation mit Chromtrioxyd in Pyridin führt zum 14-Dehydro-progesteron vom F. = 134 . Die Verbindung zeigt im UV. ein Absorptionsmaxi mum bei 242 mit (log a = 4,2).
<I>Beispiel 3</I> 0,37 Gewichtsteile d5;14;16-3ss-Oxy-20-oxo-preg- natrien werden in 10 Volumteilen n-Propanol gelöst. Zur siedenden Lösung gibt man innert 30 Minuten 0,4 Gewichtsteile blanke Natrium-Schnitzel. Nach weiteren 40 Minuten hat sich alles Natrium umge setzt. Das abgekühlte Reaktionsgemisch wird in Was ser gegossen, mit Äther extrahiert und der Äther nach dem Waschen mit Wasser und Trocknen ver dampft.
Durch chromatographische Trennung an Aluminiumoxyd und fraktionierte Kristallisation aus Methanol lassen sich das d5;14-3ss,20a-Dioxy-preg- nadien vom F. = 210-213 und das d5c14-3ss,20ss- Dioxy-pregnadien vom F. = 218-220 isolieren.
Process for the preparation of 14-dehydro-steroids It is known that steroid-14,16-dienes produce 14-iso-17-iso-steroids in the catalytic reduction, i. H. Steroids with a configuration in the 14- and 17-position that does not correspond to the normal of the corresponding natural substances.
It has now been found that the reduction of steroid 14,16-serve with nascent hydrogen 14-dehydro-steroids with the normal configuration in the 17-position. The subject of the invention is accordingly a process for the production of 14-dehydro-steroids, characterized in that steroid-14,16-dienes are reduced with nascent hydrogen.
The usual methods can be used. The formation of nasal hydrogen by reaction of an alkali or alkaline earth metal, in particular lithium, and also sodium, potassium, calcium or alkali metal amalgams, with alcohols alone and "or" or nitrogen bases is particularly suitable.
Aliphatic alcohols should be mentioned, such as methanol, ethanol, propanol, isopropanol, butanol, amyl alcohol and the like. Suitable nitrogen bases are primarily ammonia, and also aliphatic amines such as methylamine, ethylamine, propylamine, ethylenediamine, either alone or in combination.
If the reduction is carried out with the abovementioned metals and an alcohol alone, it is expedient to work at an elevated temperature, for example between 30 and 100. If a volatile nitrogen base is present during the reduction, lower temperatures must be maintained, in the case of ammonia about -40, or it can be done under pressure and higher temperature, e.g. B. room temperature, who worked. Suitable starting materials are steroids obtained from natural or by total synthesis, i. H. Optically active or racemic steroid 14,16-dienes, as well as corresponding compounds of the 19-nor- and "or D-homo series. The starting materials with a free or functionally modified acetyl, oxyacetyl or carboxyl group are particularly important in 17-position, which in 3-,
11- and / or 18- position have a free or functionally modified oxy or oxo group, for example an esterified or etherified oxy group or acetalized oxo group and, or contain further double bonds, as starting from carbon atom 5, d. H. in the 4.5 or 5.6 position. Functionally altered oxyacetyl radicals are z.
B. those with esterified or etherified oxy groups. A func tionally modified carboxyl group is primarily an esterified carboxyl group, also z. B. the acid amide or nitrile group. Such starting materials are known or can be prepared according to the method of Swiss patents 343392, 341495, 335386 and 343393.
Groups reducible under the process conditions, e.g. B. oxo groups can be protected intermediately ge, in particular by conversion into acetalized oxo groups, otherwise they are reduced at the same time, z. B. oxo to oxy groups. Oxy groups can be converted back into oxo groups by dehydrogenation according to known methods.
The products of the process can be used as medicinal products or as intermediate products for their manufacture. The 14-dehydro-progesterone obtainable according to the information in Example 1 has a progestational effect.
In the following examples, parts by weight and parts by volume have the same relationship as grams and cubic centimeters. The temperatures are given in degrees Celsius. <I> Example 1 </I> 1 part by weight of pure d5; 14;
1e-3ss_Oxy-20-oxo-pregnatrien vom F. = 186-187 is dissolved in a mixture of 40 parts by volume of dioxane and 40 parts by volume of ether and, with exclusion of moisture, at -40 to -50 with vigorous mixing into a solution of 1, 25 parts by weight of lithium in 600 parts by volume of liquid ammonia were added dropwise. After 1 hour, 55 parts by volume of n-propane oil are added dropwise to the reaction mixture.
After adding 1 part by weight of lithium, the reaction mixture is stirred for a further 1/2 hour and then poured onto ice.
Working up by extraction with ether yields 1 part by weight of crude reduction product which is oxidized in a known manner in toluene solution with aluminum tert-butoxide and cyclohexanone. Since it is known that 20-oxy-pregnane derivatives often give inconsistent reaction products in the oxidation according to Oppenauer, it is recommended to subject the crude product to post-oxidation with chromium trioxide (0.85 parts by weight) in pyridine (25 parts by volume) at 20 .
Normal work-up then yields a crude product which, after chromatographic purification, yields 14-dehydro-progesterone (0.3 parts by weight) which crystallizes from hexane in coarse prisms with a F. = 127-128. Further recrystallization increases the melting point to 134.
The constitution of this compound can be proven by the following conversions: By reducing the 14-dehydro-progesterone according to the above information and post-oxidation, e.g. B. by means of the pyridine-chromic acid complex one obtains the d14-3,20-dioxo-allopregnen from F. = 186 to 187; [a] D = -i-78 (in chloroform), which can also be obtained from the well-known dl-1-3ss-acetoxy-20-oxo-pregnene by hydrolysis and oxidation with a pyridine-chromic acid complex or according to Oppenauer .
<I> Example 2 </I> 1 part by weight of pure d5; 14; 1a-3ss-oxy-20-oxo-pregnatriene from F. = 186-187, 310 my (log a = 4.14), is used in a mixture dissolved by 20 parts by volume of dioxane and 20 parts by volume of ether and added dropwise to a solution of 0.13 parts by weight of lithium in 300 parts by volume of liquid ammonia with the exclusion of moisture and intensive mixing at -45.
After 1 hour, n-propanol is added dropwise to the reaction mixture until it becomes discolored. After adding 0.03 parts by weight of lithium, the reaction mixture is stirred for a further 30 minutes, then n-propanol is again added until it is decolorized, whereupon the last two operations (addition of lithium and decolorization with n- Propanol) repeated again who the.

Working up by extraction with ether gives 1 part by weight of crude product, from which the d5: 14-3ss, 20a-dioxy-pregnadiene from F. = 210-213 and the d5; 14_3ss, 20ss are obtained by chromatographic purification on aluminum oxide and fractional crystallization from methanol -Dioxy-preg- nadiene from F. <B> = </B> 218-220 isolate. -The 3,20-diacetate of the former compound melts at 178-179 [a] D = -32 (CHCls);
the isomeric diacetate also melts at 178-179; [a] D = -10 (CHCl1, 1). In the mixed sample, both compounds give a depression of the melting point of about 20.
Oxidation of the C-20 epimers d5; 14-3ss, 20-dioxy-pregnadienes according to Oppenauer in benzene with aluminum tert-butoxide and acetone, followed by post-oxidation with chromium trioxide in pyridine leads to 14-dehydro-progesterone from F. . = 134. <I> Example 3 </I> 0.37 parts by weight of d5; 14; 16-3ss-oxy-20-oxo-preg-natriene are dissolved in 10 parts by volume of n-propanol. 0.4 part by weight of bright sodium chips are added to the boiling solution within 30 minutes. After a further 40 minutes, all the sodium has converted. The cooled reaction mixture is poured into water, extracted with ether and the ether evaporated after washing with water and drying.
By chromatographic separation on aluminum oxide and fractional crystallization from methanol, the d5; 14-3ss, 20a-dioxy-preg- nadiene from F. = 210-213 and the d5c14-3ss, 20ss-dioxy-pregnadiene from F. = 218- 220 isolate.
