Ces dihaloacétamides substituées obtenues par le procédé objet de l'invention présentent des propriétés chimio- thérapeutiques intéressantes, par exemple une activité marquée contre les amibes.
Dans la formule précédente, le radical dé signé par<I>Ar</I> est un radical phényle substitué une à trois fois. Le ou les substituants de ce radical peuvent être des substituants halogène, alcoxy inférieur; alcoyle inférieur, alcoyl- mercapto inférieur, alcoylsulfonyle inférieur et nitro. En outre, ces substituants peuvent se trouver dans n'importe laquelle des positions disponibles du noyau phényle et, lorsqu'il y en a plus d'un, ils peuvent être identiques ou dif férents et ils peuvent se trouver dans n'importe laquelle de diverses combinaisons de positions, les uns par rapport aux autres.
Les substi- tuants halogénés peuvent être les substituants chloro, bromo, iodo et fluoro. Les substituants alcoxy inférieur, alcoyle inférieur, alcoyl- mercapto inférieur et alcoylsulfonyle inférieur comportent, de préférence, de un à six ato= mes de carbone ;
parmi ces substituants, on peut citer les groupes méthoxy, éthoxy, méthy- lènedioxy, éthylènedioxy, n-propoxy, isopro- poxy, isobutoxy, n-amoxy, n-hexoxy, etc. en ce qui concerne les substituants alcoxy infé rieurs ;
les groupes méthyle, éthyle, n-propyle, isopropyle, n-butyle, isobutyle, 2-butyle, n- amyle, n-hexyle, etc. en ce qui concerne les substituants alcoyle inférieurs ;
les groupes méthylmercapto, éthylmercapto, n-propyl- mercapto, isobutylmercapto, n-hexylmercapto, etc. en ce qui concerne les substituants alcoyl- mercapto inférieurs ;
enfin, les groupes mé- thylsulfonyle, éthylsulfonyle, n-propylsulfo- nyle, isobutylsulfonyle, n-hexylsulfonyle, etc. en ce qui concerne les substituants alcoylsulfo- nyle inférieurs.
Lorsque Y est un radical alpha,bêta-al- coylène inférieur, il comporte de 2 à 6 atomes de carbone et ses deux valences libres sont re liées à des atomes de carbone adjacents comme, par exemple, dans les radicaux

Les atomes d'halogène du radical dihalo- acétyle des composés obtenus, désignés dans la formule précédente par Hlg, peuvent être du chlore, du brome, de l'iode ou du fluor, les radicaux dihaloacétyle étant, par conséquent, des radicaux dichloracétyle, dibromacétyle, di- iodacétyle, difluoracétyle, bromchloracétyle, etc.
Dans le procédé conforme à l'invention, on prépare les composés en question en faisant réagir un amino-alcool de formule Ar-CH,-NH-Y-OH avec un agent de dihaloacétylation, <I>Ar</I> et<I>Y</I> ayant les mêmes significations que ci-dessus.
L'agent de dihaloacétylation peut être un di- haloacétate d'alcoyle inférieur ou un halogé- nure de dihaloacétyle. Lorsque l'on utilise un dihaloacétate d'al coyle inférieur, on préfère utiliser les esters méthyliques en raison de la facilité de leur pré paration et du fait qu'on se les procure facile ment ;
cependant, d'autres esters alcoyliques donnent satisfaction dans ce but comme, par exemple, les esters éthylique, n-propylique, isobutylique, 2-amylique et n-hexylique. Lors que l'on utilise un halogénure de dihaloacétyle, l'halogène de l'halogénure, c'est-à-dire l'halo gène lié à la fonction carbonyle, est de préfé rence le chlore ; cependant, on peut également utiliser les autres halogènes, par exemple le brome, l'iode et le fluor.
Lorsque l'on utilise un dihaloacétate d'al coyle inférieur, on facilite la réaction en chauf fant ensemble des réactifs sur un bain de va peur. Lorsque l'on utilise un halogénure de di- haloacétyle, on exécute la réaction de préfé rence au-dessous de la température ambiante, en refroidissant si cela est nécessaire.
Les aminoalcools de formule Ar-CH,-NH-Y-OH utilisés comme produits de départ, peuvent être préparés en traitant une benzaldéhyde substituée ayant pour formule ArCHO par une alcanolamine ayant pour formule H,,N-Y-OH Le produit de condensation de formule ARCH=N-Y-OH qui se forme, est ensuite réduit directement par catalyse, sans être isolé, ce qui donne le com posé désiré, ayant pour formule ArCH,NH-Y-OH On peut encore préparer ces composés de dé part en traitant un halogénure de benzyle substitué,
de formule Ai-CH.,-Hlg, par une al- canolamine de formule H.,N-Y-OH.
La préparation des matières premières se lon l'une ou l'autre de ces deux méthodes est illustrée par les exemples suivants <I>A.</I> 2-(4-isopropyl-benzylamino)-éthanol On chauffe sur un bain de vapeur, sous vide, pendant une heure, un mélange de 44,3 g de 4-isopropyl-benzaldéhyde et de 18,3 g d'éthannlamine. On dissout le mélange dans 125 cm2 d'éthanol chaud et on le réduit par catalyse avec 0,5 g de chlorure de palladium sur 3,5 g de charbon de bois, sous une pres sion d'environ 2 atmosphères d'hydrogène. Lorsque la réduction est achevée, on élimine le catalyseur par filtration et l'on distille l'al cool sous pression réduite.
On recristallise le résidu qui se solidifie, une fois à partir de n- heptane (43 g) et une fois à partir d'éther ; on obtient ainsi le 2-(4-isopropyl-benzylamino)- éthanol ; point de fusion : 80,911- 83,30 C (corr.).

On maintient la tempéra ture entre 00 et - 5o C en refroidissant avec un bain de glace et de sel. Lorsque l'addition est achevée, on laisse s'élever la température du mélange de réaction jusqu'à la température ambiante, tout en agitant.
On élimine par fil tration le chlorhydrate de 2-(3,4-dichloro- benzylamino)-éthanol qui se sépare, et on lave ensuite le filtrat avec de l'acide chlorhydrique 1N, puis avec de l'eau, et l'on sèche sur du sul fate de calcium anhydre.
Après avoir distillé le dichloréthylène sous pression réduite, on triture le résidu avec de l'éther et l'on obtient 5,5 g d'un produit qui est la N-(3,4-dichloro-benzyl)- N-(2-hydroxy-éthyl)-dichloracétamide ; son point de fusion est 99,40 -<B>101,50</B> C (corr.) quand on le recristallise à partir d'un mélange de benzène et de n-pentane.

Lorsque l'on élimine le solvant, on obtient une huile qui se solidifie lorsqu'on l'agite avec de l'éther. Après recris- tallisation à partir du benzène, puis ensuite à partir du dichloréthylène, on obtient la N-(2,4- dichloro-benzyl) -N-(2-hydroxy-éthyl)-dichlor- acétamide qui fond à 112,4o -113,4 C (corr.).

- éthanol. On sépare le solide par filtration et on lave le filtrat avec de l'acide chlorhydrique 1N, puis avec de l'eau. Après l'avoir séché sur du sulfate de calcium anhydre, on élimine les solvants par distillation. On dissout le résidu dans l'éther, on le filtre à l'aide de charbon de bois et on ajoute du n-pentane jusqu'à ce qu'un trouble commence à se manifester. On obtient 5 g d'un produit ayant un point de fusion compris entre 81 - 850 C.
Après recristallisa- tion avec un mélange de n-pentane et d'éther, on obtient la N-(4-n-butoxy-benzyl)-N-(2-hy- droxy- éthyl) - dichloracétamide, qui fond à 880 - 88,90 C (corr.).

Après avoir éliminé le chloroforme par distillation, on recristallise le résidu à par tir de dichloréthylène et d'une faible quantité de n-héxane ; on obtient 3,5 g de N-(2,4-di- chloro - benzyl) -N - (2 - hydroxy- éthyl) -dibrom- acétamide, qui fond à 1150 - 117,2 C. (corr.).

The dihaloacetylating agent can be a lower alkyl dihaloacetate or a dihaloacetyl halide. When using a lower alkyl dihaloacetate, it is preferred to use the methyl esters because of their ease of preparation and the fact that they are readily available;
however, other alkyl esters are satisfactory for this purpose such as, for example, the ethyl, n-propyl, isobutyl, 2-amyl and n-hexyl esters. When using a dihaloacetyl halide, the halogen of the halide, i.e. the halogen linked to the carbonyl function, is preferably chlorine; however, the other halogens can also be used, for example bromine, iodine and fluorine.
When lower alkyl dihaloacetate is used, the reaction is facilitated by heating the reagents together on a steam bath. When using a dihaloacetyl halide, the reaction is preferably carried out below room temperature, cooling if necessary.
