in welcher Arl einen durch Alkyl, Alkoxy, Halogen, Amino, Alkylamino, Dialkylamino oder Acylamino substituierten Phenylrest und Ar. dasselbe wie Arl oder einen unsubstituierten Phenylrest bedeutet, wertvolle Roborantia darstellen.
Es ist zwar bekannt, dass die Nicotinyl-amino- methylphosphonsäure und ihre Salze als Tonikum verwendetwerden können (USA-PatentNr. 2 674 599). Es ist auch bekannt, die Benzoyl-aminomethylphos- phonsäure aus Benzamid, Formaldehyd und Phos- phortrichloridherzustellen (USA-Patent Nr. 2 304156).
Die beiden bekannten Stoffe weisen nur etwa 25 der Aktivität der Verfahrensprodukte des vorliegenden Patentes auf.
Gegenstand des vorliegenden Patentes bildet ein Verfahren zur Herstellung von neuen Acyl-amino- methylphosphonsäuren der Formel

in welcher Arl einen durch Alkyl, Alkoxy, Halogen, Amino, Alkylamino, Dialkylamino oder Acylamino substituierten Phenylrest und Are dasselbe wie Ar, oder einen unsubstituierten Phenylrest bedeutet, welches dadurch gekennzeichnet ist,
dass man auf Amino- methylphosphonsäure eine einen Rest der Formel Ar.--CO- oder Ars alkylen-CO- abgebende Verbindung, worin Ara dasselbe wie Ar, bzw. Ar. oder einen Phenylrest, welcher einen durch Hydrolyse oder Reduktion in eine Aminogruppe überführbaren Substituenten aufweist, bedeutet, einwirken lässt, und gegebenenfalls den durch Hydrolyse oder Reduktion in eine Aminogruppe überführbaren Sub- stituenten in eine Aminogruppe überführt.
Die so erhaltenen Acyl-aminomethylphosphon- säuren können erforderlichenfalls in Salze anorganischer oder organischer Basen übergeführt werden. Als den Rest Ar"-CO- bzw. Ara alkylen-CO- abgebende Verbindungen kann man beispielsweise die betreffenden freien Säuren oder deren Halogenide, Anhydride oder Amide verwenden.
Die Methode der Wahl besteht aber hier darin, dass man Aminomethylphosphonsäure und eine der betreffenden freien Säuren zusammen im Schmelzfluss erhitzt, wobei die Reaktion unter Wasserabspaltung stattfindet.
Beispiel 1 20,4 g Phenylessigsäure und 11,1g Aminomethyl- phosphonsäure werden unter Rühren 15 Minuten auf 220 C erhitzt. Nach dem Abkühlen wird die erstarrte Schmelze pulverisiert, in 50 cm3 heissem Wasser gelöst, und die erhaltene Lösung mit 250 cm3 Aceton versetzt. Die gebildeten Kristalle werden mit der

Mutterlauge zusammen mit Äther zwecks Entfernung von Phenylessigsäure ausgeschüttelt. Die zurückbleibende Kristallmasse wird aus wenig Wasser umkristallisiert. Man erhält 6,5 g Phenacetyl-amino- methylphosphonsäure. Diese fällt in Form farbloser, bei 166-167 C schmelzender Kristalle an.
Beispiel 2 18,8 g p-Chlor-phenylessigsäure und 11g Amino- methylphosphonsäure werden 15 Minuten auf 230 bis 240 C erhitzt. Zu der abgekühlten Schmelze gibt man 100 cm3 heisses Dioxan, wobei 6 g unveränderte Aminomethylphosphonsäure ausfallen. Das Filtrat wird mit Ligroin versetzt und die ausgefallenen Kristalle abgenutscht. Man erhält so 11-12 g p-Chlor- phenacetylaminomethylphosphonsäure. Nach Umkristallisieren aus Wasser schmilzt die Säure bei 172 bis 173 C.
Sie löst sich in kaltem Wasser mässig, in Methanol und Äthanol gut und wenig in Aceton und Äther.
Beispiel 3 33g Aminomethylphosphonsäure werden mit 70 g 4-Chlor-benzoesäure vermischt und während 15 Minuten bei 255-26011 C geschmolzen. Die Schmelze wird nach dem Erkalten pulverisiert und mit 300 cm3 Wasser ausgekocht und filtriert. Das Filtrat wird zur Trockne verdampft, mit 200 cm3 abs. Äthanol ausgekocht und wieder filtriert. Aus dem Filtrat gewinnt man durch Einengen auf 2(3 des Volumens und Versetzen mit Äther 4-Chlorbenzoyl-aminomethylphos- phonsäure in guter Ausbeute.
Die neue Verbindung schmilzt, aus ÄthanolJÄther umkristallisiert, bei 179 bis 180 C. Sie löst sich gut in Wasser und Alkohol, schlecht in Aceton und ist praktisch unlöslich in Äther und Benzol.

Beispiel 4 60 g 4-Nitro-benzoylchlorid und 18g Amino- methylphosphonsäure werden gemischt und im COlbad erhitzt. Bei 140 C beginnt die Mischung zu schmelzen, bei 190 C erfolgt starke Chlorwasserstoffent- wicklung. Man hält 3/.t Stunde auf 195 C, lässt dann erkalten und pulverisiert die Schmelze. Diese wird mit Aceton ausgekocht (5malige Extraktion mit je 300 cm3 Aceton), das Ungelöste mit Äther gewaschen und getrocknet.
Man erhält so 34-35 g 4-Nitro- benzoyl-aminomethylphosphonsäure, die bei 206 bis 207 C schmilzt. Die neue Phosphonsäure kann aus verdünnter Natriumcarbonatlösung mit Hilfe von Salzsäure umgefällt werden. Sie ist in organischen Lösungsmitteln und auch in Wasser wenig löslich.
70 g 4-Nitro-benzoyl-aminomethylphosphonsäure, gelöst in verdünnter wässriger Natriumcarbonatlösung, werden in Gegenwart von 5%iger Palladiumkohle unter Normaldruck hydriert. Der Wasserstoff wird sehr schnell unter Wärmebildung aufgenommen. Nach dem Abfiltrieren des Katalysators wird das Filtrat schwach sauer gestellt und die ausgefallenen feinen Kristalle abgesaugt und getrocknet.
Man erhält so 52 g (das sind 84% der Theorie) an 4-Amino-benzoylamino-methylphosphonsäure. Diese schmilzt oberhalb 300 C, löst sich gut in heisser 2n-Salzsäure und sehr gut in kalter, verdünnter Natriumcarbonatlösung.
In analoger Weise, wie in den Beispielen beschrieben, lassen sich weiter herstellen: 3,4-Dichlor-benzoyl-aminomethylphosphonsäure; weisse Prismen aus Eisessig; Schmp.: 179-180 C. 4 - Methyl - benzoyl-aminomethylphosphonsäure; feine Kristalle aus Eisessig; Schmp.: 202,5-203 C.
3,4-Dimethyl-benzoyl-aminomethylphosphonsäure; Nadeln aus Eisessig; Schmp.: 206-207,5 C.

It is known that nicotinyl-aminomethylphosphonic acid and its salts can be used as a tonic (USA Patent No. 2,674,599). It is also known to produce benzoyl aminomethylphosphonic acid from benzamide, formaldehyde and phosphorus trichloride (USA patent No. 2,304,156).
The two known substances have only about 25 of the activity of the process products of the present patent.
The subject of the present patent is a process for the preparation of new acylamamethylphosphonic acids of the formula

It dissolves moderately in cold water, well in methanol and ethanol, and little in acetone and ether.
Example 3 33 g of aminomethylphosphonic acid are mixed with 70 g of 4-chloro-benzoic acid and melted at 255-26011 ° C. for 15 minutes. After cooling, the melt is pulverized, boiled with 300 cm3 of water and filtered. The filtrate is evaporated to dryness, with 200 cm3 abs. The new compound melts, recrystallized from ethanol / ether, at 179 to 180 C. It dissolves well in water and alcohol, poorly in acetone and is practically insoluble in ether and benzene.

Example 4 60 g of 4-nitrobenzoyl chloride and 18 g of aminomethylphosphonic acid are mixed and heated in a CO bath. 70 g of 4-nitro-benzoyl-aminomethylphosphonic acid, dissolved in dilute aqueous sodium carbonate solution, are hydrogenated in the presence of 5% palladium-on-carbon under normal pressure. The hydrogen is absorbed very quickly with the formation of heat. After the catalyst has been filtered off, the filtrate is made slightly acidic and the fine crystals which have precipitated are filtered off with suction and dried.
52 g (84% of theory) of 4-amino-benzoylamino-methylphosphonic acid are thus obtained. This melts above 300 C, dissolves well in hot 2N hydrochloric acid and very well in cold, dilute sodium carbonate solution.
